Effects of KH204 on the Relaxation Response of Rabbit Corpus Cavernosum and Reproductive Function in Male Rats (토끼 음경해면체평활근 이완 및 10주간 경구투여한 흰쥐의 생식기능에 미치는 KH204의 효과)

  • This study was designed to investigate the effects of KH204 on the relaxation response and reproductive function in male. Strips of rabbit corpus cavernosum were prepared for mounting and isometric tension measurement in an organ bath. On cavernosal strips contracted with $1{\times}10^{-6}$ phenylephrine and KH204 was applied in increasing concentrations from 0, 61, 183 and 549 mg/L, causing dose-dependent relaxation. Male Sprague-Dawley rats were orally administered with 61, 183 and 549 mg/kg/day of KH204 for 10 weeks. We examined organ weights, testicular sperm head counts, epididymal sperm counts, motility and morphology. KH204 relaxed rabbit corpus cavernosal strip contracted by $1{\times}10^{-6}$ phenylephrine in a dose-dependent manner. In the male rat, testicular weight was increased significantly in the KH204 treated groups compared with control group. Also in the testicular sperm head counts, epididymal sperm counts were increased significantly in the KH204-treated rats. In conclusion, the data suggest that KH204 could enhance erectile and reproductive function.

Antioxidant and Neuronal Cell Protective Effects of an Extract of Houttuynia cordata Thunb (a Culinary Herb) (어성초 추출물의 항산화 및 신경세포 보호효과)

  • The in vitro antioxidant activities and neuronal cell protective effects of 60% (w/v) methanolic extract from Houttuynia cordata were investigated. The contents of total phenolics and quercitrin in the extract were 17.71 mg/g and 75.80 ${\mu}g$/g, respectively. DPPH and ABTS radical-scavenging activities were 87.79% and 99.27%, respectively, when the extract was tested at 5 mg/ml. The FRAP (ferric reducing/antioxidant power) assay showed a dose-dependent increse in activity. In a cell viability assay using MTT, the extract protected against $H_2O_2$-induced neurotoxicity. Lactate dehydrogenase (LDH) leakage was also inhibited by the extract, as was lipid peroxidation as shown using the mouse brain homogenate test. These data indicate that a 60% (w/v) methanolic extract of Houttuynia cordata has in vitro antioxidant activities, and ingestion there of may reduce the risk of developing neurodegenerative disorders.

Antioxidative and Anti-inflammatory Activities of Ardisia arborescens Ethanol Extract (Ardisia arborescens 에탄올 추출물의 항산화 및 항염증 활성)

  • In this study, the antioxidative and anti-inflammatory activities of Ardisia arborescens ethanol extract (AAEE) were evaluated using in vitro assays and a cell culture model system. AAEE exhibited potent scavenging activity against 1,1-diphenyl-2-picryl hydrazyl (DPPH), similar to ascorbic acid, which was used as a positive control. Moreover, AAEE effectively suppressed lipopolysaccharide (LPS)- and hydrogen peroxide ($H_2O_2$)-induced reactive oxygen species (ROS) in RAW 264.7 cells. Furthermore, AAEE induced the expression of antioxidative enzymes, heme oxygenase 1 (HO-1), and thioredoxin reductase 1 (TrxR1), in addition to their upstream transcription factor, nuclear factor-E2-related factor 2 (Nrf2), in a dose-dependent manner. The upstream signaling pathways of mitogen-activated protein kinases (MAPKs) might regulate the modulation of HO-1, TrxR1, and Nrf2 expression. On the other hand, AAEE inhibited LPS-induced nitric oxide (NO) formation, without cytotoxicity. Suppression of NO formation was the result of AEEE-induced down-regulation of inducible NO synthase (iNOS). The suppression of NO and iNOS by AAEE might be modulated by their upstream transcription factor, nuclear factor (NF)-${\kappa}B$, and activator protein (AP)-1 pathways. Taken together, these results provide important new insights into the antioxidative and anti-inflammatory activities of A. arborescens. AAAEE might represent a promising material in the field of nutraceuticals.

Inhibitory Effects of Euphorbia supina Rafin on the Production of Pro-Inflammatory Mediator by LPS-Stimulated RAW 264.7 Macrophages (LPS로 활성화된 RAW 264.7 대식세포에서 애기땅빈대(Euphorbia supina Rafin)의 염증매개물질 억제효과)

  • This study was designed to evaluate the effect of hot water extract (ESW) and 70% ethanol extract (ESE) from Euphorbia supina Rafin on LPS-stimulated inflammatory response in RAW 264.7 macrophages. Upon investigation at concentrations up to $1000\;{\mu}g$/mL, ESW and ESE did not have any cytotoxic effects on RAW 264.7 macrophages. ESW induced inhibition of 21.6%~54.8% of nitric oxide (NO) production at 100~1000${\mu}g$/mL, and $PGE_2$ production was inhibited up to 25.7%~38.2% at 250~1000${\mu}g$/mL, proportional to the ESW concentrations. ESW induced inhibition of 66.1% and 54.3% of IL-6 production at 250 and $1000\;{\mu}g$/mL, respectively. ESE (100~1000${\mu}g$/mL) induced inhibition of 38.3%~77.5% of NO, 40%~94.7% of $PGE_2$, and 43.9%~89.4% of IL-6 production, proportional to the ESE concentrations. Only 44.1% of IL-10 production was inhibited at a concentration of $500\;{\mu}g$/mL. ESE induced an increase in TNF-${\alpha}$ production at a concentration of 100 and $250\;{\mu}g$/mL, whereas at high concentrations (500 and $1000\;{\mu}g$/mL), ESE induced inhibition of 19.2% and 92.4% of TNF-${\alpha}$ production, respectively. In conclusion, concentrations of more than $500\;{\mu}g$/mL ESE demonstrated effective immune-modulating activity through inhibition of NO, $PGE_2$, IL-6, IL-10, or TNF-${\alpha}$ production, as it relates to the macrophage's immuno-activity; therefore, ESE has potential as a good candidate substance for reduction of inflammatory responses.

Hepatoprotective Effects of Sumaeyaksuk (Artemisia argyi H.) Extract on LPS-mediated Inflammatory Response (LPS에 의해 유도된 염증반응에서 섬애약쑥 추출물의 간보호 효과)

  • Artemisia, a plant widely used as traditional herbal medicine in many countries, has drawn attention of the researchers. And its extracts or compounds are known to have an efficacy of antioxidant, anti-diabete, anti-cancer, anti-inflammation and neuroprotection. Sumaeyaksuk is a variant of the Artemisia argyi and major constituents are eupatilin and jaceosidin. This study was performed to investigate the effects of the sumaeyaksuk aqueous extract on inflammatory response induced by lipopolysaccharide (LPS) in human hepatoma HepG2 cells. To examine the potential hepatoprotective properties of sumaeyaksuk extract, cell viability, as well as nitric oxide (NO), reactive oxygen species (ROS), macrophage colony-stimulating factor (M-CSF), interleukin-8 (IL-8) levels, alanine transaminase (ALT), and aspartate transaminase (AST) activities, were measured. Cytotoxic activity of extracts on HepG2 cells was measured by MTT assay. Sumaeyaksuk extract did not induce cytotoxicity at concentrations of $0{\sim}400{\mu}g/mL$. NO and ROS levels significantly decreased with increasing concentration of the extract. The secretion levels of M-CSF and IL-8 were suppressed by sumaeyaksuk extract in a dose-dependent manner. Moreover, ALT (75.4%) and AST (61.6%) levels significantly decreased in sumaeyaksuk extract-treated cells at $400{\mu}g/mL$. These results suggested that the sumaeyaksuk extract attenuates the LPS-induced hepatotoxicity resulting from regulation of inflammatory factors and could potentially be used as a hepatitis therapeutic agent.

A Study on Inflammation and Itching of Cyperus rotundus Ethyl Acetate Fractions (향부자 에틸아세테이트 분획물의 염증 및 가려움증에 관한 연구)

  • In this study, Cyperus rotundus (C. rotundus) was fractionated into ethyl acetate to identify α-cyperone, a representative indicator, and its ethyl acetate fraction were evaluated to confirm the possibilityas a functional cosmetic ingredient. As a result of HPLC analysis, it was confirmed that the content of α-cyperone was 5.243%. In order to verify the inflammatory relief effect of the ethyl acetate fraction from C. rotundus (EAFC) and α-cyperone, it was confirmed that nitric oxide (NO) production inhibitory ability in RAW 264.7 macrophages induced an inflammatory reaction with lipopolysaccharide (LPS). Real-time qPCR analysis confirmed inhibition of mRNA expression level of inflammatory factors, IL-1β, IL-6, TNF-α, COX-2 and iNOS. As a results of conducting a clinical study using a simple cosmetic formulation containing EAFC, it was confirmed that the skin irritation stimulated by sodium lauryl sulfate (SLS) was calming and relieving itching. Through these results, it is believed that the C. rotundus can be used as a natural cosmetic ingredient that has the effect of inhibiting inflammation and relieving itching.

The Degradation of the Effect of Drag Reduction in Synthetic Polymer Solution (합성고분자 첨가제에 의한 마찰저항감소효과의 퇴화에 관한 연구)

  • Degradation of polymer additives is enhanced at higher temperature of the test solutions. The degradation of Co-polymer solution was investigated experimentally in a closed loop at the temperature of 6$0^{\circ}C$ and 8$0^{\circ}C$ with various polymer concentrations of 100, 200, 400, 600 ppm in order to see the effect of temperature and polymer concentration with time. The degradation effect were found to be more dependent on temperature than mechanical shear. The friction factor versus Reynolds number curves show that in the range of Reynolds number number 50,000~150,000 the friction was decreased as Reynolds number increased and the friction of solution at low temperature approached to Virk's maximum drag reduction asymptote. For constant flowrates and temperatures the degradation effect was found to be less likely in higher polymer concentration. For constant flowrates and polymer concentrations the degradation rates are affected mainly by temperature. At the temperature of 8$0^{\circ}C$ and polymer concentration of 100 ppm, drag reduction effect was disappeared after 4 hours. However, this thermal degradation could be avoided with additional materials such as surfactants which are supposed to enhance the bonding forces between polymer molecules.

Herbicidal Activity and KAPAS Inhibition of Juglone with Potential as Natural Herbicide (천연 Naphthoquinone계 Juglone의 KAPAS 저해 및 제초활성 특성)

  • The potential of juglone a plant naphthoquinone as a natural herbicide on new target, 7-keto-8-amino pelargonic acid synthetase (KAPAS) in the early step of biotin biosynthesis pathway, was performed in vitro and in vivo. Juglone effectively inhibited KAPAS activities in vitro and the $IC_{50}$ was $9.5{\mu}M$. Foliar application of juglone showed very good herbicidal activity to the eight-tested weed species. Among them, Solanum nigrum was completely controlled at a concentration of $250{\mu}g\;mL^{-1}$ with main symptoms of desiccation or burndown. Digitaria sanguinalis and Aeschynomene indica were also sensitive to juglone treatment. All eight weed species were controlled by 90~100% at a concentration of $500{\mu}g\;mL^{-1}$. However, soil application of juglone to Digitaria sanguinalis did not show any herbicidal symptoms. Cellular leakage from cucumber leaf squares treated with juglone increased depending on the concentrations increased from 6.25 to $100{\mu}M$ after 24 hours incubation with or without light. However, chlorophyll loss in cucumber leaf squares was negligible. Biotin supplements significantly rescued the inhibition of germination rate of Arabidopsis thaliana seeds previously inhibited by the juglone. Our results suggest that the juglone is a possible environmental friendly herbicide candidate with a new target KAPAS inhibiting activity.

Weight Reduction and Lipid Lowering Effects of Kimchi Lactic Acid Powder in Rats Fed High Fat Diets (고지방식이를 섭취한 흰쥐에서 김치유산균분말의 비만 억제 및 지질 저하 효과)

  • Effects of kimchi lactic acid powder (KL) on weight reduction and lipid-lowering activities were studied using rats fed high fat diet. Spraque Dawley (40 rats) were fed 20% (w/w) high fat diet far 8 weeks to induce obesity and divided equally into normal diet (ND), high fat diet (HFD), and 10 and 20% KL-added groups. Body weight of HFD was 150% (p<0.01) of ND, while those of HFK10 and HFK20 decreased by 13 and 15% compared to HFD, respectively (p<0.01). Amounts of visceral fats for HFK10 and HFK20 were decreased by 42 and 48% compared to that of HFD (198% of ND), respectively (p<0.01). B had greater effect on reducing visceral fats than body weight. Plasma triglyceride (155%), cholesterol (129%), and LDL (161%) concentrations of HFD, which had been significantly increased compared to ND, dropped by 26, 9, and 8% (p<0.01) in HFK10, and 35, 17, and 33% (p<0.05) in HFK20, respectively. Decreased HDL by high fat diet was re-increased by KL supplementation. KL showed lipid-lowering and liver-protecting effects in liver possibly by interrupting lipid absorption in the intestine either by absorbing lipids in their cell walls or degrading them rapidly. Increased excretion of TG and cholesterol in feces of HFK10 and HFK20 indicates KL might have similar role to dietary fiber.

Whitening and Anti-wrinkle Effects of Apple Extracts (사과 추출물의 미백 및 주름개선 효과)

  • The in-vitro whitening and anti-wrinkle effect of ethanol extracts from apple flesh and peel were investigated. The EtOAc fractions from the ethanol extracts of apple flesh and peel showed in-vitro antioxidant activities in a dose-dependent manner on ABTS radical-scavenging activity and ferric-reducing/antioxidant power, and had the highest total phenolic contents (84.25 and 318.25 mg GAE/g). In addition, the EtOAc fractions generally showed strong UV absorption within the UV-B range. In the cellular system, the melanin synthesis of the B16/F10 melanoma cells was decreased by the EtOAc fractions of apple peel in a concentration-dependent manner. The EtOAc fractions of apple peel also showed a great elastase inhibition of 46.40% at 100 ${\mu}g$/mL, thus showing good in-vitro anti-wrinkle characteristics. These results suggest that the EtOAc fractions from the ethanol extract of apple peel can be used as whitening and anti-wrinkle agents as well as antioxidant resources.
