In vivo Metabolism of Flupyrazofos into Plutella xylostella (Lepidoptera: Yponomeutidae) and Spodoptera exigua (Lepidoptera: Noctuidae) (배추좀나방과 파밤나방의 채내에서 Flupyrazofos의 대사)

  • In vivo metabolism study was carried out to find out the biochemical or metabolic tolerance mechanism between Diamond backmoth (DBM), Plutella xylostella and Beet armywarm (BAW), Spodoptera exigua to flupyrazofos. They showed some differences between the DBM and BAW. About 20% of flupyrazofos applied to the 3rd instar larvae of DBM was metabolized within 1 h and about 50% of that was metabolized within 4 h. The metabolites of flupyrazofos-oxon in 3rd instar larvae of DBM were increased 10 times more at 4 h than 1 h after application. The amounts of flupyrazol were nearly same between at 1 h and 4 h. The amount of unknown and origin increased 2 and 3 times more at 1 h than 4 h after application, respectively. In the 4th instar BAW larva, about 50% of flupyrazofos was metabolized within 1 h and about 70% of that was metabolized within 4 h. As metabolites, the amounts of flupyrazofos-oxon increased 2 times more at 4 h than 1 h after application. The amounts of flupyrazol increased 4 times more at 4 h than 1 h after application. The amount of unknown and origin increased 2.5 and 2 times more at 4 h than 1 h after application, respectively. From the study, it is supposed that hydrolytic enzyme, esterase, cleave the alkyl bond of flupyrazofos and conjugates with flupyrazofos. This seems to be the main tolerance mechanism of BAW to flupyrazofos.

Effects of Extracts of Persimmon Leaf, Buckwheat Leaf, and Chinese Matrimony Vine Leaf on Body Fat and Lipid Metabolism in Rats (감잎, 메밀잎 및 구기자잎 추출물의 식이보충이 흰쥐의 체지방 감량과 지질대사 조절에 미치는 효과)

  • The objective of this study was to investigate the anti-obesity and lipid-lowering effects of extracts of persimmon leaf (PWE), buckwheat leaf (BWE), and chinese matrimony vine leaf (LWE) in rats fed a high-fat diet (HFD). Male Sprague-Dawley rats (n=40) were divided into four groups: HFD (35% fat, w/w), HFD (38.5% fat, w/w) supplemented with PWE (10%, w/w), BWE (10%, w/w), and LWE (10%, w/w) groups. The epididymal, perirenal, and interscapular white adipose tissue (WAT) weights as well as plasma leptin level were lowest in the LWE group. Supplementation with PWE and BWE also tended to lower the perirenal and retroperitonal WAT weights compared to the HFD control group, and there was a significant decrease in plasma leptin concentration. Furthermore, plasma triglyceride concentration, hepatic cholesterol content, and hepatic lipid droplet accumulation were significantly lower in the PWE, BWE, and LWE groups than in the HFD group. BWE supplementation markedly lowered plasma total cholesterol concentration, although there were no significant differences in plasma HDL-cholesterol concentration and ratio of HDL-cholesterol/total cholesterol among the groups. Hepatic HMG-CoA reductase activity was significantly higher in the PWE and LWE groups than in the HFD group, and hepatic ACAT was not changed by extract supplementation. However, supplementation with PWE, BWE, and LWE significantly increased fecal acidic sterol content in rats fed a HFD. These results suggest that supplementation with PWE, BWE, and LWE may be an effective anti-obesity strategy by lowering body fat weight and improving plasma and hepatic lipid profiles in HFD-fed rats.

Antioxidant Effect of Green Tea Extracts on Enzymatic Activities of Hairless Mice Skin Induced by Ultraviolet B Light (녹차추출물에 의한 쥐표피의 효소에 대한 항산화 효과)

  • This study was carried out to investigate the acute cutaneous enzymatic antioxidant activity of green tea extracts (GTE) on hairless mice skin after a signal exposure to ultraviolet B radiation. GTE has been in corporated at concentration of 5, 25, 50 and $100\;{\mu}g$ into hairless mice skin. Under exposure conditions of $1.0\;joule/cm^{2}$, impairment of cutaneous enzymatic activity was observed. Catalase and glutathione reductase were significantly influenced in dose-dependent manner by GTE, however glutathione peroxidase and superoxide dismutase were not affected. To measure inhibition effect of the GTE on lipoxygenase. $50\;{\mu}g$ GTE extract was added in vitro to arachidonis acid. GTE showed higher inhibition effect on 5-hydroxyeicosatetraenoic acid (HETE) and 8-HETE than metalolic products of 12- or 15-HETE. The addition of 5, 25, 50 and $100\;{\mu}g$ GTE inhibited the metabolite formation of 5-HETE by 32, 52, 62 and 80%, respectively, and the metabolite formation of 8-HETE by 36, 47, 70 and 84%, respectively.

Radioprotective Effects of Cordyceps sinensis Extracts on ${\gamma}-Irradiated$ Mice (동충하초 추출물이 감마선에 조사된 생쥐에 미치는 방사선방호효과)

  • Effect of single intraperitoneal administration of Cordyceps sinensis (Cs) extract at 24 hour before whole-body ${\gamma}$ - irradiation on the survival ratio, body weight, organ weight changes and serum metabolites in the irradiated mice were investigated. The single pre-administration of Cs extract increased the 40-day survival ratio of irradiated mice from 66.7% to 83.4%. The administration of Cs extract completely prevented weight reductions of spleen and thymus produced by ${\gamma}$ - irradiation (P<0.05, P<0.01). Similar but somewhat less radioprotective effect was also found in the testis of the Cs treated mice. The administration of Cs inhibited the serum hyperglycemia produced by irradiation on the day 7th(P<0.01). However, it did not influence the serum cholesterol and protein levels on the days examined. The present study is the first report regarding Cs which was tested and found to be radioprotective.

  • As the previous report, by the administration of Sogi , Gyrophoraesculenta, to rats, cholesterol levels in liver and plasma were lowered. To know the Characteristics of the effective material contained in Iwatake, the aqueous and the alcoholic extracts were prepared, and these extracts were miked in the diet. After the feeding for 6weeks, cholesterols in liver and in plasma were determined. Either the aqueous or the alcoholic extracts administered groups showed a little low value as to these cholesterol levels compared with the value of group without the extract, but these values of alcoholic extract administered group were lower than those of aqueous extract group. From this result, it was assumed that this effective material would be the organic compound with hydrophilic groups.

Effect of Methanol Extract from Mulberry Fruit on the Lipid Metabolism and Liver Function in Cholesterol-Induced Hyperlipidemia Rats (뽕나무 품종별 오디추출물의 섭취가 흰쥐의 지질대사 및 간장기능에 미치는 영향)

  • We conducted this study to investigate the effect of methanol extract from Mulberry Fruit on the lipid metabolism and liver function in cholesterol-induced hyperlipidemia rats. We used 5 Mulberry Fruit varieties, including Daeryukppong, Kuksang 20, Subongppong, Wild variety and Cataneo. The yield of methanol extract from freezing dried Mulberry Fruits was 43.5-64.6%. Animal treated with Wild variety from Mulberry Fruit showed the decrease in body weight, food efficiency ratios, serum triglyceride (85%), and GOT value (86%). Our results suggest the hypolipidemic action of Wild variety from Mulberry Fruit.

Investigation of Varied MR Spectra by TE and Metabolite Amount in the Localized Voxel using the MR Cone-shape Phantom (자기공명분광법에서 TE와 Voxel 내의 대사물질 양에 따른 스펙트럼 변화 평가에 관한 연구)

  • The purpose of this study is to investigate the spectra of a magnetic resonance spectroscopy (MRS) in accordance with the variance of TE and the volumes of metabolites in a localized voxel for the quality assurance using a designed single voxel spectroscopy QA phantom. Because a cone-shade phantom is designed as the volume of metabolite in a localized voxel is changeable, we try to analyze the peaks of each metabolite (NAA, Cr, Cho, Lac, etc.) in accordance with metabolite volume in a localized voxel as well as echo time (TE). All data were obtained using a 3T MRI/MRS machine and analyzed using $jMRUI^{(R)}$. The results of this study show that TE is in inverse proportion to the noise of MRS and the longer TE and the less metabolite volume in the localized voxel, the peak intensities of each metabolite decrease. In case of the lactate, its peak was observed on the all TE only if the greatest metabolite is included in the localized voxel. Then, the intensity of a metabolite is more sensitive to the metabolite volume in the localized voxel than the TE. These obtained in vitro MRS data is provide the guideline that is important for in vivo metabolite quantification. But, in the edge of cone-shape vial air bubbles were observed and spectrum could not obtained. Therefore our cone-shape MRS phantom needs to be modified in order to solve these problems.

Carbohydrate Metabolism in Preimplantation Stage Embryos and the Role of Metabolites (착상전 초기 배아에서 탄수화물 대사와 그 대사물의 역할)

  • Proper development of fertilized oocyte to blastocyst is a key step in mammalian development to implantation. During development of preimplantation embryos, the mammalian embryo needs supply the energy substrate for keep viability. Usually mammalian oocyte get substrate especially energy substrate from oviduct and uterus, because it does not store much substrate into cytoplasm during oogenesis. Carbohydrates are known as a main energy substrate for preimplantation stage embryos. Glucose, lactate and pyruvate are essential component in preimplantation embryo culture media and there are stage specific preferences to them. Glucose transporter and $H^+$-monocarboxylate cotransporter are a main mediator for carbohydrate transport and those expression levels are primarily under the control of intrinsic or extrinsic factors like insulin and glucose. Other organic substances, amino acids, lipids and nucleotides are used as energy substance and cellular regulation factor. Though since 1960s, successful development of fertilized embryo to blastocyst has been accomplished with chemically defined medium for example BWW and give rise to normal offspring in mammals, the role of metabolites and the regulation of intermediary metabolism are still poorly understood. Glucose may permit expression of metabolic enzymes and transporters in compacting morula, capable of generating the energy required for blastocyst formation. In addition, it has been suggested that the cytokines can modulate the metabolic rate of carbohydrate in embryos and regulate the preimplantation embryonic development through control the metabolic rate. Recently we showed that lactate can be used as a mediator for preimplantation embryonic development. Those observations indicate that metabolites of carbohydrate are required by the early embryo, not only as an energy source, but also as a key substrate for other regulatory and biosynthetic pathways. In addition metabolites of carbohydrate may involve in cellular activity during development of preimplantation embryos. It is suggested that through these regulation and with other regulation mechanisms, embryo and uterus can prepare the embryo implantation and further development, properly.

Effects of an aqueous extract of purple sweet potato on nonalcoholic fatty liver in high fat/cholesterol-fed mice (고지방/고콜레스테롤 식이를 섭취한 마우스에서 자색고구마 열수추출물 보충이 지방간 저항성에 미치는 영향)

  • Purpose: Anthocyanins from purple sweet potato (PSP) have been investigated in vitro and in animals and found to have a protective effect against oxidative hepatic damage. In this study, we investigated that aqueous extract of PSP can ameliorate the dysfunction of lipid metabolism in mice fed a high fat/cholesterol diet. Methods: Forty C57BL/6J mice were randomly divided into 5 groups (n = 8) and fed one of the following diets for 8 weeks; normal fat (NF) diet; high fat/cholesterol (HFC) diet; HFC with 1.25% PSP (HFPL) diet; HFC with 2.5% PSP (HFPM) diet; HFC with 5% PSP (HFPH) diet. Results: Non-alcoholic fatty liver was manifested in the HFC group by showing increased levels in plasma alanine aminotransferase (ALT) activity, total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C), increased level of TC and presence of many large lipid droplets in the liver, and increased fat cell size in the HFC group compared with the NF group. However, administration of HFC induced a significant decrease in food intake, resulting in decrease in fat mass. Co-administration of PSP did not lead to reversal of body weight changes, ALT activity, and lipid levels in plasma and the liver, but suppressed excess enlargement of the fat cell size through increasing carnitine palmitoyltransferase-1 (CPT-1) gene expression in the liver. Accordingly, the number of fat droplets in the liver was reduced in PSP administered groups. Conclusion: Taken together, these results suggest that PSP may have a protective effect on the dysfunction of lipid metabolism. Conduct of further studies on the coordinated regulation of PSP for lipid metabolic homeostasis at the liver-adipose tissue axis is needed.

Effects of Water Extract from Orostachys japonicus on Lipid and Antioxidant Metabolism in the Liver of Obese Mice (와송이 고지방 식이로 유도된 비만 쥐의 간내 지질 및 항산화 대사에 미치는 영향)

  • The purpose of this study was to investigate the changes in the liver lipid and antioxidant metabolism in obese rats following ingestion of Orostachys japonicus (OJ) hot water extract. The rats were fed a high fat diet for 8 weeks to induce obesity. In the experimental group, 2.5% or 5% OJ hot water extract was added to the diets for 6 weeks. To determine the changes in the liver lipid and antioxidant metabolism in obese rats, the livers were collected and weighed, lipid content was measured, and antioxidant enzyme activity was determined. A significant decrease in liver weight was observed in the group fed with OJ hot water extracts and decreased lipid accumulation was observed in the morphological observation of liver tissue. Total cholesterol and triglyceride contents were also decreased. Both OJ extracts significantly decreased 2,2-diphenyl-1-picrylhydrazyl radical and intracellular reactive oxygen species levels. Further, the levels of the antioxidant enzymes superoxide dismutase and catalase were significantly increased in rats treated with OJ extracts. These results suggest that the intake of OJ hot water extracts may improve liver lipid metabolism by activating antioxidant enzymes.
