The Effect of Glycyrrhizic acid on Vascular Contractility (혈관수축에 대한 감초산 Glycyrrhizic acid의 효과)

  • This study aims to elucidate the effect of glycyrrhizic acid on smooth muscle contraction and to determine the detailed mechanism incorporated. We hypothesized that glycyrrhizic acid played a role in the agonist-sensitive management of smooth muscle contraction. Stripped smooth muscles of Sprague-Dawley rats were prepared in organ baths and isometric tensions were converted, stored and analyzed by using isometric transducers, a physiograph and one way ANOVA. Interestingly, glycyrrhizic acid attenuated the thick filament regulating agonist (fluoride or thromboxane mimetic)-sensitive contraction (p=0.113, 0.008, 0.004 (Student's t-test), p=0.113, 0.008, 0.004 (One way ANOVA) at 0.01, 0.03, 0.1 mM fluoride, and p=0.156, 0.004, 0.003 (Student's t-test), p=0.156, 0.004, 0.003 (One way ANOVA) at 0.01, 0.03, 0.1 mM thromboxane mimetic) and did not attenuate the thin filament regulating agonist (phorbol ester)-induced contraction (p=0.392, 0.086, 0.065 (Student's t-test), p=0.392, 0.086, 0.065 (One way ANOVA) at 0.01, 0.03, 0.1 mM phorbol ester). It is suggesting that endothelial EDRF (NO) synthesis and accessory pathways besides endothelial EDRF (NO) synthesis such as ROCK restriction might be incorporated in the glycyrrhizic acid-induced modulation of smooth muscle contraction inhibiting acto-myosin interaction.

  • Background : Leukotriene (LT) $C_4$, $D_4$, and $E_4$, the main components of slow-reacting substance of anaphylaxis (SRS-A), have been suggested to play an important role in bronchial asthma such as antigen-induced bronchoconstriction, airway hyperreactivity, and pulmonary eosinophil accumulation. The purpose of this study was to evaluate the effects of treatment with the cysteinyl-LTs (cys-LTs) antagonist, pranlukast on allergen-induced guinea pig asthma model. Methods : Guinea pigs of treatment and placebo groups were sensitized by subcutaneous injection of ovalbumin(OVA) and challenged by inhalation of aerosolized OVA (1% weight/volume OVA). Normal control group did not sensitize with OVA. Oral ingestion of pranlukast and normal saline to the treatment and placebo groups was performed. In the treatment and placebo groups, airway resistance was measured before and after oral ingestion. Serum $LTC_4$ and eosinophilic infiltration of the bronchiolar and peribronchiolar tissues were measured after ingestion in the treatment and placebo groups. Results : Allergen-induced airway constriction developed in 20 (8 in treatment group, 12 in placebo group) among 35 guinea pigs. Airway resistance was significantly decreased at 3 and 6 minutes after OVA challenge in the pranlukast treatment group. In the placebo group, there was no difference of airway resistance between before and after saline ingestion. Serum $LTC_4$ levels showed 348.4 pg/ml in the treatment group, 373.9 pg/ml in the placebo group, and 364.4 pg/ml in the control group. There were no statistically significant difference between treatment and placebo group (p=0.232), and treatment and control group (p=0.501). Eosinophilic infiltrations in the peribronchiolar region per one-microscopic field ($\times$400 high power fields) demonstrated 7.06 in the treatment group, 19.2 in the placebo group, and 4.50 in the control group. There was significant decrement of eosinophilic infiltration in the treatment group which was compared with placebo group (p=0.001). Conclusion : These results demonstrate that pranlukast, a cys-LTs receptor antagonist, can attenuate allergen induced early-phase bronchoconstriction and eosinophilic infiltration in the bronchiolar tissues.

  • 심장과 혈관의 고해상도 영상을 얻어 심질환에 관한 정보를 얻는데는 혈관조영기(Angiograph)가 가장 유용하게 사용되고 있다. 본 연구는 혈관조영기의 정량분석을 위한 영상 알고리즘을 구현하였다. 심장의 수축 이완에 따른 심벽의 부위별 움직임과 심장 체적 변화를 측정 수치화하고 혈관을 따라가면서 혈관의 두께를 기록하는 프로그램을 작성하였다. 영상획득과정에서 생기는 잡음과 위치오차를 없애는 전처리를 거친 후에 구조적 형태 인식법(Structural Pattern Recognition)으로 인식 알고리즘을 구현하였다. 14장의 심장의 수축이완 연속사진과 2장의 관상동맥 혈관조영사진을 분석한 결과 비교적 정확한 측정을 해내는 것을 관찰할 수 있었다.

The Convergence Effect of Phloretin Existent in Plants on Vascular Contractility (플로레틴(Phloretin)의 혈관내피수축 융합효과와 관련기전 연구)

  • This study tried to observe the ability to inhibit vasocontriction in phloretin - the primary ingredient of apple tree leaves and the Manchurian apricot - through ROCK(Rho-associdated, coiled-coil containing protein kinase) inactivation in rat aortae. A piece of artery that was separated from Sprague-Dawley male rats and retained or damaged the endothelium was suspended in myograph tank with two metal rings, the lower ring fixed to the bottom of the tank, and the upper ring connected to the isotonic force transducer. Interestingly, phloretin inhibited fluoride- or phorbol ester-provoked contraction implying that additional pathways dissimilar from endothelial nitric oxide synthesis such as ROCK or MEK (mitogen activated protein kinase kinase) inactivation might be involved in the vasorelaxation. Therefore, this study provides that phloretin participates in the reduction of ROCK or MEK activity in smooth muscle in addition to the endothelial-dependent action of the endotheliuim in complete blood vessels, and consequently inhibits actin-myosin interaction in smooth muscle. Furthermore, phloretin inhibited thromboxane A2-induced contraction suggesting the mechanism including inhibition of ROCK and MEK.

Change of Cardiovascular Function of Industrial Workers Apply to Lumbar Stabilization Exercise according to the Floor Type (지면의 상태에 따른 요부안정화운동 적용 시 산업체 근로자의 심혈관기능 변화)

  • This study conducted the following experiment to examine effects of cardiovascular function on lumbar stabilization exercise(LSE) in floor or swiss ball. This experiment was conducted to compare heart rate, systolic blood pressure, diastolic blood pressure and peripheral vascular oxygen saturation effects by lumbar stabilization exercise in floor or swiss ball with 18 normal adult and it divided 9 subjects. experiment group (1) is applying LSE on floor group and (2) is applying LSE on swiss ball group. Heart rate was measured by portable heart rate manometer, blood pressure was measured by hemodynamometer, and peripheral vascular oxygen concentration was measured using a computerized NURYTEC measuring apparatus analysis. These result lead us to the conclusion that systolic blood pressure and peripheral vascular oxygen concentration were influenced by LSE. but there was not differential effect between each groups. These results suggest that LSE has the capability to improve heart rate, blood pressure, peripheral vascular oxygen concentration. Consequently, LSE would be lead to increment of cardiovascular function.

The Effect of Dietary Hizikia Fusiforme on the Reduction of Blood Pressure and Total Cholesterol in Jeju Women Divers (제주 해녀의 식이성 톳 섭취를 통한 혈압 및 총콜레스테롤 감소 효과)

  • The purpose of this study was to investigate the changes of blood pressure and total cholesterol after eating Hizikia fusiforme for 8 weeks with 100g of rice per day. As a result, the systolic blood pressure of the group ingesting Hizikia fusiforme significantly decreased. Second, diastolic blood pressure decreased significantly in both groups, but there was no difference between groups. Third, the systolic - diastolic blood pressure difference showed a significant decrease in the group of ingestion of Hizikia fusiforme, while that of the group without intake of Hizikia fusiforme increased. Fourth, total cholesterol decreased significantly in the group consuming Hizikia fusiforme but increased in the group not consuming Hizikia fusiforme. The above results indicate that total intake of 100 g of dried Hizikia fusiforme per day was effective in decreasing total cholesterol and reducing systolic blood pressure.

Correlation Between Left Ventricular Peak Systolic Pressure/End-Systolic Volume Ratio and Symptomatic Improvement with Valve Replacement in Patients with Aortic Regurgitation and Enlarged End-Systolic Volume (대동맥판역류증과 좌심실수축말기용적 확장이 있는 환자에서 좌심실최고수축기압/수축말기용적비와 판막치환후의 증상적 호전과의 관계)

  • This study was designed to assess the left ventricular peak systolic pressure/end-systolic volume (PSP/ESV) ratio in predicting symptomatic improvement with valve replacement in patients with aortic regurgitation and enlarged left von'lrlcular volume. We studied 21 patients (15 men and 6 women aged 15 to 60 years) with moderate or severe aortic regur- gitation, no other cardiovascular abnormalities and left ventricular end-systolic volume over 60 m11m2. In this group we assessed the preoperative variables which routinely were measured at cardiac catheterlzation to predict symptomatic improvement with valve replacement. Six months after operation, symptoms were alleviated in 13 patients(62%), and unchanged in 8()8%). By multivariate analysis, the PSP/ESV rati was a strong predictor for functional class 6 months after surgery(p=0.005) and also for change- in functional class prior to an operation to 6 months postoperatively(p=0.0)2). By 6 months after receiving valve replacement, all patients with a ratio over 1. 71 mmHglml/m'were in functional class I or II , in contrast, of those with a ratio < 1.71 mmHg/ml/m2, 40% were in functional class III. The PSP/ESV ratio may help to predict which patients with aortic regurgitation and enlarged left ven- tricular end-systolic volume will have symptomatic improvement with valve replacement.

Changes in Anxiety and Vital Signs of Scaling Patients (스케일링 전과 후의 불안 및 Vital Sign 변화)

  • The purpose of this study was to investigate the changes in the vital signs and anxiety reactions during scaling to patients who had received the dental hygiene treatment at department of Dental Hygiene in a university. A total of 189 patients were participated in the study. The level of anxiety was assessed through a dental anxiety questionnaire consisting of dental fear survey scale and dental anxiety survey scale. The vital signs were measured before and after scaling. The obtained results were as follows. The level of anxiety before scaling was 2.16 and after scaling was 1.96. This was significantly higher in women. The pulse and systolic blood pressure after scaling significantly decreased from 77.48 times to 74.36 times per minute and from 124.96 mmHg to 122.50 mmHg respectively. These changes are within the normal physiological variations. Individual changes in respiration, pulse, blood pressure, body temperature are affected by age, gender and previous dental experience, but previous scaling experience. The level of anxiety was significantly explained by respiration before scaling and pulse after scaling. As a result, scaling was associated with dental anxiety reactions and changes in vital signs.

Comparison of Lower-Limbs Muscle Activity according to the Abdominal Co-contractive Activation (복부 동시수축 형태에 따른 하지 근활성도 비교)

  • The purpose of this study was to investigated the muscle activity difference of lower extremity by abdominal co-contractive activation. 30 healthy women without pain in the low back and lower extremity were conducted. Muscle activities of lower extremity were measured with the active straight leg raise (ASLR) in 20cm raised from the floor without bending the knee at rest, abdominal drawing-in maneuver (ADIM) and abdominal bracing maneuver (ABM), respectively. Hip adductor longus (HadL), rectus femoris (RF), and biceps femoris (BF) muscle activity in the Abdominal bracing maneuver were statistically significantly higher, also gluteus medius (Gmed) and tibialis anterior (TA) muscle activity in the ADIM were higher. We provide the evidence to the positive effects of individual abdominal co-contractive activation training which can enhance postural stability and lower extremity strengthening with volitional preemptive abdominal contraction.

The Effect of Silymarin and Ethanol Intake on Vascular Contractility (엉겅퀴 유래 Silymarin의 단독 및 알코올 병용 시 혈압 조절 효과)

  • In the study, we endeavored to assess the convergence effect of Silybum marianum-derived silymarin and epidemiologically-correlated alcohol intake on vascular contractility and to determine the mechanism involved. There were few reports addressing the question whether thin or thick filament modulation is included in ethanol and silymarin-induced regulation. We hypothesized that ethanol at a low concentration and silymarin play a role in agonist-dependent regulation of vascular contractility. Denuded arterial muscles of Sprague-Dawley male rats were suspended in organ baths and isometric tensions were transduced and recorded using isometric transducers and an automatic data acquisition system. Interestingly, both silymarin and ethanol didn't encourage silymarin alone-induced inhibition in agonists-induced contraction suggesting that endothelial nitric oxide synthesis might be involved in ethanol or silymarin-induced modulation of vascular contractility and additional pathways besides endothelial nitric oxide synthesis such as ROCK inactivation might be involved in the silymarin-induced modulation of vascular contractility.
