Objectives The purpose of this study is to investigate effectiveness of the Ha-Taedok treatment and discuss how to improve the Ha-Taedok method Methods We analyzed the medical records of 20 children who were treated by Ha-Taedok method. 20 questionnaires were administered to the mothers of those children and statistically analyzed by using PASW Statistics 18. Results The children who were treated by Ha-Taedok method weighed at birth ranged from a minimum of 2.7 kg to a maximum of 3.86 kg, and the children's weight after treated by Ha-Taedok method were varied from a minimum of 2.8 kg to a maximum of 4.7 kg. The Ha-Taedok treatment was initiated to children from 3 to 23 days after their birth. 15 out of 20 mothers of the children treated by the Ha-Taedok method were satisfied with Ha-Taedok method (75%). All of them answered that they were willing to repeat the treatment to their children again or recommend the treatment to others. 10 out of 20 mothers told that the main factor/point for improvement of Ha-Taedok method was publicity. Conclusions If the effectiveness of Ha-Taedok method is scientifically proven and recommended, the scope of Ha-Taedok method for newborn infants will be widely used.
Objectives Hataedock is an orally-administered herbal extract that used to treat toxic heat and help excrete meconium accumulates in the fetus from pregnancy. This study compared the effects of Hataedock using the extract of Douchi and omega-3 to control inflammation in the colonic mucosa. Methods NC/Nga mice were divided into three groups; Control group (no treatment), FGT group (given the extract of Douchi), and O3T group (given omega-3). The 3-week-old mouse from each group were given the extract of Douchi and omega-3. After 2 weeks, the colonic mucosa tissues of the mice were observed via Immunohistochemical staining for IL-4, IL-13, CD40, $Fc{\varepsilon}RI$, $p-I{\kappa}B$, EGF in the intestinal mucosa. Results In both FGT and O3T group, IL-4, IL-13, STAT-6 levels and CD40-positive reactions were decreased. However, EGF-positive responses were increased. FGT group showed 35% decrease in IL-4, 45% decrease in IL-13, 48% decrease in CD40, 50% decrease in $Fc{\varepsilon}RI$, 54% decrease in $p-I{\kappa}B$, 52% increase in EGF compared to the control group. O3T group also showed 14% decrease in IL-4, 23% decrease in IL-13, 22% decrease in CD40, 29% decrease in $Fc{\varepsilon}RI$, 41% decrease in $p-I{\kappa}B$, 39% increase in EGF compared to the control group. Conclusions Both early administration of Douchi extract and omega-3 were effective in regulating Th2, relieving inflammation and developing colonic mucosal tissues. Hataedock with Douchi extract may be more effective in modulating immune system in colonic mucosa than omega-3.
Objectives Hataedock (HTD) is an oral Korean herbal medical oral treatment that removes fetal toxic heat and meconium from new born babies. The purpose of this study is to evaluate whether Hataedock treatment of Duchi extracts has anti-inflammation effects in atopic dermatitis-like skin lesions in House Dust Mite-Induced NC/Nga Mice. Methods The mice were divided into 3 groups (n=10 per group) as follows: the control group (Ctrl group), AD-induced group (AE group), AD-induced with HTD treatment group (DT group). 3-week-old NC/Nga mice were introduced to Hataedock treatment, made of Duchi extract. After 4 weeks, House Dust Mite-Induced application was used six times per week for 3 weeks to induce the first atopic dermatitis, and second AD in 7 weeks after. To examine skin injuries and anti-inflammatory effect, PKC, MMP-9, iNOS immunohistochemistry were used. Results The alleviate effect of the skin damage and angiogenesis was observed in DT group. The damage of stratum corneum, hyperplasia, edema, infiltration of lymphocytes and distribution of capillary were decreased in DT group. Also, the study results suggested that Hataedock treatment made of Duchi extracts in DT group remarkably decreased skin damages by 51% (p < 0.001), as well as PKC by 91%, MMP-9 by 48% (p < 0.001), iNOS by 51% (p < 0.001). Conclusions Based on the study results, we observed that Hataedock treatment of Duchi extracts alleviates AD by diminishing various inflammatory cytokines, initial steps of AD development, in the skin lesions. Potential applications for prevention and treatment of atopic dermatitis are expected.
Objectives: This study investigated the effects of Douchi Hataedock on Th2-skewed conditions to control allergic rhinitis. Methods: NC/Nga mice were divided into three groups: 10 mice were assigned to the control group (CTRL; no treatment), 10 mice to allergic rhinitis-induced (ARE) without treatment group, and 10 mice to the allergic rhinitis-induced (FGT) after Douchi Hataedock treatment group. The 3-week-old mice of the FGT group were given one 10 mg/kg dose of Douchi Hataedock extract and resensitized to allergic antigens at weeks four, five, and six. Allergic rhinitis was induced primarily in mice nasal cavities for five days after one week of final sensitization. The second induction used the same method one week after the first induction was completed. After one week, the nasal mucosal tissues of each group were observed. Immunohistochemical staining for IL-4, STAT6, CD40, $Fc{\varepsilon}RI$, substance P, MMP-9, $NF-{\kappa}B$ p65, p-IkB, and iNOS in the nasal mucosa was also performed. Results: The FGT group had less respiratory epithelial damage and less mucin secretion in goblet cells than the ARE group and showed a 62% decrease in IL-4, 85% decrease in STAT6, 71% decrease in CD40, 69% decrease in $Fc{\varepsilon}RI$, 43% decrease in substance P, 49% decrease in MMP-9, 43% decrease in NF-kB p65, 38% decrease in p-IkB, and 73% decrease in iNOS compared to the ARE group. Conclusions: Douchi Hataedock lessens inflammation in epithelial and goblet cells and reduces inflammatory mediator secretion in a mouse allergic rhinitis model.

Methods After Hataedock treatment of Duchi extracts on days 0, 3-week-old NC/Nga mice were sensitized on days 28, 35, 42 by exposure of DNFB (dinitrochlorobenzene) and were induced to have AD. Immunohistochemistry of NF-${\kappa}B$ p65, iNOS, COX-2 and TUNEL assay of apoptotic body was used to identify changes of skin damages and anti-inflammation effects. Results The alleviate effect of the skin damage and angiogenesis was observed in DT group. The damage of stratum corneum, hyperplasia, edema, infiltration of lymphocytes and distribution of capillary were decreased in DT group. Also, the study results suggested that Hataedock treatment of Duchi extracts in DT group remarkably downregulated levels of NF-${\kappa}B$ p65 by 70% (p < 0.001), as well as COX-2 by 51%, iNOS by 62% (p < 0.001). Additionally, Hataedock treatment of Duchi extracts in DT group up-regulated apoptosis of inflammatory cells by 68% in atopic dermatitis-like skin lesion. Conclusions From the study results, we observed that Hataedock treatment of Duchi extracts alleviates AD through diminishing various inflammatory cytokines in the skin lesions, which are involved in the initial steps of AD development. It is anticipated to have potential applications for prevention and treatment of atopic dermatitis.
Objectives Hataedock is a treatment that dispels toxic heat and meconium which has been accumulated to the fetus from a pregnant mother via orally administering herbal extracts to a newborn baby. This study was conducted to compare the efficacy of Hataedock, with using the extract of Coptis japonica & Glycyrrhiza uralensis, to the early administration of probiotics for immunomodulation in the intestinal mucosa. Methods NC/Nga mice were divided into three groups; Control group (no treatment), CGT group (3-week-old mice given the extract of Coptis japonica & Glycyrrhiza uralensis), and MBT group (3-week-old mice given a Bifidobacterium). After 2 weeks, the intestinal mucosa tissues of each group of mice were observed. Immunohistochemical staining for IL-4, IL-13, CD40, $Fc{\varepsilon}RI$, $p-I{\kappa}B$, EGF, and VEGF in the intestinal mucosa was performed. Results CGT group showed 65% decrease in IL-4, 67% decrease in IL-13, 58% decrease in CD40, 72% decrease in $Fc{\varepsilon}RI$, 76% decrease in $p-I{\kappa}B$, 41% increase in EGF and 100% increase in VEGF compared to the control group. MBT group also showed 50% decrease in IL-4, 63% decrease in IL-13, 33% decrease in CD40, 53% decrease in $Fc{\varepsilon}RI$, 46% decrease in $p-I{\kappa}B$, 23% increase in EGF and 151% increase in VEGF compared to the control group. Conclusions These results suggest that both Hataedock, with using the extract of Coptis japonica & Glycyrrhiza uralensis, and early administration of probiotics were effective in regulating Th2, relieving inflammation and developing intestinal mucosal tissues. Hataedock with extract of Coptis japonica & Glycyrrhiza uralensis may be more effective for immunomodulation in intestinal mucosa than probiotics.
Objectives This study investigated the effects of Hataedock treatment with Douchi on induction of allergic rhinitis in obese induced NC/Nga mice. Methods NC/Nga mice were divided into control group (Ctrl), allergic rhinitis induced obese mice group (ARE), and allergic rhinitis induced obese mice group with Douchi Hataedock treatment (FGT). The 3-week-old mice of the FGT group were given one 10 mg/kg dose of Douchi Hataedock extract and sensitized with allergic antigens at weeks 4, 5, and 6. After 1 week of final sensitization, allergic rhinitis was induced primarily in mice nasal cavities for five days. After one week of the completion with the first induction, the second induction was introduced by the same method. After 1 week, few samples of the nasal mucosal tissues of each group were prepared. The factor of Th2 differentiation and inflammation control such that IL-4, STAT6, CD40, $Fc{\varepsilon}RI$, substance P, MMP-9, $NF-{\kappa}B$ p65, p-IkB, iNOS and COX-2 were observed by immunohistochemistry. Also, the difference in nasal mucosal injury was observed by histochemical method (PAS staining). Results The FGT group showed that reduced IL-4 production, STAT6 expression and CD40 expression by regulating excessive Th2 differentiation. Also, production of substance P and MMP-9 and activity of $Fc{\varepsilon}RI$ in mast cells were decreased. Inhibition of $NF-{\kappa}B$ p65 activity was induced by inhibition of p-IkB, and the production of inflammatory enzymes iNOS and COX-2 were decreased. In addition, the damage of intramural respiratory epithelium was low and excessive mucin secretion in goblet cells was low. Conclusions This study confirmed the possibility of controlling the allergic rhinitis in obese children who are expected to have an overactive inflammation.
The aim of this study is to evaluate the anti-inflammatory effect of Hataedock treatment using Coptidis Rhizome and Glycyrrhiza Uralensis (CG) mixed extract in allergic rhinitis induced NC/Nga mice. We divided NC/Nga mice into 3 groups as follows; allergic rhinitis-induced group after CG Hataedock treatment (CGT, n=10), no treatment group (Ctrl), allergic rhinitis elicited group (ARE). To induce allergic rhinitis, NC/Nga mice of 3 weeks age were sensitized on 7, 8 and 9week by Ovalbumin (OVA) antigen in intranasal space. Hataedock using CG extract was administered on week 3 in allergic rhinitis-induced group (CGT) after Hataedock treatment. To identify distribution of Interlukin (IL)-4, Cluster of differentiation 40 (CD40), high-affinity IgE receptor ($Fc{\varepsilon}RI$), substance P, Matrix metallopeptidase 9 (MMP-9), Nuclear $factor-{\kappa}B$ ($NF-{\kappa}B$) p65, Inducible nitric oxide synthase (iNOS) and Cycloxygenase-2 (COX-2), we used histological examination. CGT significantly inhibited IL-4 and CD40 response compared with ARE. The reduction of Th2 cytokine expression decreased inflammatory mediators such as $Fc{\varepsilon}RI$, substance P, MMP-9, $NF-{\kappa}B$ p65, iNOS and COX-2. Such immunological improvement induced reduction of respiratory epithelial damage and mucin secretion in goblet cell. These results indicate that Hataedock treatment suppresses allergic rhinitis through modulating of Th2 responses and diminishing various inflammatory mediators in nasal mucosal tissue. It might have potential applications for prevention and treatment of allergic rhinitis.
Objective This study is to learn the effects of Hataedock method using Coptidis rhizoma and Glycyrrhiza uralensis mixed extract on inflammatory response in allergic rhinitis-induced obese NC/Nga mice. Materials and Methods The mice were fed with high fat-diet to be obese, and were divided into 3 groups as follows; allergic rhinitis-induced obese mice group with Hataedock method (CGT, n=10), no treatment group (Ctrl), allergic rhinitis elicited obese mice group (ARE). To induce allergic rhinitis, NC/Nga mice of 3 weeks age were sensitized on 7, 8 and 9 weeks by ovalbumin antigen in intraperitoneal space. After 7 days of final sensitization, allergic rhinitis was initially induced in mice through nasal cavities for 5 days. After 1-week, allergic rhinitis was induced again by the same method. Histological examination was used to identify distribution of IL-4, CD40, STAT6, $Fc{\varepsilon}RI$, substance P, MMP-9, NF-${\kappa}B$ p65, iNOS and COX-2. Results Hataedock method significantly reduced IL-4, STAT6 and CD40 response (p<0.05). In CGT, the inhibition of Th2 differentiation decreased inflammatory mediators such as $Fc{\varepsilon}RI$, substance P, MMP-9, NF-${\kappa}B$ p65, iNOS and COX-2 (all p<0.05). The immunological improvement led reduction of respiratory epithelial damage and mucin secretion in goblet cell. Conclusion The results of this study show that the Hataedock method suppresses the expression of allergic rhinitis by decreasing the inflammatory mediators through the regulation of Th2 differentiation even when the inflammation reaction is increased by obesity. Therefore, Hataedock may have potential preventive measure of allergic rhinitis accompanied by obese.
