Three Novel Endophytic Fungal Species Isolated from Roots of Medicinal Crops in Korea (약용작물의 뿌리에서 분리된 3종의 국내 미기록 내생균)

  • We isolated endophytic fungal strains from the roots of three medicinal crops, Ligusticum chuanxiong, Angelica gigas, and Cnidium officinale, cultivated at Yeongju, Korea. The fungal strains were identified based on their morphological characteristics and molecular analyses of their internal transcribed spacer, large subunit rDNA, and beta-tubulin regions. Thereby, we identified three previously unrecorded endophytic fungal species, Dactylonectria pauciseptata, Rhizopycnis vagum, and Sistotrema sernanderi, in Korea. In this report, we describe the morphological characteristics and phylogenetic analyses of these three fungal species.

Unrecorded Endophytic Fungi Belonging to Genus Phyllosticta Isolated from Leaves of Woody Plants (목본식물 잎에서 분리된 Phyllosticta 속의 국내 미기록종 내생균)

  • We isolated endophytic fungi from Smilax china and Cryptomeria japonica. These fungal strains were identified based on their morphological characteristics and phylogenetic analyses of their internal transcribed spacer, large subunit rDNA, and translation elongation factor 1-α DNA. Among them, we discovered two fungi belonging to the genus Phyllosticta, which have not been previously recorded in Korea. We have described these two fungal strains, Phyllosticta ericarum 19E458 and Phyllosticta philoprina 19E012 in this study.

  • We isolated endophytic fungal strains from roots of Rhododendron schlippenbachii growing in Minjujisan mountain, Korea. The two strains were identified as Engyodontium album and Phaeosphaeria microscopica based on their morphological characteristics and sequence analysis of the internal transcribed spacer (ITS) and large subunit regions of the ribosomal DNA. These two species, E. album and P. microscopica, have not been previously reported in Korea.

  • We isolated endophytic fungi from the conifer leaves of Pinus densiflora inhabiting Buan-gun, Jeollabuk-do, Korea. We identified the isolated fungal strains based on phylogenetic analysis performed using the nucleotide sequences of the internal transcribed spacer, large subunit, and beta-tubulin. We confirmed the presence of three novel endophytic fungi in Korea, namely Paracamarosporium hawaiiense, Tubakia dryina, and Zasmidium fructigenum. In this report, we described the morphological characteristics of these fungal strains and the results of their phylogenetic analysis.

  • Diverse endophytic fungi were isolated from surface-sterilized leaves of three species of conifers inhabiting various sites in Korea: Abies nephrolepis, Pinus koraiensis and Taxus cuspidate. The isolates were identified based on morphological characteristics and sequences analysis of both internal transcribed spacer and large subunit regions of rDNA. In this paper, we report on five previously unreported species of endophytic fungi isolated from conifers: Biscogniauxia maritime, Nemania diffusa, Pezicula carpinea, Phomopsis juglandina and Sydowia polyspora.

Diversity of Fungal Endophytes from Pinus koraiensis Leaves in Korea (한국의 잣나무에서 분리한 내생균의 다양성)

  • The composition of endophytic fungal species from Pinus koraiensis was studied in two areas (Yeongwol and Jincheon), Korea. To identify 113 isolates, rDNA ITS regions were sequenced. On the basis of the ITS sequence, Pestalotiopsis sp. was the most frequently isolated species in both areas. Pestalotiopsis sp. (68.5%) and Lambertella sp. (12.4%) were isolated frequently in Yeongwol, whereas Pestalotiopsis sp. (41.7%), Hypoxylon sp. (20.8%) and Phomopsis sp. (12.4%) were dominant in Jincheon. Hypoxylon sp. and Phomopsis sp. were not found in Yeongwol.

  • Endophytic fungi were isolated from surface-sterilized roots of a species of orchid, Oreorchis patens, in Korea. The isolated strains were identified based on morphological characteristics and sequence analyses of the internal transcribed spacer (ITS) and large subunit (LSU) of rDNA regions. Two species, Thelonectria veuillotiana and Phialocephala humicola, are first reported in Korea with their descriptions.

  • In this study, endophytic fungi were isolated from the surface-sterilized roots of Calanthe discolor and Cephalanthera longibracteata collected from the Chungnam, Jeju, Kyungnam and Chungbuk provinces in Korea. The morphological characteristics of the obtained isolates were examined and their sequences of the internal transcribed spacer (ITS) rDNA region were analyzed using the ITS1F and ITS4 primers for species identification. Leptodontidium orchidcola showed the highest species abundance and frequency among the isolated endophytic fungi. Additionally, the community analysis revealed a high specificity between the host plants and the endophytic fungal species.

  • Diversity of endophytic fungi of two species associated with host plants, Pinus densiflora and Larix kaempferi in Mt. Oser were investigated. Endophytic fungi were isolated from surface sterilized leaves of the 13 host plants and identified by morphological traits and rDNA ITS sequences analysis. Totally 37 isolates were discovered and identified into 17 taxa. Of them, 59% isolates belonged to Leotiomycetes, 30% isolates Sordariomyetes, 8% isolates Dothideomycetes and 3% isolates Agaricomycetes. Results showed a similar diversity pattern with previous studies. Also, species diversity (H') of endophytic fungi isolated from L. kaempferi was higher than that from P. densiflora. The endophytic species composition showed significant differences between host plants. Particularly, taxon of Lophodermium is a major genus in the host plants. However, critical research of the endophytic species diversities within the genus in Korea is required. The communities of endophytic fungi isolated in this study showed differences in diversities and species composition between host plants.

Plant Growth-Promoting Activity of Endophytic Fungi Isolated from the Roots of Native Plants in Dokdo Islands (독도 자생식물의 뿌리로부터 분리된 내생균의 식물생장촉진 활성)

  • Endophytic fungal strains were isolated from the roots of six species plants in the Dokdo islands. Native plant samples, such as Artemisia japonica, Chenopodium album and Solanum nigrum were isolated from Dongdo, and those such as Cyrtomium falcatum, Dianthus longicalyx and Tetragonia tetragonoides were isolated from Seodo. In total, thirty two fungal strains were isolated from these native plants. To identify the fungal strains, polymerase chain reaction (PCR) amplification of internal transcribed spacer (ITS: containing ITS1, 5.8s and ITS2 region) regions was done with universal primers ITS1 and ITS4. Endophytic fungi of four species were isolated from A. japonica, eight species from C. album, three species from S. nigrum, three species from C. falcatum, three species from D. longicalyx and eleven species from T. tetragonoides. Culture filtrates (CF) of isolated endophytic fungi were used to treatwaito-c rice seedlings to test plant growth-promoting activity. As a result of bioassay, Ca-5-2-2 strain isolated from C. album expressed highest plant growth-promotion activity. Of all the endophytic fungi isolated, Penicillium sp., Fusarium sp. and Aspergillus sp. were the most abundantly distributed fungal strains in the six plants used in this study.
