Diversity of Mitochondrial DNA Cytochrome b Gene in Roe Deer (Capreolus pygargus tianschanicus Satunin) from Jejudo Island, Korea (한국의 제주도에 서식하고 있는 노루(Capreolus pygargus tuanschanicus Satunin)의 미토콘드리아 DNA cytochrome b 유전자의 다양성)

  • As one of researches to clarify the taxonomic status of roe deer from Jeju island (C. pygargus tianschanicus), we analyzed partial sequences of mtDNA cytochrome b gene from six roe deers at Jeju island in Korea. Maximum nucleotide Tamura & Nei's distance among three haplotypes was 0.005, and this distance was comparable to the diversity within other roe deer subspecies: it is suggested that roe deers from the mainland dispersed rarely to Jejudo Island, although further analyses are ne-cessary to decide whether or not it was occurred by human introduction. Furthermore, nucleotide distance between cytochrome b sequences of roe deers from Jeju (C. pygargus tianschanicus) and the sequence of roe deer from west Siberia(C. p. pygargus), obtained from GenBank, was average 0.013, and it is suggested that C. p. tianschanicus diverged from C. p. pygargus of west Siberia 0.65 Myr ago.

Modeling the Spatial Distribution of Roe Deer (Capreolus pygargus) in Jeju Island (제주 노루(Capreolus pygargus)의 서식지 선호도 분석)

  • The habitat preference of roe deers(Capreolus pygargus) in Jeju island, South Korea was analyzed by using their occurrence probability in MaxEnt model in this study. Totally 490 surveying data were gathered and 15 environmental variables were chosen for the model in which 6 variables out of 15 ones were filtered and finally removed because of there being higher correlation(over 0.7 in correlation coefficient). According to the modeling, roe deers were known to prefer the area ranging from 200 to 700 meter and over 1,500 meter in sea level, where there were not many dominant tree and/or dominant vegetation with low density so that understory vegetation can grow well with plentiful sunlight and can be used as a food of herbivore like roe deers. Otherwise, the region ranging from 700 to 1,500 meter was mostly covered with high density vegetation which cut off sunlight trying to penetrate through the dominant vegetation. It can cause a lower density of vegetation on surface, which can not attract to roe deers.

Palynotaxonomic Study on the Korean Pyrolaceae (한국산 노루발과(Pyrolaceae)의 화분분류학적 연구)

  • Pollen grains of seven taxa on the family Pyrolaceae, known to be distributed in Korea (Pyrola dahurica(Andreses) Kom., P. incarnata (DC.) Fisch. ex Kom., P. japonica Klenze ex Alefeld, P. minor L., P. renifolia Max., Orthilia secunda (L.) House, Chimaphila japonica Miq.), were examined by the light and the scanning electron microscope in order to evaluate their taxonomic significances. The pollen units of genus Pyrola, Orthilia, Chimaphila were tetrad, monad, polyad respectively. This characteristics was good character for delimiting the genus, and Orthilia secunda (L.) House was more proper scientific name than Pyrola secunda L. based upon this character. Pollen size, tricolporate length, surface sculpturing pattern were different slightly among the treated taxa, and surface sculpturing patterns of Pyrola dahurica, P. incarnate, P. renifolia, Chimaphiza japonica were different from previous reports. Evolutionary trends of pollen grains in Pyrolaceae were inferred from the pollen units and aperture shapes.

Cultural characteristics and antioxidant activity of wild-type collections of Hericium erinaceus (노루궁뎅이버섯 야생 수집균주의 배양적 특성과 항산화 활성)

  • This study aimed to characterize the mycelial morphologies of Hericium erinaceus isolates, including 18 wild-type collections and cultivatable species 'Noru 1' and 'Noru 2', in Korea. The morphological characteristics were used to classify the species based on aerial or branched mycelia and their brownish or whitish colors when grown on potato dextrose agar. Of the wild-type collections, the isolates KFRI 509, KFRI 1091, KFRI 1093, and KFRI 1623 showed faster mycelial growth than the cultivars 'Noru 1' and 'Noru 2'. Moreover, 60% ethanol extracts of the fruiting body of the mushroom showed the highest phenol content and DPPH and ABTS radical scavenging activity. The DPPH and ABTS radical scavenging activities of KFRI 507, KFRI 508, KFRI 842, and KFRI 1623 were 10-60% higher than those of 'Noru 1' and 'Noru 2', depending on the extract concentrations. Thus, results suggest that these wild-type collections could be useful for breeding genetic sources or processed food materials with high antioxidant activity.

  • 연구목적: 생약으로 부터 항암제 개발연구의 일환으로 민간에서 조경. 지혈 등에 사용되고있는 노루발(Pyrola japonica)의 세포독성 물질과 그의 화학적구조를 규명하기 위해서 본 연구를 실시하였다. 실험 방법 : 옴건한 노루발의 지하부를 methanol로 환류 추출하여 얻은 methanol extract를 n-hexane, ethylacetate, n-butanol 순으로 분획하였다. 그 후 각각의 분획을 L1210 세포에 대하여 in vitro에서 실험한 결과 n-heane, ethylacetate 분획에서 강한 활성이 나타남을 알 수 있었다. 따라서 n-hexane, ethylacetate 분획을 반복하여 silica gel column chromatography 하여 1종의 활성물질과 4종의 비활성물질을 단리하였다. 실험결과 : 단리된 활성물질은 각종 spectral data를 검토하여 chimaphilin (L1210의 ED$_{50}$ : 0.6 $\mu\textrm{g}$/ml, K562의 ED$_{50}$ :< 0.6 $\mu\textrm{g}$/ml) 임을 확인하였고 이 물질은 5,8-dihydro-2.7-dimethyl-4-naphtoquione의 변성물질 임을 알 수 있었다.있었다.

Molecular phylogeny of Astilbe: Implications for phylogeography and morphological evolution (노루오줌속(Astilbe)의 분자 계통: 계통지리 및 형질 진화에 대한 고찰)

  • Astilbe (Saxifragaceae) is a genus well known for its disjunctive distribution in Asia and eastern North America. In this study, we reconstructed a molecular phylogeny of the genus using the sequences of ITS regions of nuclear ribosomal DNA. A total of 17 species representing major lineage of Astilbe and closely related taxa were included in the phylogenetic analyses. We obtained a Bayesian phylogenetic tree in which Saxifragopsis was positioned as a sister group to Astilbe. The Japanese endemic species, A.platyphylla was the most basal lineage within the genus. This species is well known for its distinct morphological features such as unisexual flowers, apetaly, and calyx with 7-11 lobes. Two species, A. biternata, a New World representative of the genus, and A. rivularis widely distributed in S. Asia, branched off early in the evolution of Astilbe. The remaining species formed a strongly supported core clade, which diverged into two robust geographical lineages: the first ("Japonica" clade) of species distributed in Japan, Taiwan, and Philippines and the other ("Rubra" clade), of taxa in China and Korea. The ITS phylogeny indicates that the Bering land bridges were the major route for the origin and dispersal of A. biternata. The two Taiwanese taxa and A. philippinensis were found to derive from the Japanese member, as the genus advanced southwards. The ITS phylogeny suggests that apetaly originated independently at least two times within the genus. Our results do not support Engler's classification system of the genus based on the leaf type (simple vs. compound), but reaffirm Hara's taxonomic idea which primarily considered the features of calyx.

The Current Number of Its Individuals and Management Plan of Roe Deer(Capreolus pygarus tianschanicus Satunin) Inhabiting Mt. Halla(Hallasan), Jeju Island, Korea (한라산에 서식하는 노루(Capreolus pygarus tianschanicus Satunin)의 개체수 현황과 관리방안)

  • This study researched the number of individuals of Roe deer (Capreolus pygarus tianschanicus Satunin), which inhabit Mt. Halla (Hallasan) National Park and its neighboring areas ranging about $270km^2$ areas. The research was done from November(late fall), 1998 to March(early spring), 2001, and additional research was carried out from November 2006 to January 2007. As a result of this, we identified a total of 1,444 deer, and found out the fact that there lived 5.33 deer per $km^2$. By region, we identified a maximum of 270 deer at Donnaeko hiking path and a minimum of 12 deer at the adjacent areas to the second Sanrok-road. The frequency of Roe deer group sighting was higher at the edge of Hallasan National Park than any other surveyed region. Thus, it is estimated that the distribution of Roe deer of Hallasan is more influenced by the quantity and quality of food than human-made interference during wintry season.
