DNA 검정우에서 FSH 호로몬 투여가 난포란의 채란율에 미치는 영향

  • 초음파기기를 이용한 난포란의 채란 기술은 유전적으로 우수한 수정란의 다량확보가 가능함으로써 가축개량에 널리 응용할 수 있다. 그러나 초음파유도 난포란의 채란은 무엇보다도 회수율과 회수한 난포란의 질적 등급의 개선이 이루어져야 할 것이다. 본 연구는 등지방층두께, 일당증체량, 근내지방도 및 배최장근 단면적에 연관된 DNA marker가 검정된 한우로부터 FSH(FOLLTROPIN-V, Vetrepharm, Canada)를 투여하여 FSH 투여전.후의 포의 발달율과 등급별 회수율을 조사하였다. 공시동물의 발정주기에 관계없이 매주 무처리와 채란 12시간 전에 FSH를 각각 l00mg 과 200mg을 근육 주사하여 난포란의 채취를 유토하였다. 난소내 난포란의 채란은 SONOACE-600형 (Medison CO., 한국) 초음파기기를 사용하여 모니터 상에 검게 나타나는 난포를 17-gauge needle (Cook, Australia) 로써 70mmHg로 유지된 regulated vacuum pump를 이용하여 모두 채란하였으며, 소난포는 손가락 촉지를 이용하여 채란하였다. 난포의 크기는(large : $\geq$6mm, medium : 2-6mm, small $\leq$2mm) 무처리구에서 small(56.6%), medium(33.0%), large follicles(10.4%) 순으로 나타났으며, 전체 난포수는 106(8.2$\pm$3.6개) 였다. FSH 200mg 투여구에서는 medium(47.5%), small(32.5%), large follicles(20.0%) 순으로 나타났으며, 전체난포수는 40(10.0$\pm$2.2개) 였다. FSH l00mg 투여구에서는 medium(45.6%), small(43.5%), large follicles (10.9%) 순으로 나타났으며, 전체 난포는 92(7.7$\pm$2.5개) 였다. FSH 투여전 초음파 image 상으로 관찰한 난포수와 FSH 투여 후 초음파기기와 손가락 촉지를 이용하여 난포의 발달 및 난포란의 채란율은 200mg 투여 구에서는 투여 전에 4.3$\pm$0.6개였으나 투여 후에는 9.3$\pm$2.1개로 증가하였으며, 채란율은 75.0% 였다. l00mg 투여구에서도 투여 전후에 각각 5.3$\pm$1.2개 및 8.2$\pm$2.3개였으며, 채란율은 81.6%였다. 난포란의 채란율은 무처리구에서 88.7%(7.2$\pm$3.9개)로 나타났으며, 회수한 난포란의 등급은 각각 0.0%(GI), 5.3%(GII), 28.T%(GIII), 66.0%(GIV)로 나타났다. FSH 200mg 투여구의 채란율은 87.5%(8.8$\pm$2.6개)로 나타났으며, 등급별회수율은 GI(2.8%), GII(8.6%), GIII(34.3%), GIV(54.3%)으로 나타났다. 뿐만 아니라 FSH 100mg 투여구에서도 회수율은 89.1%(6.8$\pm$5.2개)였으며, 등급별 회수율은 각각 GI(8.5%), GII(13.4%), GIII(43.9%), GIV(34.2%)로 나타났다.

Study on Metagonimus yokogawai(Katsurada, 1912) in Korea VII. Susceptibility of Various Strains of Mice to Metagonimus Infection and Effect of Prednisolone (요꼬가의흡충에 관한 연구 Vll. 마우스 Strain별 감자성 및 Prednisolone의 영향)

  • An experimental study was undertaken to observe the difference in susceptibility of mouse strains to Metagonimus yokogawai infection by estimating it from worm recovery rate and dimension of worms. It was also studied the effects of prednisolone injection on the chronological pattern of worm recovery in ICR mice. The metacercariae were obtained from sweetfish and 300 in each number were given to 5 strains (CBH, A, DBA, $C^{57}BL$ and KK) of mice, and after 7 days period, the worms were collected from their intestine. Prednisolone at the dose of 10 mg/kg was injected to ICR mice every other day from 7 days prior to infection until sacrificed at 6 hours to 35th post-infection day. ICR mice infected with M. yokogawai but untreated were used for controls. The success rate in infection of mice ranged 25.0-83. 3% by strains, the worm recovery rate 1. 2-18. 9%, and the average size of worms O. 554-0. 683 mm long and 0.214-0.244 mm wide. The higher rates and larger size of worms were observed in KK and $C^{57}BL$ strains than others and the difference was statistically significant. In ICR mice for control, the worm recovery rate until 1 day after infection was relatively high (38-66%) but it became much lower (less than 0.7%) during 1-35 days. However, prednisolone injection brought about persistently high recovery rates (16-80%) until 21 days. It was concluded that the susceptibility to M. yokogawai infection is different by strains of mice but it can be elevated by prednisolone injection probably due to suppression of Immune respon3e3 in ICR mice.

Migration and Distribution of Spargana in Body of Experimentally Infected Mice (실험감염 스파르가눔의 마우스체내 이행경로 및 분포)

  • The migration and distribution pattern of spargana in mouse body was observed after experimental infection through mouth. The spargana were obtained from the snake, Natris tigrina lateralis, caught in Hoengseong-gun, Kangwon-do. A total of 28 male mice (ICR strain), 21∼259 in body weight, were fed each with 5 scolices (and necks) of spargana and killed after 10 minutes to 14 days. Systemic autopsy was performed on each mouse to recover the spargana. The results are as follows: 1. The spargana were found to penetrate into the stomach or duodenal wall of mice as early as 10 minutes after infection. They completed the penetration within 30 minutes and appeared in abdominal cavity. It was observed that spargana did not migrate tangentially along the gut wall but directly perforated the wall. 2. After 1 hour to 1 day the majority of spargana distributed in abdominal cavity of mice except a few which migrated to muscles or subcutaneous tissues. 3. It was within 7 days that nearly all of the spargana migrated to subcutaneous tissues. Out of total 28 in number found from subcutaneous tissues, 13 distributed around neck region, 12 around trunk and other 3 on head of mice and the most common sites were submandibular and subscapular areas. There was nearly no host tissue reaction to migrating spargana. 4. The initial length of spargana given was 4 mm in average but it increased to 12 mm after 7 days and to 35 mm after 14 days. The results suggest that spargana orally given to mice penetrate the gut wall within 30 minutes followed by escaping into abdominal cavity, and after passing through thoracic cavity or abdominal wall they anally Localize in subcutaneous tissues chieay around neck region within 7 days.

The Effect of Electroacupuncture on Reactive Gliosis Expressing GFAP in Rat with Transient Global Cerebral Ischemia (흰쥐 일과성 뇌허혈 시 GFAP으로 표지되는 반응성 신경아교세포증에 대한 전침의 효과)

  • The purpose of this study was carried out to investigate the effect of electroacupuncture on reactive gliosis expressing GFAP in rat with transient global cerebral ischemia. Subjects were randomly divided into two groups, a control group and a electroacupuncture group on ST36, LI11 and SP9 with 2 Hz and 1 mA. The rats were sacrificed on 1, 3 and 7 days after transient cerebral ischemia using ligation of left common carotid artery. After making brain slide sections, they were immunostained with GFAP antisera(1:2,500). The results were as follows: The numbers of astrocytes of electroacupuncture group were decreased than those of control group at every 1, 2 and 7 days. Especially, the numbers of astrocytes at 3 days(p<0.01) and 8 days(p<0.05) were different statistically. And astrocytes had resting, hypertrophic and moving types on cerebral cortex. The decrease of numbers of astrocytes expressing GFAP showed that electroacupuncture could localise and minimize the brain damage by transient cerebral ischemia and cause brain cell plasticity.

Effect of PNF Combination Patterns on Muscle Activity of the Lower Extremities and Gait Ability in Stroke Patients (PNF 결합패턴이 뇌졸중 환자의 하지 근 활성도 및 보행능력에 미치는 영향)

  • The purpose of present study was to determine effects of a PNF combination pattern training on muscle activity of lower extremities and gait ability in hemiparetic subjects. Twenty chronic stroke patients participated. Participants were randomly divided into either control group and experimental group. Experimental group received PNF combination pattern training, four times per a week for six weeks and control group received general exercise training. For the lower limbs muscle activity, RMS of action potential were analyzed and gait ability tests was conducted with 10MWT, DGI and F8WT. For the significance test of control and experimental group for measuring time by exercise application method, two-way repeated measure ANOVA. As the result, muscle activity of RF(p<.05), VM, TA, LH, LG was significantly increased between measurement period(p<.001), In VM, TA, LH, LG, there were interactions each measurement time and between group. 10MWT, DGI, F8WT in gait ability was significantly improved between measurement periods(p<.001) and In 10MWT, DGI, F8W, there were interaction each measurement time and between group. In conclusion, the PNF combination pattern application will be effective strategy to increasing the muscle activity and improving gait ability in the rehabilitation of stroke patients.

Facial Expression Animation which Applies a Motion Data in the Vector based Caricature (벡터 기반 캐리커처에 모션 데이터를 적용한 얼굴 표정 애니메이션)

  • This paper describes methodology which enables user in order to generate facial expression animation of caricature which applies a facial motion data in the vector based caricature. This method which sees was embodied with the plug-in of illustrator. And It is equipping the user interface of separate way. The data which is used in experiment attaches 28 small-sized markers in important muscular part of the actor face and captured the multiple many expression which is various with Facial Tracker. The caricature was produced in the bezier curve form which has a respectively control point from location of the important marker which attaches in the face of the actor when motion capturing to connection with motion data and the region which is identical. The facial motion data compares in the caricature and the spatial scale went through a motion calibration process too because of size. And with the user letting the control did possibly at any time. In order connecting the caricature and the markers also, we did possibly with the click the corresponding region of the caricature, after the user selects each name of the face region from the menu. Finally, this paper used a user interface of illustrator and in order for the caricature facial expression animation generation which applies a facial motion data in the vector based caricature to be possible.

Effects of integrated obesity management program on obese elementary school children (초등학교 비만아동을 위한 통합비만관리프로그램의 효과)

  • The purpose of this study was to develop a integrated obesity management program and measure the effects of the program on obesity, life style, dietary self-efficacy and exercise self-efficacy among obese elementary school children. The integrated obesity management program was provided for the experimental group of 4~6th grade elementary school students for 8 weeks. The program included obesity related health educations based on behavior modification and self-efficacy once a week, exercise classes three times a week. Method: There were 30 children with obesity in the experimental and 28 in the control groups. The collected data were analyzed by the 𝑥2-test, independent t-test and repeated measure ANOVA. Results: After 8weeks of participation in the program, the experimental group showed significantly lower body mass index (p<.01), percent body fat(p<.05) and obesity index(p<.01), significantly higher life style (p<.05)), dietary self-efficacy(p<.001), exercise self-efficacy(p<.05) than those in the control group. Conclusion: This study provides evidence that integrated obesity management program is effective nursing intervention for managing child obesity.

Long-term Feeding Effects of Different Dietary L-ascorbic Acid Levels-on Growth and Tissue Vitamin C Concentrations in Juvenile Korean Rockfish (조피볼락 치어의 장기간 사육에 있어서 사료내 L-ascorbic acid 농도가 성장과 조직내 Vitamin C 농도에 미치는 영향)

  • A long term feeding trial was conducted to study the effects of different dietary vitamin C levels on growth and its tissue distributions in juvenile Korean rockfish, Sebastes schlegeli. Prior to the start of feeding trial, fish were fed the basal diet supplementing no L-ascorbic acid for four weeks to minimize their body reserves of vitamin C. Then fish were divided into six groups with triplicates and given one of the laboratory semipurified diets supplementing either 0, 25, 50, 75, 150, or 1500 mg L-ascovbic acid (AA)/kg diet $(C_0,\;C_{25},\;C_{50},\;C_{75},\;C_{150},\;&\;C_{1500})$. Fish fed the $C_0$ diet had lower percent weight gain, feed conversion ratio, specific growth rate, and protein efficiency ratio than did fish fed the other diets (P<0.05). After 28 weeks of feeding trial, tissue AA concentrations of fish fed $C_0$ diet were lower than those of fish fed $C_{1500}$ diet (P<0.05). A large amount of total tissue Ah may be reserved in muscle, but the unit AA concentration seemed to be higher in brain than did the other tissues. The growth performances of fish fed $C_{25}$ diet were not different compared to those of fish fed $C_{50}-C_{1500}$ diets (P>0.05), and diet analysis of vitamin C showed that the $C_{25}$ diet had 65 mg AA/kg diet. Therefore, the present long-term study may suggest that the dietary vitamin C requirement is approximately 65 mg AA/kg diet in juvenile Korean rockfish.

Effect of Gender-specific Bovine Serum Supplemented Medium on Cell Culture (성별 특이 소 혈청이 세포 배양에 미치는 영향)

  • The research has been aimed to investigate the effect of different sera including fetal bovine serum (FBS), male bovine serum (MS), female bovine serum (FS), and castrated-male bovine serum (C-MS) on cell proliferation, follicular maturation and ovulation in vitro. Established cell lines and primary cells were cultured in the culture media supplemented with different sera and cells proliferation was observed by cell counting and MTT assay. The results indicated that cell proliferation was significantly different for different serum source. Proliferation of bovine and human myogenic satellite cells was highest in MS. In contrast, proliferation of breast cancer cells and immune cells were the highest in FS and FBS, respectively. There was no difference in the rate of follicular growth, whereas the rate of ovulation was higher in FBS and C-MS. Finally, the wound healing effect and cell proliferation of 3T3-L1 cells showed that wound healing was fastest in FS and cell proliferation was higher in MS. These results suggest the importance of an optimal serum selection in the experiments involving cell culture system, and gender-specific Hanwoo sera may be used as a substitute to FBS.

Association between Myofascial Pain Syndrome and the Assessment of Pain and the related Function tests in female telephone directory assistance operators using VDT (VDT 사용 여성 전화교환원들의 근막동통증후군과 동통 및 기능평가 검사와의 관련성)

  • The Association between myofascial pain syndrome and some tests was assessed in a cross-sectional study of 904 female telephone operators using video display terminals(VDTs). 105 cases were diagnosed as Myofascial pain syndrome with symptom questionnaire, laboratory examination and physician's physical examination and 550 controls were defined to show only musculoskeletal symptoms using NIOSH symptom criteria. Data on demographics, musculoskeletal symptom and visual analogue scale(VAS) were obtained by questionnaire. Anthropometric measurements, pressure pain threshold, Grip & Pinch strength and laboratory test were conducted. No significant difference between case and control at comparing of demographics, occupational history and body measurements were observed. But, Grip strength, pressure pain threshold and VAS showed the statistical difference between case and controt. Age, pressure pain threshold of Rt. Upper trapezius and VAS were associated with myofascial pain syndrome in multivariate logistic regression analysis. This results suggest that, to diagnose of myofascial pain syndrome, the consideration of the objective aspects among patients' subjective symptom complaints through the VAS and pressure pain threshold is required.
