On the Effective Thermal Diffusivity of Water-Protein-Fat Food System ($\cdot$단백질$\cdot$지질계(불균질계) 식품의 유효열확산율)

  • One dimensional unsteady heat conduction was studied on soybean curd as a water-protein-fat food system. This heterogeneous soybean curd could be treated as homogeneous material as for the unsteady heat conduction by 'effectiver' thermal diffusivity. Measurements were made with a apparatus designed and constructed by the author and also made with ice to establish the reliability, the reproducibility and the accuracy of the apparatus. It was found that the effective thermal diffusivity of soybean curd was substantially dependent on the fraction of protein and fat as well as water.

However, developments in the field of molecular biology have led to an increase in the knowledge of biological systems and their interactions. Proteins are biomolecules that are essential in determining the structure and carrying out most of the functions in living cells. The future of pharmacy belongs to a special category of protein therapeutics. Based on the result of industry analysis, it is suggested how to commercialize protein therapeutics for biotech-enterprises.

  • 해양생명체인 홍합(mussel)은 접착단백질(adhesive protein)을 생산, 분비함으로써 홍합 자신을 바다 속의 바위와 같은 젖은 고체표면에 단단히 부착할 수 있도록 하여 강한 파도의 충격이나 바닷속의 부력 효과에 대하여도 저항할 수 있다. 홍합접착단백질은 현재 알려진 어떠한 화학합성 접착제보다도 강력한 자연적인 접착제로 알려져 있으며 대부분의 에폭시 수지보다 두 배 정도의 커다란 인장강도를 지니고 있으면서도 휘어질 수 있는 유연성을 가지고 있는 혁신적인 물질이므로 접착제 산업계에서 매우 커다란 관심을 끌고 있다.   • Vitellogenesis, an important reproductive process in oviparous animals, includes the processes of hormonally regulated synthesis of yolk precursor protein, vitellogenin (Vg), and their deposition in ovarian oocytes as a vitellin which is an important energy source as well as buoyancy regulator of the egg. Vg genes consist of a gene family that encompasses a large number of lipoproteins and produce different Mr. Vg proteins in liver. The expression of Vg is largely dependent on the estrogen, and both reproductive cycle and temperature also influence Vg synthesis. Synthetic estrogens or estrogenic pollutants was sufficient to induce Vg in both sexes of oviparous vertebrates. Therefore, the estrogenic induction of vitellogenesis in male has been used for biological marker in the screening of estrogenicity of certain endocrine disrupting compounds and the monitoring the world-wide contamination of estrogenic compounds in wild life. In the studies on the biological hazard and influence of endocrine disrupting chemicals using the Vg induction in oviparous males, it is important to consider the reproductive cycle, zoogeography and biodiversity of the wild life animals in Korea.

  • 환경오염을 신속하게 모니터링하기 위한 생물지표의 개발은 증가하고 있는 오염의 심각성에 비추어 매우 중요한 과제로 여겨지고 있다. 본 연구에서는 독성물질처리에 의하여 선택적으로 발현이 조절되는 단백질의 동정을 통하여 독성물질에 대한 단백질 생물지표를 발굴하고자 시도하였다. 즉, 송사리(Oryzias latipes)를 유기인계 살충제인 다이아지논(diazinon)에 0, 0.1, 1, 5 mg/L 농도로 24시간 노출시킨 후, 머리와 몸통부분으로 나누어 단백질 발현패턴을 분석하였다. 본 시스템에서 다이아지논 처리에 의하여 유의적으로 발현이 증가된 단백질로서 alpha-tubulin, ribonuclease pancreatic precursor, protein hfq 등을 동정하였으며, 이 가운데 alpha-tubulin과 $hsp90{\beta}$의 발현이 다이아지논 농도에 의존적으로 증가하는 것을 semi-quantitative RT-PCR방법으로 확인하였다. 이와 같이 다이아지논 처리에 특이적으로 발현이 증가된 송사리 단백질들은 노출평가를 위한 생물지표로서 개발에 응용될 수 있을 것으로 평가된다.

  • As a mussel-inspired surface independent modification chemistry using catecholamine family molecule was suggested on 2007, there are tremendous efforts being done by researchers from around the world to adjust and develop diverse applications using catecholamine family. Accordingly, we will discuss about the novel method to extend catecholamine applications, which is through the functional group substitution of catecholamine molecules.

Characterizing Fluorescence Properties of Dissolved Organic Matter for Water Quality Management of Rivers and Lakes (하천 및 호소 수질관리를 위한 용존 자연유기물질 형광특성 분석)

  • Fluorescence measurements of dissolved organic matter(DOM) have the superior advantages over other analysis tools for applying to water quality management. They are simple and fast and require minimal pretreatment of samples. Fluorescence index($F_{450}/F_{500}$), synchronous spectra, and fluorescence excitation-emission matrices(EEM) of various DOM samples were investigated to discriminate autochthonous/allochthonous composition, protein-like fluorescence, fulvic-like fluorescence, humic-like fluorescence, terestrial humic-like fluorescence by comparing among the real DOM samples of different origins with the help of literature. The samples used included standard purified DOM, lake, river and wastewater treatment effluent. The relative distribution of various DOM composition was derived from the ratios of each fluorescence region. The results were very consistent with those expected from the sample properties. Allochthonous and terrestrial humic-like fluorescence were more prominent in the samples with abundant soil-derived DOM components. In addition, the protein-like fluorescence property was more pronounced in the samples where strong algal or microbial activities were expected. It was also shown that the ratio of protein-like/terrestrial humic-like fluorescence obtained from synchronous spectrum and fluorescence EEM could be used as an indicator for the evaluation of wastewater treatment on the downstream water quality of rivers and for the prediction of the degree of algal/microbial activities in lakes. It is expected that the results of this study will provide the basic information to develop the future water quality management techniques using DOM fluorescence measurements.

  • It is well known that the native conformation of many proteins can be stabilized by carbohydrates or polyalcohols. However, the mechanism of the stabilization still remains unclear. In the present studies, to characterize the cryoprotective mechanism of corn starch enzyme hydrolysates on fish protin, solubility of hydrolysates, thermal behavior of hydrolysates and actomyosin solution, and enzyme kinetics in frozen system were investigated. The solubility of the hydrolysates increased with the increase in D.E. value. The $T_g^{'}$ of the hydrolysates were linearly correlated with D.E. value and the T-g value of the hydrolysates (D.E. 5,10,15,20) were reported to be $-7.2^{\circ}C\;-8.8^{\circ}C\;-11.9^{\circ}C$, and $-14.3^{\circ}C$, respectively. The results of enzyme experiments showed that the higher the D.E. value, the higher was the rate of reaction in frozen storage ($-12^{\circ}C$). It is found to support the cryostabilization mechanism that the hydrolysats act to enmesh the protein in a glass state where all deteriorative processes are greatly slowed down.

Production of Immunospecific Egg Yolk Antibody with Recombinant Staphylococcal Enterotoxin B (SEB) Protein (포도상구균에서 분비하는 장내독소 B(SEB)에 대한 재조합 단백질을 이용한 면역특이적 난황항체 생산)

  • Staphylococcal enterotoxin B (SEB), which is a bacterial superantigen produced by Staphylococcus aureus, is associated with serious diseases, including food poisoning and atopic dermatitis. This study was performed to produce about 30 kDa of recombinant SEB protein and to immunize in chickens to acquire the specific egg yolk antibody (IgY) against the recombinant SEB. Chickens were immunized with the recombinant SEB intramuscularly in the breast muscle by injection 3 times at intervals of two weeks. Serum- and egg yolk-antibody titers of hens against SEB were highest at 4 weeks after first immunization. In western blot, anti-recombinant SEB IgY was reacted immunospecifically against the recombinant SEB and commercialized SEB. These results suggested that the recombinant SEB antigen could be used as an immunogen to elicit antibody (IgY) against SEB and the anti-recombinant SEB IgY could neutralized staphylococcal enterotoxin B effectively.
