Fungicide Selection for Control of Lycium chinense Anthracnose Caused by Colletotrichum spp. (구기자 탄저병 방제를 위한 살균제 선발)

  • Anthracnose caused by Colletotrichum spp. has been known as a significant disease which commonly infects to the fruits of Chinese matrimony (Lycium chinese) in the field conditions. To select effective fungicides for the control of Chinese matrimony anthracnose, the antifungal activity of 15 fungicides were evaluated with 13 different strains of Colletotrichum spp. in the laboratory condition. Six fungicides (Dithianon WG, Tebuconazole SC, Tebuconazole WG, Trifloxystrobin + Tebuconazole SC, Azoxystrobin SC and Polyoxin D zinc salt + Carbendazim WP) out of them, showed effective suppression with the mycelium growth of pathogenic fungus, and were selected to test in vivo of the field condition. Five fungicides, Dithianon WG, Tebuconazole WG, Trifloxystrobin + Tebuconazole SC, Tebuconazole SC and Azoxystrobin SC, were significantly effective to protect anthracnose of Chinese matrimony, the variety "Chungyang Jerae".

Functional Genomic Analysis of Bacillus thuringiensis C25 Reveals the Potential Genes Regulating Antifungal Activity against Rosellinia necatrix (Bacillus thuringiensis C25의 흰날개무늬병 Rosellinia necatrix에 대한 항진균 활성에 관여하는 유전자 특성 및 기능 유전체학적 연구)

  • Biocontrol agents (BCAs) are widely used to protect plants from diverse biotic and abiotic stresses in agricultural and ecological fields. Among the various microbes, many subspecies of the gram-positive genus, Bacillus, have been successfully industrialized as eco-friendly biological pesticides and fertilizers. In the current study, we demonstrated that Bacillus thuringiensis C25 exhibited antagonistic effects on the mycelial growth of Rosellinia necatrix, a fungal phytopathogen. Scanning electron microscopy analysis revealed that B. thuringiensis C25 degraded the cell wall structures of R. necatrix mycelia. In the functional genomic analysis of B. thuringiensis C25, we annotated 5,683 genes and selected the gene sets that potentially encoded fungal cell wall degrading enzymes (CWDEs). The growth inhibition effects on R. necatrix were highly correlated with the transcriptional activity of the mycelial cell wall degrading genes of B. thuringiensis C25. The transcript levels of CWDEs, including CshiA, B, and Glycos_transf_2 genes in B. thuringiensis C25, were enhanced following co-cultivation with R. necatrix. In conclusion, our study suggested that B. thuringiensis C25 could serve as a suitable candidate for controlling R. necatrix and could facilitate elucidating the mechanisms underlying the antifungal activities of BCAs against phytopathogens.

Studies on the Germination Characters of Korean Ginseng (Panax ginseng C.A. Meyer) Seed (고려인삼종자(高麗人蔘種子)의 발아특성(發芽特性)에 관(關)한 연구(硏究))

  • This study was conducted to define the optimal conditions for embryo growth during seed stratification and for breaking dormancy as well as seed germination of stratified ginseng seeds. The experiments were also carried out to detect some materials which were expected to induce seed dormancy in the ginseng seeds. The results summarized as follows; 1. The growth of embryo during seed stratification was significantly inhibited by the existence of endocarp. The fastest embryo growth was resulted at $15^{\circ}C$ and an estimated optimal temperature for embryo growth was about $18^{\circ}C$. 2. There was no significant difference between the embryo growth and germination ratio of ginseng seeds which were sown in seed bed at Aug-5 without seed stratification and that of artificial seed stratification. 3. Embryo growth and germination ratio was significantly inhibited by high temperature treatment at $30^{\circ}C$ for 24 hours or respiration stress by immersing seeds in water for 10 days or more. 4. When the seed stratification was started at $10^{\circ}C$, growth of embryo in the ginseng seeds were almost stopped. But, when the seeds were stratified first at $20^{\circ}C$ for 50 days and next at $10^{\circ}C$ for 50 days, the embryo growth was significantly promoted compared with the embryo growth in the seeds which were stratified at $20^{\circ}C$ for 100 days. 5. The successive embryo growth after seed stratification was significantly accelerated at $10^{\circ}C$ but the seeds chilled at $5^{\circ}C$ for 100 days were resulted in the highest germination ratio as well as the shortest days for germination. 6. The successive embryo growth during chilling treatment and seed germination were significantly inhibited by immersing seeds in water just before chilling treatment or during chilling treatment and by interruption of chilling treatment with raising temperature to $20^{\circ}C$ for 20 days during chilling treatment. 7. The germination ratio of ginseng seeds which finished chilling treatment was highest at $10^{\circ}C$ and 62.5% was the estimated soil moisture for the best germination of ginseng seeds. The ginseng seeds were found to require high amount of oxygen for germination. 8. Only water soluble material in homogenized ginseng seeds showed a significant inhibiting effect on the seed germination of sesame, millet and soybean. Water soluble material dissolved from undehisced ginseng seeds showed stronger inhibiting effect on the seedling growth of sesame than material from dehisced ginseng seeds. Extraction temperature did not influence the inhibiting effect of the material dissolved from ginseng seeds on the seedling growth of sesame. 9. Water soluble materials dissolved from the berry pulps, leaves, fresh roots and dried roots also showed a significant inhibiting effect on the seedling growth of sesame. 10. Water soluble materials dissolved from the ginseng seeds, leaves and fresh roots showed a significant inhibiting effect on the germination of true fungi and the growth of spawn but the growth of phytopathogenic bacteria was not. 11. Among the water soluble materials dissolved from ginseng seeds, the materials of low molecular weight less than 3,000 were resulted a significant inhibiting effect on the seedling growth of sesame and the materials of high molecular weight also showed an inhibiting effect.

Cross-resistance of Colletotrichum acutatum s. lat. to Strobilurin Fungicides and Inhibitory Effect of Fungicides with Other Mechanisms on C. acutatum s. lat. Resistant to Pyraclostrobin (Strobilurin계 살균제에 대한 고추탄저병균의 교차저항성과 Pyraclostrobin 저항성균에 대한 다른 기작 살균제의 억제 효과)

  • Colletotrichum acutatum s. lat. 20JDS8 sensitive and 20CDJ6 resistant to pylaclostrobin were used to investigate the cross-resistance with fungicides belonging to the strobilurins and the characteristics of fungicidal controlling activities with different mechanisms against the isolate resistant to the fungicide. The resistant isolate of 20CDJ6 also showed the resistance to azoxystrobin, trifloxystrobin, and kresoxim-methyl, suggesting that there is a cross-resistance relationship. All fungicides with different action mechanisms inhibited mycelial growth of both susceptible and resistant isolates of C. acutatum s. lat., but their disease control effects in fruits were different according to the fungicides. The disease control effect of isopyrazam against 20JDS8 and 20CDJ6 was very low, and fluazinam showed a control effect of 91.9% and 88.1% against 20JDS8 and 20CDJ6 only when it was treated before inoculation by spraying spore suspensions on pepper fruits without wounds. Tebuconazole and prochloraz effectively inhibited not only the mycelial growth of 20JDS8 and 20CDJ6 on potato dextrose agar medium, but also disease incidence in red pepper fruits. As a result of this study, C. acutatum s. lat. 20CDJ6 resistant to pyraclostrobin showed cross-resistance with other strobilurin fungicides. In addition, we think that fluazinam, tebuconazole, and prochloraz can be recommended as alternative fungicides for the control of red-pepper pyranthracnose pathogens resistant pyraclostrobin. However, fluazinam can be effective only if it is treated protectively before the occurrence of the disease.

Biological Control of White Rot in Apple Using Bacillus spp. (Bacillus spp.를 이용한 사과 겹무늬썩음병의 생물학적 방제)

  • Apple white rot, caused by Botryosphaeria dothidea, is one of the important diseases in Korea. B. dothidea can cause pre- and postharvest decay on apple fruit as well as canker and dieback of apple trees. In this study, we isolated bacteria from the trunk of apple trees and tested their antagonistic activity against B. dothidea. Five bacterial isolates (23-168, 23-169, 23-170, 23-172, and 23-173) were selected that were most effective at inhibiting the mycelial growth of the pathogens. The isolate 23-172 was identified as Bacillus amyloliquefaciens and four isolates 23-168, 23-169, 23-170, and 23-173 were identified as Bacillus velezensis by RNA polymerase beta subunit (rpoB) and DNA gyraseA subunit (gyrA) gene sequencing. All isolates showed strong antagonistic activity against B. dothidiea as well as Colletotrichum fructicola and Diaporthe eres. All isolates exhibited cellulolytic, proteolytic and phosphate solubilizing activities. In particular, two isolates 23-168, 23-169 were shown to significantly reduce the size of white rot lesions in pretreated apple fruits. These results will provide the basis for the development of a fungicide alternative for the control of white rot of apple.

Biological Control of Soybean Anthracnose by Pseudomonas sp. (Pseudomonas sp.를 이용한 콩 탄저병의 생물학적 방제)

  • Pseudomonas sp. antagonistic to Colletotrichum truncatum and C. gloesporioies was selected as a biological control agent for soybean anthracnose. Pseudomonas sp. inhibited the mycelial growth of pathogens effectively as the funhicides such as benomyl and fluazinam in vitro tests without any adverse effects on soybean. Seed treatment with Pseudomonas sp. increased emergence rate of soybean seeds significantly after inoculaton with C. truncatum. When the suspension of Pseudomonas sp. was sprayed on soybean plants, the control efficacy was not different from that of fungicides, benomyl and fluazinam two weeks after treatment, however the efficacy did not last long enough after three weeks.

Response of Systemic Fungicides of Rhizoctonia spp. Causing Rhizoctonia Blight on Turfgrass (잔디에 Rhizoctonia 마름병을 유발하는 Rhizoctonia spp.의 침투성 살균제에 대한 반응)

  • The Rhizoctonia blight causing by Rhizoctonia spp. is an important disease of turfgrass, requiring fungicide application to maintain acceptable conditions for turfgrass good qualities in the golf course. The experiment was conducted to determine the mean 50% effective concentration inhibiting mycelial growth ($EC_{50}$) value of to flutolanil, pyraclostrobine and hexaconazole to Rhizoctonia solani AG-1 IB, Rhizoctonia cerealis and Rhizoctonia solani AG2-2IV isolated from Gyeongbuk province of Korea in vitro. Five discriminatory concentrations of each fungicide were used to detect in vitro sensitivity. The mean of $EC_{50}$ values to three systemic fungicides was the lowest isolate of R. solani AG-1 IB. However, the sensitivity of fungicides to Rhizoctonia solani AG2-2IV were higher mean $EC_{50}$ value of 0.026 ${\mu}g\;a.i.\;ml^{-1}$ of pyraclostrobine and 0.044 ${\mu}g\;a.i.\;ml^{-1}$ of flutolanil. R. cerealis was the lowest sensitivity to hexaconazole which was an average $EC_{50}$ value of 0.022 ${\mu}g\;a.i.\;ml^{-1}$. Inhibition of mycelial growth rate (%) by three combine fungicides using the $EC_{50}$ value of each fungicide was the highest R. solani AG2-2IV. Results of this study were may confirmed in vitro response fungicide of three Rhizoctonia species for control of Rhizoctonia blight in the field.

Characteristics of Phytophthora capsici Causing Pepper Phytophthora Blight Resistant to Metalaxyl (Metalaxyl에 대한 저항성 고추 역병균의 특성)

  • Isolation frequency of resistant isolates of Phytophthora capsici to metalaxyl was reported to be 38.9% through the resistance monitoring for metalaxyl in P. capsici causing pepper Phytophthora blight. Metalaxyl was very effective to mycelium growth, while not to zoosporangium germination and zoospore release. $EC_{50}$ values of metalaxyl in the inhibition of mycelium growth were 0.204, 0.151, 0.379, and $0.215\;{\mu}g\;mL^{-1}$ against each isolate sensitive to the fungicide as P. capsici 06-119, 06-143, P08-7, and P08-31, respectively, whilst those were 5.242, 5.724, 6.621, and $5.377\;{\mu}g\;mL^{-1}$ in P. capsici 06-125, 06-155, P08-50, and P08-60. For the field fitness, several factors, which were mycelium growth, zoosporangium germination, zoospore release, virulence to pepper plants, and the zoosporangium and the oospore production, were investigated with 4 sensitive isolates and 4 resistant isolates. Between 2 groups differentiated by the sensitivity of metalaxyl, there was no significance in mycelium growth, zoosporangium germination, zoospore release, and virulence to pepper plants. However, the zoosporangium and the oospore production in each resistant isolate, which were related to survival of P. capsici in fields, were superior to those of sensitive isolates. Based on results of this study, it was suggested that the increase of the percentage of resistant isolates to metalaxyl resulted from the high capacities of the zoosporangium and the oospore production.

Anti-melanogenesis Effect of Ganoderma lucidum Mycelial Extract Supplemented with Oriental Raisin Tree (Hovenia dulcis) Extract (헛개나무 추출물이 첨가된 영지버섯균사 추출물의 멜라닌 생성 억제효과)

  • The aim of this study is to investigate the effect of Hovenia dulcis (oriental raisin tree) extract on ganodermanondiol (GN) contents in Ganoderma lucidum (G. lucidum) mycelia. GN has a triterpenoid structure and is one of the major active components of G. lucidum. Furthermore, we previously proved its inhibitory effects on tyrosinase activity and melanin biosynthesis in B16F10 melanoma cells. In this study, we observed significantly increased GN contents in G. lucidum mycelial extracts supplemented with 15% (v/v) oriental raisin tree extract (ORTE) by HPLC analysis. In addition, melanogenesis was significantly inhibited by G. lucidum extract supplemented with 15% ORTE when compared to G. lucidum extract without ORTE supplementation. Furthermore, mycelial growth of G. lucidum was increased by ORTE supplementation in both solid and liquid cultivation. These results suggest that the oriental raisin tree is useful as natural ingredient for increasing GN biosynthesis as well as whitening effect of G. lucidum.

Antifungal Activity of Bacillus Subtilis HK2 against Trichothecium Roseum Causing Pink Rot of Melon and White Stain Symptom on Grape (멜론 분홍빛썩음병과 포도 흰얼룩병의 원인균인 Trichothecium Roseum에 대한 Bacillus Subtilis HK2의 항균활성)

  • Pink Rot on melon and White Stain Symptom on grape are caused by Trichothecium roseum, one of the most important diseases of grape and melon. These diseases have been occurred in national-wide in Korea and causes irreversible damage on the grape and the melon at harvest season. This research presents the evaluation of the capacity of Bacillus subtillis HK2 to protect both melon and grape against T. reseum and establishes its role as a biocontrol agent. In this study, we isolated a Bacillus strain HK2 from rhizosphere soil, identified it as Bacillus subtillis by 16S rRNA analysis and demonstrated its antifungal activity against T. roseum. Under I-plate assay it was observed that the effect of hyphal growth inhibition was not due to production of volatile compounds. The optimum culture condition of HK2 was found at 30℃ and initial pH of 7.0. Application of HK2 culture suspension reduced 90.2% of white stain symptom on grape as compared to control, resulting in greater protection to grape against T. roseum infestation. Butanol extract of HK2 culture purified using flash column chromatography. The antifungal material was a polar substance as it showed antifungal activity in polar elute. Therefore, our results indicated a clear potential of B. subtilis HK2 to be used for biocontrol of Pink rot in melon and white stain symptom on grape caused by T. roseum.
