Effects of Pine Needle Ethyl Acetate Fraction on Acctylcholine(ACh) and Its Related Enzymes in Brain of Rats (뇌 조직의 아세틸콜린 및 그 관련효소에 미치는 솔잎(Pine Needle) 에틸아세테이트획분의 영향)

  • This study was designed to investigate the effects of ethyl acetate(EtOAc) fraction of pine (Pinus densiflora Sieb et Zucc) needle on cholesterol and lipofuscin(LF) accumulations, acetylcholine(ACh) and its related enzyme activities such as choline acetyltransferase(ChAT) and acetylcholinesterase(AChE), and monoamine oxidase-B(MAO-B) activity, which destroyed the catecholamine related neurotransmitters in brain membranes of Sprague- Dawley (SD) rats. Male SD rats were fed basic diets (control group) and experimental diets (EtOAc-25, EtOAc-50 and EtOAc-100) for 45 days. Cholesterol accumulations in mitocholndria and microsomes were significantly inhibited (11.8-12.1% and 9.6-13.0%, respectvely) in EtOAc-50 and EtOAc-100 groups. ACh levels and ChAT activities were significantly increased about 10% in membranes of EtOAc-100 group compared with control group. AChE activities were significantly increased about 8 -12% in membranes of EtOAc-50 and EtOAc-100 groups compared with control group. MAO-B activities were significantly inhibited about 10% in membrane of EtOAc-l00 group compared with control group. These results suggest that ethyl acetate fraction of pine needle may play an effective role in inhibiting cholesterol and improving a membrane fluidity, and learning and memory impairments. Activation Differences of Superior Parietal Lobule and Cerebellum Areas While Inferring Geometrical Figures per Intellectual Category in Adolescents (도형 과제 수행 때 나타나는 청소년의 지능별 대뇌 및 소뇌의 활성도 차이 분석)

  • The relationship between the cerebral cortex and human intelligence has been studied using various methods, and related brain areas involved in intellectual manifestation have been discovered individually. Such studies have also shown the cerebellum is closely involved in various cognitive functions such as language, memory, and information processing. However, studies showing an activity difference between the cerebral cortex and cerebellum when performing specific tasks are hard to find. This study searched and analyzed the active regions of the cerebral cortex and cerebellum seen while performing the inference of geometrical figures. A WAIS intelligence test was conducted using 81 healthy boys (16.3 years of age on average), and five categories were classified. While performing the inference of shapes, their brain images were taken using functional magnetic resonance imaging (fMRI). As a result, the activity in 12 brain regions was observed, including in the cerebral cortex, the bilateral inferior parietal, the visual cortex, bilateral superior parietal, frontal-Inf-Tri-R, and bilateral caudate, while activities in 5 discrete areas were seen in the cerebellum. In particular, the higher the intelligence (IQ) of the subject, the stronger their activity. Among those with the most superior intelligence, subjects with an IQ of 140-147 showed significantly higher activity compared to the other groups. Such results seem to represent a very high utilization of intelligence in a highly gifted group, and we can expect to use this to determine the super gifted.

Study on Biochemical Pollutant Markers for Diagnosis of Marine Pollution II. Changes in Oxygen Radicals and Their Scavenger Enzymes of the Flounder(Paralichthys olivaceus) in the Yellow Sea (해양오염의 진단을 위한 생화학적 오염지표에 관한 연구 II. 황해산 넙치(Paralichthys olivaceus)의 산소라디칼 및 제거효소의 변화)

  • This study was designed as a part of efforts to investigate the biochemical pollutant markers for diagnosis of marine pollutions by changes in oxygen radicals and their scavenger enzymes of the flounder (Paralichthys olivaceus)in Yellow Sea of Kores. Protein contents in brian and muscle of cultured flounder in Yellow Sea were remarkably lower(30-45% and 25-45%, respectively) than those of wild flounders in Pohang(control) of East Sea. Lipid peroxide(LPO) levels in serum of cultured and wild flounders in Yellow Sea were significanltly higher (30-80% and 125-145%, respectively)than those of wild flounder in Pohang. Hydroxide radical formations and superoxide dismutase(SOD) activities in serum of cultured flounders in Yellow Sea were significantly 15-30% and 15-35% lower than those of wild flounders in Pohang, but glutathione peroxidase (GSHPx) activities in brain of cultured flounders in Yellow Sea were significantly 15-25% higher than those of wild flounders in Pohang. It is believed that significantly decreases of protein contents in brain anad muscle, remakable increases of malondialdehyde(LPO) in serum and glutathione peroxidase (GSHPx)in brain of cultured flounders of Yellow Sea may be used as a biochemical pollutant markers for diagnosis of marine pollutions.

Histological study of neurosecretory cells in the brain and the thoracic ganglion with gonadal development of Palaemon serrifer (줄새우아재비, Palaemon serrifer의 생식소발달에 따른 뇌와 흉부신경절 신경분비세포의 조직학적 연구)

  • The present paper is a histological study of neurosecretory cells in the brain and the thoracic ganglion with the gonadal development in Palaemon serrifer. The reproductive cycle includes the successive stages of the growing period (February-March), the mature period(April-May), the ripe and spent periods(June-August) and the degenerative and resting periods(September-January). The neurosecretory cells are grouped into four types based on Matsumoto(1958) : A-,A'-, B- and E-cells. A- and A'-cells are $80-90{\mu}m,\;B-cell\;is\;30-40{\mu}m$ and E-cell is $10-15{\mu}m$. A- and B-cells are the positive to CHP and AF, while B-cell is the positive only to AF. The secretory grannules of a A-cell are transported to the axon, and at the same time they are discharged through the peripheral membrane. Of the four neurosecretory cells, A- and I-cells show the difference of secretory activity according to the gonad developmental process. In the female, A-cells show secretory activity for the ripe and spent periods, while I-cells show for the mature, ripe and spent periods. In the male, A-cells show secretory activity for the mature, ripe and spent periods, while I-cells show for the growing, mature, ripe and spent periods.

Effects of Dandelion on Oxygen Free Radical Generating and Scavenging System of Brain in Streptozotocin-Induced Diabetic Rats (서양민들레가 Streptozotocin으로 유발한 당뇨 횐쥐의 뇌조직 중 유해 활성산소 생성 및 제거 효소계에 미치는 영향)

  • Many studies have shown that hyperglycemia leads to an increase of lipid peroxidation in diabetic patients and animals, reflecting a rise reactive oxygen species production. It is increasingly recognized that brain is another site of diabetic organ damage. Accordingly, this study was to investigate the effect of dandelion on oxygen free radical generating and scavenging system of brain in streptozotocin (STZ)-induced diabetic rats. Male Wistar rats were divided into diabetic (control) and diabetic-dandelion supplemented groups. Dandelion was supplemented for 4 weeks with dandelion leaf and root powder (DLP, DRP) or dandelion leaf and root water extract (DLW, DRW) based on 11.4 g of raw dandelion/kg diet. Diabetes was induced by single injection STZ (55 mg/kg B.W., i.p.)in a citrate buffer. Oxygen free radical generating enzymes, cytochrome P-450, amino-pyrine N-demethylase, aniline hydroxylase and xanthine oxidase, were lowered in dandelion supplemented-groups compared to the control group. Superoxide dismutase, catalase and gluthathione peroxidase activities of brain were also lower in dandelion leaf and root supplemented-group than in the control group, whereas glutathione S-transferase activity and gluthathione content were increased in dandelion supplemented-groups compared to the control group. With regard to the lipid peroxidation products, the malondialdehyde content of brain was lower in dandelion supplemented groups. Therefore, it could be suggested that powder and water extract of dandelion leaf or root are beneficial in preventing diabetic complication from lipid peroxidation and free radical in brain of diabetic rat brain.

The Effect of Puerariae thubergiana Bentham Extract on Brain Tissue in Alcohol-Treated Rats (칡추출물이 알코올을 급여한 흰쥐의 뇌조직에 미치는 영향)

  • This study investigated the effect of Puerariae Flos (PF; flower of Puerariae plant) and Puerariae Radix (PR; root of Puerariae plant) water extracts on the activities on the activities of ethanol-metabolizing enzymes and free radical generating/scavenging enzymes of brain in ethanol-treated rats. Five groups of male Sprague-Dawley rats were orally administered ethanol (25%, v/v) 5 g/kg body weight/day, and sacrificed 5 weeks post treatment. PF and PR water extracts were supplemented in a diet based on 1.2g (I) or 2.4 g (II) raw PF or PR/kg body weight/day. Alcohol dehydrogenase activity of brain was significantly lowered in PF of PR groups, whereas aldehyde dehydrogenase activity was significantly higher in PR groups than those of control and PF groups. Cytochrome P-450 content, aminopyrine D-methylase and aniline hydroxylase activities were decreased in both PF and PR groups compared to control group. Aldehyde oxidase and xanthine oxidase activities tended to decrease by Puerariae plant extract supplemented goups and degree of decrease predominated in PRI. Superoxide dismutase and glutathione S-transferase activities were increased in PF or PR groups, whereas glutathione peroxidase and catalase activities were significantly decrased by Puerariae plant extracts supplement. These results indicated that supplementation of PF or PR lowers free radical generating enzymes activities. It was suggested that the activities of ethanol metabolizing emzymes and antioxidant enzymes in brain can be enhanced by PF or PR supplement in ethanol-treated rats.

  • Weanling female Sprague Dawley rate were fed 1.2mg pyridoxine' HCl/kg diet Cdepleted diet) and 2 22mg pyridoxine' HCI/kg dietCcontrol diet). The control and one depleted group were fed their diets throughout growth, gestation, and lactation. Other three depleted groups were fed the depleted diet throught growth and gestation, and then pyridoxine w was supplemented by feeding control diet at 5, 10, and 21 days postpartum. The brains were analyzed for proteolipid protein, cholesterol, and cerebrosi­d de. Percentage stimulation of erythrocyte alanine aminotransferase activity was also determined. Body and brain weight were significantly lower at all ages in depleted group than the control and depleted group showed inadequacy of B6 at all ages. PProteolipid protein and cholesterol were significa­n ntlylower in the depleted group at 10, 21, 35 and 5 50 days. The postnatal development of cerebroside in brain was delayed in depleted groups suppleme­I nted at 5, 10, and 21 days. When supplementation was initiateo at 5 days postparturn, contents of cho­lesterol proteolipid protein were reversed. But some differences in brain development of pups we­re evident when supplementation of dams was de­layed to 10 days or 21 days.
