The Variation of Oxygen and Hydrogen Isotopic Composition in Precipitation and Geothermal Waters from the Yuseong Catchment (유성 소유역의 강수 및 지열수의 산소.수소 안정동위원소 조성 변화)

  • The oxygen and hydrogen isotopic composition in precipitation, groundwater and geothermal water were monitored over four-year period from 2001 to 2004 at the Yuseong catchment, Daejeon. By analyzing the long term rainfall pattern, we found out the drought cycle of 6 or 7 year. We fortunately revealed that the oxygen and hydrogen isotopic composition of rain has progressively changed to heavier isotopic ratios from 2001 to 2004. The weighted mean values of ${\delta}^{18}O\;and\;{\delta}D$ of rain are calculated to be $-7.7%o\;and\;-51%o$, respectively. These isotopic values are much heavier than those of groundwater and geothermal water collected at the same period, which indicates that the rain or snow of the study area would not immediately affect the isotopic composition of groundwater or geothermal water. Comparing with the previous data, the groundwater and geothermal water collected at 1990 and 1992 year has the heaviest isotopic composition and afterwards their isotopic composition has been progressively shifted to the direction of lighter composition field.

Effects of Supplementation of Ruminally Protected Amino Acids on In vitro Ruminal Parameters and Milk Yield and Milk Composition of Dairy Cows in Mid-lactation (보호아미노산의 추가 공급이 반추위 발효성상 및 비유중기 착유우의 유량 및 유성분에 미치는 영향)

  • This study was undertaken to investigate the effects of ruminally protected amino acids (Methionine and Lysine) on in vitro ruminal parameters, and in vivo milk yield and milk composition in mid-lactating cows. In the first in vitro experiment, there were no statistical significances between treatments in ruminal pH and dry matter digestibility during various incubation times. In the second in vivo experiment, milk yield decreased by 11.92% in control and 5.68% in the treatment respectively, but decrease rate of milk yield in the treatment was lower than control. Milk yields naturally decreased as time goes by since the DIMs(Days in milk) of the cows in experiment were in mid-lactation period. 4% FCM(Fat corrected milk) and milk protein yields also, respectively, decreased by 11.25% and 11.09% in control and 6.16% and 5.47% in the treatment as compared with the intial. Milk protein and milk fat production were higher in the treatment(0.90kg, 1.10kg) than those of control(0.66kg, 0.79kg). Milk fat content significantly increased with supplementing protected amino acids as compared to control(P<0.05). From the above results, protected amino acids were positively utilized in the performances of mid-lactating cows without inhibiting rumen fermentation. Further investigation is suggested for essential amino acid composition and intestinal digestion rate out of rumen bypass protein in dietary protein to be estimated.

Distribution and Alteration of Mating Type of Phytophthora capsici Population from Red Pepper in Korea (국내 고추역병균 Phytophthora capsici 집단의 교배형 분포 및 변화)

  • Distribution and alteration of mating type of Korean population for Phytophthora capsici were monitored from 1995 to 1998. Total 973 isolates collected from 75 pepper fields throughout the country were divided into A1 mating type 573 isolates and A2 mating type 400 isolates. Both mating types were founded in all provinces examined, but the distribution patterns differed between the fields, The single mating type of A1 or A2 isolates was found in some fields, whereas various combinations of both mating types were in all years and provinces surveyed. Ratios of A1 and A2 were varied with years and fields. However, only single mating type either A1 or A2 was detected in some fields. While, in another field located in Gongju, only A2 isolates were detected in 1995 and 1997, but A1 isolates were appeared in 1998. Varied ratios and alterations of A1 and A2 mating type in fields indicate a potential sexual recombination of the fungus, which may cause genetic diversity.

  • At fertilization, sperm penetrates into oocyte, male and female pronuclei are fused together, and mitotic division follows. However, little information is available on the interactive roles and dynamic processes between cytoplasmic and nuclear components during the pronuclear formation, migration and cell division. The assisted reproductive technologies such as, intracytoplasmic sperm injection (ICSI) and round spermatid injection(ROSI) could provides new treatments for the male infertility as well as tools for the study of basic mechanism during fertilization. Nuclear transfer can also provide a mechanism on the interactive roles between nucleus and cytoplasm since the process includes nuclear reprogrammming of differentiated cells in the enucleated oocytes. Recently, I have investigated developmental processes in porcine oocytes following fertilization parthenogenesis, ICSI, ROSI and nuclear transfer using indirect immunocytochemical and electron microscopic studies. The results could provide an insight into biological questions related with epigenesis as well as strategies for the enhancement of embryology in general such as ICSI and nuclear transfer.

  • In order to apply thermoplastic composites using PETG resin to various industrial fields such as bicycle frames and industrial parts, it is necessary to verify the impact resistance, durability, mechanical properties and 3D analysis of the manufactured composite materials. To improve the mechanical properties, durability and impact resistance of PETG resin, an amorphous resin, in this study, compound and injection molding process were carried out enhanced various weight percent POE(polyolefin elastomer). The thermal and mechanical properties of the thermoplastic composites, and the charpy impact strength, The analysis was performed to evaluate the characteristics according to weight percent of POE. Charpy impact strength test was conducted to analyze the impact characteristics, and the fracture section was analyzed after the impact strength test. In the case of POE material-added thermoplastic composites, thermal and mechanical properties tend to decrease, but workability and impact resistance tend to be superior to those of PETG materials.

Genetic diversity and population structure of Atractylodes japonica $K_{OIDZ}.$ in Korea (한국내 삽주의 유전적 다양성과 집단구조)

  • The study of genetic diversity was carried out in Atractylodes japonica $K_{OIDZ}$. Although this species has been regarded as medically important one, there is no report on population structure in Korea. Starch gel electrophoresis was used to investigate the allozyme variation and genetic structure of eight Korean populations of this species. Of the 15 genetic loci surveyed, nine (60.0%) was polymorphic in at least one population. Genetic diversity was high at the species level $(H_{es}=0.144)$, whereas, that of the population level was relatively low $(H_{ep}=0.128)$. Nearly 87% of the total genetic diversity in A. japonica was apportioned within populations. The sexual reproduction, high fecundity, and perennials are proposed as possible factors contributing to high genetic diversity. The indirect estimated of gene flow based on Gst was 1.69.

THE EARLY EFFECTS OF SODIUM FLUORIDE ON THE RAPID PALATAL EXPANSION IN GROWING DOGS (유성견의 정중구개봉합 급속확대시 투여된 불화나트륨의 초기 효과에 관한 연구)

  • The purpose of this study was to observe the effects of sodium fluoride on the bony repair and regeneration processes after the rapid palatal expansion in the growing dogs. Eighteen dogs were divided into experimental and control groups. They were in the late mixed dentition. The rapid Palatal expansion was undertaken in all the animals($180^{\circ}$ turn/day) for ten days. The animals were sacrificed on 0, 15 and 45 days after the finish of expansion. One mg NaF/kg of body weight/day were given orally to the experimental group. Blood samples were drawn before and after expansion and the se겨m calcium, phosphate and alkaline phosphatase level were measured. The undecalcified bone section of midpalatal suture area was made, and observed under the light microscopy The results were as follows ; 1. The day after expansion, the infiltration of inflammatory cells were prominent and the new bone formation started at the edges of the two palatal plates bodering the midpalatal suture in both groups. Especially, the newly formed osteoid were very extensive and the osteoblasts lining the osteoid were very active in the experimental group. 2. At fifteen days after expansion, the active osteoblasts lining the osteold at the surface of trabecular bony spicules and active new bone formation were observed in the both groups. However, the cellular activity and new bone formation were more prominent In the experimental group. 3. At forty five days after expansion, the continuous osteoid and new bone formation and active osteoblasts were observed in the experimental group. But these phenomena were not observed in the control group. In the control group, the numerous osteoclasts were adjacent midpalatal suture and the bony remodeling process was begun. The serum alkaline phosphatase level was maintained highly in the experimental group, but decreased in the control. According to the above results, the author reached the conclusion that sodium fluoride has the stimulation effects on the osteoid production of the osteoblasts during the healing process after the rapid Palatal expansion more continuously.

  • A numerical analysis for temperature distribution of four different materials such as iron, silicon, aluminum and PVC has been performed in this study to predict thermal behaviors of combined weapon systems in a large environmental tester. Thus, experimental conditions have been proposed using a calculating software (SolidWorks 2007 COSMOS FloWorks) to prepare for field tests and analyze heat flow inside the environmental tester and temperature distributions of materials. The boundary conditions of the analysis are composed of inlet and outlet conditions of the environmental tester with different pressures and the limit of low temperature of -30$^{\circ}C$. The soaking time of the system in the environmental tester has been calculated by this commercial program in this study to carry out the experiment.

In silico Analysis of PERVs Based on the Porcine Genomic Sequence Information (돼지 유전체 염기서열을 이용한 내인성 리트로 바이러스 분석에 관한 연구)

  • This study was conducted to identify the PERV (Porcine Endogenous Retrovirus) integration sites and their characterizations using the porcine genomic sequence information. Total 114 Mb (4.2%) sequence of the 2.7 Gb pig genome was investigated for the PERV sequences. As the results, 8 PERV sequences were identified and their genomic structures were deduced from the BLAST searches against previously known PERV genes. Seven PERVs have internal deletions in the protein coding region and they will not be functional. The other one also has internal deletions in the gag and env genes, indicating this PERV is also defective. Even though we could not identify the functional PERVs in this study, the results presented here can be used for the fundamental research materials for controlling PERV infections in relation to xenotransplantation using porcine organs and tissues.
