인체의 귀는 물리적인 크기에 비해 탁월한 능력을 가지고 있는 감각기관이다. 귓바퀴에 모여진 미소한 압력변동은 4cm의 길이에 놓여져 있는 귓구멍, 중이 및 내이 를 통해 작게는 수백 배, 크게는 1,000배까지 기계적으로 증푹시킨다. 불과 완두콩 만한 크기의 달팽이관에서는 소리의 스펙트럼을 순간적으로 분석 처리하며, 비선형 적으로 신호를 압축시키는 응답특성을 가지고 있다. 또, 수많은 청신경 세포가 내재 되어 있어, 들은 소리를 대뇌에서 이용할 수 있는 신경 신호로 변환시켜 준다. 귀가 가지고 있는 신비로운 능력중에서 우리가 알고 있는 지식은 극히 일부에 머무르고 있으며 아직도 많은 의문점이 해답을 기다리고 있다. 국내에서도 하루빨리 공학과 의학 분야가 협력하여 우리가 모르고 있는 청감의 신비를 규명하고 소음이 인체에 미치는 영향에 대한 한국판 연구 결과가 축적될 수 있기를 기대한다.
This study was conducted to examine the effects of electric stimulation conditions on in vitro developmental ability of caprine embryos after somatic cell nuclear transfer. Recipient oocytes were surgically collected after superovulation by using CIDR and FSH, PMSG, hCG and estrous synchronization in Korean native goats. The caprine ear cells were cultured in vitro in serum-starvation condition (TCM-l99 + 0.5% FBS) for 3 to 5 days of cell confluence. The zona pellucida of in vivo and in vitro matured oocytes were partially drilled using laser system. Single somatic cell was individually transferred into the enucleated oocyte. The reconstructed oocytes were electrically fused with 0.3M mannitol. After the electofusion, embryos were activated by electric stimulation or Ionomycin + 6-DMAP. Nuclear transfer embryos were cultured in mSOF medium supplemented with 0.8% BSA 6∼7 days at 39 , 5% $CO_2$, 5% $O_2$, 90% $N_2$. The fusion rate of donor cells was 60.4% and 40.3 % in ear cell and fetal fibroblast, and cleavage rate were 40.6% and 48.2%, respectively. No significant difference was found in the fusion and cleavage rate in different donor cells. Nuclear transferred oocytes were fused by electric pulses of 1.30∼1.40, 2.30∼2.39 and 2.40∼2.46 ㎸/cm. There was no significant difference among different electric pulses in fusion rates (26.7, 34.8 and 43.8%). The cleavage rate was higher (p<0.05) in 1.30∼1.40 ㎸/cm (82.9%) than 2.30∼2.39 ㎸/cm (43.8%) and 2.40∼2.46 ㎸/cm. (51.8%). The fusion rates of recipient oocyte source were 1st (43.5% and 23.6%), 2nd (55.7% and 39.2%) and 3rd (66.1% and 52.8%) in in vivo and in vitro oocytes. However, fusion ratee were significantly higher (p<0.05) in in vivo than in vitro oocyte. The cleavage rate of fused oocytes from in vivo and in vitro sources were 52.6% and 54.4%, respectively. No significant difference was found in the cleavage rate according to the recipient oocyte source. These results suggest that factors such as field pulse of electric stimulation and oocyte source could affect in vitro developmental ability of nuclear transplanted caprine oocytes.
Analysis of random amplified polymorphic DNAs(RAPDs) and internal morphological features were performed using three species of medicinal plants in the genus of Angelica(A. gigas Nakai, A. sinensis(Oliv.) Diels., A. acutiloba Kitagawa) to distinguish between these three species. Fifty decarmer oligonucleotide primers were screened for the RAPDs of the herbal plant species. Five primers generated distinct RAPD markers specific to the species of Angelica, In analysis of the degree of similarity, A. sinensis(Oliv.) Diels is more closely related to A. acutiloba Kitagawa than to A. gigas Nakai. Furthermore, we proved the usefulness of RAPD analysis for the discrimination of the species using dry roots and commercial plant materials. Mast cell distribution was quantified in acidified toluidine blue sections of normal skin from 8 different sites in 10 dogs and compared to the predilection sites of canine atopic dermatitis. Mast cell counts varied significantly from site to site (p<0.0001) and counts in the superficial dennis were significantly higher than the deeper dennis (p<0.05). The highest mast cells distribution sites were the concave surface of the ear (mean $74.88{\pm}17.93\;per\;mm^{2}$) and the interdigital skin of the forefeet (mean $28.326{\pm}6.24\;per\;mm^{2}$). Counts in these sites were $280\%$ higher than all the other sites. Our results may provide some evidence that cutaneous mast cell distribution may be a factor in the frequent occurrence of ear and foot pruritus in atopic dermatitis. However, the low mast cell count in the predilection sites of atopic dermatitis did not explain the common occurrence of atopic lesions. Therefore, other factors or more complicated pathogenesis may be correlated with these predilection sites.

In this study, the anti-inflammatory effect of Polyopes affinis ethanol extract (PAEE) was investigated using LPS-stimulated RAW 264.7 cells and a croton oil-induced ICR mice model. Treatment with PAEE significantly reduced production of nitric oxide (NO) and pro-inflammatory cytokines [interleukin (IL)-6, tumor necrosis factor $(TNF)-{\alpha}$, and $IL-1{\beta}$] in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. PAEE treatment also reduced expression of inducible NO synthase, cyclooxygenase-2, nuclear $factor-{\kappa}B$, and mitogen-activated protein kinases in LPS-stimulated RAW 264.7 cells. In the croton oil-induced ear edema test, application of PAEE (10~250 mg/kg body weight) reduced ear edema in a dose-dependent manner, and PAEE treatment at 50 mg/kg body weight showed similar inhibitory effects compared with prednisolone (10 mg/kg body weight). Histological analysis revealed reduced dermal thickness and lower number of infiltrated mast cells. These results suggest that PAEE might be used as a promising anti-inflammatory agent for inhibition of LPS-induced inflammation and ear edema formation.
This study was conducted to investigate the effect of 70% ethanol extract (DR) on the mast cell-mediated allergic contact dermatitis induced by dinitrofluorobenzene in BALB / c mice, which affects the cell activity by antigen in RBL-2H3 mast cells Respectively. The ethanol extracts of RBL-2H3 cells activated by DNP-HSA and anti-DNP IgE antibodies inhibited the secretion of ${\beta}$-hexosaminidase, histamine, and IL-4 and $TNF-{\alpha}$ Production was suppressed. In the DNFB-induced contact allergic dermatitis animal model, treatment with ethanol extract reduced ear swelling and inhibited serum histamine and IL-4 secretion, and DR treatment effectively prevented mast cell infiltration in dermatitis-induced areas. As a result, the ethanol extract may be used as a therapeutic agent for mast cell-mediated allergic diseases such as atopic dermatitis.
Ultra-fine powder (FA) extracts from persimmon were investigated as potential food materials in industrial processes because such extracts have various useful biological activities. We examined the antioxidant and anti-allergic activity of FA extracts using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and ferric reducing activity of plasma (FRAP) models, and by measuring interleukin 4 (IL-4) expression in spleen cells and in an atopy animal model. FA extracts were prepared using solvents including distilled water and 50% (v/v) acetone. The results showed that FA extracts had potent DPPH and FRAP activities, decreased IL-4 expression in spleen cells, and inhibited ear thickness and inflammation in a mouse animal model. The data indicate that a FA extract could be used as an antioxidant and/or anti-inflammation agent, and in industrial food preparation processes.
Inflammatory polyps in feline ear are nonneoplastic, inflammatory growths that arise from the middle ear or the eustachian tube and extended into the pharynx or external ear canal. Two 2-year-old female Russian blue cats showed 2-3 weeks history of aural discharge, crust formation in external ear, and head or ear shaking. Two masses were surgically excised from ear canal, and submitted for diagnosis. Histopathologically, these masses were covered with hyperplastic ciliated epithelium or nonkeratinizing squamous epithelium with partial erosion and ulceration. The core of masses was consisted of proliferated connective tissue and massive infiltration of mononuclear cells. Immunohistochemically, about 90% of infiltrated mononuclear cells demonstrated CD3 positive T cell. According to both polymerase chain reaction (PCR) and reverse transcriptase-PCR, tissues samples were negative for feline viral pathogens. Based on the clinical, gross, histopathologic findings, these two cases were diagnosed as inflammatory polyps originated from the middle ear in cats.

In this study, we investigated antimicrobial and cytotoxicity effects of Undaria pinnatifida Sporophyll, which using methanol, dichloromethane and ethanol were extracted and fractionated into four different types: methanol (UPMM), hexane (UPMH), butanol (UPMB) and aqueous (UPMA). The antimicrobial activity was increased in proportion to its concentration by the paper disc method. Among the solvent fractions, UPMM and UPMB showed relatively strong antimicrobial activities in the order. Among various partition layers, the methanol partition layer (UPMM) was showed the strongest cytotoxic effects on all cancer cell lines. We also observed quinone reductase (QR) induced effects in all fraction layers of UP on HepG2 cells. The QR induced effects of UPMH on HepG2 cells at $320\mu g/mL$ concentration indicated 2.36 with a control value of 1.0.

This study was conducted to determine developmental ability of reconstructed embryos by nuclear transfer using somatic cell of Korean bull with high genetic value. Fibroblast cells obtained from ear biopsy of the bull were cultured in Dulbecco's Modified Eagle's medium (DMEM) at 37$^{\circ}C$ in air containing 5% $CO_2$. The cummulus-oocyte complexes were collected from slaughterhouse and were matured in vitro for 20 h in TCM 199 culture medium and the oocytes were then enucleated in modified phosphate buffered saline with cytochalasin B. Matured bovine oocytes were enucleated by aspirating the first polar body and metaphase chromatin using a beveled pipette in modified phosphate buffered saline. The ear fibroblast cells were fused into enucleated oocyte by electrical stimulation. The reconstructed oocytes were activated with ionomycine and 6-dimethylaminopurine, and then cultured in CR1aa medium for 7.5 days. Out of 524 bovine eggs reconstructed by nuclear transfer 65.6%(277/422) embryos were cleaved, and 30.7% (85/277) cleaved embryos were developed to the morula to blastocysts. There was no difference of developmental ability in vitro of reconstructed embryos regardless of donor cell passages. In order to determine fate of foreign mitochondria of donor nucleus, the Mito Tracker stained cells were fused into enucleated oocytes. The donor mitochondria were detected early stage of embryos, but disappeared rapidly. The developmental ability of reconstructed embryos was not impaired by Mito Tracker treatments. The results indicate that viable reconstructed embryos can be producted by nuclear transfer using somatic cell of Korean bulls.bulls.
