Diversity of Endophytic Fungi Isolated from Hydrophytes in Wetland of Nakdong River (낙동강 지류의 하천 습지에 자생하는 수생식물에서 분리된 내생균류의 다양성)

  • Endophytic fungal strains were isolated from 5 aquatic plant species (Trapa japonica, Miscanthus sacchariflorus, Oenanthe javanica, Typha orientalis and Zizania latifolia) native to river wetland of tributary in Nakdong river. Total 34 strains were purely isolated, and then its internal transcribed spacer (ITS) regions were amplified. After that, phylogenetic analysis based on ITS sequences and deduction of diversity indices were done. Fungal isolates were belonged to 17 genera, concretely in Acremonium, Alternaria, Aspergillus, Cladosporium, Emericellopsis, Fusarium, Galactomyces, Leptosphaeria, Microsphaeropsis, Penicillium, Peyronellaea, Phoma, Pseudeurotium, Rhizomucor, Talaromyces, Trematosphaeria and Zalerion. Especially, fungal isolates were distributed intensively in genera Alternaria and Talaromyces. This study deals with the diversity of endophytic fungal species that showing symbiotic relationship with their host aquatic plants.

Detection of Endolichenic Fungi Producing Antifungal Compound (항진균성 물질을 생산하는 지의류 내생 곰팡이의 탐색)

  • To isolate a novel antifungal compound, we obtained 100 kinds of endolichenic fungi from Korean Lichen & Allied Bioresources Center and examined their antifungal capability. Three fungi Usnea rigidula (2326), Parmotrema pseudotinctorum (2202) and Myelochroa sp. (2292) showed high antifungal activity against Candida albicans when they grew in both liquid and solid media. We extracted the culture supernatants of these three fungi with chloroform and then with ethyl acetate. Chloroform fraction exhibited the highest antifungal activities when those fractions were examined for the growth inhibition of Candida albicans with disc diffusion method. The chloroform faction was on further analysis with $C_{18}$ column chromatography to see whether the inhibitors are already known or not. Two peak fractions were collected from 4-day culture extract for Usnea rigidula and from 6-day culture extract for Parmotrema pseudotinctorum on the HPLC. A peak fraction from chloroform extracts of 4-day culture filtrate of Parmotrema pseudotinctorum showed higher antifungal activities against C. albicans and C. glabrata than another peak fraction. It appears that the antifungal materials are relatively nonpolar as usnic acid often found in lichenic fungi.

  • Sixty five soil specimens were collected from the nineteen leguminous plant roots throughout 9 sites in four Provinces from Augast 12 to November 21, 1991. VA-mycorrhizal spores isolated from the collected soils were identified and classified into four genera 21 species. However, six spores were newly found and described here: Acaulospora elegans, A. undulata, Glomus laminated spores of. macrocarpus var. macrocarpus, Gl. WUMI%3, Gl. scintillans, Scutellospora verrucosa. Species of three spores that belong to 3 genera; Acaulospora sp., Glomus sp., Scutellospora sp. were unidentified.

Genetic Diversity of Endophytic Fungi Isolated from the Roots of Halophytes Naturally Growing in Suncheon Bay (순천만에 자생하는 염생식물의 뿌리로부터 분리된 내생진균의 유전적 다양성)

  • Endophytic fungi were isolated from the roots of halophytes, Suaeda japonica and Carex scabrifolia in the Suncheonbay. The ITS region in rDNA of 15 endophytic fungal strains were amplified using PCR with universal primers ITS1 and ITS4, and those amplified fragments were sequenced. Based on ITS sequence, five fungal genera were identified in S. japonica and seven fungal genera were identified in C. scabrifolia. The Shannon's diversity index (H') of endophytic fungi isolated from S. japonica and C. scabrifolia was 1.561 and 1.889, respectively. In phylogenetic analysis, it was shown that Ascomycota and Pezizomycotina was widely distributed both in S. japonica and C. scabrifolia. Also, Sordariomycetes, Dothideomycetes and Eurotiomycetes were shown to be distributed in these halophytes used in this experiment.

Plant Growth-Promoting Activity and Genetic Diversity of Endophytic Fungi Isolated from Native Plants in Dokdo Islands for Restoration of a Coastal Ecosystem (해안 생태계의 복원을 위하여 독도에 자생하는 식물로부터 분리된 내생진균류의 식물생장촉진활성과 유전학적 다양성)

  • In this study, plant samples of five species were collected from the Dokdo islands in South Korea. Plant samples such as Asparagus schoberioides, Corydalis platycarpa, Festuca rubra, Sedum oryzifolium, and Setaria viridis were collected from the Dongdo and Seodo. Endophytic fungal strains were isolated from the roots of five plants from the Dokdo islands. Thirty-three fungal strains were isolated from these native plants. All the endophytic fungi were analyzed by internal transcribed spacer (ITS) sequencing (ITS containing ITS1, 5.8s, and the ITS2 region). Waito-c rice seedlings were treated with fungal culture filtrates to test their plant growth-promoting activity. A bioassay of the D-So-1-1 fungal strain isolated from S. oryzifolium confirmed that it has the highest plant growth-promoting activity. All the endophytic fungi belong to four orders: Eurotiales (86%), Capnodiales (3%), Hypocreales (4%), and Incertae sedis (7%). The endophytic fungi were classified as Ascomycota, which contained Aspergillus (12%), Cladosporium (3%), Eurotium (3%), Fusarium (18%), Microsphaeropsis (6%), and Penicillium (58%) at the genus level.

Secondary Metabolites Produced by an Endophytic Fungus Arthrinium sp. JS420 of Halophytic Plant Suaeda japonica (칠면초 유래 내생균(Arthrinium sp. JS420)이 생산하는 이차대사산물 규명)

  • Four polyketides, tyrosol (1), (3R,4R)-(-)-4-hydroxymellein (2), (3R,4S)-(-)-4-hydroxymellein (3), and 1-phenyl-1,2-ethanediol (4) were isolated from organic extracts of cultures of an endophytic fungus Arthrinium sp. (JS420) isolated from stem of a halophyte Suaeda japonica Makico. Chemical structures of the isolated compounds were elucidated by comparison of their spectral data such as NMR and ESIMS with reported literature values. Among the isolated compounds, 3 and 4 were isolated for the first time from this fungus.

  • Endophytic fungi have yielded a variety of secondary metabolites so far. In the course of the project to find bioactive secondary metabolites from cultures of endophytic fungi, an isolate of Arthrinium phaeospermum (JS 0567) was selected for chemical investigation. A large scale culture of this strain in rice media was extracted with an organic solvent and the extract was subjected to a serious of chromatography, which led to six metabolites. Their chemical structures were elucidated as 2,3,6,8-tetrahydroxy-1-methylxanthone(1), 2,3,4,6,8-pentahydroxy-1-methylxanthone(2), 3,4,6,8-tetrahydroxy-1-methylxanthone(3), 3,6,8-trihydroxy-1-methylxanthone(4), 2,4,2',4',6'-pentahydroxy-6-methylbenzophenone(5), and 5,7-di hydroxy-3-methylphthalide(6) on the basis of spectroscopic data. To the best of our knowledge, this is the first study on the secondary metabolites from Arthrinium phaeospermum.

  • Vesicular-arbuscular mycorrhizal fungi were isolated from the soils of three different plant communities (Cassia mimsoides var. nomame, Themeda triandra var. japonica, Miscanthus sinensis var. purpurascens). Eight azygospores or chlamydospores of VA-mycorrhizal fungi were identified (eight species of four genera), and six among them newly described in Korea (six species of two genera); Glomus aggregatum, Gl. ambisporum, Gl. geosporum, Gl. mosseae, Scutellospora coralloidea, Sc. heterogama.
