Effects of amprolium hydrochloride on expression of drug metabolizing enzyme genes in olive flounder Paralichthys olivaceus (Amprolium hydrochloride가 넙치 Paralichthys olivaceus의 약물대사 유전자 발현에 미치는 영향)

  • This study was undertaken to evaluate the effect of amprolium hydrochloride on detoxification process of olive flounder Paralichthys olivaceus. A series of two experiment was performed based on the LD50 value obtained for amprolium. First, thirty flounder (average weight 230.27 g; average length 27.99 cm) was randomly allocated into five groups. Treatment was carried out using intra-muscular injection of amprolium at the dose levels of 4, 8, 16, and 32 mg/kg body weight. At 8, 24 and 48 h post injection, liver and kidney were collected for expression assay of drug metabolizing enzymes and pro-inflammatory cytokine genes. We found that the interleukin-1β (IL-1β) mRNA level were induced at 32 mg/kg and CYP1A genes showed the opposite pattern, while UDP-glucuronosyl-transferase (UGT1A7) and GST were significantly reduced in the liver. Moreover, the suppression of drug metabolizing enzymes and cytokine gene in the kidney was observed after treatment. Another treatment was carried out using intramuscular injection with 4, 8, 16, and 32 mg/kg and 60, 80, 100, 120 mg/kg body weight. At 6 days post injection, liver was collected. The IL-1β expression was markedly induced in the experimental group treated with 4 mg/kg. In addition, glutathione S-transferase (GST) mRNA level was higher in the group with 4 mg/kg. In conclusion, our data suggests that amprolium seem to cause direct or indirect physical, or biological toxicity of flounders, although this drug is considered one of the safest synthetic anticoccidial drugs of the livestock industry.

The Change of Glutathione Metabolism in Liver and Kidney of Cisplatin treated Rats (Cisplatin 투여 후 백서의 간 및 신장에서 Glutathione 대사의 변화)

  • Glutathione (GSH) is a well-known antioxidative cellular component which is ubiquitous in nature. Several enzymes involved in GSH metabolism and recycling have been found to play important roles in detoxification of xenobiotics and free radicals. In this study, total GSH content, activity of GSH peroxidase and GSH reductase were measured in liver and kidney of cisplatin treated rats. Total GSH content (mM/g protein) of liver was higher in cisplatin treated rats ($1.51{\pm}0.28$) than of nontreated control ($0.95{\pm}0.28$), and in kidney, it was also higher in cisplatin treated rats ($0.87{\pm}0.20$) than that of control ($0.68{\pm}0.14$). The activity of GSH peroxidase (${\mu}M/mg$ protein/min) was lower in liver of cisplatin treated rats ($348.0{\pm}18.54$) than that of control ($415.5{\pm}53.15$), in kidney it was increase din cisplatin treated rats ($380.5{\pm}51.86$) compared to control ($327.3{\pm}20.36$). The activity of GSH reductase (${\mu}M/mg$ protein/min) was higher in liver of cisplatin treated rats ($3.09{\pm}0.88$) than that of control ($2.28{\pm}0.61$), in kidney it was also higher in cisplatin treated rats ($8.50{\pm}2.62$) than that of control ($3.30{\pm}1.10$). In summary, detoxification of ciplatin was revealed lesser effect in kidney as show increasion of GSH peroxidase and reductase and detoxification of cisplatin was expressed effectively in liver by increasing of GSH content and decreasing GSH peroxidase.

Effects of Phenolic Compounds of Persimmon Leaves on Antioxidative System and Miscellaneous Enzyme Activities Related to Liver Function in Ethanol-Induced Hepatotoxicity of Rats (감잎의 Phenolic Compounds가 에탄올을 투여한 흰쥐의 항산화계 및 기타 효소활성에 미치는 영향)

  • To investigate antioxidative effects of phenolic compounds separated from persimmon leaves(PL)(Diospyros kaki Thunb.) on the ethanol-induced hepatotoxicity in rat, Sprague-Dawley rats weighing 100-150 g were divided into 5 groups; control group(CON), PL(70 mg/kg) administered group(PEl), ethanol(5 mL/kg, 25%) administered group(ETH), PL(70 mg/kg) and ethanol administered group (PE2), and PL(140 mg/kg) and ethanol administered group(PE3), respectively. The antioxidative activity of persimmon leaves decreased in order of ethylacetate>interphase materials>n-butanol>chloroform>n-hexane>water fraction. The growth rate and feed efficiency ratio decreased by ethanol were gradually increased to the adjacent level of CON by administering PL. The serum activities of ALT, alkaline phosphatase and lactic acid dehydrogenase elevated by ethanol were decreased significantly. It was also observed that the activities of SOD, catalase, and GSH-Px of rat liver increased by ethanol were markedly decreased in PL administered group as compared to ETH. The GSH content of liver was decreased by ethanol, but that was increased in PE1 and PE2 compared with ETH as a dose-dependant manner. These results suggested that phenolic compounds separated from persimmon leaves have a possible protective and relievable effect on the ethanol-induced hepatotoxicity in rats.

Effect of Selenium Yeast on MPTP (1-methyl-4-phenyl-propion-oxypiperidine)-Induced Neurotoxicity in Mice (Selenium이 MPTP(1-methy-4-phenyl-1,2,3,6-tetrahydropyridine)에 의해 유도된 생쥐의 신경독성에 미치는 영향)

  • This study is investigated the effect of selenium against neurotoxicity induced by MPTP(1-methy-4-phenyl-propion-oxypiperidine) in mice. In order to demonstrate neuroprotective activity of selenium, mice were administrated orally with selenium(25, 50, 100 ${\mu}g/kg$, once/day) for 10 days, and MPTP(10 mg/kg) was injected subcutaneously into the mice for 6 days from the beginning 1hr before selenium treatment. Test of rota road activity was inhibited by treatment with selenium in MPTP-induced neurotoxicity group when compared to MPTP treatment group in normal mice. Monoamine oxidase(MAO)-B activity and cerebral lipid peroxide content were significantly decreased in the treatment of selenium in MPTP-induced neurotoxicity group when compared to MPTP treatment group in normal mice and MAO-A was not affected. Activities of cerebral superoxide dismutase, catalase and glutathione peroxidase were significantly increased in the treatment of selenium in MPTP-induced neurotoxicity group when compared to MPTP treatment group in normal mice. These results suggest that selenium might be estimated the result from the cooperative action of its inhibitory effect on monoamine oxidase-B with that of the enhancement of antioxidant(SOD, catalase, GSH-Px) defence ability.

Effect of Dietary Monascus Koji on the Liver Damage Induced by Bromobenzene in Rats (식이성 홍국이 Bromobenzene에 의한 간 손상의 해독에 미치는 영향)

  • In the present study, it is observed that Monascus diet may have a hepatoprotective effect on the liver damage induced by bromobenzene in rats. By treatment with bromobenzene (400 mg/kg, i.p.) once a day for 3 consecutive days, the liver damage was reduced in rats fed 2% Monascus diet, based on the liver functional and histopathological findings. Furthermore, retreatment of bromobenzene to the animals with damaged liver showed higher decreasing rate of hepatic glutathione content and increasing rate of cytochrome P450 dependent aniline hydroxylase activity at 4 h in rats fed 2% Monascus diet than those fed STD diet, and V$_{max}$ in glutathione S-transferase was higher in liver of rats fed 2% Monascus diet than those fed STD diet. On the other hand, activities of antioxidant enzymes such as hepatic glutathione S-transferase, catalase and superoxide dismutase were generally higher both in bromobenzene and 2% Monascus diet treated group than those fed STD diet. In conclusion, the rats fed 2% Monascus diet showed lower liver damage than those fed STD diet, which may be due to the acceleration of bromobenzene metabolism and detoxication of oxygen free radicals.s.

Effect of Ethanol Extract from Portulaca oleracea L. on the Lipid Metabolism in Rats fed Hypercholesterol Diet (고콜레스테롤혈증 유발식이에서의 쇠비름 에탄올 추출물이 흰쥐의 지질대사에 미치는 영향)

  • 쇠비름(Portulaca oleracea L.)은 쇠비름과(Portulacaceae)에 속하는 일년생 초본으로 우리나라 각지에 5~9월에 걸쳐 자생하고 있다. 한국보다는 중국에서 식용으로 애용되고 있으며, 해열, 이뇨, 산혈 등의 효능이 있어 임질, 요도염, 유종, 대하증, 임파선염 등에 약용으로 사용하고 있다. 쇠비름의 성분으로는 vitamin $B_1$ 및 C, saponin, tannin, flavonoid, dopamine, noradrenalin 등이 보고되었다. 쇠비름에 관한 연구로는 항산화효과, 항균효과 및 해독효과가 발표되었으나, 고지혈증에 관한 연구는 미흡한 실정이다. 따라서 본 연구에서는 고콜레스테를 유발식이에서 쇠비름 에탄을 추출물이 혈청 중 지질에 미치는 영향에 대해 검토하고자 하였다. 실험동물은 6주령의 Sprague Dawley 흰쥐 수컷을 각 실험군당 10마리씩, AIN-93M을 기본으로 하여 대조군(고지방식이), 쇠비름 에탄올 추출물 투여군의 3군(고지방식이+25mg/kg, 75mg/kg, 125mg/kg)으로 나누어 4주간 실험 식이를 급여하고 사육기간 중 사료 섭취량, 체중증가율, 식이효율을 측정하였다. 실험 종료 후, 혈액을 채취하여 혈액에서의 total lipid, total cholesterol, triglyceride, HDL-cholesterol은 효소법을 이용한 kit(아산제약)을 사용하여 측정하였고, 심혈관계 질환의 위험도에 이용되는 동맥경화지수는(atherogenic index, AI) 계산식을 이용하여 산출하였다. 실험결과, 최종체중, 체중증가율, 식이효율 및 장기무게는 실험군 간의 차이를 나타내지 않았다. 그러나 고지방식이에 쇠비름 에탄올 추출물을 125mg/kg의 농도로 병합 투여했을 때 혈중의 total lipid, total cholesterol 및 triglyceride는 대조군에 비해 유의적으로 감소하였으며, 고지방식이에 의해 증가된 동맥경화지수도 대조군에 비하여 유의적으로 저하되었다. 그리고 고지방식이로 증가된 ALT 및 AST 활성이 쇠비름 에탄올 추출물 투여로 유의적으로 감소하였다. 따라서 쇠비름 에탄올 추출물은 혈청 중 지질 개선 효과와 간 기능 개선효과가 있는 것으로 사료되나, 어떤 성분이 콜레스테롤 저하 효과를 나타내는지 보다 자세한 연구가 요구되어 진다.

Effects of the Omija (Schizandra chinensis Baillon) extract on the metabolism and renal Cadmium contents in Cadmium administered rats (오미자 추출물이 카드뮴을 급여한 흰쥐의 대사와 신장내 카드뮴 함량에 미치는 영향)

  • This study was conducted to investigate the effects of Korean Omija extract on the hepatic and renal function in cadmium intoxicated rats. Male Sprague Dawley of 4 weeks old, weighing 100$\pm$10 g, were randomly assingned to four groups which received over four weeks one of the followings : deionized water for control (CW),3% Omija extract (OE),50 ppm cadmium water (CD) and 50 ppm cadmium water plus 3% Omija extract (CDOE). The results are as followes: there ware no significant differences between CD and CDOE in the body weight gain and food efficiency ratio. But Cadmium contents of kidney, GPT and LDH activities were significantly reduced in CDOE as compared to CD. Weight gain of kidney in CDOE, significantly higher than that of CD, increased to nearly normal level. GOT activities in CDOE, significantly different from that of CD, also considerably lowered to same level as that of tile normal rat group, CW The results suggested that Omija extract may have some protective effects from cadmium intoxication by reducing cadmium accumulation in kidney.

Effect of Soybean Supplementation on Murine Drug-metabolizing Enzymes and Benzo(a)pyrene-induced Lung Cancer Develpoment (콩보충식이가 생쥐의 해독효소계 및 Benzo(a)pyrene에 의해서 유도된 폐암발생에 미치는 영향)

  • Soybean has drawn much attention mainly due to its chemopreventive action as well as antiestrogenic effect. Although suppression of breast and prostate cancers were believed to be exerted via antiestrogenic or antiandrogenic activity of genistein, its mechanism of prevention against other cancers has not been clearly demonstrated. We proposed that prevention by soybean from other cancers than sex hormone -related cancers was achieved via modulation of drug-metabolizing enzymes. Addition of acid hydrolysate of 80% methanol extract of soyflour to diet caused a significant induction of quinone reductase, an anticarcinogenic marker enzyme and one of drug-metabolizing enzymes, in mouse lung while it suppressed arylhydrocarbon hydroxylase, involved in bioactivation of procarcinogens, in kidney and small intestine. It is likely that active components exist in a conjugated form and released by acid hydrolysis to be able to affect drug-metabolizing enzyme and exert chemopreventive activity. Benzo(a)pyrene-induced tumor development in mouse lung was greatly reduced by soybean extract supplementation, which is consistent with the extract's capability to modulate favorably arylhydrocarbon hydroxylase and quinone reductase towards chemoprevention.

  • Preventive effect of azalea pollen extracts against aniline-induced hepatic toxicity in mice was investigated in this experiment. When the biochemical and histological changes were measured, preventive effect was more striking by treatment with water extract. After treatment with azalea pollen extracts, hepatic microsomal aniline hydroxylase activity increased as compared to control. Whereas, aniline level in serum and liver significantly decreased. The Vmax value without affecting Km value increased by the water extract treatment, the results obtained suggest that the characteristics of increase in the aniline hydroxylase activity may include induction of enzyme proteins. These data indicate that the observed preventive effects of azalea pollen extracts against hepatotoxicity is due to the induction of aniline metabolizing enzyme.

Ultrastructural Studies on Mercury Poisoning in the Liver, Kidney and Gills of Carassius carassius L. (수은중독에 의한 붕어(Carassius carassiusr L.) 장기의 미세구조 변화)

  • Electron microscopic studies were made to investigate changes in the fine structure of the liver, kidney and gills of Carassius carassius L. following exposure to 1 and 2.5 ppm of $HgCl_2$. The following results were obtained: 1. In the mercury-treated liver cells, an increase in the number of lysosomes were noticed. These lysosomes appeared to be of two types; round ones containing some crystalline structures and others with phagocytosed glycogen granules and mitochondria. Also observed were mitochondrial swelling where the matrix appeared less electrondense, and segregation of the nucleoli in the nucleus. 2. In the kidney, mercury treatment resulted in thickening of the basement membrane of the glomerulus, and appearance of vacuoles and cytoplasmic bodies in the proximal convoluted tubule. The vacuoles seemed to be formed from mitochondria. Nuclear shrinkage was also noticed at 2.5 ppm of $HgCl_2$. 3. Many large and small lysosomes appeared in response to mercury in the epithelial cells of the gill lamella. Also the lamellar membrane became fuzzy in appearance. 4. It can be concluded from these results that mercury-induced changes in the fine structure are associated with activation of detoxication processes and impairment of energy metabolism.
