Studies on the Chemical Resistance of Phytopathogenic Bacteria II. Selective Effect of Chemical Resistance on the Rice Bacterial Leaf Blight Pathogen, Xanthomonas oryzae (Uyeda et Ishiyama) Dowson, to Agrepto (식물병원성 세균의 약제저항성에 관한 연구 II. 벼흰빛잎마름병균의 Agrepto에 대한 저항성의 선발효과)

  • The experiments were carried out to investigate the selection effects on the drug resistance to Agrepto in Xanthomonas oryzae, the causal bacteria of rice bacterial leaf blight. The results obtained were as follows. 1, The Agrepto was stable at the heat treatment of $105^{\circ}C$ for 20 minutes, when the drug was added in the media. 2. The local isolates of the bacteria, 75-6 and 75-9, showed the different resistante reaction, when they were selected by the Agrepto contained media which concentrations of 10r9/m1 and 100ug/ml. 3. The individuals shelving high degree of resistance, which can grow on the media contained 10,000ug/m1 of Agrepto, could selected by the concentrations as low as 10ug/ml and 100ug/m1, in one generation. 4. The highly resistant isolates which selected by 3-stepwise selections such as 100ug/ml, 3,000ug/ml and 10,000/ml plots, showed nearly normal growth at the media contained 100ug/ml of Agrepto 5. When the isolate 75-9 was selected at the 100ug/m1 of concentration, showed various degrees of resistance, indicating that the isolate may be composed of resistance groups that lower than 500ug/ml, between 500-1,000ug/ml and 1,000-3,000ug/ml, to the Agrept.

Screening and Characterization of Probiotic Strains for Prevention of Bacterial Fish Diseases (어류의 세균성 질병 예방을 위한 Probiotic균주의 선발 및 특성)

  • The purpose of the present study was to screen the effective of lactic acid bacteria (LAB) as probiotics, which are able to protect aquacultural fish pathogenic bacteria, and investigate their characterization. Twenty strains of lactic acid bacteria were isolated from fish intestine, fermented fish foods and kimchis. These bacteria were screened for antagonistic activity against fish pathogenic bacteria. Seven tested LAB strains were able to inhibit the fish pathogenic bacteria, including Vibrio anguillarum, Edwardsiella tarda, and Streptococcus sp.. Of the probiotic candidates, BK19 strain isolated from fermented pollack viscera indicated the largest inhibition activity. Moreover, this strain showed a resistance over low pH and antibiotic agents. Therefore this probiotic candidate BK19 was finally selected and identified as a probiotic strain. This particular probiotic bacteria was identified as Lactobacillus sakei BK19 by biochemical characteristics and 165 rRNA PCR amplification.

Selection of Antagonistic Soil Actinomycetes Against Both Colletotrichum orbiculare and Botrytis cinerea in Cucumber Plants (오이 탄저병원균과 잿빛곰팡이병원균에 억제 효과가 있는 방선균 선발)

  • The purpose of this study is to select actinomycetes with antifungal activity against Botrytis cinerea and Colletotrichum orbiculare, which are airborne pathogens in cucumber plants. In 2020, 560 species of actinomycetes were isolated from rhizome soils of various vegetables in Korea; 7 strains showing simultaneous antifungal activity against two pathogens were selected. Finally, strain S20-465 was selected through dual culture and plant assay. This strain was identified as Streptomyces sp. based on 16S rRNA analysis. The culture filtrate of strain S20-465 inhibited mycelial growth of both pathogens by more than 60%. In addition, when cucumber plants were treated with 20-fold and 40-fold diluted culture filtrates of S20-465, lesions caused by B. cinerea and C. orbiculare on cucumber leaves were significantly reduced compared to the control. This results suggest that strain S20-465 produces specific secondary metabolites with antifungal activity against both pathogens.

  • Fifty eight isolates of R. solani collected from rice plants infected to sheath blight disease at seven different locations through the country were evaluated for their cultural characteristics on media. Degree of resistance of nine rice varieties were tested by the seven isolates representing each culture type from the grouped pathogene-city. Great variation of cultural characterisics on PSA media was observed among isolates, and 58 isolates tested were grouped into 7 culture types. Locational distribution of isolates belong to each culture type was different between types, and only isolate type 'Ia' was distributed in all locations collected. The degree of pathogene-city of each isolate to rice adult plant was different between 7 culture types grouped, and no isolates infected severely on all tester rice varieties. The degree of resistance to the disease showed great variation among 9 tester varieties, and varietal reaction to the disease was varied along with isolates inoculated. An isolate, la, shown strong pathogenecity and distributed through 7 locations was selected for the screening of varietal resistance to rice sheath blight disease.

Selection of Bacillus amyloliquefaciens M27 for Biocontrol on Lettuce Sclerotinia Rot (상추균핵병의 생물적 방제를 위한 Bacillus amyloliquefaciens M27 선발)

  • For selection of effective antagonists for control Sclerotinia rot of lettuce, 29 bacteria were isolated from soil in Korea. The bacterial isolates M27 and RM43 identified as Bacillus sp, were selected as prospective agents for inhibiting mycelial growth of Sclerotinia sclerotiorum and Sclerotinia minor. Among the selected isolates, Bacillus amyloliquefaciens M27 effectively suppressed incidence of Sclerotinia rot by Sclerotinia sclerotiorum and Sclerotinia minor in the lettuce nursery.

  • This study was investigated the physiological properties of auxin-producing bacteria that have plant growth promoting activity and plant pathogen antagonistic ability. Auxin-producing bacteria were isolated from field soils of Gyeongsan, Korea. Selected strains were identified as a Pseudumonas fulva N21 and a Pantoea agglomerans; K35 by morphological and physiological test, and Biolog (Microlog) system. Auxins were determined by Salkowski in vitro test and mungbean adventitious root induction bioassay. Also produced indole-3-acetic acid (IAA) was identified by TLC. During cell growth, auxin production were highest in their idiophase after log phase and $35^{\circ}C$ at pH 7.5.

Development of Repetitive DNA Probes for Genetic Analysis of Phytophthora capsici (Phytophthora capsici의 유전적 특성 분석을 위한 Repetitive DNA Probe의 개발)

  • To develop DNA markers for analysis of genetic characteristics of Phytophthora capsici population, randomly selected clones from HindIII-digested genomic DNA library of P. capsici 95CY3119 were surveyed by hybridizing to Southern blots of HindIII-digested total genomic DNA of P. capsici. Probe DNAs inserted into selected individual clones strongly hybridized with HindIII digests of P. capsici. Among probes examined, PC9 revealed the repetitive and highly polymorphic bands to HindIII digests of inter-and intra-field P. capsici isolates. Genetic diversity of individual isolates was also clearly revealed in cluster analysis based on its band patterns. The other probe, PC22, was hybridized only to DNA from P. capsici and this was highly repetitive. However, there was no response to other Phytophthora species and Pythium sp. These DNA probes could be used as very useful markers in analysing genetic diversity and identification for P. capsici population throughout the world.

Selection of Probiotic Bacteria from Yulmoo Kimchi Using a Stimulated Human Intestinal Model System (인체장모델시스템에 의한 열무김치로부터 프로바이오틱스 균주 선발)

  • To select potent probiotics from lactic acid bacteria in Yulmoo Kimchi, an in vitro and stimulated human intestinal model system (SHIMS) test were performed. One Leuconostoc mesenteroides strain from five strains of Yulmoo Kimchi and one Lactobacillus plantarum from 12 strains of KCTC and KCCM were selected according to survival in acidic and bile salts conditions. Between the two species, Leu. mesentroides displayed higher survival activity in a SHIMS test. The strain was identified as Leu. mesentroides by 16S rRNA sequencing and was designated as Leu. mesentroides K01.

Isolation of Biogenic Amines-Degrading Strains of Bacillus subtilis and Bacillus amyloliquefaciens from Traditionally Fermented Soybean Products (전통장류에서 Biogenic Amines 분해 능력을 가지는 Bacillus subtilis 및 Bacillus amyloliquefaciens 균주의 분리)

  • In order to reduce harmful biogenic amines in the traditionally fermented soybean products, five isolates with biogenic amines-degrading property were obtained from 83 traditionally fermented soybean products. The strains were found to reduce biogenic amines including histamine, tyramine, putrescine, and cadaverine by 27 to 92% in the cooked soybean containing 5.3% of each biogenic amine over 10 days of incubation. The morphological and biochemical tests and the phylogenetic relationships based on 16S rRNA gene sequences indicated that the five isolates were most closely related to Bacillus subtilis or B. amyloliquefaciens. The use of selected strains would be a potential control measure in manufacturing traditionally fermented soybean products that are difficult to control biogenic amine levels.

  • After treatment of basidiospores of P. cornucopiae with ultraviolet light, 84 putative mutants from 4671 isolates were obtained. The highest proportion of auxotrophic mutants was obtained from the isolates irradiated to give $0.4{\sim}1.0%$ survival. Fourteen auxotrophs were selected for protoplast fusion and each genetic marker was identified.
