Effects of Inoculation with Mycorrhizal Fungi, Pisolithus tinctorius and Glomus sp. on the Rooting of Quercus acutissima Carr. Cuttings at Various Ortet Ages (모래밭 버섯균(菌)과 Glomus 균근균(菌根菌)의 인공접종(人工接種)이 연령(年齡)이 다른 상수리나무에서 채취(採取)한 삽수(揷穗)의 삽목발근(揷木發根)에 미치는 영향(影響))

  • This study was conducted to determine the effects of mycorrhizal inoculation and ortet ages on the rooting of Quercus acutissima cuttings. The cuttings taken from 12-week-, 2-year-, 4-year-, and 20-year-old plants were rooted in mid summer in the rooting medium (vermiculite 2 : peatmoss 1 by volume) with or without Pisolithus tinctorius (Pt) and Glomus sp. inocula and IBA under intermittent misting system in an open shed. The average percentages of rooting were about 82%, 49%, 29%, and 13% for cuttings taken from 12-week-, 2-year-. 4-year-, and 20-year-old seedlings, respectively. Pt inoculation, enhanced rooting of cuttings at all age classes, except 12-week-old seedlings, with the highest enhancement(22%) observed in cuttings taken from 20-year-old trees. The highest percentage of rooting in each age group eras 88.9% in 12-week-old seedlings treated with Pt plus 3.000ppm IBA, 75% in 2-Year-old plants with 1,000ppm IBA, 58.3% in 4-year-old plants with 3.000ppm IBA and 22% in 20-year-old plants. The addition of Glomus sp. fungus inoculum failed to enhance rooting. Pt mycorrhizal inoculation enhanced root dry weight, length, and diamter of adventitious roots at cuttings taken from 12-week- and 20-year-old trees, except the cuttings taken from 4-year-old seedlings. Rooted cuttings had more total nitrogen content in the leaves than unrooted cuttings, and the greater rooting response was associated with the higher phosphorus content in the leaves.

  • This experiment was conducted to determine the safety and feasibility of using compost-promoting-bacteria. Compost-promoting-bacteria was isolated from livestock compost containing sawdust. The isolated bacteria was identified as Bacillus subtilis LYH201 by the method of the composition of the fatty acid with MIDI system and Bergey's manual. This Bacillus subtilis LYH201 had the following characteristics : Gram-positive, straight rod ($0.5{\sim}0.7{\mu}m$ width, $2.5{\sim}3.0{\mu}m$ length), facultatively aerobic and product of xylanase, CMCase, catalase, oxidase, protease and $0.5{\sim}0.7{\mu}m$-amylase. Growth of Bacillus subtilis LYH201 at saccharose as carbon source(0.5%) was faster than other carbon source. Activity of cellulase. $0.5{\sim}0.7{\mu}m$-amylase and protease from Bacillus subtilis LYH201 after 24 hours at $50^{\circ}C$ by agar diffusion method was higher than that of low temperature. Optimum growth condition of Bacillus subtilis LYH201 was $50^{\circ}C$ and pH 6.

  • Microscopic observations of arbuscular mycorrhizae (AM) were done with the colonizations of AM and the determinations of chitin in the plant roots of Sorghum bicolor, Cassia mimosoides, Capsicum annuum and Allium fistulosum. The intercellular and intracellular hyphae, arbuscules and vesicles were microscopically observed, according to increases of colonization of AM in the roots of four plants.   • This study was attempted to isolate and identify the strain revealing high angiotensin converting enzyme (ACE) inhibitory activity from various soy fermented foods, i.e. meju, soybean paste and soy sauce. Forty-two strains with morphologically different characteristics were selected and the ACE inhibitory and proteolytic activities were examined. Of the strains tested, SS103 which was isolated from soy sauce showed the highest ACE inhibitory and proteolytic activities and was finally selected for further studies. The SS103 strain showed motility, rod form and ellipsoidal spores. The shape of colonies on the agar media was irregular, mucoidal and surface dull. The strain could grow under aerobic conditions of pH 5~9 and 10~$50^{\circ}C$. Main cellular fatty acid was $C_{15:0}$ anteiso, $C_{17:0}$ cis and $C_{17:0}$ iso, which was 33.9%, 18.8% and 16.5%, respectively. Based upon these morphological, biochemical and cultural properties, SS103 was identified as a Bacillus subtilis. Optimum cultural condition of Bacillus subtilis SS103 was pH 8.0, $37^{\circ}C$ and 48 hr.48 hr.

Assessment of Probiotic Potential of Bacillus spp. Isolated from Ulleungdo, Korea (울릉도 토양에서 분리한 Bacillus 속 균주의 프로바이오틱 잠재성 평가)

  • Probiotics have been isolated from various environments and Bacillus species are advantageous among the probiotic bacteria due to their ability to form endospores that can compensate for the limitation of typical probiotics such as lactic acid bacteria. The aim of this study is to investigate the probiotic potential of Bacillus species from Ulleungdo soil, known as an unpolluted environment in Korea. Soil samples were collected from various areas of Ulleungdo, and Bacillus spp. were isolated, and assessed for antibiotic resistance and enzymatic activity. Six Bacillus spp. were not resistant to all tested antibiotics and subsequently tested for enzyme activity. We found the six Bacillus spp. were all inactive β-glucuronidase enzyme, which can have detrimental effects on human health, and one of Bacillus spp. showed an activity of Leucine arylamidase suggesting its probiotic potential.

  • Five sporocarpic species of Glomus were found at the four sites of the different plant's cultivations in Korea ; G. clavisporum, G. liquidambaris, G. rubiforme, G. sinuosum, and G. taiwanense. The morphological characteristics of these species collected were observed and compared under light microscopes. Three species G. clavisporum, G. liquidambaris and G. taiwanense were similar with each other in the aspects of having no peridium and the clavate spores orderly arranged within sporocarps. G. sinuosum was different from the other in the aspects of having a thick and sinuous peridium and the orderly arranged spores within sporocarps. However G. rubtforme was different from the other four species in aspects of sporocarps haying ovoid spores arranged irregulary.

Xylanase-producing Bacillus subtilis isolated from spent mushroom (Pleurotus eryngii) substrates (큰느타리버섯 폐배지에서 분리한 Xylanase 생성 Bacillus subtilis CS9)

  • A Gram-positive, endospore-forming, rod-shaped bacterial strain was isolated from spent mushroom (Pleurotus eryngii) substrates taken from the Hoamfarm located in Keyollgnam, Korea. The isolate, designated CS9, was facultatively anaerobic, motile rod and produced xylanase. The strain grew optimally at $40^{\circ}C$ and pH 6.0. The major cellular fatty acids were anteiso-$C_{15:0}$, anteiso-$C_{17:0}$, and iso-$C_{17:0}$. The genomic DNA G+C content was 45 mol%. Comparative 16S-rDNA sequence analysis showed that the isolate CS9 formed a distinct phyletic line within the genus Bacillus and was most closely related to Bacillus subtilis YB1, with 16S DNA sequence similarity of 96.8%. Sequence similarities to other type strains were 92-94%. On the based of physiological and molecular properties, the isolate CS9 was classified within the genus Bacillus as Bacillus subtilis CS9.

Isolation and Characterization of an Antagonistic Endophytic Bacterium Bacillus velezensis CB3 the Control of Citrus Green Mold Pathogen Penicillium digitatum (감귤저장병 병원균 Penicillium digitatum 방제를 위한 길항 내생세균 Bacillus velezensis CB3의 분리 및 특성 규명)

  • In order to develop environment friendly fungicide for the control of citrus green mold (Penicillium digitatum) using endophytic bacteria, the 21 bacterial isolates were isolated from citrus leaves in seven different orchards in Jeju Province. Among the 21 bacterial isolates, 5 bacterial isolates presented antifungal activity against green mold pathogen P. digitatum. The CB3 isolate, which showed the most strong antagonistic effect, was selected through opposite culture against the pathogen. The rod-shaped, gram-positive bacterium CB3 was identified as Bacillus velezensis based on morphological, physiological characteristics, 16S rDNA, and gyr A gene sequence analysis. The isolate CB3 showed strong antifungal activity against two citrus postharvest pathogen P. digitatum. Citrus fruits were treated by wound inoculation with P. digitatum pathogen, and the control efficacy of CB3 culture broth was 66.7% ($1{\times}10^8$ cfu/ml). In conclusion, The stability of CB3 and its strong antifungal activity also lead us to believe that it has potential for application as an environment friendly biological control agent.

Identification and Characterization of an Endophytic Strain of Streptomyces from Rice Roots (Orysa sativa L.) (벼(Orysa sativa L.) 뿌리로부터 분리된 내생 Streptomyces 균주의 동정 및 특성)

  • We isolated an endophytic actionmycete from root tissues of rice plant collected from paddy field near Dankook University, Cheonan, Korea. Surface sterilized roots were laid on the selective agar plates and incubated. The powdery actinomycete colonies appeared on the root surface after four weeks incubation. We isolated a strain JK-5 among them and could determine its taxonomical position as Streptomyces diastaticus subsp. ardesiacus by using 16S ribosomal DNA sequencing. The chemotaxonomical and morphological studies confirmed the taxonomical position of the strain JK-5. The shape of aerial hyphae was flexible and they contained spore chains with more than 30 smooth spherical spores per chain. Cell walls contained LL-diaminopimelic acid. There was no characteristic sugar in whole-cell hydrolysates. The major fatty acids were anteiso-15:0, anteiso-17:0 and iso-16:0. The specific menaquinones, MK-9 ($H_6$), MK-9 ($H_8$), were detected. The GC content was 72%. Antifungal activities of the strain JK-5 were relatively strong against fungal plant pathogens. The endophytic growth of the strain JK-5 was confirmed by SEM observation of the root and stem of the infected rice plant.

Entomopathogenic Fungi-mediated Pest Management and R&D Strategy (곤충병원성 진균을 활용한 해충 관리와 개발 전략)

  • Entomopathogenic fungi can be used to control a variety of sucking and chewing insects, with little effect on beneficial insects and natural enemies. Approximately 170 entomopathogenic fungal insecticides have been registered and used worldwide, with the recent focus being on the mode of action and mechanism of insect-fungal interactions. During the initial period of research and development, the industrialization of entomopathogenic fungi focused on the selection of strains with high virulence. However, improvement in productivity, including securing resistance to environmental stressors, is a major issue that needs to be solved. Although conidia are the primary application propagules, efforts are being made to overcome the limitations of blastospores to improve the economic feasibility of the production procedure. Fungal transformation is also being conducted to enhance insecticidal activity, and molecular biology is being used to investigate functions of various genes. In the fungi-based pest management market, global companies are setting up cooperative platforms with specialized biological companies in the form of M&As or partnerships with the aim of implementing a tank-mix strategy by combining chemical pesticides and entomopathogenic fungi. In this regard, understanding insect ecology in the field helps in providing more effective fungal applications in pest management, which can be used complementary to chemicals. In the future, when fungal applications are combined with digital farming technology, above-ground applications to control leaf-dwelling pests will be more effective. Therefore, for practical industrialization, it is necessary to secure clear research data on intellectual property rights.
