A Novel Screening Strategy for Salt-resistant Alpha-helical Antimicrobial Peptides from a Phage Display Library (Phage Display Library를 이용한 Salt-Resistant Alpha-Helical 항균 펩타이드의 새로운 탐색방법)

  • A novel screening strategy for salt-resistant antimicrobial peptides from a M13 peptide library was developed. Fusion of MSI-344, a magainin derivative and indolicidin to pIII coat proteins did not significantly affect viability of the recombinant phages, which indicated that the pIII could neutralize toxicity of the antimicrobial peptides and therefore it is possible to construct antimicrobial peptide library in Escherichia coli. On the basis of the conserved sequence of ${\alpha}$-helical antimicrobial peptides, a semi-combinatorial peptide library was constructed in which the peptides were displayed by pIII. To remove hemolytic activity from the library, the phages bound to red blood cells were removed, and the subtracted phage library was screened for binding to target bacteria Pseudomonas aeruginosa and Staphylococcus aureus under high salt concentrations. The screened peptides showed relatively low antimicrobial activity against the target bacteria. However, antimicrobial activities of the screened peptides P06 and S18 were not affected by the cation concentrations of 150 mM $Na^+$, 2 mM $Mg^{2+}$ and 2 mM $Ca^{2+}$ without significant hemolytic activity. This screening strategy that is based on binding capacity to target cells provides new potential to develop salt-tolerant antimicrobial peptides.

A Case Report of a Patient with Active Ulcerative Colitis Treated with Do-Che Decoction-based Korean Medicine (도체탕(導滯湯) 위주의 한방치료로 호전된 활동성 궤양성 대장염 환자 치험 1례)

  • Background : Sulfasalazine for treating ulcerative colitis has a clinical limitation due to its adverse effects. This case is about a patient with ulcerative colitis who could not tolerate sulfasalazine and was improved by Do-che decoction-based Korean medicine. Method and Results : Purulent and bloody diarrhea more than 20 times a day, mild fever, chilling and tenesmus were main symptoms of this patient who was diagnosed as ulcerative colitis 10 years ago. Do-che decoction-based Korean medicine which was recommended for purulent and bloody diarrhea in Dong-Ui-Bo-Gam, the classic literature of Traditional Korean Medicine was given to the patient for more than 1 month. After treatment, the frequency of purulent and bloody diarrhea, tenesmus and fever was decreased remarkably, and C-reactive protein, erythrocyte sedimentation rate, white blood cell count and maximum temperature also dropped to the normal range. Conclusion : Do-che decoction-based Korean medicine has a potential effect and may be a alternative therapeutic option in patients with ulcerative colitis who cannot tolerate sulfasalazine due to its adverse effects.

Anti-Inflammatory Effects of Glycyrrhiza glabra Linne Extract in a Dextran Sulfate Sodium-Induced Colitis Mouse Model (감초 추출물의 Dextran Sulfate Sodium 유도 마우스 궤양성 대장염 억제 효과)

  • The aim of this study was to evaluate the anti-inlfammatory effects of Glycyrrhiza glabra Linne extract on ulcerative colitis induced by 3% dextran sulfate sodium in mice. The experimental animals were divided into six groups: control(normal), DSS-induced colitis(control), 1 mg/kg, 10 mg/kg, and 100 mg/kg of Glycyrrhiza glabra Linne extract, and 150 mg/kg 5-aminosalicylic acid(5-ASA)(positive control). We evaluated the pathological disease activity index(DAI), change in weight, colon mucosa damage and myeloperoxidase(MPO) in colon mucosa. Treatment with 10 mg/kg and 100 mg/kg of Glycyrrhiza glabra Linne extract led to significant loss of body weight, the decrease of MPO activity and clinical symptoms such as DAI and histological change. In particular, 100 mg/kg Glycyrrhiza glabra Linne extract led to markedly greater improvement than 150 mg/kg 5-ASA treatment. These results suggest that Glycyrrhiza glabra mediated anti-inflammatory action on colorectal sites may be a useful therapeutic approach to ulcerative colitis.

Effects of Goihwa-san on Ulcerative Colitis Induced by Dextran Sulfate Sodium in Mice (괴화산(槐花散)이 Dextran Sulfate Sodium으로 유도된 생쥐의 궤양성 대장염에 미치는 영향)

  • Objectives : This study was carried out to investigate the effects of Goihwa-san(GHS) against ulcerative colitis induced by DSS (dextran sulfate sodium). Method : The sample group was divided into three. The control group consisted of mice that were not inflammation-induced. The pathological group was composed of untreated colitis elicited mice. The experimental group was administered GHS after colitis elicitation. The effects on ulcerative colitis were evaluated by the morphological change of colonic mucosa, the anti-oxidant effect, HSP 70, NF-${\kappa}$B, COX-1, COX-2 and iNOS. Results : In terms of immunohistochemical changes, the distribution of COX-1 in mice treated with GHS increased noticeably more than that in the pathological group. The distributions of HSP70, NF-${\kappa}$B, COX-2, iNOS in mice treated with GHS more decreased than those in the pathological group. Regeneration of surface epithelial cell and goblet cell in mucosa was observed by optical microscope. The colonic lengths in GHS-treated mice were more elongated than those of DSS only treated mice. Conclusion : GHS is a candidate treatment for ulcerative colitis.

  • This study is designed to evaluate the effects of an oriental herbal medication namely Danggwihwahyeolsan-gamibang and Gami-Jeonssibaekchulsan, on ulcerative colitis. Subject: A patient diagnosed ulcerative colitis who're not improved through western medication. Methods: The Clinical data was analyzed on a pastient with ulcerative clitis due to Changpung(腸風) resulted from Hanyeolchakjab(寒熱錯雜), whose symptoms were combined diarrhea, abdominal pain et al. The patient visited at the internal medicine department of Daegu Haany University affiliated Daegu Oriental Hospital on May 18. 2007 to June 25. 2007. The improvement of symptoms were checked by Seo Colitis Activity Index(AI).

Effects of Hwangryunhaedoktang on DSS-induced Colitis (황련해독탕이 DSS로 유발된 흰쥐의 궤양성 대장염에 미치는 영향)

  • Objectives : We examined the effect of Hwangryunhaedoktang(HH) on the experimental ulcerative colitis induced by dextran sulfate sodium (DSS). Methods : Experimental colitis was induced in rats by daily treatment with 5% DSS in the drinking water for 5 days. Afterward, the mts were divided into two groups: the control group was administered water and the sample group was administered HH for 7 days. Results : The sample group provided HH for 7 days demonstrated fast recovery of body weight compared with the control group. Histologic change showed fast regeneration of crypt and surface epithelial cells and decreased edema of the submucosa and decreased lymphatic follicle of mucosa compared with the control group. Immunohistochemical stain usingCOX-2 gene was decreased and there was localized Ki-67 positive reaction. Regeneration of surface epithelial cell and goblet cell in mucosa was observed by transmission electron microscope. These results indicate therapeutic effect of HH on DSS-induced colitis in rats.

  • The present study was performed to elucidate the ability of anti-$TNF-{\alpha}$ antibodies produced in sheep colostrums to neutralise $TNF-{\alpha}$ action in a cell-based bioassay and in a small animal model of intestinal inflammation. Colostrums from sheep immunized against $TNF-{\alpha}$ significantly inhibited $TNF-{\alpha}$ bioactivity in the cell based assay. The higher than anticipated variability in the two animal models precluded assessment of the ability of antibody to prevent $TNF-{\alpha}$ induced intestinal damage in the intact animal.

  • Pseudomembranous colitis, thought to be uncommon in children, is a bacterial, toxin-mediated inflammatory process resulting in acute or chronic diarrhea and is characterized by colonic pseudomembranes. It is mediated by toxins produced by Clostridium difficile and is increasingly recognized in pediatric population. Diagnosis is based on positive culture of C. difficile in selective media and positive test of C. difficile toxin. Oral metronidazole or vancomycin are the main treatment options but avoidance of further antibiotics should also be encouraged where possible. We have experienced a case of pseudomembranous colitis in a 4-year-old female presented with septic shock and colitis. This case was diagnosed with positive test of C. difficile toxin B and confirmed by isolation of the organism on cultire in selective media. Symptoms have been ameliorated by discontinuation of antibiotics and administration of metronidazole and oral vancomycin, and ICU care.

  • Ulcerative colitis, a diffuse inflammatory disease of the mucosal lining of the colon and rectum, is characterized by bloody diarrhea that waxes and wanes without apparent cause. In oriental medicine, Jakyaktang has been used to treat the disease, diagnosed as Damp-Heat dysentery. We prescribed Jakyaktang-gamibang to two patients who had suffered from ulcerative colitis diagnosed as Damp-Heat dysentery. The two patients were treated with Jakyaktang-gamibang and improved in consciousness of symptoms. We suggest that oriental medicine therapy is effective for treatment of ulcerative colitis.
