Purification and Characterization of the Red Carotenoprotein from the Muscle of Blue Mussel, Mytilus edulis (진주담치 근육중의 적색 Carotenoprotein의 정제 및 특성)

  • Most of carotenoprotein complexes have been extracted by using buffered solutions. However, in this study carotenoprotein from the muscle of Blue mussel(Mytilus edulis) was extracted by a detergent such as Triton X-100. It was purified and characterized by $20\%$ (w/v) $(NH_4)_2SO_4$, DEAE-cellulose ion exchange and Sephacryl S-300 gel filtration. The carotenoprotein(${\lambda}_{max}=462nm$) had an approximate M. W. of 372KDa(gel filtration). SDS-PAGE analysis of the carotenoprotein indicated the presence of four polypeptides of 60KDa($23.70\%$), 46.9KDa($9.14\%$), 26KDa($49.14\%$) and 13KDa($18.02\%$). Carotenoprotein denaturated by treatment with SDS to a final concentration of $0.2\%$ (w/v) caused a hypsochromic shift of ${\lambda}_{max}$ from 462nm to 456nm. The carotenoprotein contained lipids as structure units. The amino acid composition of the carotenoprotein contained large essential amino acid amounts of $62.8\%$, and the content of threonine($35.9\%$) was higher than other amino acids, but histidine, methionine and proline were not present. In the carotenoprotein, the major fatty acids were $C_{16:4},\;C_{16:0},\;C_{20:5}\;and\;C_{22:6}$. The percentages of polyunsaturated fatty acids($62.4\%$) were higher compared to other fatty acids(saturated fatty acids $19.6\%$, monounsaturated fatty acids $18.0\%$). Carotenoid was extracted from the carotenoprotein by acetone and it was separated into five different components by preparative TLC(benzene:petroleum ether:acetone=69:17:14). The major components of carotenoid were mytiloxanthin($74.79\%$) and 3,4,3'- trihydroxy-7',8'-didehydro-${\beta}$-carotene($18.26\%$), and they were at least presented as prosthetic groups of carotenoprotein.

Effects of Ridging Times on Agronomic Characteristics, Yield and Feed Value of Corn Hybrid for Silage in Paddy Field Cultivation (논 토양에서 사일리지용 옥수수 재배시 배토시기가 생육특성 및 영양성분에 미치는 영향)

  • This study was carried out to investigate the growth characteristics, yield and chemical compositions of corn hybrids for silage according to ridging times in the paddy field cultivation. The experimental design was arranged in a randomized block design with five treatments and three replication. The ridging times of five treatments were untreated (C), 7 leaves (T1: 31 days), 8 leaves (T2: 37 days), 9 leaves (T3: 42 days) and 10 leaves stage (T4: 48 days after sowing) of silage corn. The ridging treatments (T1, T2, T3, T4) were higher (p<0.01) dry matter yield and TDN yield than the unridging treatment (C). However, there were no significant differences among T1, T2, T3 and T4 treatments. The Crude protein content of unridging treatment was decreased (p<0.01), while NDF content was increased (p<0.01) compared to ridging treatments. The contents of mineral and amino acid in ridging treatments were higher than unridging treatment (p<0.05, p<0.01). In particular, mineral and amino acid contents were the highest in T4 and T1, respectively (p<0.01). Based on the above results, ridging work is to be increased the yield quantity and nutritional value of silage corn. And the best result was obtained in T4 treatment of ridging treatments.

Antitumor Effect of Polysaccharide Produced from a Mutant of Acetobacter pasteurianus IFO 13751-5 (Acetobacter pasteurianus IFO 13751-5 변이주가 생산하는 다당류의 항암효과)

  • The polysaccharide was predominantly composed of glucose (79.60%), galactose (9.68%), mannose(8.67%), fructose (1.96%;) and xylose (0.09%). Major amino acid in protein was consisted of cystein, aspartic acid, valine and glutamic acid. The tumor growth inhibition ratio of the polysaccharide against Sarcoma 180 was at highest level of 64.96%, when administerd at the concentration of 50 mg/kg and life prologation ratio showed 28.91% at the concentration of 50 mg/kg. The direct cytotoxic effect of polysaccharide was not observed in the tumor bearing mice with Sarcoma 180 in vitro.

  • A $Ca^{2+}-independent$ microbial transglutaminase (mTGase) from the actinomycete Streptomyces mobaraensis IFO13819 is a useful enzyme in the food industry. It is consists 406 amino acid residues, which comprised leader and pro region of 75 amino acid residues and the structure region of 331 amino acid residues. Pro and structure gene of TGase were cloned into the yeast shuttle vector pYAEG-TER and then used to transform Saccharomyces cerevisiae 2805. Expression of mTGase in recombinant was confirmed with Northern hybridization and the maximal activity of TGase was shown 26 mU/ml.

Cloning and Expression of A Bacillus licheniformis Cellulase Gene (Bacillus licheniformis WL-12의 cellulase 유전자 클로닝과 발현)

  • A thermophilic bacterium producing the extracellular cellulase was isolated from soybean paste, and the isolate WL-12 has been identified as Bacillus licheniformis on the basis on its 16S rRNA sequence, morphology and biochemical properties. A gene encoding the cellulase of B. licheniformis WL-12 was cloned and its nucleotide sequence was determined. This cellulase gene, designated celA, consisted of 1,551 nucleotides, encoding a polypeptide of 517 amino acid residues. The gene product contained catalytic domain and cellulose binding domain. The deduced amino acid sequence was highly homologous to those of cellulases of B. licheniformis, B. subtilis and B. amytoliquefaciens belonging to the glycosyl hydrolase family 5. When the celA gene was highly expressed using a strong B. subtilis promoter, the extracellular cellulase was produced up to 7.0 units/ml in B. subtilis WB700.

흑마늘의 이화학적 성분 및 항산화활성

  • 흑마늘의 이화학적 성분의 변화 및 항산화 활성에 대한 연구를 분석한 결과, 흑마늘의 색은 제조공정을 거치면서 L, a 및 b값이 모두 감소하였는데 내부색의 경우 가공 초기에 크게 감소하였다. 수분은 가공 중 계속해서 감소하였고, pH도 점차 산성화되어 가공이 끝난 시료에서 pH $4.22{\pm}0.02$였다. 총 페놀 화합물과 플라보노이드는 1단계시료에 비해 최종단계에서 각각 약 1.9배, 2.6배 증가하였다. Total pyruvate와 thiosulfinate도 비슷한 패턴으로 각각 약 1.6배, 5.8배 증가하였다. 유리당 중 가장 함량이 높은 fructose는 1단계에서는 $1,403.03{\pm}6.24\;mg/100\;g$ 였던 것이 4단계에서는 $2,454.45{\pm}4.20\;mg/100\;g$으로 증가하였으나, 반면에 이당류인 sucrose와 maltose는 흑마늘의 숙성과 더불어 서서히 감소하는 경향이다. 흑마늘의 구성아미노산은 glutamic acid의 함량이 가장 많았고, 다음으로 proline과 aspartic acid였으며, 유리아미노산은 taurine, homocysteine 및 $\delta$-hydroxylysine은 1단계 혹은 2단계 시료에서는 검출되지 않았으나, 2단계 혹은 3단계 이후시료에서 미량 검출되었다. 흑마늘 가공중 물추출물과 에탄올 추출물의 DPPH에 대한 전자공여능은 숙성이 진행될수록, 추출물의 첨가 농도를 높일수록 점차 증가하여 4단계 시료의 $1,000\;{\mu}g/mL$ 농도에서 에탄올 추출물은 $45.63{\pm}0.43%$, 물추출물은 $67.40{\pm}0.21%$로 시험된 시료 중 가장 높은 활성을 보였다. 환원력은 100 및 $500\;{\mu}g/mL$ 농도에서 1, 2단계 시료를 제외한 모든 시료에서 물추출물이 에탄올 추출물 보다 높은 활성을 보였다.

The effect of bean sprouts by-product addition on the growth of Pleurotus ostreatus (콩나물 부산물 첨가가 느타리 생육에 미치는 영향)

  • In this study, we attempted to find alternative materials to replace Cotten seed meal(CM) using the wastes of bean sprouts by-product in bottle cultivation of Pleurotus ostreatus. In Proximate analysis, the protein content of BW was slightly lower than that of CM, but the growth characteristics of mushrooms were similar to those of control when byproduct of bean sprouts was used. In addition, the amino acid composition of fruit body did not show a significant difference in the treatment section. Therefore, it is judged that BW could be a suitable substitute for CM in the cultivation of Pleurotus ostreatus.

Effects of Cultured Oyster Powder on Food Quality of Soybean Pastes (양식산 굴 첨가가 된장의 품질에 미치는 영향)

  • As a part of a study on the effective use of cultured oyster, oyster powder-added soybean pastes were prepared and its characteristics were examined. As the concentration of oyster powder added to commercial soybean paste was increased, the moisture level decreased, the crude and amino nitrogen content increased with no change in the salinity and titratable acidities. Also, the color of the soybean paste changed to a darker color as the concentration of oyster powder added to commercial soybean paste increased. Judging from the results of the above experiment and sensory evaluation, the optimal additive concentration of oyster powder to commercial soybean paste was revealed as 9%. The total amino acid content of oyster powder-added soybean paste was 2% higher than that of commercial soybean paste. The ratio of calcium and phosphorus of oyster powder-added soybean paste was about 1:2-2:1, an improvement in calcium absorption as compared to commercial soybean paste. Functional fatty acids, such as 20:5n-3 and 22:6n-3, were not detected in commercial soybean paste but detected in oyster powder-added soybean paste. The major free amino acids in oyster powder-added soybean paste were glutamic acid and aspartic acids. According to the above results, the nutritional, functional and taste properties of commercial soybean paste were improved while reducing preparation time of the housewife.

  • A carotenoprotein from the muscle of Ascidian (Halocynthia roretzi) was purified by ion exchange chromatography and gel filtration chromatography, and analyzed molecular weight, stability of pH and heat, effect of denaturing agents, amino acids and fatty acids composition. The purified carotenoprotein had absorption maxima of 463 nm and 439 nm. The carotenoprotein had an approxmimate molecular weight of 64.4 kDa in polyacrylamide gel electrophoresis. The amino acid compositions of carotenoprotein were mainly Gly (15.39%), Asn (11.31%), Gln (10.62%) and Ser (13.35%). The major fatty acids composition of carotenoprotein were $C_{16:1(n-7)}\;(15.4%)$, $C_{22:1(n-9)}\;(14.5%)$ and $C_{20:1(n-11)}\;(11.4%)$. The monounsaturated fatty acids (45.2%) contained abundant content compared to other saturated (38.1%) and polyunsaturated (11.7%) fatty acids.

Analysis of Nutritional Components of Lepista nuda (민자주방망이버섯(L. nuda)의 영양성분 분석)

  • This study was carried out to analyze the nutritional component of Lepista nuda in order to estimate its nutritional and functional values. The content of moisture were $90.90{\pm}0.09%$ carbohydrate $4.34{\pm}0.07%$, crude protein $3.70{\pm}0.03%$, ash $0.55{\pm}0.04%$, and crude fat $0.47{\pm}0.01%$, respectively The reducing sugar was $636.17{\pm}7.50mg$. Total amount of free sugar was 97.32 mg, which were mainly composed of galactose (51.98 mg) and trehalose (22.48 mg). In the results of mined analysis, the content of K was the highest as $103.10{\pm}0.18mg$, followed by $56.01{\pm}0.20mg$ of Mg. The total content of hydrolyzed amino acid was 310.39 mg which consist of essential amino acids including valine (25.40 mg), leucine (22.52 mg), lysine (21.34 mg) and 198.89 mg of non-essential amino acids including glutamic acid (60.72 mg), aspartic acid (30.17 mg), and alanine (29.72 mg), as the major amino acids. free amino acids were 220.86 mg which were composed of essential amino acids (36.21 mg) inclusive of isoleucine (21.57 mg) and threonine (11.20 mg) and non-essential amino acids (184.65 mg) including glutamic acid (42.78 mg) and aspartic acid (32.44 mg) as major amino acids. Total content of amino acid derivatives was 187.85 me and that of hydroxyproline was highest as 121.50 mg.
