Acute toxicity of lambda-cyhalothrin to Chironomus riparius in water alone and sediment-water systems (저니토독성시험법(sediment toxicity test)을 이용한 lambda-cyhalothrin의 깔따구 (Chironomus riparius)에 대한 급성독성)

  • The midge Chironomus riparius is a member of widespread dipteran insect which has been used for sediment toxicity test. The 2nd-3rd larvae Chironomus riparius were exposed to $^{14}C$-lambda-cyhalothrin in laboratory water only system(A) and sediment-water system(B) at $20{\pm}2^{\circ}C$. The concentration was nominally in the range of 0.16 to $20{\mu}g/{\ell}$ and 62.5 to 8000 ${\mu}g/kg$, respectively. System(A) was prepared by applying the chemical to 250 ml water and introducing Chironomus riparius larvae into the system(day 0). System(B) was prepared by applying the chemical to the 25 g of soil and 250 ml water and shaking and rolling them for two hours. After allowing the system to settle for two days, Chironomus riparius larvae were introduced into the system. After 72 hours, the two systems were terminated and dead larvae were observed. The measurement of residue in water was conducted at the start(day 0) and the end of the study(day 3). The residue of $^{14}C$-lambda-cyhalothrin in water was accounted for approximately 65% of nominal concentration in system(A) and $0.2{\sim}0.4%$ in system) on 3 days. $LC_{50}$ was less than $0.156{\mu}g/{\ell}$ in system(A) and 889 ${\mu}g/kg$ in system(B), based on the nominally applied concentration.

Identification and characterization of allergens of Chironomus fkavuoynys adults (Chironomidae, Diptera) in mice (노랑털깔따구(Chironomus flaviplumus) 성충의 알레르기 항원단백 분석)

  • Non-biting midges fchironomidae, Dipteral are one of the largest insect families, which are distributed worldwidely and are found in nearly all types of inland waters. They are known to be aggressive inhalant allergens which cause allergenic diseases. In this study, the crude antigens of Chironomus SavipLumn adults which are most widely distributed in Korea were extracted. and their allergens were analysed with the sera from experimentally sensitized mice. The mice were immunized with $1{\;}\mu\textrm{g}{\;}or{\;}10{\;}\mu\textrm{g}$ of the crude antigens, respectively, and the specific serum IgE levels were measured by both ELISA and passive cutaneous anaphylaxis (PCA) techniques. The highest levels of both total IgE and chironomid-specific IgE were found in the mouse sera obtained after 9 weeks of the first infection with $1{\;}\mu\textrm{g}$ crude antigen. The crude antigen was separated into 16-18 protein bands on gel by SDS-PAGE. The crude extract was assessed by SDS-PAGE/immunoblot analysis. One IgE-binding band (65 kDa) was detected by developing with colorimetric substrate, and 4 IgE-binding bands (65, 52, 35 and 25 kDa) by developing with CSPD chemiluminescent substrate. The SDS-PAGE gel of the crude extract of chironomid adults was equally cut into 30 pieces and each of them was eluted to isolate proteins by molecular weight, and the allergenicity of each eluate was assessed by applying P-K test on rats. Proteins of 65, 35 and 15 KDa showed the highest P-K titer (${\times}512$) which was 16 times higher than that of the crude extract (${\times32}$). The P-K titer of 52 kDa protein was also 4 times higher ($128{\times}$) than that of the crude extract, whereas the 25 kDa protein poorly responded, which seemed not antigenic. In conclusion, the present result in mice demonstrated that adults of Chironomus fcuiplumus, a predominent species in Korea, cause allergenic diseases and the main allergens are 65, 52, 35 and 15 kDa proteins, of which 65 kDa protein seems to be a main allergen.

Expression of Heat Shock Protein 70 Gene and Body Color Changes in Non-biting Midge Larvae (Glyptotendipes tokunagai) Effected by O3 Treatment (오존(O3) 노출에 의한 조각깔따구(Glyptotendipes tokunagai)의 체색 변화 및 heat shock protein 70 발현 변화)

  • Ozone (O3) is a general disinfectant to remove micro-pollutants in water treatment system. Previous studies have reported effect of ozone to bacteria and pathogens removal, but its effect to the relatively large organisms has little known. In this study, we investigated potential effects of ozone toxicity to the non-bite midge larvae (Glyptotendipes tokunagai) with accumulate mortality, coloration change and expression of heat shock protein 70 (HSP70). The accumulate mortality rate of G. tokunagai increased in a dose-time dependent manner and the highest mortality rate was observed to 75% at 30 minute of exposure duration with 2.0 ppm of ozone concentration. Exposure to ozone was a factor increasing body color of the larvae. The tendency of HSP70 mRNA expression showed up-regulation in ozone exposure at 20 minute. After that time, the expression of HSP70 in exposed group decreased to a similar level of control group. Our results clearly showed that ozone toxicity affects physical and molecular activity of G. tokunagai, implying the potential hazardous of ozone in the aquatic ecosystem including macroinvertebrates.

  • Over the past few years, many studies have been conducted on the flow, sediment movement, pollution transportation and scour etc. However, very few attempts have been made at the hydraulic studies reflecting upon the ecological function. The objective of this study is to examine the structures of the flow and turbulence in an open circular channel and their relationship to distribution of the organisms and chironomids. Under different flow conditions, the organic matter and some chironomids were injected into the channel. Using the obtained velocity data, the flow mechanisms and the turbulent shear stresses were analyzed. Organic matters and chironomids were distributed on the region that the velocity was slower and the turbulent shear stresses were smaller. Some habitat moved even though chironomids were inhabited. This phenomenon has relationship with the flow mechanism. Some chironomids have distributed around the habitat structure of a hemisphere. The secondary flow has affected the deposition of the organic matters and the distribution of chironomids.

Water distribution system contamination source estimation based on trace analysis (Trace Analysis 기능을 활용한 상수도 관망 내 오염물 유입 지점 추정)

  • 지난 2019년 인천시, 서울시 문래동, 포항시 등에서 발생한 수질사고로 인해 국민의 상수도에 대한 신뢰도가 최악의 상황에 있으며, 이후로도 깔따구 유충이 발견되는 등 상수도 관망 내 체계적인 수질 관리 및 빠르고 정확한 수질 사고 발생 지점의 추정이 중요해 지고 있는 실정이다. 오염물 유입 추정은 수리학적 사고로 고려되는 누수와는 달리 상대적으로 그 지점 추정이 어렵다. 대게의 경우 수리해석을 진행하여 유량과 유향을 파악한 뒤 계측 지점에서부터 동일 시간대로 역으로 흐름을 거슬러 올라가며 확률상 높은 지점을 추정하는 것이 일반적인 방법이다. 본 연구에서는 범용 수리해석 프로그램인 EPANET2.2에 내장된 Trace Analysis (이후 trace 분석) 옵션을 사용한 오염물 유입 지점 추정 방법론을 소개한다. 본 연구에서는 방법론의 검증을 위해 오염물 유입지점은 한 곳으로 가정하였다. 해당 방법론은 먼저 절점별 trace 분석을 실시하여 모든 지점에서 수질 관측 지점까지 물이 도달하는데 소요되는 시간을 산정한다. 해당 시간과 오염물 관측 데이터와의 비교를 통해 유입 확률이 높은 지점을 추출한다. 이를 위해 실측 데이터가 필요하며, 결과는 지점별 확률로 나타난다. 모의 결과 1개의 수질 관측 지점으로도 개략적인 지점을 선정할 수 있는 것으로 나타났다. 다만, 수질 관측 지점의 수에 따라 분석 결과의 정확도가 향상한다. 마지막으로 유입 지점 추정 확률이 낮은 경우, 유입 지점 추정 확률을 향상시킬 수 있는 추가 수질 분석 지점을 결정하였다. 본 연구에서 소개한 방법론은 향후 수질 사고 발생 시 최초 확산 방지를 위한 격리 지점 선정에 근거를 제시할 수 있을 것으로 기대하며, 나아가 수질 관측 지점을 결정 및 대응 방안 수립 가이드라인으로 활용할 수 있을 것이다.

  • The digestive apparatus and food of the Korean bitterling, Acheilognathus signifer, from the upper reaches of the Hongcheon River were investigated by examining their gill rakers, pharyngeal teeth, and digestive-tube contents. Gill rakers of the first arch were relatively few (17 to 21, mean = 19) and short (mean = 0.36 mm, SD = 0.035). The 2nd to 5th pharyngeal teeth were well developed, while the first tooth was poorly developed. Gill rakers and pharyngeal teeth were completely formed early in life. The digestive tube was long and arranged in many coils. The digestive-tube contents of Korean bitterling mainly consisted of phytoplankton and detrital microorganisms.

Hydraulic Habitat Analysis of Benthic Macroinvertebrates at Gapyeong Stream (가평천의 저서성 대형무척추동물 서식처의 수리 특성 분석)

  • Hydraulic habitat analysis of Benthic Macroinvertebrates was performed at Gapyeong stream. Among the kinds of the Benthic Macroinvertebrates, the most representative ones are the Ecdyonurus kibunensis from the clingers, Paraleptophlebia cocorata from the swimmers, Chironomidae spp from the burrowers, Psilotreta kisoensis iwata from the sprawlers. They showed different habitat types by the hydraulic conditions such as flow velocity, depth and particle size of riverbed materials. Habitat conditions of swimmers were determined mainly by the flow velocity rather than flow depth or riverbed materials. Burrowers prefer sand and silt, and inhabited at the riverbed. Sprawlers prefer cobble or boulder and inhabited for velocity of 0.05~0.15 m/s. Clingers prefer pebble or cobble and inhabited for velocity of 0.06~0.15 m/s. Although the habitats of each groups are different by the hydraulic properties, they were found to be different mainly by the water velocity.

  • In order to extract and analyze complex features of the behavior of animals in response to external stimuli such as toxic chemicals, we implemented an adaptive computational method to characterize changes in the behavior of chironomids in response to treatment with the insecticide, diazinon. In this paper, we propose an energy minimization model to extract the features of response behavior of chironomids under toxic treatment, which is applied on the image of velocity vectors. It is based on the improved active contour model and the variations of the energy functional, which are produced by the evolving active contour. The movement tracks of individual chironomid larvae were continuously measured in 0.25 second intervals during the survey period of 4 days before and after the treatment. Velocity on each sample track at 0.25 second intervals was collected in 15-20 minute periods and was subsequently checked to effectively reveal behavioral states of the specimens tested. Active contour was formed around each collection of velocities to gradually evolve to find the optimal boundaries of velocity collections through processes of energy minimization. The active contour which is improved by T. Chan and L. Vese is used in this paper. The energy minimization model effectively revealed characteristic patterns of behavior for the treatment versus no treatment, and identified changes in behavioral states .is the time progressed.

Stress Evaluation to Heavy Metal Exposure using Molecular Marker in Chironomus riparius (분자지표 유전자 발현을 통한 Chironomus riparius 중금속 노출 스트레스 평가)

  • Heavy metals are common pollutants in the freshwater environment and have toxicological effect in habitat organisms. The heavy metals highly accumulated in sediment and organism, and observed various physiological responses. In this study, we investigated the molecular response to heavy metal toxicity (Al, Aluminum; Cr, Chromium; Cu, copper; Mn, Manganese; Zn, Zinc) through expression of heat shock protein 40, 70, 90 (HSP40, 70, 90), cytochrome 450 (CYP450), Glutathione S-transferase (GST) and Serine-type endopeptidase (SP). HSPs showed up-regulation in Cu and Zn exposures. Furthermore, HSPs expression in treated groups tended to be higher than the control group. The tendency of CYP450 and GST mRNA expression was higher for Cr and Cu than for other exposure group. The expression of SP gene was low at Al exposure and other group were measured to be similar to control. These results suggest that heavy metal toxicity in freshwater ecosystem may affect physiological and molecular process. Also, the comprehensive gene expression in the aquatic midge Chironomus riparius give useful information to potential molecular biomarkers for assessing heavy metal toxicity.

Seasonal Prevalence and Integrated Control Method of Insect Pests in the Dredged Soil Dumping Area in Masan, Korea (마산시 준설토 투기장에서 발생하는 유해곤충의 계절적 발생 소장과 통합적 저감방법)

  • This study was conducted to monitor and prevent outbreak of insect pestsin dredged soil dumping area after completion of dredging construction in Masan City. Monitoring was carried out using tent trap, colored sticky trap, and CDC light trap. A total of 217,073 individuals belonging to 23 species from 10 families in 3 orders were collected. In overwintering survey using tent trap, 3 species were collected. 2 species (Leptocera fuscipennis (Haliday) and Ephydra japonica Miyagi) of them were outbreak species. In color sticky trap, more than 96% of total individuals were comprised of five species: Urolepis maritima Walker (43%), E. japonica (19%), Fucellia sp. 1 (13%), Philotelma sp. 1 (10%), and Homalometopus sp. 1 (9%). In CDC light trap, three dominant species were Homalometopus sp. 1 (91%), Glyptotendipes tokunagai Sasa (6%), and L. fuscipennis (1%), representing about 98% of the total. To prevent damage caused by outbreak of insect pests, we carried out ecological control methods such as covering the fresh soil in outbreak area, using light trap, pumping up water and so on, minimizing use of thermal fogging and insect growth regulatorwhen the insect pest population was rapidly increasing.
