This study presents the effects of micro-geometries on the swimming behavior of Pseudomonas aeruginosa. First, we have measured parameters of single-cell motility including cell speed, run duration time, and tumble angle under two dimensional space. The results are used to calculate motility coefficients in the width of microchannels ranging from 10 to $100{\mu}m$. Since the single-cell motility parameters measured depend on the interaction of flagella with the microchannel wall, the duration time of the running cell in restricted geometries is distinctively different. Therefore, the motility of bacteria is decreased by restricted geometries. This study suggests that microfluidic approach is useful tool for the analysis of bacterial motility under the restricted space and rapid analytical tool.

광양만내 해양바이러스양은 2.0$\times$$10^{8}$ particles/ml로 매우 풍부했다. 각 계절별 바이러스의 밀도는 여름에 최대 9.0$\times$$10^{8}$ particles/ml에서 겨울에는 최소값인 0.7$\times$$10^{8}$ particles/ml을 기록했다. 광양만 내의 바이러스, 박테리아, 식물플랑크톤 생물량의 공간적 분포는 open ocean system에 해당하는 정점 28, 38, 42, 46, 51에서 보다 closed estuarine system에 해당하는 정점 2, 5, 10 ,12, 16, 20에서 많은 것으로 나타났다. 그리고, 다른 closed estuarine system에 해당하는 정점 22, 26, 32, 34는 높은 바이러스 밀도를 보였지만 상대적으로 open ocean system에 비해서 낮은 박테리아와 식물플랑크톤의 생물량을 나타냈다. 몇몇 정점의 수심 깊이에서는 박테리아의 밀도가 바이러스의 밀도를 100배정도 초과했다. 해양 바이러스와 그들의 host system의 계절별 밀도는 역동적으로 번화했으며, 그들의 계절별 변화는 밀집한 상호연관성을 가졌다. 여름에 바이러스와 박레리아의 밀도는 증가된 반면 식물플랑크톤의 엽록소-a 농도는 평균값을 유지하였다. 겨울에는 바이러스와 박테리아의 밀도가 급속하게 줄어들었고, 마찬가지로 엽록소-a 농도 역시 감소하다가, 곧 다시 증가했다. 바이러스의 밀도는 2001년 8월에 최고점에 도달했으며, 박테리아의 밀도는 2001년 8월과 2002년 6월에 최고값을 가졌다. 반면에 엽록소-a의 농도는 2002년 4월에 최대치에 도달했다. 전체 host와 바이러스 밀도로 볼 때, 그들의 pool은 바이러스에 의한 mortality에 의해서 steady-state 상태로 유지되고, 바이러스의 밀도 또한 steady-state를 유지하는 것으로 보였다. 이러한 시도는 우리 나라 광양만 내에 존재하는 해양 바이러스의 생태적 분포 연구를 다루는 첫 번째 실험으로 사료된다.

To deal with issues from global climate changes, renewable bioenergy has become important. Algae have been regarded as a good resource for biorefinery and bioenergy, and also have potential capability to remove nutrient and non-decompositional pollutants for wastewater advanced treatment. Although algal-bacterial ecological interaction would be a crucially important factor in using algae for wastewater advanced treatment and resource recovery from wastewater, very little is known about ecological interaction between algae and bacteria in a real wastewater environment. In this study, under a real municipal wastewater condition, we characterized wastewater pollutant treatability and bacterial communities in response to growth of Ankistrodesmus gracilis SAG278-2, which can grow in wastewater and has a high lipid contents. The growth of algal population using the wastewater was inhibited by increase in wastewater bacteria while bacterial survival and cellular decay rate were not influenced by the algal growth. Removals of recalcitrant organic matters and total nitrogen were improved in the presence of algal growth. According to T-RFLP and statistical analysis, algal growth affected time-course changes in bacterial community structures. The following 16S rRNA gene amplicon, cloning results showed that the algal growth changes in bacterial community structure, and that bacterial populations belonging to Sediminibacterium, Sphingobacterium, Mucilaginibacter genera were identified as cooperative with the algal growth in the wastewater.

라오스의 Houay Pano 유역은 상업적 조림으로 인해 2011년부터 2013년까지 급속한 토지이용 변화를 겪어왔다. 본 연구는 이러한 토지이용변화가 박테리아 거동에 어떠한 영향을 주는지 이해하기 위해 Soil and Water Assessment Tool (SWAT) 모형을 활용한 박테리아 거동 모델링을 수행하였다. SWAT 모형은 수치 표고 모델, 토양 특성, 토지 이용 등의 정보를 종합하여, 유역 내수량 및 수질의 변화를 모의할 수 있는 모형으로, 본 연구는 대표적인 분원성 지표 세균 (Fecal Indicator Bacteria)인 대장균 (Escheichia coli, E. coli)을 대상으로 모델링을 수행하였다. SWAT 모형은 지표면 위 박테리아를 1)식물 위, 2)토양 용액상, 3)토양 입자상으로 구분하여 모의한다. 각 상태로 분할된 박테리아는 소멸 (die-off), 씻김 (wash-off), 침투, 표면 유출을 통한 수계로의 이동 등의 단계를 통하여 유역 내에서 거동한다. 본 연구는 서로 다른 기후의 영향을 배제하기 위해 각 토지이용 시나리오를 (2011, 2012, 2013) 실제 기후 조건과 동일 기후(2011-2013 평균) 조건으로 분류하여 분석하였다. 실제 기후 조건에서 SWAT 모형은 표면 유출, 토사 유출, E. coli 거동에 대해 2011년부터 2012년까지 감소, 2012년부터 2013년까지 증가로 모두 동일한 양상을 모의하였다. 이는 강수량의 양상과 동일한 것으로, 강수량이 표면 유출의 양을 결정하고, 달라진 표면 유출에 따라 토사 유출과 E. coli 거동이 결정되기 때문이다. 하지만 동일 기후 조건에서는, E. coli 거동 동인인 표면 유출과 토사 유출이 비교적 일정해짐에 따라, 각 상태로 분할된 박테리아의 초기 부하량값이 E. coli 거동을 결정하는 주된 요인임을 확인 할 수 있었다. 따라서 초기 부하량 분할에 활용되는 엽면적 지수 (Leaf Area Index)와 분배계수 (BACTKDDB)의 정확도가 요구된다. 추가적으로 본 연구는 박테리아의 유입원인 비료 모델링과, LAI를 활용한 박테리아 초기 부하량 산정, 토양 특성 변수와 토지 이용 변수의 분리, 지하수를 통한 박테리아 거동 등을 중심으로 SWAT 모형의 한계점과 개선점을 제시하였으며, 본 연구 결과는 토지이용변화가 박테리아 거동에 주는 영향을 모형적으로 이해하고, 또한 추후 박테리아 모델링 개발에 도움을 줄 것으로 예상된다.

When microorganism is injected into porous medium such as soils along with appropriate substrate and nutrients, biomass retained in the soil pore. Soil pore size and shape are varied from the initial condition as a result of biofilm formation, which make hydraulic conductivity reduced. In this research, hydraulic conductivity reduction was measured after microorganism are inoculated and cultured with synthetic substrates and nutrients. Biomass-soil mixture was evaluated its applicability to the field condition as an alternative liner material in landfill by measuring hydraulic conductivity change after repetitive freeze-thaw cycles. Resistance of biofilm to chemical solution and degree of biodegradation were measured through column test.
Photosynthetic bacteria, Rhodospirillum rubrum, have been reported to change their metabolic patterns depend-ing on the light condition. The genetic approach for such a metabolic change is one of main subject in pho-tosynthetic bacteria. It has been reported that the extrachromosomal plasmid might be related to this metabolic regulation. In this study, we have determined the partial sequences of R. rubrum plasmid pKYl with HindIII fragments and the predicted pKYl ORFs and physical map. We found the 8 putative proteins related to the genetic recombination of bacterium, which is reported to the alternative gene expression. Our results suggest that the genes located in pKYl are possibly involved in the metabolic switch according to the photocondition.

본 연구는 군부대 사격장의 중금속 오염토양에 대하여 생물학적 용출기술(BT)과 전기동력학적 기술(ET)의 통합공정의 적용성 평가 연구에 대한 것이다. 사격장 오염 토양의 경우 사격에 의해 탄두가 피탄지에 박히면서 오염토양 내에 잔존하여 탄두를 구성하는 주성분인 납과 구리 등에 의해 지속적인 오염원으로 작용하는 특징을 가진다. 따라서 사격장 토양오염정화를 위해서는 이 탄두를 물리적으로 선별하는 물리적 선별공정을 전처리공정으로 수행한 후 인공적으로 조성된 셀에 통합공정 적용성 평가를 위한 현장실증시험을 수행하였다. 생물학적 용출을 통해 토양내 잔류하는 중금속을 이온화시켜 이동성을 크게 한후 전기동력학적 기술을 통해 토양내에서 전해질로 이동시켜 최종적으로 전해질을 처리하는 시스템으로써 공정 모니터링결과 납과 구리 모두 주목할 만한 제거효율을 얻을수 있었다. 오염물질별 공정 적용성 평가결과 납의 경우 황산화박테리아에 의해 이온화가 되지만 황산화박테리아의 생장 부산물인 황산염이온(${SO_4}^{2-}$)과 반응하여 안정성이 큰 Anglesite($PbSO_4 $)를 형성하므로 전체적인 제거효율이 저하되는 것을 확인하였고 기타 미생물을 이용한 생물학적 용출기술 연구의 필요성을 확인하였다. 구리의 경우 황산염박테리아를 이용한 생물학적 용출공정 및 전기동력학적 처리공정의 통합공정을 통해 주목할 만한 제거효율을 얻을수 있었으며 통합공정의 효율성을 확인할 수 있었다. 본 연구를 통하여 미생물학적 용출기술과 전기동력학적 기술의 통합공정은 현장특이성(Site-specific) 확인후 적용가능성이 있음을 확인하였다.
The bio-electrode for L-asparagine, excellent in the reproducibility of responsibility, has been constructed by immobilizing the bacterium Proteus mirabilis on an ammonia gas sensor. This electrode was investigated for the effects of pH, temperature, buffer solution, bacterial amounts and interferences, and stability with the lapse of time. The response of the bacterial electrode was linear in the range of $9.0{\times}10^{-5}$ ∼ $1.0{\times}10^{-2}M$ L-asparagine with a slope of 58.9mV/decade in pH 7.8, 0.05M phosphate buffer solution at $30^{\circ}C$. The bacterial amounts used for this electrode was 3 mg and response time was 7∼9 min. Therefore, this assembly can be used for the determination of L-asparagine.
The aim of this study was tofind out the effects of supplementation of vitamin A to the diets of high or low amounts of concentrates for ruminants. In the first experiment, ruminal fermentation patterns with the data of pH, VFA production and cellulose disappearance rates in the rumen in vitro were investigated. In the second experiment, enzyme activities, gas production and dry matter degradabilities using cellulolytic bacteria, Ruminococcus flavefaciens were investigated. Ruminal pH was higher in low amounts of concentrates than in high amounts of concentrates as expected, however, no significant differences were found. Cellulose disappearance rates improved in vitamin A addition particularly in early incubation time (before 24h) and also the production of volatile fatty acids increased in vitamin A addition. These trends were more evident in diets containing high amounts of concentrates than in low amounts of concentrates and it may indicate that vitamin A is more required in the diets of high amounts of concentrates. In the second experiment, gas production, enzyme activities and dry matter degradabilities using cellulolytic bacteria, Ruminococcus flavefaciens were not different between vitamin A added and non-added diets. Ruminococcus flavefaciens may not require additional vitamin A for its own growth.
