Studies on Rapid Microbiological Testing Method of Fresh Pork by Applied Resazurin Reduction Test(RRT) Method (Resazurin 환원법을 응용한 돈육의 신속 미생물 검사법에 관한 연구)

  • In order to search for reliable rapid methods of estimating bacterial counts in pork, this study was tried to measure resazurin reduction time which is simple in experimental method, low in analytical cost, able to estimate bacterial count within short time. The results were summarized as follows; Correlation coefficient between initial bacterial log count(25$^{\circ}C$/72hr, Y) and resazurin reduction time(X) from blue color to pink color during incubation at 25$^{\circ}C$ and 30$^{\circ}C$ was higher than other conditions as -0.95 and -0.94, respectively. Considering correlation coefficient and reduction time, incubation temperature was compatible at 30$^{\circ}C$, and regression equation(RE) was Y = -0.4386X + 7.7870. At a bacterial load of $10^2$cfu/$cm^2$, $10^3$cfu/$cm^2$ and $10^4$cfu/$cm^2$ in pork, reduction time was 13.2hr, 10.9hr and 8.6hr, respectively. Correlation coefficient between initial bacterial log count(30$^{\circ}C$/72hr, Y) and resazurin reduction time(X) from blue color to pink color during incubation at 30$^{\circ}C$ was highest among other conditions as -0.93, and RE was Y = -0.4171X + 7.5540. At a bacterial load of $10^2$cfu/$cm^2$, $10^3cfu/$cm^2$ and $10^4cfu/$cm^2$ in pork, reduction time was 13.3hr, 10.9hr and 8.5hr, respectively. Correlation coefficient between initial bacterial log count(35$^{\circ}C$/72hr, Y) and resazurin reduction time(X) from blue color to pink color during incubation at 30$^{\circ}C$ was highest among other conditions as -0.93, and RE was Y = -0.3514X + 6.7513. At a bacterial load of $10^2$cfu/$cm^2$, $10^3$cfu/$cm^2$ and $10^4$cfu/$cm^2$ in pork, reduction time was 13.5hr, 10.7hr and 7.8hr, respectively.

Estimation of Evaporation Rate of Swine Slurry Using the Natural Evaporation System(NES) in summer (여름철 자연증발시스템(NES)의 腞슬러리 증발효율 평가)

  • The purpose of this study was to establish the optimal operation condition of the natural evaporation system(NES) which was used for reducing swine slurry. Especially the main point of this study is to estimate the effect of climate condition(clear & rainy) and spray type(batch & flow) for the evaporation rate of swine slurry applying the NES in summer. Experiment was performed from June to August, which was generally regarded as summer in Korea, with the spray type of batch in 2000 and that of flow in 2001. As a result of experiment for batch and flow type, the averaged evaporation rate was measured into 2.71 and 3.59 l/ton . $m^2$ . day on clear days and 0.62 and 0.66 l/ton . $m^2$ . day on raint days, respectively. Based on the calculated evaporation rate by the climate condition and the spray type, it was proved that the averaged reduction rate for total input(1t/day) were 15.99% and 3.19% on clear and rainy days and the evaporation rate of the flow type was superior to that of the batch type by 5%, approximately. Therefore, it was concluded that the supplementary equipment, such as fan, should by operated in rainy days and the spray type of flow rather than that of batch should be recommended to increase the evaporation rate in the natural evaporation system(NES).

Morphological Characteristics and Germination as Affected by Low Temperature and GA in Orostachys 'Jirisan' and 'Jejuyeonhwa' Seeds, Korea Native Plant (지리산바위솔과 제주연화바위솔 종자의 형태특성 및 저온과 GA에 대한 발아 반응)

  • This study was conducted to improve the seed germination by low temperature and $GA_3$ in $Orostachys$ $japonicus$ A. Berger (Jirisan) and $O.$ $iwarenge$ (Mak.) Hara (Jejuyeonhwa), Korean native plants. Observation of the seeds using a stereo microscope showed that all seeds of the two species have wrinkled surface and oblong shape. Seed size ranged 0.77-1.00/0.25-0.37 mm (length/width), indicating that the seeds are minute seeds. When the seeds of two $Orostachsis$ species were sown into petri-dish and placed in a plant growth chamber of 10, 15, 20, or $25^{\circ}C$, 'Jirisan' showed seed germination below 20% at all temperatures and 'Jejuyeonhwa' 80% at only $10^{\circ}C$. Seed germination of 'Jirisan' increased up to 44% at $10^{\circ}C$ by low temperature ($4^{\circ}C$) storage for 10 days, but decreased again at storage for more than 20 days. The seeds of 'Jejuyeonhwa' showed a large increase in seed germination by low temperature for 20-30 days, which was 95% at $10^{\circ}C$, but low temperature for more than 40 days significantly decreased seed germination. Dipping treatment in GA3 solution of $50-400mg{\cdot}L^{-1}$ for different periods (3, 6, 12, and 24 hrs) remarkably improved germination rate and speed in both species, 80-100% in 'Jirisan' and 90-100% in 'Jejuyeonhwa' at all concentrations and dipping times used in this study.

HACCP Application of Instant Ablactation Baby Food Processing (즉석 이유식 제조시 HACCP의 적용)

  • In this study. HACCP system was applied to improve the quality of instant ablactation baby food with rising sanitary problem among SunSig(powder of roasted grains and vegetables) . It took about 9min 30 second to produce and the temperature status of raw materials were 30.9∼31.8$\^{C}$. Raw material and production phase goods had average 6.2 in pH and 0.23∼0.63 in water activity. The average number of total aerobic bacteria(TAB) and coliform were 1.4$\times$10$^4$ CFU/g, 5.5$\times$10 CFU/g in raw materials. And the number became 8.7$\times$10$^2$ CFU/g, 6.9$\times$10 CFU/g after 1st grinding step. The distributions of TAB and coliform in equipments were 3.8$\times$10$^4$ CFU/㎠, 8.0$\times$10 CFU/㎠ on the average. According to storage temperature, variations of number of TAB coliform were slight at 4$\^{C}$ but increased at 20$\^{C}$ and 30$\^{C}$ . Therefore following subjects can be issued on the basis of results. Minimize the storage time and keep at cool temperature. 1st and 2nd grinders should be managed hygienically. Establish the heating step during the production of instant action baby food. Instant ablactation baby food produced should be stored at cool and freeze condition.

Effect of Conjugated Linoleic Acid(CLA) Feeding Levels and Periods on Textural Property and Fatty Acid Composition of Pork (Conjugated Linoleic Acid(CLA) 급여수준과 급여기간이 돈육의 조직감과 지방산 조성에 미치는영향)

  • To investigate the effects of conjugated linoleic acid added diet feeding on CLA accumulation and quality characteristics of pork meat. The CLA used to add in diet was chemically synthesized by alkaline isomerization method with corn oil. Pigs were divided into 5 treatment groups(4 pigs/group) and subjected to one of five treatment diets(0, 1.25% CLA for 2 weeks, 2.5% CLA for 2 weeks, 1.25% CLA for 4 weeks and 2.5% CLA for 4 weeks, CLA diets; total fed diets) before slaughter. Pork loin were collected from the animals(110 kg body weight) slaughtering at the commercial slaughter house. Pork loin meat were aerobic packaged and then stored during 2, 5, 8, 11 and 14 days at 4$^{\circ}C$. Samples were analyzed for shear force value, texture, TBARS, fatty acid composition, cholesterol and CLA content. CLA treatment groups showed significantly(p〈0.05) higher shear force value compared to those of control group at 11, 14 days of cold storage. All treatments were decreased significantly as the storage period passed. There was a not significantly difference in texture between control and CLA treatment groups. All CLA treatment groups showed significantly(p〈0.05) lower TBARS value than the control. TBARS value was increased significantly during storage in all treatment. CLA treatment groups showed significantly(p〈0.05) lower cholesterol content compared to those of control group. As dietary CLA was increased in feed, the content of CLA was increased, but the control was almost not detected. The contents of CLA were not significantly changed during chilled storage for 14 days. In the change of fatty acid composition, the contents of oleic, linoleic and arachidonic were decreased by dietary CLA-supplementation, whereas the increase level of CLA-supplementation resulted in the higher palmitic and stearic acid. In all results, CLA could be accumulated in pork meat and its antioxidant capability has been indicated. It was suggested that dietary CLA-supplementation could be produced high quality pork.

Quality Characteristics of Gat Kimchi Added with Cheonnyuncho Water Extract during Cold Storage (천년초 물 추출물을 이용한 갓김치의 저온저장 중 품질 특성)

  • In this study, quality characteristics of three different kinds of Gat Kimchi, added with Cheonnyuncho fruit (C-fruit) water extract, Cheonnyuncho cladodes (C-cladodes) water extract, and without Cheonnyuncho water extract (control group), were investigated during storage for 80 days at $5{\pm}1^{\circ}C$. In terms of mineral content of Gat Kimchi, C-fruit group showed higher calcium and zinc contents than the control group, whereas Cheonnyuncho groups showed higher iron content, and C-cladodes showed higher potassium content than the control group. The pH was mostly higher in Cheonnyuncho groups than in the control group until 40 days of storage, whereas the pH of the C-cladodes group significantly decreased after 60 days. On the other hand, titratable acidity showed the reverse tendency during storage. Hunter's color L, a, and b values all increased during the storage period. The hardness value was significantly higher in Cheonnyuncho groups than in the control group after 40 days of storage, and hardness value of the C-fruit group was significantly higher than those of the other groups after 80 days of storage. In terms of sensory evaluation, there was no significant difference between the control group and Cheonnyuncho group during fermentation. As a result, the study suggests that addition of C-fruit extracts increase nutrition and function of Gat Kimchi during storage.

Effects of Conjugated Linoleic Acid Accumulated Pork on Fatty Acid Composition and Quality Characteristics of Press Ham (CLA가 축적된 돈육으로 제조된 Press Ham의 지방산 조성 및 품질 특성에 미치는 영향)

It was not clearly changed by storage time. No remarkable differences were found in sensory properties (color, flavor, texture and acceptability) between control and CLA treatment groups. It was not clearly changed by the passage of storage time. In the change of fatty acid composition, the contents of oleic, linoleic and arachidonic were decreased by dietary CLA-supplementation, whereas the increase level of CLA-supplementation resulted in the higher palmitic and stearic acid than control. As dietary CLA was increased in feed, the content of CLA in pork was increased.

Quality Characteristics of Pressed Ham Containing Grape Seed Oil (포도씨유가 함유된 프레스햄의 품질특성)

  • Pressed ham was manufactured to investigate the effects of grape seed oil on the quality characteristics of pressed ham. Five treatments were divided based on differences in the amount of grape seed oil added into the pressed ham. For control, 10% of back fat was only added without grape seed oil. For the first treatment, 10% of grape seed oil among the lard component added into the pressed ham was replaced. For the 2nd, 3rd and 4rd treatments, 20%, 30% and 40% of grape seed oil was respectively replaced. Pressed ham manufactured using grape seed oil was vacuum packaged and then stored for 1, 7, 14, 21 and 28 days at 4℃. Samples were analyzed for shear force value, sensory properties, TBARS and fatty acid composition. In the 1, 21 and 28 days of storage, shear force value of grape seed oil treatment (T4) was significantly lower than that of control (P<0.05). No remarkable differences were found in sensory properties among control and grape seed oil treatment groups. The TBARS value was significantly higher in control than in grape seed oil treatment group(T4) at 28 days of storage (P<0.05). The TBARS of control and grape seed oil treatment groups increased significantly as the storage period increased(P<0.05). The linoleic acid(C18:2) content of grape seed oil treatment groups was significantly higher than that of control(P<0.05). But the contents of C10:0~C20:4 were decreased significantly by grape seed oil additive (P<0.05). Saturated and monounsaturated fatty acid content of control was significantly higher than that of grape seed oil treatment groups(P<0.05). Whereas the increase level of grape seed oil additive resulted in the significantly higher polyunsaturated fatty acid content(P<0.05). Based on these findings, we conclude that the sensory properties and lipid oxidation(TBARS) of manufactured pressed ham were not affected by grape seed oil addition. Also, our results indicate that high-quality pressed ham can be manufactured with strengthen of polyunsaturated fatty acid content.

Evaluation for Cleanness of Kitchen and Hall of Restaurants in Seoul (서울지역 음식점 주방 및 식당의 청결도 평가)

  • The cleanness of Korean(n=292), Chinese(n=46), Japanese(n=114) and Western(n=74) styles restaurants in 25 districts in Seoul was evaluated in terms of kitchen area(6 criteria), handling of raw materials(3 criteria), hygiene practice for cook(1 criteria), treatment of wastes(2 criteria) and hall(2 criteria). The western-style restaurants were scored the highest points in all criteria. The other three restaurants were poorly evaluated compared with western-style ones in kitchen area, including cleanness of inside of the kitchen, hygiene practice, ventilation and working environment, drainage, and storage of utensils. The most critical criterion which represents the overall evaluation was the ventilation and working environment of the kitchen for Chinese, Japanese and Western-style restaurants, and was the cleanness of inside the kitchen for Korean-style restaurants.

Quality Characteristics of Press Ham Containing Conjugated Linoleic Acid-Triglyceride (Conjugated linoleic acid-triglyceride가 함유된 프레스햄 품질특성)

  • CLA was chemically synthesized by alkaline isomerization method using corn oil. CLA-TG was synthesized by chemical reaction using sodium methoxide. For the control, 10% of back fat among the total component was only added without the annex of CLA-TG. For the first treatment, 5% of CLA-TG among the lard component added into the press ham was replaced. For the 2nd, 3rd and 4rd treatments, 10%, 15% and 20% of CLA-TG was respectively replaced. Manufacture press ham using CLA-TG were vacuum packaged and then stored during 1, 7, 14, 21 and 28 days at 4℃. Samples were analyzed for shear force value, sensory evaluation, TBARS, fatty acid composition and CLA content. Shear force value of control was significantly higher than that of CLA-TG treatment groups(P<0.05). All treatments were increased by the passage of storage time. No remarkable differences were found in sensory properties among control and CLA-TG treatment groups. CLA-TG treatment groups showed significantly(P<0.05) lower TBARS value than the control. TBARS value was increased significantly during storage in all treatment. In the change of fatty acid composition, the contents of C14:0~C20:4 were decreased significantly by CLA-TG additive. Whereas the increase level of CLA-TG additive resulted in the significantly higher unsaturated fatty acid and CLA content. Summing up the a forementioned results, press ham manufacturing with CLA-TG additive was not affected in sensory evaluation. Also, it may be assumed that the high quality press ham can be manufactured with the extent of storage period and CLA accumulation.
