Effect of Heating Time on Contents of Amino Acids and Related Compounds in the Muscle Extract of Snakehead (가열시간별 가물치육 엑스중의 아미노산 및 그 관련화합물의 변화)

  • As a fundamental study of fresh water fish on a suitable cooking method and on flavor components from the view point of food science, changes in the free amino acid composition of the muscle extracts of snakehead (Channa argus) during heating in boiling water were investigated. The muscle extract of raw fish was featured a very high content of glycine, taurine, glutamic acid and histidine, and a large amount of urine was also determined in the extract; the former four components comprised about $53\%$ of the total free amino acids. The total extractable nitrogen was greatly increased with the heating time till 120 minutes of heating, while it gradually decreased thereafter. The apparently increased components on heating of 120 minutes were taurine, glycine, alanine, hydroxyproline, and ${\beta}$-aminoisobutyric acid, etc. including urea. After hydrolysis of the extracts, some of the amino acids were increased, the content of ethanolamine, lysine, 1-methyl histidine, phenylalanine, glutamic acid and taurine, etc. were apparently increased.

Freshwater Fish Fauna in the Seomjin River, Gokseong-gun, Korea (섬진강 수계의 곡성군 어류상)

  • The fauna of freshwater fish and the structure of fish community were investigated from March to October in 2001 at six stations in the middle reach of the Seomjin River, Gokseong-gun. The collected fishes (1,786 individuals) were identified into 30 species of 22 genera belonging to S families. Of them, cyprinid fish occupied 26 species, only one species (Rhinogobius brunneus) was secondary freshwater fish. Dominant species was Zacco platypus (dominance index: 24.2%) and subdominant species was Pungtungia herzi (10.7%). The endemic species of Korea were 11 species including Rhodeus uyekii (36.7%) about the ichthyofauna. In estimation of water quality by diversity indices, the general conditions of water quality of all survey times and all stations were relatively fine $(\beta$-mesosaprobic, diversity index: 2.70). But four survey sites except St. 2 and St. 3 showed severe water pollution with one or move times in total survey periods.

  • Crucian carp Carassius carassius L., Skate-fish Parasilurus asolus, snake head Ophicephalus argus and loaches Misgrunus anguillicaudatus have been esteemed as the tasty fresh water fish in Korea. However, a little has been studied on their taste compounds. Amino acids, nucleotides and their related compounds as the taste giving substance in the dorsal muscle of crucian carp, skate-fish, snake head and loaches were analyzed. Hypoxanthine (4.6-30.3 ${\mu}mole/g$, on dry base) was dominant among the nucleotide materials in the dorsal muscle of crucian carp, skate-fish, and snake head while IMP (12.8 ${\mu}mole/g$) was detected with the highest amount in loaches meat. IMP was also high in the muscle of skate-fish (13.5 ${\mu}mole/g$) and snake head (5.6 ${\mu}mole/g$). The other nucleotide materials, CMP, UMP, GMP, AMP, and inosine were present less than 2.0 ${\mu}mole/g$ in all the fish meat. The amino acid composition showed that glutamic acid and aspartic acid were the most abundant in the dorsal muscle of crucian carp, skate-fish, snake head and loaches. The analysis of free amino acids showed that histidine, lysine, glycine, and serine were the leading amino acids in the dorsal muscle of crucian carp, alanine, serine, and lysine were of skatefish, glycine, lysine, alanine, and serine were of snake head, while alanine, serine, and glycine were of loaches. Isoleucine and leucine were detected in trace amount in the muscle of skate-fish, snake head, and loaches. It is believed that alanine, Iysine, serine, glycine and histidine along with IMP and hypoxanthine may play an important role as the taste compounds in fresh water fish.

Evaluation of Water Quality in the Keumho River System According to the Freshwater Fishes (담수어를 이용한 금호강수계의 수질판정)

  • The fish species collected in the Keumho River basin are 42 species 31 genera belonging to 15 Families. This report was investigated for the evaluation of water quality in the Keumho River system which is a tributary of Nakdong River in Korea on september in 1999. The fishes collected were 42 species, 31 genera belonging to 15 Families. The dominant species were 5 species; Zacco platypus, Zacco temmincki, Squalidus chankaensis tsuchigae, Moroco oxycephalus, Squalidus gracilis majimae, and 8 species; Hemibarbus longirostris, Pseudogobio esocinus, Culter brevicauda, Cobitis rotundicaudata, Pseudobagrus fulvidraco, Pungitius sinensis kaibarae, Monopterus albus, Channa argus were rare species. The relationship among the GPI, EC and BOD by the organic pollutants were over 0.9. The group pollution index(GPI) was lowest at St. 1(0.85) and highest at St. 1(0.85) and highest at St. 5(2.33). The water quality of the Keumho river divided into 3 parts; the water of upper reaches in river(St. 1) was 1st class(oligotrophic condition), middle parts(St. 2, 3, 4) were 3rd class($\alpha$-mesosaprobic condition) and lower part(St. 5) was 4th class(Polysaprobic condition) as the source of tap water, respectively. And the tributary which are the Sinryeong Stream(St. 6), the Sincheon(St. 7) and the Donghwa Stream(St. 9) in Keumho river were 2nd class as the source of tap water. The results in this study was represented same patterns as the result by the use of indicator species like as algae and invertebrates for the discrimination of water quality. So, some freshwater fish species can be use applicant for the discrimination of water quality.

  • Experimental infection of rainbow trout Oncorhynchus mykiss with viral hemorrhagic septicemia virus (VHSV, genotype IVa) from olive flounder Paralichtys olivaceus was examined. The cumulative mortalities of three different lot of rainbow trout fry challenged with VHSV ($10^{6.3-7.3}TCID_{50}$/fish) were less than 15%. No difference of virulence was observed in experimental infection using 5 in vivo passaged VHSV and original VHSV. No mortality was observed in seawater-reared rainbow trout (adult) challenged with VHSV ($10^{5.3-6.3}TCID_{50}$/fish) and virus was not detected in the fish. We thus concluded that VHSV from olive flounder has low virulence to rainbow trout fry, but not pathogenic to seawater-rainbow trout (adult).

  • Sensory evaluations of synthetic extracts prepared on the analytical data in the previous papers (Yang and Lee, 1979, 1930-a, 1980-b, 1982) were undertaken to determine the origin of the taste of wild common carp, Cyprinus carpio, and Korean snakehead: Channa argus. Taste panel assessments of synthetic extracts prepared with each extractive component omitted were carried out by a triangle difference test, and changes in taste profile were assessed. The synthetic extracts, prepared with about 40 pure chemicals based on the analytical data from the species satisfactorily revealed the natural taste of the original extracts except slight difference in meaty taste and mildness. From the results of omission test the major components which contribute to produce the taste of the species were assessed as follows: glycine, glutamic acid, arginine, lysine, threonine, alanine, IMP, inosine, hypoxanthine, glucose, succinic acid, $Na^{+},\;Cl^{-}\;and\;PO_{4}^{3-}$ in common carp: glycine, alanine, glutamic acid, IMP, Na+ and $PO_{4}^{3-}$ in Korean snakehead.

Changes in Prolactin and Growth Hormone Gene Expression in Three Freshwater Teleosts with Rapid Changes in Salinity (급격한 염분변화에 따른 담수어 3종의 프로락틴 및 성장호르몬 유전자 발현변화)

  • The changes in osmolality and the gene expression profiles of prolactin (PRL) and growth hormone (GH) with rapid changes in salinity were compared in the eel (Anguilla japonica), crucian carp (Carassius carassius), and masu salmon (Oncorhynchus masou). Fish stocked in freshwater (FW) were abruptly transferred to experimental tanks containing FW, 50% seawater (50% SW), or 100% SW (SW). Blood samples and pituitary glands were collected 2 and 24 hrs after the exposure. No mortality was observed in SW eel (n=6), whereas all of the crucian carp (n=6) and two masu salmon (n=6) exposed to SW died after land 24 hrs, respectively. The PRL mRNA levels of the eel and masu salmon decreased in 50% SW and SW compared to those of the fish kept in FW after 24 hrs, whereas the PRL levels of crucian carp were higher in 50% SW than in FW. Unlike the PRL mRNA levels, the GH mRNA levels of the eel did not differ significantly among three different salinities, while the GH mRNA levels of crucian carp and masu salmon increased significantly in 50% SW and SW after 24 hrs. The serum osmolalities increased marginally in the eel and masu salmon in 50% SW at 24 hrs (19% and 9%, respectively), whereas those of crucian carp increased abruptly in 50% SW (50% increase). These results suggest that the synthesis of PRL and GH is important in relation to the osmoregulatory system with environmental changes in salinity.

  • Some of ecological factors and the early life history of endangered freshwater fish, Pseusobagrus brevicorpus, were investigated to proliferate artificially and to restore the population by release seedlings. The density of population in restricted area by small scale dam was 1.25 ind. $m^{-2}$, and the low capacity for reproduction having 200$\sim$250 eggs ind$^{-1}$. were thought to be a reason of this endangered situation. From the result of induction of natural spawning in aquarium after hormone injection it was considered that the spawning substrate would be aquatic plants with slender branches. The hatching was observed after 50 hrs after fertilization and the larvae showed aggressive behavior. Yolk was almost absorbed 3 days after hatching, and the larvae shaped tad-pole grew up to fry after 30 days. We discussed on the strategies for conservation of this species according to those results from investigation on ecology and early life history.

An Epidemilolgical Survey of Echinostoma hortense Infection in Chongsong-gun, Kyongbuk Province (경북 청송군 일부 기역의 호르틴스극구흡충 유행양상)

  • An epidemiological study was performed to know the infection status of 5. hortense among the human residents and fish intermediate hosts in Chongsong·gun, Kyongsangbuk·do. Out of 263 people examined, 59(22.4%) were positive for the eggs of E. hortense. Total 152 fresh water fishes (9 species) were ellamined, and Odentobutis obscura interrupts, Misgurnus an- guiIZicaudatus and Squalidus coreanus were found to harbour the metacercariae of E. hortense. Juvenile or adult E. hertense were expelled from 35 out of 54 cases treated with pragiquantel. The number of worms Per individual ranged from 1 to 649, with the average burden of 51. The eosinophil count of infected persons ranged 1∼24% (6% in average). The number of eggs per day Per worm (EPDPW) in human host was estimated about 1,478. The results showed that Chongsong-gun is an endemic area of E. hortense.

The Fish Migration through Fishlock at Youngam Lake (영암호 갑문식어도를 통한 어류의 회유)

  • The dimension of the fishlock at the Youngam Lake is 6.6m wide and 30m long, and it has been used for both fish migration and ship passage. The capacity of attraction freshwater is $0.172\;\textrm{m}^3/s$, and the water depth of the fishlock varies between 1.9m - 5.7m due to the tide. Fishes were sampled weekly by using 3m wide maze net from April to June, 1999, and by using purse seine covering the entire space of fishlock from July to November, 1999. 116,014 samples were comprised of 25 species, and 99.5% of the samples were 10 kinds of diadromous fishes. The order of the diadromous fishes in abundance was Hemirhamphus kurumeus, Hypomesus olidus, Coilia ectens, and Mugil cephalu, where these four species consist 99% of the individuals, and the other six species contribute only a few of individuals. Each species had unique peak migration period. The peak migration of Hypomesus olidus, Mugil cephalus, Coilia ectens, and Hemirhamphus kurumeus happened on July 20, July 15, May 10, and May 1, respectively.tively.
