This study was conducted to select a desiccant and determine its concentration for safe usage to improve the harvesting efficiency of soybeans. Soybeans were treated with a desiccant (non-selective herbicide) before and after the maturation stage. The drying effect of the desiccant was higher at earlier treatment times than at the maturation stage, but the difference was not statistically significant. The higher efficacy might be related to the drying process of the leaves and stems, with most of the leaves and stems having already been dried by the time of hand harvesting. Desiccant treatments had no adverse effects on soybean yield, weight of 100 grains, seed quality, or seed germination rate compared with the untreated control. Pesticide residue analysis showed minimum residue concentration to be lower than the tolerance level of pesticide residues. In conclusion, it was confirmed that the desiccant was effective in drying soybean, and that there was no damage to the quality of soybean seeds. In addition to the drying effect, the dessicant treatment also facilitates the removal of weeds that interfere with the mechanical harvest and improves harvesting efficiency through the drying of the growth imbalanced individual. The desiccant treatment is expected to shorten the mechanical harvesting time by 1-2 weeks. It is thought that the selection of the proper cultivation period for other crops after soybean cultivation will be more advantageous.
Sulfoxaflor is a new active ingredient within the sulfoximine insecticide class that acts via a unique interaction with the nicotinic receptor. The MRLs (maximun residue limit) of sulfoxaflor in apple and pear are set at 0.4 mg/kg and that in pepper is set at 0.5 mg/kg. The purpose of this study was to develop an analytical method for the determination of sulfoxaflor residues in agricultural commodities using HPLC-UVD and LC-MS. The analysis of sulfoxaflor was performed by reverse phase-HPLC using an UV detector. Acetone and methanol were used for the extraction and aminopropyl ($NH_2$) cartridge was used for the clean-up in the samples. Recovery experiments were conducted on 7 representative agricultural products to validate the analytical method. The recoveries of the proposed method ranged from 82.8% to 108.2% and relative standard deviations were less than 10%. Finally, LC-MS with selected ion monitoring was also applied to confirm the suspected residues of sulfoxaflor in agricultural commodities.
To control effectively and safely Phytophthora root rot caused by Phytophthora capsici on tomato in hydroponic culture, tank-mixing method was considered with two pesticides, metalaxyl copper oxychloride 50% WP and dimethomorph dithianon 38% WP. Forty days after transplanting of tomato seedlings, 4 mL of sporangia of P. capsici (about 25 sporangi/mL) per plot was inoculated around tomato plant roots, and at 5 days after inoculation, the pesticides tank-mixed at three dilution levels, 12,500, 25,000 and 50,000, were drenched 1, 2 or 3 times per plot on the culture cube every 15 days for metalaxyl copper oxychloride 50% WP and every 10 days for dimethomorph dithianon 38% WP. During the drenching period, the residue levels of metalaxyl and dimethomorph in hydroponic culture solution were similar to the initial levels but the level of dithianon was drastically decreased from one day after tank-mixing. In tomato drenched with metalaxyl copper oxychloride 50% WP, metalaxyl was detected $0.02\sim0.04$ mg/kg in all diluted plots. Dimethomorph was detected $0.012\sim0.021$, $0.001\sim0.006$ and $0.001\sim0.003$ mg/kg in 12,500, 25,000 and 50,000 times diluted plots, respectively, while dithianon was detected 0.005, 0.003 mg/kg in 12,500 and 50,000 times diluted plots, respectively. The detection levels of three pesticides were far below compared with the levels of Korean MRLs. Incidences of Phytophthora root rot were not found in all the plots, but phytotoxic responses were recognized in the 12,500 times diluted plots of both pesticides. Based on the above results, the drenching of the culture solution tank-mixed with these pesticides could be recommended as a very safe and effective method to control Phytophthora root rot in tomato in hydroponic culture.
The effects of herbicides on biochemical processes in soil environment were studied by examining the effects of the chemical structure of each herbicides on soil enzyme activities and pesticides residue revealed when soil treated with urea was incubated at $28{\pm}1^{\circ}C$ for 56 days. The inhibition effects of herbicides on soil enzyme activites in soil decreased in the order of urea group>dinoseb>propanil>diphenyl eter group>acid amide group for urease, and dinoseb>urea group>diphenyl ether group>acid amide group for L-glutaminase and protease, dinoseb>diphenyl ether group>urea group>acid amide group for phosphatase. Herbicides inhibited the activities of soil enzyme in the early stage of treatment but increased the activities of urease, L-glutaminase and protease in the late stage. When herbicides were treated in soil together with urea the degradation of insecticides was accelerated.

In the present study, the metabolism of [$^{14}C$]butachlor was investigated in rice plant according to the OECD test guideline No. 501. [$^{14}C$]Butachlor was treated as granule to paddy water by application of 1.5 kg ingredient (a.i.)/ha at the 3~4 leave stage of rice plant. At 85 days after treatment (DAT), samples of panicle, foliage, and roots were taken for radioactivity analysis. Upon harvest at 126 DAT, rice plants were separated into brown rice, husk, straw, and root parts. Amounts of total radioactivity absorbed by rice plant ranged from 8.6 to 9.8% of applied radioactivity (AR). Total radioactive residues (TRRs) of rice plant at 126 DAT was the highest as 4.0421 mg/kg (7.3% AR) in the straw followed by 1.4595 mg/kg (2.4% AR) in the root, 0.7257 mg/kg (0.1% AR) in the husk. The lowest level recording 0.1020 mg/kg (0.1% AR) was found in brown rice. Each part was extracted with various solvents and solvent/water mixtures. Greater than 70% of TRRs was readily extractable from foliage, panicle, husk and straw. Only 34.0% of the brown rice and 43% of root based on TRRs were extractable showing that the residues were completely assimilated in the plant tissue. The level of non-extractable radioactivity was ranged from 26.2 to 66.0% of TRRs. From this study, five tentative major metabolites (M1, M2, M3, M4 and M5) were observed in rice extracts. Among the metabolites, 2,6-diethylaniline assigned as M4 was identified in rice plant by comparing to retention time of reference standard. Un-metabolized butachlor was not detected in any fractions. In soil extracts, N-(butoxymethyl)-N-(2,6-diethyl phenyl)acetamide, 2,6-diethylaniline, M2, M3 and M5 were observed. And the concentration of butachlor was low level (ca. 0.03 mg/kg).
The purpose of this study was to develop a methodology to detect spinosad which are difficult to analyze by multi-component simultaneous analysis of pesticide residues. We monitored spinosad due to the paucity of related information. The spinosad was determined using HPLC with UV detector at 250 nm. Correlation coefficient ($r^2$) for standard curve of spinosad A and D at standard concentration of 0.1-5.0 mg/kg were 0.999, respectively. Limit of quantitation (LOQ) of HPLC analysis was 0.005 mg/kg while limit of detection (LOD) was 0.001 mg/kg. Recovery experiments were conducted on five representative agricultural products to validate the analytical method. The recovery of proposed methods ranged from 74.9% to 104.0% and relative standard deviations were less than 10%. Spinosad residues were investigated in 16 commodities collected from 22 provinces. In this study, residues on all samples were not detected.
In order to elucidate the leaching behaviour of the herbicide bentazon in soil, soil columns(5cm ID ${\times}$ 34 cm L) were packed with three different soils up to 30 cm height, followed by the treatment of [$^{14}C$]bentazon, and rice plants(Oryza sativa L.) were grown for 9 weeks on these columns, with the columns without growing rice plants as the control for comparison. The amounts of $^{14}C$ activities percolated were about 92% of the originally applied $^{14}C$ irrespective of the physicochemical properties of the soils in the absence of rice plants, whereas $21{\sim}50%$ of the originally applied $^{14}C$ was percolated in the presence of rice plants, suggesting that the amounts of $^{14}C$ leached decreased remarkably in rice-cultivating soils. Bentazon leached faster in soil with higher pH and with lower organic matter content in the presence of growing rice plants. The amounts of $^{14}CO_{2}$ evolved from the soil columns were less than 0.2% of the originally applied $^{14}C$. Smaller amounts of $^{14}C$ were translocated into shoots via roots in soils with higher organic matter content. $^{14}C$ activities distributed into the aqueous phase of the leachate collected from the soil columns increased with leaching period and by rice cultivation, whereas the physico-chemical properties of soils did not exhibit any effect. The amounts of soil-bound residues increased remarkably by cultivating rice plants.
In order to investigate the behavior of the herbicide metolachlor [2-chloro-6'-ethyl-N-(2-methoxy-1-methylethyl)aceto-O-toluidide] in fish, carps (Cyprinus carpio L.) were exposed to the herbicide at $LC_{10}$(1.93 mg/L) for 4 days. Metolachlor dissolved in water was absorbed rapidly into carps to mark the maximum concentration 6 hours after exposure. The amounts of the $^{14}C$-metolachlor and its metabolites absorbed in gall were much higher than those in the other parts, strongly suggesting that biliary excretion involving enterohepatic recirculation could be an important route for the elimination of metolachlor. The $^{14}C$-radioactivity distributed into aqueous phase fraction in test water and in carp extract was increased in time-dependent manner. Extraction rate of $^{14}C$ absorbed in carp tissues was decreased remarkably up to 6 hours after exposure, suggesting that the possible polar metabolites of metolachlor were transformed into the conjugates to form non-extractable bound residues.
In order to establish a whitefly control system using conventional groups of insecticide(carbamate, organophosphorus and pyrethroid insecticides), three alternative application methods were designed on the medium growth stage of cucumber. To discriminate the effectiveness of these sequences observed were the residual activity of insecticides, frequency of insecticide applications, residue of insecticides in cucumber leaves, development of insecticide resistance in whitefly, and yield of fruits. Spraying furathiocarb, a carbamate insecticide, was very effective in reducing the frequency of application for the control of white flies. The effectiveness of furathiocarb was enhanced by the potentiation process to carbofuran, the long residual activity, and the lower development rate of insecticide resistance. Methion, an organophosphorus insecticide, did not show resistance development after successive use but resulted in short residual activity. However, other organophosphates, profenofos and phenthoate, lost their activity by the resistance development. Decreasing activity was common to pyrethroids, deltamethrin and zetacypermethrin due to resistance. From these results it could be drawn a conclusion that furathiocarb, a carbamate insecticide, was the most desirable among conventional insecticide groups for the management of greenhouse whitefly population on the cucumber. To prevent an outbreak of the insect pest by various cause, it was recommended to choose acetamiprid, a nicotinoid, which showed very good control efficacy to the resistance insects to conventional insecticides.
As the pine wilt disease spread rapidly over Korea in recent, aerial pesticide spraying to the infected pine stands increased abruptly. The increasing aerial pesticide application for control of the disease would result in disturbance of soil ecosystem in pine stands. This study was conducted to assess the disturbance, using field experiment in pine stands of Hongleong experimental forests in Seoul with BACI experimental design with three sprayed plots and three unsprayed plots of $25m^2$. We sprayed fenitrothion of 0.0335 kg a.i./ha over each of the sprayed plots, being the same as the pesticide residues of litters after aerial spraying. The pesticide had been applied on 4 June, 11 June, and 29 June in 2002, comparable with the dates of the aerial spraying in the previous year in Busan. We monitored population of soil organisms including arthropods, fungi, bacteria, and actinomycetes at two sampling days before pesticide application, at two sampling days during the application season, and at three sampling days after last application for two months since late May in 2002. The pesticide applications did not make any significant effect on the population of arthropods, fungi, bacteria, and actinomycetes in soils and on the structure of soil organism community, showing low probability of significant effects of the aerial pesticide application on soil organisms in pine stands.
