Effects of Bombusae concretio Salicea on $Amyloid-{\beta}$-induced Neuronal Cell Toxicity and Lipid Peroxidation in Cultured Rat Astrocytes (흰쥐 astrocyte에 있어서 $amyloid-{\beta}$에 의한 독성과 지질과산화에 미치는 천축황(天竺黃)의 영향)

  • The present study was done to investigate the effects of Bombusae concretio Salicea (BCS) on cultured astrocyte cell system and lipid peroxidation in $A{\beta}25-35$ treatment conditions. Cell killing was significantly enhanced by addition of increasing concentrations of $A{\beta}25-35$. Pretreatment of BCS attenuated in cell killing enhanced by increasing concentrations of $A{\beta}25-35$. MDA level induced by $A{\beta}25-35$ treatment was significantly increased and the level was slightly reduced by pretreatment of BCS. The present study showed that $A{\beta}25-35$ strongly increased MDA level and the level was enhanced by addition of increasing concentrations of In conclusion, it was shown that $A{\beta}25-35$ is not only potent lipid peroxide inducer, but also cause protection of neurodegeneration induced by $A{\beta}25-35$. It can be concluded that the activation of antioxidative enzymes may be related to the inhibition of lipid peroxidative reactions. We cannot fully explain to effects of BCS at present; however, the ability of BCS to reduce cell killing and MDA level induced by $A{\beta}25-35$ suggest that BCS may be a protective agent for free radical generating compounds such as $A{\beta}25-35$.

Antioxidant Properties and Protective Effects of Inula britannica var. chinensis Regel on Oxidative Stress-induced Neuronal Cell Damage (금불초 추출물의 항산화 효과 및 산화 스트레스에 대한 신경세포 보호작용)

  • The antioxidant properties and protective effects of Inula britannica on ${H_2}{O_2}$-induced SH-SY5Y neuroblastoma cell damage were investigated. A series of solvent fractions, including hexane(Fr.H), petroleum ether, chloroform, ethyl acetate(Fr.EA), and water fraction(Fr.W), were prepared from the 70% methanol extracts of Inula britannica. Fr.W had the highest total contents of phenolics and flavonoids, followed by Fr.EA. The antioxidant properties of the fractions were also evaluated by analyzing their scavenging activities on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radicals, 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals, and nitric oxide. Fr.W showed the strongest activities in all assays. The concentrations of Fr.W that resulted in 50% reductions of the DPPH and ABTS radicals were 20.7 ${\mu}g$/mL and 39.4 ${\mu}g$/mL, respectively. Fr.W showed the weakest cytotoxic activities on the SH-SY5Y cells, whereas it effectively protected ${H_2}{O_2}$-induced cell death, increasing cell survival by 35.0-77.0% at a concentration range of 62.5-250 ${\mu}g$/mL. In this range, Fr.W also significantly decreased intracellular ROS levels by 34-39%. Overall, the antioxidant properties of Inula britannica can contribute to rescuring neuronal cells from oxidative stress-induced cell injury.

Antioxidant and Neuronal Cell Protective Effects of an Extract of Houttuynia cordata Thunb (a Culinary Herb) (어성초 추출물의 항산화 및 신경세포 보호효과)

  • The in vitro antioxidant activities and neuronal cell protective effects of 60% (w/v) methanolic extract from Houttuynia cordata were investigated. The contents of total phenolics and quercitrin in the extract were 17.71 mg/g and 75.80 ${\mu}g$/g, respectively. DPPH and ABTS radical-scavenging activities were 87.79% and 99.27%, respectively, when the extract was tested at 5 mg/ml. The FRAP (ferric reducing/antioxidant power) assay showed a dose-dependent increse in activity. In a cell viability assay using MTT, the extract protected against $H_2O_2$-induced neurotoxicity. Lactate dehydrogenase (LDH) leakage was also inhibited by the extract, as was lipid peroxidation as shown using the mouse brain homogenate test. These data indicate that a 60% (w/v) methanolic extract of Houttuynia cordata has in vitro antioxidant activities, and ingestion there of may reduce the risk of developing neurodegenerative disorders.

Neuroprotective Effects of Kaempferol, Quercetin, and Its Glycosides by Regulation of Apoptosis (Kaempferol, quercetin 및 그 배당체들의 apoptosis 조절을 통한 신경세포 보호 효과)

  • Alzheimer's disease (AD) is a neurodegenerative disease caused by accumulation of amyloid beta ($A{\beta}$) in the brain. In the present study, we investigated the neuroprotective effects of four flavonoids such as kaempferol, kaempferol-3-O-glucoside, quercetin, and quercetin-3-${\beta}$-D-glucoside against neuronal apoptosis induced by $A{\beta}$ in SH-SY5Y neuronal cells. Treatment with $A{\beta}$ decreased cell viability compared to the non-treated normal group. However, treatment with the four flavonoids increased cell viability in SH-SY5Y cells treated with $A{\beta}$. In addition, we measured the expression of apoptosis-related proteins such as Bcl-2-associated X protein (Bax) and cleaved caspase-9. Treatment with the four flavonoids down-regulated Bax and cleaved caspase-9 in $A{\beta}$-treated SH-SY5Y neuronal cells. Overall, the results of the present study demonstrated the neuroprotective effect of flavonoids by anti-apoptotic activity in $A{\beta}$-induced SH-SY5Y neuronal cells. These results suggest that these four flavonoids would be useful therapeutic and prevention agents for AD.

Vitamin E protects neurons against kainic acid-induced neurotoxicity in organotypic hippocampal slice culture (뇌 해마 절편 배양 모델에서 흥분 독성에 대한 비타민 E의 신경 보호 효과)

  • Kainic acid (KA), an agonist for kainate and AMPA receptors, is an excitatory neurotoxic substance. Vitamin E such as alpha-tocopherol and alpha-tocotrienol is a chain-breaking antioxidant, preventing the chain propagation step during lipid peroxidation. In the present study, we have investigated the neuroprotective effects of alphatocopherol and alpha-tocotrienol on KA-induced neuronal death using organotypic hippocampal slice culture (OHSC). After 15h KA treatment, delayed neuronal death was detected in CA3 region. Alpha-tocopherol and alpha-tocotrienol increased cell survival and reduced the number of TUNEL-positive cells in CA3 region. These data suggest that alpha-tocopherol and alpha-tocotrienol treatment have protective effects on KA-induced cell death

The Protective Effects of Sasim-tang on the Brain Cell Damage in Photothrombotic Ischemia Mouse Model (광화학적 허혈성 뇌졸중 모델에서 사심탕(瀉心湯)의 뇌세포 손상 보호효과)

  • Objectives : This study was designed to investigate the effects of Sasim-tang (SST) on proinflammatory cytokine production in a photothrombotic ischemia mouse model. Methods : Photothrombotic ischemia was induced in stereotactically held male Balb/c mice using rose bengal (10 mg/kg) and cold light. The target of photothrombotic ischemic lesion was 1 mm anterior to bregma and 3 mm lateral to midline with 2 mm in diameter, which are decreased by oral administration of SST. Results : SST protected ischemic death of brain cells through inhibition of pro-inflammatory cytokines production and catalytic activation of caspase-3 protease in photothrombotic ischemia mouse model. Conclusions : The results of this study suggest that SST can have protective effects on brain cell damage in a photothrombotic ischemia mouse model.

Neuroprotective Effect of Ethyl Acetate Fraction of Portulaca oleracea L. (마치현 에틸아세테이트 분획물의 뇌세포 보호효과)

  • Portulaca oleracea L. is known to have many biological benefits such as anti-oxidant, anti-inflammatory, anti-allergic and anti-tumor. The objective of this study is to explore the neuroprotective effect of P. oleracea L. against glutamate-induced oxidative stress in mouse hippocampal HT22 cells. P. oleracea L. 70% ethanol extract and solvent fractions have the potent neroprotective effects on glutamate-induced nerotoxicity by induced the expression of heme oxygenase (HO)-1 in HT22 cells. Especially, ethyl acetate fraction showed higher protective effect. In HT22 cell, P. oleracea L. treatment with ERK inhibitor (PD98059) and c-JUN N-terminal kinase (JNK) inhibitor (SP600125) reduced P. oleracea L. ethyl acetate fraction induced HO-1 expression and P. oleracea L. ethyl acetate fraction also increased ERK and JNK phosphorylation. Furthermore, we found that treatment of P. oleracea L. caused the nuclear accumulation of Nrf2. In conclusion, the ethyl acetate fraction of 70% ethanol extract of P. oleracea L. significantly protect glutamate-induced oxidative damage by induction of HO-1 via Nrf2, ERK and JNK pathway in mouse hippocampal HT22. Taken together these finding suggest that P. oleracea L. ethyl acetate fraction is good source for taking active compounds and may be a potential therapeutic agent for brain disorder that induced by oxidative stress and neuronal damage.

  • Ths paper describes the transient response characteristics of the grounding systems for electric power facilities against impluse currents. In this work, when the impulse current was injected through the grounding conductor for arrester, the investigations treasuring and analyzing potential rises induced at the cormron cormection trint and other grounding conductors were conducted. The lightning impulse current was awliErl so as to simulate the on-set of arrester due to lightning surges.   • The grounding system of low voltage power systems is TT grounding system in Korea. In order to follow the international standard, TN grounding system is adopted. However, the performance of grounding systems has not been evaluated. This paper deals with the experimental results of protection ability of grounding system when lightning surge invades to the neutral line of low voltage power system. As a result, the TT grounding system is most frail for the lightning surge and it does not protect the electrical devices. On the other hand, the TN grounding system perfectly protects the electrical equipment and prevents the electric shock for human through the equipotential bonding. In case of TN system with supplement grounding, it is very important to lower the supplement grounding resistance to protect the electrical equipment and electric shock for human.

Effects of Allium hookeri Extracts on Glutamate-induced Neurotoxicity in HT22 Cells (글루타메이트로 유발한 세포독성에 대한 삼채추출물의 뇌세포 보호 효과)

  • Glutamate-induced oxidative stress results in neuro-degenerative disorders in many central nervous system (CNS) such as Alzheimer's disease, ischemia, Huntington's disease, and Parkinson's disease. Our study was performed to investigate neuroprotective effects of Allium hookeri extracts (leaf, root, and whole) on glutamate-induced HT22 cells. In this study, ethanol extract of A. hookeri showed the outstanding neuroprotective effect in HT22 cells. In addition, we found that ethanol extract of A. hookeri root increased heme oxygenase (HO)-1 in HT22 cells. Moreover, ethanol extract of A. hookeri root also upregulated nuclear accumulation of nuclear factor E2-related factor 2 (Nrf2) in HT22 cells. These results demonstrate that ethanol extract of A. hookeri root contributes neuroprotective effects against glutamate-induced oxidative stress in HT22 cells, via Nrf2-mediated HO-1 expression. Our study suggests that ethanol extract of A. hookeri root could be the potential agent for the treatment of many neuro-degenerative diseases.
