Effects of Red-Yeast-Rice Supplementation on Blood Lipids and Relations among Cholesterol, Bone Markers, and Hormones in Overiectomized Rats (난소 절제 쥐에서 홍국의 첨가가 혈중 지질에 미치는 영향과 혈중 콜레스테롤과 골대사 지표 및 호르몬과의 관련성)

  • This study was performed to evaluate the effect of red-yeast-rice on blood lipids and also to investigated the relation of blood lipids, bone related hormones, and bone mineral density in overiectomized rats. Forty female rats (body weight $210\pm5$g) were divided into two groups. The rats in the first group were ovariectomized which the others received sham operation (SHAM). And then each rat group was further divided into control diet $(casein\;20\%)$ and red-yeast-rice powder supplemented $(0.1\%)$ diet group. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone mineral density (BMD) and bone mineral content (BMC) were measured using PIXImus in spine and femur. Bone formation and bone resorption were measured by serum osteocalcin and urinary deoxypyridinoline (DPD) crosslinks value. Serum growth hormone, IGF-1 and calcitonin were analyzed using radioimmunoassay kits. The red-yeast-rice group had significantly lower the blood triglyceride concentration and had lower the blood total cholesterol concentration than casein group did in ovariectomized rats. And the red-yeast-rice group than casein group in ovariectomized rats. The red-yeast-rice group had significantly lower the atherogenic index than control group within the ovariectomized groups. In the experimental rats, serum total cholesterol concentration was negatively correlated with femur BMC and crosslink value. Crosslink value was negatively correlated with spine BMC and IGF-1. Osteocalcin concentration was negatively correlated with crosslink value. Growth hormone was negatively correlated with crosslink value and positively correlated with spine BMC. The findings of this study suggest that red-yeast-rices are beneficial for blood lipid in ovariectomized rats. And when considering a population-based public health approach to lowering cholesterol and bone loss, and preventing coronary heart disease and osteoporosis, the red-yeast-rice dietary supplement could provide a new approach for the maintenance of healthier cholesterol and bone.

The Effect of Dietary Calcium Level on Biochemical Variables of Bone Metabolism in Ovariectomized Female Rats (식이 칼슘량이 난소 절제한 흰쥐에서 골대사 지료에 미치는 영향)

  • This study was done to evaluate the effectiveness of dietary calcium level(a diet which met 100% or twice the calcium level in AIN-76 diet) on preventing bone loss in ovariectomized rats. Forty female Sprauge-Dawley rats(body weight 200$\pm$5g) were divided into two groups. One group were ovariectomized(Ovx) while the others received sham operation(Sham). Thereafter, each rat group was further divided into normal calcium diet(NCD, 0.52%) and high calcium diet(HCD, 1.04%) sub-groups. All rats were fed on experimental diet and deionized water ad libitum for 8 weeks. Urinary pyridinoline & creatinine and serum estradiol, luteinizing hormone, calcium, phosphate, total protein, albumin, alkaline phosphatase and osteocalcin were determined. There were no significant differences in serum calcium. total protein and albumin in the two groups(Ovx vs Sham) of rats. Ovariectomized rats had significantly lower estradiol than sham operated rats. There was a highly significant correlation between total bone mineral density(TBMD) and overall level of esteradiol(r=0.59, p<0.05). Total bone mineral density did not correlate significantly with ALP or osteocalcin, although a negative trend was evident. However, the rats fed high calcium diet had a lower crosslinks value and osteocalcine than the rats fed normal calcium diet. An increased rate of bone turnover is usually associated with a decrease in bone mass bexause bone formation at each remodeling site is never as great as resorption. Ovariectomized rats fed high calcium diet had a lower crosslink value and osteocalcin; it means high cacium diet decreased bone turnover rate. The findings from the present study demonstrated that bone loss due to ovarian hormonal deficiency can be partially prevented by a high calcium diet. Futhermore, these findings support the strategy of the use of a high calcium diet in the prevention of estrogen depletion bone loss (postmenopausal osteoporosis).

Effects of Nutrient Intake, Bone Mineral Density and Bone Mineral Content in Ovariectomized Women (난소 절제 여성의 영양소 섭취 상태가 골밀도 및 골무기질 함량에 미치는 영향)

  • This study investigated associations between nutrient intake, lumbar bone mineral density (BMD) , and bone mineral content (BMC) among 33 ovariectomized women (mean age =47.2 y) . Forty-five premenopausal women participated as a control group. The BMD and BMC of the lumbar spine (L$_2$-L$_4$) were measured by dual energy x-ray absorptiometry. Nutrient intake was estimated by the convenient method and a quantitative food frequency questionnaire was designed for this study that included the most commonly consumed foods sources of calcium. Participants were asked to identify all daily physical activities, and the number of hours per activity. The participants were also grouped by calcium intake. The total calcium intake of all participants was estimated by dietary calcium intake and then the subjects were divided into quartiles to assess the lumbar BMD and BMC of the highest quartile and the lowest quartile of calcium intake. The ovariectomized women consumed 602 mg/d of calcium which is 86% of RDA. There were significant differences in lumbar BMD and BMC between control and ovariectomized group. Within ovariectomized group the highest quartile calcium intake group had significantly greater lumbar bone mineral density and bone mineral content than the lowest quartile calcium intake group. Correlation analysis revealed that the ALP was positively associated with calcium index in control women, while ALP was positively associated with energy intake in ovariectomized women. And body weight was positively correlated with the spinal BMD and BMC in all women. The spinal BMD was negatively associated with menarche age, number of child, and the age of last child delivery, and age in control women. However, neither menarche age nor the age of last child delivery were associated with both spinal BMD in ovariectomized women. These results confirmed that ovariectomized and low calcium intake is associated with poor bone mineral density. Energy and calcium intake and adequate body weight should be recommended in ovariectomized women to prevent osteoporosis.

Effects of Oolong Tea Supplementation on Bone Mineral Density and Bone-Related Markers in Ovariectomized Rats (난소 절제 쥐에서 우롱차의 섭취가 골밀도 및 골대사 지표에 미치는 영향)

  • Polyphenols may be primarily responsible for the health benefits associated with tea consumption. However, the benefits of the tea polyphenols to bone health have not been studied well. The purpose of this study was to investigate the effects of Oolong tea on bone mineral density and bone-related markers in ovariectomized rats. Sprague-Dawley female rats were randomly assigned to Sham-Control, Sham-Oolongtea, Ovx-Control, and Ovx-Oolongtea groups. Bone mineral density (BMD) and bone mineral content (BMC) were estimated by using PIXImus (GE Lunar Co, Wisconsin.) in the spine and femur. After 6 weeks of the experimental diets, body weight gain, food intake, and food efficiency ratio (FER) were significantly higher in Ovx groups than in Sham groups regardless of diet. The serum concentration of calcium, phosphorus, ALP, and calcitonin were not significantly different according to Oolong tea supplementation. There were no significant differences in urinary calcium and phosphorus excretion between all groups. The urinary DPD crosslinks value was significantly higher in the Ovx-Control group than in the Sham-Oolongtea group. Spine BMD, femur BMD, and spine BMD per body weight were significantly lower in the Ovx groups than in the Sham groups regardless of diet. In the OVX group, spine BMC per body weight, femur BMD per body weight and femur BMC per body weight were significantly higher in the Oolong tea groups than in Control groups. It was concluded that Oolong tea supplementation positively influenced bone health in ovariectomized rats.

THE HISTOLOGIC STUDY OF THE RAT TEMPOROMANDIBULAR JOINT CHANGE FOLLOWING OVARIECTOMY (자성백서의 난소절제시 악관절 변화에 관한 조직학적 연구)

  • This investigation studied the effect of ovariectomy on the temporomandibular joint (TMJ) of rat. Forty female Sprauge-Dawley strain white rats, each weighing about 200grams were used and divided into two groups. The one group was performed ovariectomy, and the other was performed sham operation. Both group were killed at the intervals of 3 days, 1, 2, 4, and 8 weeks postoperatively, paraffin section were stained with H&E, MT and histomorphometric study of the TMJ was performed in a synchronous manner with an sham operated control group The result were as follows: 1. At 4 weeks after ovariectomy, an adhesion between the inferior surface of the articular disc and the condyle was observed in the central portion. At 8 weeks after ovariectomy, a focal thickening of the bone (osteophyte) was observed in anterior portion and there was a marked decrease in the trabecular bone in the posterior region. 2. The thickness of cartilage zone was increased at 2 weeks postopratively in the central portion, and decreased at 4 weeks postopratively in the posterior portion. The thickness of transitional zone was decreased at 1 week postoperatively in the anterior and proliferating zone was decreased postoperatively. The thickness of articular zone was decreased at 3 days postopratively in the anterior portion, and decreased at 1 week postoperatively in the central portion. From the above finding, we could conclue that estrogen deficiency in rats causes alteration of TMJ in marked early stage.

RELATIONSHIPS BETWEEN THE MANDIBLE AND THE FEMUR IN OSTEOPOROSIS INDUCED BY OOPHORECTOMY AND EFFECT OF PREVENTIONS BY CALCITONIN IN THE ALBINO RATS (난소 절제한 흰쥐에서 골다공증이 하악골에 미치는 영향과 칼시토닌의 예방효과)

  • Osteoporosis, a well known metabolic bone disease, occurs due to imbalance of bone formings and resorptions. In an attempt investigate if postmenopausal osteoporosis would have influence on the mandible, histomorphometric analysis was performed in the mandible and the femur of the albino rats. Osteoporosis was induced by bilateral oophorectomy. To determine the effect of calcitonin, calcitonin(Asahi Chemical Co., Japan) was injected in the oophorectomized rats. The results were as follows : 1. The width of cortical bone of the femur was decreased in the oophorectomized group compared to the normal control and calcitonin injected groups and the porosity of cortical bone of the femur was increased in the oophorectomized group compared to the normal control and calcitonin injected groups. 2. The osteoid tissue and resorption lacuna of the femur was increased in both oophorectormized and calcitonin injected groups than the normal control group. 3. The width of cortical bone of the mandible was decreased in the oophorectomized group compared to the normal control and calcitonin injected groups. 4. The porosity of cortical bone of the mandible was decreased in both oophorectomized and calcitonin injected groups than the normal control group. 5. The relative volume of trabecular bone of the mandible was decreased in the oophorectomized group compared to the normal control and calcitonin injected groups. 6. The osteoid tissue of the mandible was increased in the calcitonin injected group than the normal control and oophorectomized groups. 7. The resorption lacuna of the mandible was increased in both oophorectomized and calcitonin injected groups, particularly in the oophorectomized group.

Trigonella Goenum-Graecum Seed Extract Alleviates Menopausal Symptoms and Osteoporosis in Ovariectomized Rats (호로파(葫蘆巴) 종자 추출물이 난소 절제술을 받은 쥐에서 갱년기 증상 및 골다공증에 미치는 영향)

  • Objectives : We proposed the hypothesis that administering Trigonella foenum-graecum seed extract (TSE) could alleviate menopausal symptoms and osteoporosis resulting from estrogen deficiency. Methods : Ovariectomized (OVX) rats were administered TSE at doses of 300 or 600 mg/kg body weight for 8 weeks, followed by measurement of serum lipid profile and serum bone markers using ELISA kits. Additionally, analysis of related genes in the femur and uterus was performed using Western blot and real-time PCR. Additionally, micro-CT analysis was performed to investigate the protective effect of TSE against bone loss due to oophorectomy. Results : The administration of TSE led to significant reductions in triglyceride (TG), total cholesterol (TC), low-density lipoprotein (LDL) cholesterol, and glucose levels in the serum of OVX rats. Furthermore, TSE increased estradiol levels in the serum and notably improved the levels of biochemical markers associated with bone metabolism. Additionally, TSE exerted significant regulatory effects on the mRNA levels of alkaline phosphatase (ALP) and receptor activator of nuclear factor kappa-B ligand (RANKL)-genes closely associated with bone metabolism in the femur. TSE also demonstrated pronounced effects on uterine tissue, with improvements observed in gene expression related to estrogen receptors. Conclusion : Our findings confirm the efficacy of TSE in ameliorating menopause symptoms by modulating elements associated with both bone and lipid metabolism in the serum, uterine tissue, and femur of OVX rats. The present findings suggest that TSE may offer potential therapeutic effects for symptoms related to menopause and osteoporosis in females.

  • Since 2019, ultrasound and magnetic resonance imaging for evaluation of urogenital disorders have been covered by the National Health Insurance (NHI) in Korea. Patients with urogenital malignancies were already insured by NHI for ultrasound and MRI. With the expansion of NHI coverage, patients with suspected prostate or gynecologic cancer, uterine fibroids before myomectomy and some other benign disease such as congenital anomaly can receive benefits of NHI. In consideration of these changes, radiologists and other clinicians should be aware of the indications and standard images of each examination and the required reporting forms. Clinical application based upon thorough understanding of the NHI guidelines will aid in improving the standard care of patients.

The Effects of Pueraria and Rehmannia Glutinosa Intake and Exercise on Epigenetic Modification in Ovariectomized Rat Skeletal Muscle (난소 절제 쥐의 골격근에서 갈근 및 지황 섭취와 운동이 후성 유전적 변화에 미치는 영향)

  • The purpose of this study was to determine the effect of Pueraria lobate-root based combination supplementation containing Rehmannia glutinosa and exercise on histone modification in ovariectomized rat hindlimb skeletal muscle. Sixty rats were fed with high fat diet and randomly assigned into the following groups for 8 weeks: 1)HSV; High fat+Sedentary+Vehicle, 2)HSP; High fat+Sedentary+PR, 3)HSH; High fat+Sedentary+Estradiol, 4)HEV; High fat+Ex+Vehicle, 5)HEP; High fat+Ex+PR, 6)HEH; High fat+Ex+Estradiol. Exercise consisted of low intensity treadmill exercise(1-4th wk:15 m/min for 30 min, 5-8th wk: 18 m/min for 40 min, 5 times/week). The result of this study showed that exercise and Pueraria and Rehmannia glutinosa intake suppressed weight gain. Furthermore, exercise and Pueraria and Rehmannia glutinosa intake increased muscle mass. This study observed H3K9 acetylation and demethylation in plantaris muscle in exercised group, but no difference in soleus muscle. To test whether the decrease in HDAC4, HDAC5 and G9a mRNA levels after exercise and Pueraria/Rehmannia glutinosa intake, HDAC4, HDAC5 and G9a mRNA levels were determined by real-time PCR. Only exercise induced HDAC5 and G9a mRNA reduction in plantaris muscle, but not in soleus muscle. In conclusion, these data demonstrates that exercise and Pueraria/Rehmannia glutinosa intake effect on body compositions. These changes are regulated by epigenetic modifications, such as histone acetylation and methylation. Future studies should focus on gene-specific epigenetics and other epigenetic mechanism for Pueraria/Rehmannia glutinosa intake.

The Effect of $OPB-K^{(R)}$ on the Osseointegration and Stability of Implant ($OPB-K^{(R)}$가 임플랜트의 골유착 및 안정성에 미치는 영향에 관한 연구)

  • Statement of problem: It has been proved that Pleurotus eryngii Quel and Eleutherococcus senticosus have antiinflammatory action and not only stimulates the proliferation and activity of osteoblast but inhibits the generation and activity of osteoclast in vitro. Pleurotus eryngii Quel and Eleutherococcus senticosus are the main component of $OPB-K^{(R)}$. Purpose: The purpose of this study was to evaluate $OPB-K^{(R)}$ which enhances the healing rate of peri-implant bone and the bone mineral density. Materials and methods: Thirty six specially designed implants were installed in the tibia of rats. The group medicated with $OPB-K^{(R)}$ was the experimental group, and that without was the control group. hen the implant stability was measured by $Periotest^{(R)}$. Bone mineral density and histological measurement were conducted at the 2nd, 4th and 6th week $Periotest^{(R)}$ and bone mineral density values were analyzed statistically with independent t-test at 95% confidence level(p<0.05). Results: The results of this study were as follows : 1. There was no statistically significant difference in $Periotest^{(R)}$. values between the experimental group and control group at the 2nd week, however, on the 4th and 6th week there was significant difference(p<0.05). 2. There was no statistically significant difference in bone mineral density between the experimental group and control group at the 2nd and 4th week, however on the 6th week there was significant difference(p<0.05). 3. Histological analysis showed difference in osseointegration on the 4th and 6th week between the groups. Conclusion: From the results, it is concluded that the $OPB-K^{(R)}$ medicated group showed statistically better results in bone density and stability than the control group. Clinically it would be better to medicate $OPB-K^{(R)}$ to patients for a long period of time after implantation to get superior results.
