Effect of Pepsin-HC$\ell$ Concentration and Digestion Time on the Protein Digestibility of the Cattle Skin Meal (Pepsin농도와 소화시간이 우피분의 단백질 소화율에 미치는 영향)

  • Protein digestibitlities of hydrolyzed cattle skin meals were examined at a constant pepsin-MC$\ell$ concentration (0.2%) for varying lengths of incubation time (from 4 to 20 hours) and at varying concentrations of pepsin-HC$\ell$ (from 0.0125 to 0.2%) for 16 hours at 45$^{\circ}C$. The results are summarized as follows: 1. Protein digestibilities of hydrolyzed cattle skin meals in 0.2% pepsin-HCl were 66.31%, 80.69%, 83.72%, 84.65% and 81.45% for 4, 8, 12, 16 and 20 hours incubation, respectively. Protein digestibilities were maintained above 80% for 8-hour incubation and were increasing incubation time. 2. Protein digestibilities of hydrolyzed cattle skin meal incubated for 16 hours at 0.2%, 0.1%, 0.05%, 0.025% and 0.0125% pepsin-HC$\ell$ solution were 85.10%, 82.08%, 76.18%, 74.67% and 64.82%, respectively. Protein digestibilities were decreased with decreasing pepsin concentration.

  • This study has been carried out to investigate the effect of protein components of a diet on the increasing of larval body weight and diet efficiency, as well as the amylase activity of larval digestive juices during the 5th instar. Defending on the amounts of soybean meal as a protein source, the six different kind of artificial diets containing mulberry leaf powder fed to the larvae. The results obtained may be summarized as follows: 1. As the protein components of the diet were increased, the amount of increased larval body weight was also increased. 2. As the protein components of the diet were increased, both the cocoon weight and cocoon layer weight were also increased. 3. As the protein components of the diet were increased, both the amount of diet digested and coefficient of digestibility were also increased. 4. As the protein components of the diet were increased, the diet efficiency of larvae were also increased. 5. But the experimenters could not observe any correlation between the increase of protein components of a diet and the amylase activity of the digestive juices.

  • The haemolymph protein, digestive fluid proteins and digestive fluid amylase activity of wild silkworm, Theophila mandarina those of the were studied by polyacrylamide gel electrophoresis. In addition, they was also compared with silkworm. 1. 6 main protein bands in female and 7 main protein bands in male were detected in the larval haemolymph of T. mandarina where as 8 and 7 main protein bands in female and male of B. mori were observed. Some differences in the haemolymph protein ands of T. mandarina and B. mori were observed. 2. 15 protein bands and 12 protein bands were found in the larval digestive fluid of T. mandarina and B. mori respectively. Some differences in the mobility of digestive fluid proteins of T. mandarina and B. mori were noticed. 3. Larval digestive fluid amylases were anionic and moved near the tracking dye in both T. mandarina and B. mori. Mobility of the digestive fluid amylases relative to bromophenol blue were 0.019 and 0.020 in T. mandarina and B. mori respectively.

Analysis of Evaluation Methods for the Efficacy of Protein Removal Agents for Soft Contact Lens (소프트콘택트렌즈 단백질제거제의 효능 평가법 분석)

  • Purpose: The present study was conducted to establish the experimental condition for the proper evaluation of protein removal efficacy when developing protein removal agents. Its protein removal efficacy was further analyzed and compared with the result from protein removal efficacy against protein deposition on contact lens to suggest the evaluation method for efficacy of protein removal agents. Methods: Protein digestibility assay presented in the Korean pharmacopoeia was selected to establish the evaluation method for efficacy of papain, pancreatin, subtilisin A and protease itself as a ingredient and protein removal tablets or solution containing those enzymes and find a suitable test conditions. Furthermore, the cleaning efficacy of commercially available protein removal tablets and solution on balafilcon A lens deposited with protein artificially was measured and the correlation between two evaluation methods was further analyzed. Results: When pancreatin itself and the product containing pancreatin was evaluated by protein digestibility assay, both reached 28 IU/mg, the standard value of protein digestibility suggested by the Korean pharmacopoeia. In case of protease and subtilisin A tested with trichloroacetic acid B solution, both of them met the enzyme activity level proposed by the manufacturers when they were evaluated by protein digestibility assay however, papain and subtilisin A tested with trichloroacetic acid A solution were not reached the enzyme activity level. Among protein removal agents, three products except a product containing pancreatin did not meet the enzyme activity value specified by the manufacturer when they were evaluated by protein digestibility assay. However, actual protein removal efficacy of three products except a papain-containing product on the lens was greater than 90% protein removal. In the case of papain-containing protein removal product, its effect was not measured by protein digestibility assay however, its actual protein removal efficacy on the lens reached 73.72%. Conclusions: From the results, it was confirmed that the efficacy of protein removal agents for contact lens should be evaluated by different method according to the type of proteolytic enzyme contained. That is, the protein removal agents containing pancreatin, protease and subtilisin A can be evaluated by protein digestibility assay and protein removal efficiency evaluation and the products containing papain can be effectively evaluated by only the evaluation method for protein removal efficiency employing the lens.

Evaluation of the Nutrient Digestibilities by Different Fecal Collection Methods in Juvenile and Adult Korean Rockfish (Sebastes schlegeli) (조피볼락 치어 및 성어에서 분 수집 방법에 따른 영양소 소화율)

  • The apparent nutrient digestibilities were examined by using chromic oxide indicator according to the various fecal collection methods in juvenile and adult Korean rockfish (Sebastes schlegeli). Feces were collected from three replicate groups of fish by dissection, stripping or decantation using fecal collector attached to fish rearing tank, respectively. The digestibilities of dry matter, protein, lipid, and energy were affected by fecal collection methods (P<0.01), but not affected by fish size. The digestibilities of nutrient determined by stripping or decantation methods were significantly higher than those determined by dissection method (P<0.01). No significant differences in the digestibilities of protein, lipid and energy were found between the stripping and decantation methods in adult fish (P>0.01). The digestibilities of dry matter, protein, lipid, energy, nitrogen-free extract, and total amino acids in juvenile and adult fish were 58, 93, 94, 79, 32, and $93\%$, and 61, 94, 96, 80, 29, and $94\%$, respectively, when they were measured by decantation method. Methionine, cystine and valine digestibilities were significantly lower than those of other amino acids in both juvenile and adult fish (P<0.01). Results indicate that stripping or decantation with fecal collector could be a reliable digestibility procedure for measuring the nutrient digestibilities in Korean rockfish.

Identification and Characterization of Protease-Resistant Proteins from Adzuki Beans (소화 효소 저항성을 지니는 팥 단백질의 성질 규명)

  • It is already known that adzuki beans (Vigna angularis) are able to control appetite. Therefore, this study tested the proteins isolated from adzuki beans for their protease resistance and interaction with the intestinal mucosa. The major proteins from adzuki beans were found to be resistant to the digestive enzymes pepsin and pancreatin, and were identified using 2D-SDS-polyacrylamide gel electrophoresis and mass spectrometry. The major adzuki proteins were easily fractionated by treating the soluble protein extract with 10mM $CaCl_2$, and were found to contain lactotransferrin, a homologous protein to the dynein light chain domain, proteinase inhibitor, and proteins with unknown functions. From a tissue binding assay using mouse intestinal tissue sections, the major protein fraction showed weak, yet significant and specific binding to the mucosa layer of the small intestine. Thus, the current results suggest that adzuki proteins are resistant to digestive enzymes, which enables them to survive protease digestion in the intestinal tract, plus they may interact with the intestinal mucosa layer. Therefore, the molecules responsible for controlling appetite in adzuki beans are presumably protease-resistant proteins that interact with the intestinal mucosa or delay digestion in the digestive tract.

Effects of Dietary Supplementation of Scoria on Growth and Protein Digestibility in Juvenile Parrot Fish Oplegnathus fasciatus and Olive Flounder Paralichthys olivaceus (사료 내 송이의 첨가가 돌돔 Oplegnathus Fasciatus과 넙치 치어 Paralichthys olivaceus의 성장 및 단백질 소화율에 미치는 영향)

  • We evaluated the effects of dietary supplementation of scoria on growth performances and apparent protein digestibility in juvenile parrot fish Oplegnathus fasciatus and olive flounder Paralichthys olivaceus. In experiment, parrot fish at initial average weight of $7.6{\pm}0.01g(mean{\pm}SD)$ were divided into 9 groups(three groups per dietary treatment) and fed one of three isonitrogenous diets which contained 0%, 1% and 2% of scoria, respectively. Experiment was conducted with juvenile olive flounder to demonstrate the improved apparent digestibility of protein found in the experiment. Five experimental diets were formulated to contain different levels of scoria: Diet 1(control), 0% scoria; diet 2, 1% scoria; diet 3, 2% scoria; diet 4, 3% scoria; and diet 5, 4% scoria. After 9 weeks of feeding trial(Exp), no significant differences were observed on growth performances between Scoria 2% and the control group. However, the growth was significantly lower in scoria 1% compared to that of the control group. Hematological parameters were not affected by the supplementation of scoria. Interestingly, apparent protein digestibility was improved by dietary supplementation of scoria both in parrot fish and olive flounder. The findings suggest that dietary supplementation of 2-3% scoria is optimum levels in order to improve the protein digestibility in the diets for juvenile parrot fish and olive flounder.

꽃사슴의 단백질 요구량에 관한 연구

  • 본 시험은 3마리의 2년생 famale sika deer에게 동일한 에너지 수준하에 단백질 함량($7\%,\;12\%\;17\%$)이 각기 다른 사료를 급여하였을 때, 사료 영양소의 이용성 변화 및 체내 N 대사, 유지를 위한 단백질 요구량, 대사성 분질소 함량 등을 규명하기 위하여 실시하였으며, 결과를 요약하면 다음과 같다. 1. DM, OM, ADF의 소화율을 사료내 단백질 함량에 따른 차이를 보이지 않았으며(P>.1), CP의 소화율은 사료내 단백질 수준이 증가함에 따라 향상되었다(P<.05). 2. 사료내 단백질 함량이 증가함에 따라 분과 뇨로의 N 배출량 모두 증가하였으며(P<.05), 사료내 단백질 수준이 7, 12, $17\%$ 이었을 때 N retention은 각각 26.9, 31, 8, $38.9\%$였다. 3. 혈증 urea-N 함량은 사료내 단백질 함량의 증가에 따라 현저하게 증가하였다.(P<.05) 4. Famale sika deer의 유지수준의 단백질요구량(protein requirement for maintenance)은 $1.81g/kgW^{0.75}/d$다. 5. 대사성 분 질소(metabolic facal N)함량은 $0.32g/kgW^{0.75}/d$였다.

Effect of Garlic on the Digestion of Beef Protein during Storage (쇠고기에 첨가한 마늘의 소화효과)

  • Chopped garlic was added to beef to determine its effect on the protein digestion during storage and heat treatment. The digestibility of raw beef without garlic was not significantly changed during storage at $4^{\circ}C$, but increased as garlic added and aging time increased. The optimal aging time and amount of garlic added was varied with heating time. Trypsin inhibitor did not change the digestibility of beef due to its thermal inactivation. Gel chromatography revealed that the lower molecular weight peptides(2,200~6,150 dalton) were shown in beef-garlic mixture through aging and heating procedure. When aged beef with garlic was digested with four-enzyme system, the soluble portion was increased significantly in comparison with that from raw beef without garlic. Protein quality of beef, as measured by computed PER(C-PER), was improved from 2.14 of raw beef to 2.50 of aged beef with chopped garlic.

Growth Inhibition of Rats Fed Raw or Heated Korean Beans and the Effect of Methionine or Protein Supplementation (한국산 생두류 및 익힌두류를 섭취한 흰쥐의 성장저해와 Methionine 및 단백질 첨가의 영향)

  • A study was made on the effect of anti-nutritional factors found in some Korean beans : soybean, red bead, mung bean and kidney bean. Two animal experiments were conducted to investigate the nutritional value of the beans. The first experiment, in which the diet contained 15% protein from raw beans, compared the intensity of inhibition caused by methionine deficiency or a total amino acid deficiency. In the second experiment, the conditions were the same as in experiment I, except that heated beans were substituted for raw beans. Severe growth inhibition and high mortality was found in the raw kidney bean and red bean groups than in the soybean and mung bean groups. As no effect on the growth inhibition of raw bean groups was shown by methionine and protein supplementation, the inhibition could be ascribed mainly to the low feed intake and the low protein digestibility caused by toxic factors. Pancreatic enlargement was obserbed in all the raw bean groups. A increase in body weight, a decrease in mortality and a decrease in the weight pancreases were found in the heated bean groups. But the digestility of the diet and of the protein and the PER by heating did not increase as markedly as weight, except in the heated red bean groups. Even with heat treatment, the whole inhibitory action could not be eliminated.
