Isolation and production of soymilk-clotting enzymes from Bacillus sp. K-324-7 (대두유 응고효소 생산에 관한 연구)

  • A bacterial strain which was capable of producing extracellular soymilk-clotting enzyme was isolated from soil samples during the course of screening test. The characteristics of the isolated strain K-324-7, indicated that the strain belonged to species of Bacillus cereus. The crude purification of this enzyme was precipitated by salting out with ammonium sulfate of 0.8 saturation. The optimal pH for the enzyme activity was at $6.1{\sim}7.0$ and below $50^{\circ}C$. The optimal culture medium for the production of soymilk-clotting enzyme were consisted of 0.2% glucose, 0.2% peptone, and 0.5% $KH_2PO_4$ with initial pH value of 6.5. The activity of enzyme was maximum when the microbe was cultured for 3 days at $35^{\circ}C$.

Characteristics of Pichia anomala K15 Producing Killer Toxin Isolated from Traditional Nuruk (전통 누룩으로부터 분리된 Killer Toxin 생산 균주 Pichia anomala K15의 특성)

  • In this study, killer yeasts were isolated from traditional Nuruk to improve storage and suppress contaminant in food industry. Among killer yeasts, yeast K15 showed strong killer toxin activity and inhibited growth of Salmonella Typhimurium and Vibrio parahaemolyticus. Killer yeast K15 was identified with Pichia anomala by the Microlog TM 4.0 identification system and homology of the ITS sequence. Killer toxin generated from P. anomala K15 was inactivated by pronase E and suggested to be a protein. Therefore killer toxin of P. anomala K15 was thought to be safe in human such as bacteriocin. P. anomala K15 was sufficient for growth in 50% glucose and could be used to prevent contaminant in initial stages of alcohol beverage fermentation.

  • Glucosinolates, containing indole glucosinates in Chinese cabbage, cabbage, radish and kale were analyzed as 6, 5, 5 and 2 types of glucosinolates, respectively, Four types of glucosinolates from mustard was identified, but indole glucosinolates were not detectable. In Chinese cabbage, the content of glucosinolates showed higher amount in the bottom, stalk and leaf part in the order, Total amounts of glucosinolates, in dry basis, were higher amount in Chinese cabbage, cabbage, radish, kale and mustard in the Order, and also in processing conditions, showed higher amount in fresh, sun dried, oven dried and cooked samples in the order. During the storage in Chinese cabbage and cabbage, total amounts of glucosinolate decreased gradually as time goes by.

Extraction of Nonlinear Dynamical Component by Wavelet Transform in Hydro-meteorological Data (수문기상자료의 웨이블렛 변환에 의한 비선형 동역학적 성분의 추출)

  • In the present study, we applied wavelet transform to decompose the hydro-meteorological data such as precipitation and temperature into the components with different return periods with a primary objective for extraction of nonlinear dynamical component. For the transform, we used the Daubechies wavelet of order 9 ('db9') as a basis function. Also, we applied the correlation dimension analysis to determine whether or not the detail and approximation components at the respective decomposition stage with the increasing of scale in the wavelet transform reveal the nonlinear dynamical characteristics. In other words, we proposed the combined use of the wavelet transform and the correlation dimension analysis as methodology to extract the nonlinear dynamical component from the hydro-meteorological data. The derived result has shown the method proposed in the present study is suitable for the segregation and extraction of the nonlinear dynamical component which is, in general, difficult to reveal by using the raw data.

Effect of Dietary Lipid on Fatty acid Pattern in Developing Brain Mitochondria in Rats (식이 중 지방 함량이 성장기 쥐의 뇌 미토콘드리아 지방산 조성에 미치는 영향)

  • 식이 중 지방함량이 성장기 뇌의 미토콘드리아 지방산 조성에 미치는 영향을 살펴보기 위해 옥수수기름을 각각 10, 2, 0.5% 포함한 3종류의 식이를 임신한 Sprague Dawley종의 흰쥐에게 임신말기부터 제공했다. 수유기간 중의 영향을 살펴보기 위해 대조군(10%옥수수기름 섭취)의 일부에 분만 후 0.5% 결핍식이를 섭취시켰고, 또 이유후부터 0.5% 결핍식이군의 일부에 10% 정상식이를 주어 회복효과를 관찰하고자 하였다. 전 실험기간 동안 체중과 뇌 무게, 단백질 함량을 측정하였고 뇌에서의 미토콘드리아를 분리하여 지방산 조성을 관찰하였다. 출생후 수유기간 동안에는 대조군에 비해 지방결핍식이군에서 체중과 뇌의 무게가 다소 높았으나 이유 후 부터 점점 감소하여 유의적인 차이를 볼 수 없었고 특히 0.5% 식이군의 체중은 생후 6주와 7주에 유의적인 감소를 보였다. 그러나 체중에 대한 뇌무게와 뇌의 단백질 함량에 있어에는 대조군과 결핍군 사이에 유의적인 차이를 볼 수 없었다. 따라서 지방결핍이 뇌 성장에 미치는 영향은 다른 영양소에 비해 크지 않음을 알 수 있었다. 뇌 미토콘드리아의 지방산 조성은 나이와 식이에 따라 변하였다. 나이를 먹음에 따라 단쇄 포화 지방산은 감소하고 다 불포화 지방산의 함량은 증가하였다. 또한 결핍식이군과 대조군 사이에 뇌 미토콘드리아 지방산조성을 살펴 본 결과 지방 결핍의 좋은 지표가 되고 불포화도, triene 과 tetraene의 비, 그리고 ${\omega}-6$ 계열과 ${\omega}-9$ 계열의 비에 있어서 큰 차이를 볼 수 있었다. 이들로 부터 성장초기에 지방함량을 조절한 식이에 의해 뇌 미토콘드리아 지질 조성이 크게 변하고 이에따라 미토콘드리아의 기능도 유의적인 영향을 받고 있음을 알 수 있었다.

Characteristics and breeding of a new brown variety 'Geumhyang' with short cultivation period in Flammulina velutipes (재배기간이 짧은 갈색 팽이버섯 신품종 '금향'의 육성 및 특성)

  • 'Geumhyang', a new variety with short cultivation period and yellowish ivory color in Flammulina velutipes, was bred by crossing between monokaryons isolated from 'Garlmoe' and wild strain 'CBMFV-33'. In bottle cultivation, the period of mycelial growth, primordia1 formation, and growth of fruit body was required 23 days, 7 days, and 17 days, respectively. Total culvivation period was 47 days, showing 7 days shorter than that of 'Garlmoe'. Pileus diameter was greater than that of 'Garlmoe', and pileus color showed yellowish ivory. Fruit body production per 850 ml bottle was 160 g, which was 8% higher than that of 'Garlmoe'. The taste, derived from 'Garlmoe' and wild strain, was excellent because of better chewing feeling than white ones. 'Geumhyang' with differentiated color and taste, has an advantage to target a niche market of white ones.

  • The present investigation demonstrates HLA-DR and PGP9.5 double positive cells accumulate thymus cortical region in normal baboon thymus and baboon lung. But, these cells disappeared in thymus and lung of bronchopulmonary dysplasia (BPD) animals. 125d GC animal model is more suitable for BPD than 140d GC animal. Anti-bombesin antibody, 2A11 treated baboon recover normal level of HLA-DR positive cells from BPD animal. In addition, thymocytes show responsiveness for bombesin. These observation suggest that blocking BLPs protects a chronic lung injury by BPD and 2A11 is possible agent for passive therapy of BPD.

Seed Structure and Effects of Storage on Germination of Astragalus membranaceus (황기 종자구조 및 저장이 휴면에 미치는 영향)

  • This experiment was conducted to examine the seed structure and to investigate the changes of seed vigor and germinability according to storage condition of Astragalus membranaceus. In case of the hard seed, there observed a gap between a testa and an embryo with abnormal shapes of an hilum and its micropyle. Seeds stored at room temperature maintained vitality for 4 months but continued to lose its vitality, while increase the hand seeds ratio as storing time longer. The content of Ca increased in proportion to the length of storing time.

Effect of Short-term High $CO_2$ on Growth of Botrytis cinerea (고농도 이산화탄소의 단기 처리가 Botrytis cinerea 생장에 미치는 영향)

  • This study was conducted to evaluate the inhibition efficacy of high CO$_2$ atmosphere (35, 60, and 100% ${\times}$ 24, 48, and 72 h) on growth of Botrytis cinerea in vitro in order to offer sterilizing method of horticultural crops including peach fruits. Botrytis cinerea was isolated from a naturally infected peach fruits. Growth of the fungus at 25$^{\circ}C$ declined with increased CO$_2$ concentration and treatment duration. Especially, 100% CO$_2$ provided completely inhibition effect of growth of the fungus for 72 h. After removal of high CO$_2$ condition, however, the fungus showed normal growth speed. The growth of fungus at low temperature was completely inhibited temporarily by short-term 100% CO$_2$ treatment, but resumed right after transferring to normal atmosphere at 25$^{\circ}C$.

Use of Visible Storage for Display and Conservation Treatment of a Large-sized Jar Coffin (대형 옹관의 보존처리 과정과 이를 활용한 개방형 수장고의 운용)

  • This paper describes the restoration and display of a large jar coffin at the Naju National Museum. Excavated from Tomb No. 3 at Bokam-ri, Naju, the large jar is 194 cm in height and its rim diameter is 108 cm. The jar had been broken into approximately forty fragments prior to its restoration, and a supporting structure was installed to ensure the safety of the relic during the restoration process. The restoration was conducted inside a visible storage for large jar coffins for approximately three weeks from February 5 to February 27, 2018. Over two weeks during this period, the restoration efforts inside the storage were visible to the public in the permanent exhibition space so that visitors could observe the process in person. This was planned to help visitors understand and arouse their interest in large-sized relics such as jar coffins and the restoration process. This study also investigates the practices of overseas institutions in order to suggest plans for raising public awareness of conservation treatment at conservation science centers.
