맛집탐방 - 이명닭발 -문화의 거리 속 '이명닭발'

  • 장마가 기승을 부리던 지난 7월 어느날 상수역 근처 닭발 전문점을 찾았다. 예로부터 닭발은 퇴행성관절염에 이용되는 한약 처방과 같이 달여서 복용하는 경우가 많았다. 근래에는 미용 식품으로도 환영받고 있다. 닭발은 피부나 관절을 구성하는 콜라겐이나 콘드로이친 성분이 다량 함유되어 있으며 또 다수의 단백질 성분이 함유 되어 있어 소화흡수도 잘되고 영양학적으로 우수한 식품이다.

Production of Yeast Extract by a Combined Method of Autolysis and Enzymatic Hydrolysis (자기소화와 효소가수분해 방법을 병용한 효모 추출물의 제조)

  • A combined method of autolysis and enzymatic hydrolysis of baker's yeast was developed for the production of yeast extract, which is widely used as a natural food ingredient. From statistical analysis, NaCl and ethanol addition were found to be significantly effective factors in autolysis of yeast. The optimum dosages of salt and ethanol were 3% and 1%, respectively. Heat treatment and the use of cell lytic enzyme were not significantly effecting on the autolysis. Yeast hydrolysate was prepared by autolysis, followed by enzymatic hydrolysis using proteases, nuclease and deaminase. Additionally, the hydrolysate was processed by downstream process including Maillard reaction and debittering. The total dry matter yield and total nitrogen yield for the process were 76% and 59%, respectively. Compared to a process using brewer's yeast, when baker's yeast was used as a raw material, a higher recovery yield was obtained.

Additivity of Ileal Crude Protein Digestibility and Comparison of Digestibility with Methodological Consideration in Broilers (육계에서의 회장 조단백질 소화율 가산성 평가 및 방법론적 소화율 비교)

  • This study was aimed at evaluating the additivity of crude protein digestibility in mixed diets of corn and soybean meal (SBM), and comparing direct and indirect methods for evaluating crude protein (CP) digestibility. Totally, five hundred and twenty-five 18-day-old broiler chickens were grouped into 7 blocks based on body weight, and randomly allocated to 6 treatment groups in a randomized complete block design. The basal diet, diet 3, was corn-SBM-based, containing 65% corn and 28% SBM. Diets 1 and 5 contained corn and SBM, respectively, as the sole CP source. To use the difference method, 2 diets, diets 2 and 4, were prepared by mixing corn and SBM at the expense of the basal diet, respectively. Diet 2 contained 79% corn and 14% SBM, and diet 4 contained 32.5% corn and 34% SBM. To evaluate the additivity of digestibility values, the difference between measured values for the mixed diets (diets 2, 3, and 4) and predicted values calculated using the measured values for diets 1 and 5 was examined. The apparent (AID) and standardized (SID) ileal digestibility of CP in SBM differed between the direct and indirect methods; however, corresponding digestibility did not differ for corn. Additionally, the predicted and measured digestibility of both AID and SID differed in diets 2 and 3, implying that the digestibility values obtained from diets 1 and 5 were not additive for mixed diets. In conclusion, this study showed that digestibility evaluated by direct and indirect methods depends on the ingredients having different CP concentrations, and this finding may be considered to improve the accuracy of feed formulation for broiler chickens.

호흡기,소화기계 감염환자로부터 전염성 바이러스 분리 및 특성

  • 호흡기계 및 소화기계에 감염된 전염성 바이러스에 대한 역학적 기초자료로 이용하고자 1999년 1월부터 12월까지 부산지역에서 분리된 전염성 바이러스의 특징과 계절적 발생추이, 환자의 성별, 연령별 발생에 대해 조사한 결과는 다음과 같다. 1. 1999년도 바이러스 가검물 2261건에서 분리한 호흡기계감염 바이러스 279건과 소화기계 감염 바이러스 83주를 분리하였으며, 이중 인플루엔자 바이러스 A형이 96주(29.6%), B형이 107주(33.0%)로 대부분을 차지하였다. 2. 1999년의 바이러스 분리의 성별 분포는 총 360명의 환자 중 179명(49.7%)의 남성 및 181명(50.3%)의 여성으로 비슷한 양상을 나타내었다. 이중 호흡기계의 경우 279명의 감염환자 중 남성이 130명(46.6%), 여성이 149명(53.4%)으로 여성의 감염율이 비교적 높았으나, 소화기계의 경우 83명의 감염환자 중 남성이 51명(61.4%), 여성이 32명(38.5%)으로 남성의 감염율이 거의 2배정도 높게 나타났다. 3. 1999년의 연령별 분포는 10세 이하의 어린이가 194명(59.9%)으로 대부분을 차지하였으며, 이중 인플루엔자 바이러스가 99명(30.6%)으로 가장 높은 감염을을 나타내었다 유행성이하선염 바이러스의 감염어린이 중에 $l1{\sim}15$세의 연령층이 15명으로(53.3%)로 가장 높게 나타났다. 4. 1999년 월별 감염율은 호흡기계 감염증 바이러스의 경우 1월부터 4월까지, 그리고 12월에 증가 추세를 보이면서 4월에 가장 높은 감염율을 나타내었다 소화기계 감염증 바이러스의 경우 9, 10, 11월을 제외한 모든 월별에 관찰되었으며, echo와 coxsackie 바이러스는 무균성 수막염 환자에서 하절기에 집중적으로 발생하였다. 동절기에 유행하는 설사 바이러스는 12월에 비교적 높은 양상을 나타내었다. 5. 인플루엔자 바이러스는 MDCK 세포에서, 아데노 바이러스와 유행성 이하선염 바이러스는 HEp-2 세포에서, 파라인플루엔자 바이러스는 Vero 세포에서, 그리고 echo, coxsackie B 바이러스와 장내 바이러스는 HEp-2, Vero, BGM 세포에서 뚜렷한 세포병변 효과를 나타내었다. 6. 분리한 바이러스는 전자현미경으로 관찰한 결과 인플루엔자 바이러스 A 형(HINI, H3N2)은 95nm, B 형은 70nm크기의 구형을 나타내었으며, 바이러스표면의 지질 이중층이 뚜렷하게 관찰되었다. 아데노 바이러스는 외피가 관찰되지 않았으며, nucleocapsid는 symmetry이고 크기는 71nm로서 바이러스 입자 표면에 icosaheral capsomer의 배열이 명확하게 관찰되었고, 파라인플루엔자 바이러스와 유행성 이하선염 바이러스는 외피가 있는 구형의 큰 viron으로180, 170nm 크기이었다. 7. Echo와 coxsackie B group 바이러스는 모두 외피가 없는 isometric 형으로 크기는 $30{\sim}45nm$ 이었고, enteric adeno 바이러스는 84nm 크기로서 외피가 없고, 입자 표면에 capsomer의 배열이 명확하게 관찰되었고, rotavirus는 크기가 70nm이며 외층 capsid 단백질과 내층 capsid 단백질이 두층으로 되어 있는 전형적인 수레바퀴 모양을 나타내었다. 이상의 결과로 보아 호흡기계 및 소화기계에 감염되는 전염성 바이러스는 연중 지속적으로 분리되고 있으며 전염성이 강하여 집단 발생은 일으키는 경우도 많고 최근 들어 유행성 이하선염과 흥역 바이러스의 발생률이 높은 추이를 나타내고 있지만 아직은 특이한 바이러스 치료제가 개발되어 있지 않았으므로 지속적인 대책과 아울러 장기적인 발생 가능에 대한 예방책을 흥보하여야 할 것으로 보이며 계속적인 바이러스성 전염병 유행예측조사 및 역학조사가 적극적으로 이루어져야 할 것으로 사료된다.

  • The effects of divalant metal ions on the ATPase activity were studied by using my of ibrillar protein of rabbit (Chin-Chilla species) fed with vegetable oils. The results obtained were as follows : 1. The ATPase activity in myofibrillar protein of the is not significant to Rabbit exhibited a common biohapsic respnse, such as the ATPase activity is high at a lower ionic strength and low at a higher ionic strength. 2. The effect of EDTA on the ATPase activity of Myofibrillar protein extracted from Rabbit fed vegetable oils was tested by using various concentrations. The ATPase activity was inhibited from 0.2mM and over concentration of EDTA. 3. The ATPase activity in Myofibrillar protein was decreased remarkably in 0.2mM and over concentration for $Mg^{2+}$, and in 1.0mM and over concentration for $Ca^{2+}$. 4. in vitro, the digestibilities in A, B, C and D groups of Rabbit muscle treated with Papsin and Trypsin for 30 minutes at 36$^{\circ}C$ water bath were 71.66%, 73.87% ; 70.62%, 77.93% ; 67.93%, 76.52% ; and 86.79%, 90.22%, respectively.y.

Extraction of protein from defatted sesame meal using the enzyme from Bacillus sp. CW-1121 (Bacillus sp. CW-1121이 생성하는 단백 분해 효소를 이용한 참깨박 단백질의 용출)

  • To extract insoluble proteins of sesame meal residue by using microorganism, the sesame meal residue was treated with crude enzyme solution from Bacillus sp. CW-1121. It was found that the solubility reached to maximum at pH 7.5, $45^{\circ}C$. Under optimum condition, the nitrogen solubility with the enzyme solution from Bacillus sp. CW-1121 reached to 60% in 2 hours. Nitrogen solubility of protein from sesame meal showed minimum value at pH 4.5 and significantly increased above pH 6.0. When the protein from sesame meal extracted with Bacillus sp. CW-1121 was subjected to sodium dodecyl sulfate polyacrylamide gel electrophoresis, water soluble protein was showed 4 bands and salt soluble protein was showed 2 bands. The amino acid composition of water soluble protein, salt soluble protein and free amino acid indicated relatively high contents of serine (17.24 mg/g), glutamic acid (10.77 mg/g) and glutamic acid (6.55 mg/g). Specially, the contents of essential amino acids were high.

  • The characteristics of parasporal inclusion body from Bacillus thuringiensis subsp. kurstaki KB099 isolate which is high bioactive to the tobacco cutworm, Spodoptera litura, were examined. Parasporal inclusion of B. thuringiensis subsp. kurstaki KB099 isolate showed only 1 band at 130 kDa compared with B. thuringiensis subsp. kurstaki HD-l isolate producing 2 protein bands at 130 kDa and 60 kDa from by SDS-PAGE analysis without any enzyme treatment. Also, we confirmed that gut extract of sensitive S. litura KB099 isolate had digested only 60 kDa ${\delta}$-endotoxin protein. When the digestive enzyme of sensitive insect responsible for parasporal inclusion from KB099 and HD-l isolate was treated to each of them, protein band 60 KDa of KB099 was maintained up to 12 hours but all bands of HD-l were disappeared within 6 hours. In KB099 isolate, 6 genes (Cry1Aa, Cry1Ab, Cry1Ac, Cry1C, Cry1D and Cry1I) were identified by PCR analysis. Also, $Cry^-$ mutant of KB099 isolate was investigated by phase- contrast microscope, SDS-PAGE and PCR.

Characteristics of Tenebrio molitor larvae and Bombyx mori pupae sequentially fermented with Aspergillus oryzae and Bacillus subtilis (Aspergillus oryzae와 Bacillus subtilis를 순차접종 발효한 갈색거저리유충과 누에번데기 발효물의 품질특성)

  • Defatted soybean, larvae of brown mealworm (Tenebrio molitor), and powdered pupae of silkworm (Bombyx mori) were fermented in solid and liquid forms using Aspergillus oryzae and Bacillus subtilis. The protein degradation rate (NDR) through solid fermentation was the highest in the fermented soybean control sample (54.69±6.54%), followed by silkworm pupae (34.82±5.99%) and brown mealworm larvae (30.54±3.80%). When these edible insects were fermented in liquid form, solid extraction yield was 37.73-46.88%, and protein yield was 47.47-63.02%. NDR of fermented liquid form products increased to 58.90, 52.62, and 50.13% for soybean, brown mealworm larvae, and silkworm pupae, respectively. SDS-PAGE of the liquid fermented products confirmed that microbial fermentation decomposed higher-molecular-weight proteins into small polypeptides. In vitro digestibility of liquid forms of edible insects increased by 1.26 to 1.53 times after fermentation. The protein solubility, foaming ability, and foam stability of liquid-fermented edible insects all tended to increase through fermentation.
