A Review of Dose-response Models in Microbial Risk Assessment (미생물 위해성 평가의 용량-반응 모델에 대한 고찰)

  • Dose-response models in microbial risk assessment can be divided into biologically plausible models and empirical models. Biologically plausible models are formed by the assumptions in dose distribution of microbes, host sensitivity to microbes, and minimal infectious dose of microbes : there are Exponential model and $\beta$-Poisson model, representatively. Empirical models are mainly used to express the toxicity of chemicals : there are Weibull-Gamma model etc. Deviance function (Y) is used to fit available data to dose-response models, and some dose-response models for food-borne pathogens are developed in humans and experimental animals.

Anti-Obesity Effect of Krill Oil by Regulation of Adipokines in High Fat Diet-Induced Mouse Model (고지방식이 동물모델에서 크릴오일의 아디포카인 조절을 통한 항비만 효과)

  • This study examined the anti-obesity effect of krill oil (KO) by regulating adipokines in a high-fat diet (HFD)-induced obese mouse model. The mice were fed a 60 kcal% HFD for 16 weeks, and KO was then administered at an oral dose of 100, 200, and 500 mg/kg/day for four weeks before the end of the experiment. The administration of KO at concentrations of 200 and 500 mg/kg/day decreased body weight gain significantly compared with the HFD-fed control group. In addition, the HFD-fed control group showed the abnormal release of adipokines by an increase in leptin and decrease in adiponectin, compared to the normal diet-fed normal group. On the other hand, KO (500 mg/kg/day)-administered group attenuated the abnormal release of adipokines by the down-regulation of leptin and the up-regulation of adiponectin. Therefore, KO could be a promising therapeutic agent for obesity by the regulation of adipokines.

Neuroprotective and Anti-inflammatory Effects of Bee Venom Acupuncture on MPTP-induced Mouse (MPTP 유발 파킨슨병 동물 모델에 대한 봉독약침의 신경보호 효과 및 항염증 효과)

  • 목적 : 이 연구는 MPTP 유발 파킨슨병 동물 모델에서 봉독약침의 신경보호 효과 및 항염증 효과를 확인하기 위해 시행되었다. 방법 : C57BL/6 mice에 신경독소인 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)를 하루에 2시간 간격으로 MPTP-HCl(20mg/kg per dose)을 4번 복강 내 주입하여 중뇌 흑질의 도파민 신경세포를 파괴한 파킨슨병 동물 모델을 유발하였다. 실험군은 MPTP군, MPTP 현종 BVA군, MPTP 곡지 BVA군, MPTP 신수 BVA군의 4군으로 하였다. 마지막 MPTP 투여 2시간 후에 1차로 봉독약침을 시술하고, 그 후 48시간 간격으로 총 5차 연속 시술하였다. 봉독약침액의 농도는 0.2mg/Kg으로 하였고, 경혈은 양측 현종($GB_{39}$), 곡지($LI_{11}$), 신수($BL_{23}$)를 사용했고, 주입량은 각 경혈당 양측으로 각 $20{\mu\ell}$씩 주입하였다. 항염증작용을 알아보기 위해 TH, MAC-1, iNOS HSP70을, 세포사멸에 대한 신경세포의 보호효과를 알아보기 위해 caspase-3을 면역조직화학법을 사용하여 실시하였다. 결과 : 실험 결과 MPTP 유발 파킨슨병 동물 모델에서 현종 곡지 신수혈에 대한 봉독약침은 TH-Immunoreactivity neuron의 감소와 microglial activation을 억제하였다. 봉독약침군 모두 효과를 보였으나 그 중 현종과 신수혈에서 특히 억제작용이 컸다. MAC-1에서는 현종혈이 억제작용이 컸다. HSP70-IR neuron은 곡지에서 유의한 억제작용을 보였으나, iNOS neuron은 모든 군에서 유의한 차이를 보이지 않았다. 또한 세포사멸억제여부 실험에서 봉독약침은 모두 억제작용을 보였으나 특히 곡지자침군에서 caspase-3 발현을 유의하게 억제하였다. 결론 : 이러한 결과는 봉독약침이 MPTP 투여로 인한 중뇌 흑질의 염증에 의한 도파민 신경세포 손상을, 염증을 억제함으로써 항염 효과를 나타냄을 알 수 있으며, 신경세포를 보호하는 활성이 있음을 보여줌과 동시에 세포사멸을 억제하는 활성이 있다고 사료된다.

$H_2^{15}$O 심근 PET애서 혈류분포의 비균일성과 분배계수

  • H$_2$$^{15}$ O PET을 이용하여 심근혈류를 측정할 때 물의 분포가 조직마다 불균일한 특성을 나타내고자 관류 가능한 조직분획(perfusable tissue index; PTI)$^{1)}$ 개념을 도입한 연구가 보고된 바 있으나 PTI로는 비균일성을 정량적으로 다루지 못하였다$^{2)}$ . 반면에 단일 구획 모델에서 분배계수(partition coefficient: λ)를 변수로 추정하여 혈류분포의 비 균일성을 나타낼 수 있음을 컴퓨터 모의실험으로 보인 연구가 있다$^{3)}$ . 이 연구에서는 이미 앞서 발표한 연구의 혈류측정 모델을 정상인 실험용 개에 적용하였으며 그 결과 혈류와 분배계수 모두가 정상 범위 안에 들어옴을 확인하였다. 또한 컴퓨터 모의실험을 통해 얻은 분배계수와 비균일성 지표인 구성비균일성(constitution heterogeneity; CH) 와의 관계를 이용하여 동물실험에서 측정한 분배계수로부터 CH를 유추해 보았다.

Hepatoprotcetive Effects of Oyster (Crassostrea gigas) Extract in a Rat Model of Alcohol-Induced Oxidative Stress (알코올로 유도된 간 손상 동물모델에서 굴 추출물의 간 보호 효과)

  • This study was conducted to investigate the protective effects of water extract from Crassostrea gigas (CGW) against ethanol-induced hepatic toxicity in rats. Seventy-two male Wistar rats (6-week-old) were divided into six groups of 12 animals each: control group (1 mL saline/d), ethanol-treated group, positive control group (ethanol+Hovenia dulcis Thunb extract), CGWL group (ethanol+low dosage of CGW), CGWM group (ethanol+medium dosage of CGW), and CGWH group (ethanol+high dosage of CGW). All groups except the control group received ethanol (40% ethanol 5 g/kg) orally. CGW administration with ethanol resulted in prevention of ethanol-induced hepatotoxicity by increasing levels of serum alanine aminotransferase and ${\gamma}-glutamyltransferase$. CGW supplementation significantly reduced formation of malonaldehyde and inhibited reduction of hepatic glutathione and peroxidase levels, as compared with the ethanol-administration group. Further, CGW suppressed expression of CYP2E1, which was elevated by ethanol administration. Consequently, our results indicate that Crassostrea gigas may exert hepatoprotective effects against alcohol-induced hepatocyte injury by intensifying the anti-oxidative defense system.

The Production of Transgenic Mouse Harboring Mutated Pig Rhodopsin Gene (돌연변이가 야기된 돼지 로돕신 유전자를 지닌 형질전환동물의 생산)

  • It is generally known that mutations in any of several genes encoding photoreceptor-specific proteins have resulted in retinitis pigmentosa (RP), a disease characterized by losing photoreceptor function with progressive degeneration of photoreceptor cells and eventually leading to blindness. To study the procure and cure of photoreceptor degeneration, we produced transgenic mice. Transgene consisted of a 12.5kb genomic DNA fragment that contains mutated pig rhodopsin gene (Pro-347-Ser) including both the 5'-franking (4.0 kb) and the 3'-franking (2.9 kb) sequences. This gene was used for the production of transgenic mouse. The mutated rhodopsin DNA was microinjected into male pronuclei of fertilized mouse (C57BL /6]) embryos. We detected transgenic animals harboring mutated rhodopsin gene by PCR and Southern blot analysis. These transgenic mice showed stable transmission of microinjected rhodopsin gene into their offspring. Therefore these animals will provide a novel approach to study the mechanism of the photoreceptor degeneration and be provided as a disease model for the treatment of the blind in human.

Hypolipidemic Effect of Onion Peel Extract in Rats Exposed to Cigarette Smoke Extract with a High-Fat Diet (고지방식이를 섭취한 흡연 흰쥐모델에서 양파껍질 추출물의 혈액지질 대사 개선 효과)

  • This study was designed to investigate whether or not onion peel extract can lower blood lipid levels in rats exposed to cigarette smoke (CS) extract with a high-fat diet. Initially, male Sprague-Dawley rats were housed individually in a stainless steel, wire-bottomed cage with free access to AIN-93G diet. Rats were weight-matched and assigned to the following five groups: 1) control rats (CT) fed standard AIN-93G diet alone, 2) control rats exposed to CS extract (CT+CS), 3) high-fat group (HF) fed standard AIN-93 diet supplemented with 3% lard and 0.2% cholesterol, 4) high-fat group exposed to CS extract (HF+CS) fed standard AIN-93 diet supplemented with 3% lard and 0.2% cholesterol plus CS extract, and 5) high-fat plus onion peel (OP) extract group exposed to CS extract (HF+CS+OP) fed standard AIN-93 diet supplemented with 3% lard, 0.2% cholesterol, and onion peel extract (20 mg/17 g diet) plus CS extract. Using this feeding protocol, all animals completely consumed their respective diets throughout the 6 week duration. Blood was collected via the orbital sinus at weeks 0, 3, and 6, following overnight food deprivation. OP extract feeding resulted in significant reductions in blood triglyceride, total cholesterol, and non-HDL-cholesterol. Further, serum activities of aspartate transaminase and alanine transaminase were significantly reduced by OP extract at 6 weeks. These results provide clear evidence that onion peel extract has a profound inhibitory effect on blood lipids in rats exposed to CS extract. These findings suggest that OP extract can be used as an effective means in alleviating the serum lipid concentration after CS exposure.

  • 본 연구에서는 건축물 내장 재료에 대한 연소독성평가와 독성지수연구로써 FT-IR을 이용한 연소가스분석실험을 하였다. 독성지수를 산정하기 위한 실험의 화재모델로 콘 칼로리미터 화재모델(KS F ISO/TR 9122-4)을 사용하였으며 ISO 19702의 절차에 따라 FT-IR을 이용하여 건축 재료의 연소로부터 발생되는 가스의 분석을 수행하였다. 국제규격에서 제시하고 있는 몇 가지 독성지수 산정법 중 ISO 13344에서 규정하는 방법에 따라 FED 값을 산정하였으며, 30분간 시험동물에 노출 시 대상의 50%가 사망하는 농도인 $LC_{50}$을 기준으로 하여 3가지 재료의 독성지수화를 통해 상대적인 독성 위험도를 평가하였다.

A Fundamental Study on the Effect of Ocean Fertilization by Deep Sea Water (해양심층수에 의한 해역 비옥화 효과에 관한 기초 연구)

  • In this study, we investigated the effect of ocean fertilization by deep sea water, using an ecosystem model which contains not only phytoplankton but also zooplankton. The model is based on NEMURO which consists of eleven compartments - two species of phytoplankton, three species of zooplankton, $NO_3$, $NH_4$, $Si(OH)_4$, particulate organic nitrogen, dissolved organic nitrogen and particulate silicon. We introduced nitrogen cell quota in the both species of phytoplankton, and silicon cell quota in the large phytoplankton in addition to the eleven compartments of NEMURO. We made the experiment at Izu Oshima Island in order to investigate the effect of ocean fertilization. In this experiment, we could not find clear differences between the cases with and without deep sea water. We investigated the causes of the experiment results by the model simulations. One of the causes was high concentrations of nutrients in surface seawater used in the experiment. Another was that the increase of total concentration of inorganic nitrogen does not necessarily accelerate the photosynthetic rate because inorganic nitrogen uptake rate is related to the ratio of $NO_3$ to $NH_4$. Because the model can represent the results of the experiment, we investigated the effect of ocean fertilization by deep sea water using this model. We found that the effect of ocean fertilization hardly appeared when the interval of the addition of deep sea water was too short, or the amount of deep sea water was too much. It is supposed that if the addition of deep sea water is too frequent or too much, the dilution of plankton's concentrations will exceed the effect of promoting phytoplankton's photosynthesis.
