Organic Precipitate Flotation of Trace Metallic Elements with Ammonium Pyrrolidinedithiocarbamate(Ⅰ). Determination of Bismuth, Cadmium, Cobalt and Lead in Water Samples by Coprecipitation-Flotation with Cu-pyrrolidinedithiocarbamate (Ammonium Pyrrolidinedithiocarbamate에 의한 극미량 금속원소의 유기침전 부선에 관한 연구(제1보) Cu-pyrrolidinedithiocarbamate 공침부선에 의한 물시료중 비스무트, 카드뮴, 코발트 및 납의 정량)

  • The organic precipitate flotation using Cu(II)-pyrrolidinedithiocarbamate complex as a coprecipitant was studied for the preconcentration and determination of trace Cd, Pb, Bi and Co in several water samples. Experimental conditions such as pH of solution, amounts of Cu(II) and ammonium pyrrolidinedithiocarbamate(APDC), stirring time, the type and amount of surfactant, etc. were optimized for the effective flotation of analytes. After 3.0 mL of 1,000 ${\mu}g/mL$ Cu(II) solution was added to 1.00 L water sample, the pH of the solution was adjusted to 2.5 with HNO3 solution. Trace amounts of analytes were coprecipitated by adding 2.0% APDC solution. And the precipitates were flotated onto the surface of solution with the aid of nitrogen gas and sodium lauryl sulfate. The floats were collected from mother liquor, and filtered through the micropore glass filter by suction. The precipitates were dissolved with 4 mL conc. HNO3, and then diluted to 25.00 mL with deionized water. The analytes were determined by graphite furnace atomic absorption spectrophotometry. This flotation technique was applied to the analysis of some water samples, and the 90 to 120% of recoveries were obtained from the spiked samples, this procedure could be concluded to be simple and applicable for the trace element analysis in various kinds of water.

Activation Mechanism of Protease in the Germination of Mulberry Seeds (뽕나무 종자 발아시의 Protease 발현기구)

  • The activity change of mulberry seeds protease was compared during germination for 5 days at 28$^{\circ}C$ in the dark place after daily hormone injection of different concentration. The protease from germinated mulberry seeds for 4 days was partially purified and the enzyme characteristics was investigated. The protease activity of mulberry seeds treated by hormone was highest with 10 $\mu$m GA3 followed by 10 $\mu$M zeatin and 10 $\mu$M kinetin. The protease activity of mulberry seeds was increased by 14% with 10ml agar culture that control at 4th day of germination. The protease from mulberry seeds was purified 313 fold by DEAE-Toyo-pearl 650M, Butyl-Toyopearl, Hydrozylapatite and Toyopearl HW 55M. After purification, the specific activity of the enzyme was 175 units/mg. Optimum pH and temperature of protease from mulberry seeds was 5.0 and 37$^{\circ}C$, respectively. The protease was stable below 37$^{\circ}C$ and the enzyme activity was decreased by 50%, when incubated at 52$^{\circ}C$ for 10minutes. The protease activity of mulberry seeds was inhibited by metal ions such as mercury, iron, zinc, copper, but activited by magnesium, choromium, aluminium ions. The Km value of the protease was 0.89mM with azocasein as a subscribe.

Bronze Production Technology in the Early Iron Age: A comparative study of bronze artifacts recovered from the Hoam-dong site in Chungju and Chongsong-ri in Buyeo (초기철기시대 청동기의 제작기술 - 충주 호암동유적과 부여 청송리유적 출토 청동기의 비교 연구-)

  • Thirty-three Early Iron Age bronzes at the sites of Hoam-dong in Chungju and Cheongsong-ri in Buyeo were investigated in order to study the manufacturing technique and the provenance of lead. Chemical analysis using X-ray fluorescence showed that 33 bronzes consist of copper(Cu), tin(Sn) and lead(Pb) served as major elements. Major and minor elemental analyses by EPMA were performed on two mirrors and 2 weapons of the bronzes investigated. The results shows that bronze mirrors from Chungju and Buyeo were high-tin bronzes(> 30 wt%). And 20% of tin and 5% of lead were founded in bronze weapons. Iron, zinc, arsenic, silver, nickel, sulfur and cobalt detected in four bronzes as minor and trace elements. The four bronzes were alloyed considering their function and were not heat treated after casting due to their high tin content. Lead isotope analysis using TIMS indicates that thirty-three bronzes were distributed southern Korea peninsula except Zone 1. As a result, lead raw materials came from various regions in Korean Peninsula not from Gyeongsang-do regions. The manufacturing techniques of bronze ware generalized at this age, and bronze was produced in various sites using raw materials from various sources.

Analysis of Dry Process Products for Recycling of Spent Secondary Batteries (폐 이차전지 리사이클링을 위한 건식공정 생성물 분석)

  • The purpose of this study is to recover valuable metals from spent batteries using a dry process. We focused on the effect of the smelting temperature on the composition of recovered solid and liquid products and collected gaseous products. After removal of the cover, the spent battery was left in NaCl solution and discharged. Then, the spent battery was made into a powder form through a crushing process. The smelting of the spent battery was performed in a tubular electric furnace in an oxygen atmosphere. For spent lithium-ion batteries, the recovery yield of the solid product was 80.1 wt% at a reaction temperature of 850 ℃, and the final product had 27.2 wt% of cobalt as well as other metals such as lithium, copper, and aluminum. Spent nickel-hydrogen batteries had a recovery yield of 99.2 wt% at a reaction temperature of 850 ℃ with about 37.6 wt% of nickel and other metals including iron. For spent nickel-cadmium batteries, the yield decreased to 65.4 wt% because of evaporation with increasing temperature. At 1050 ℃, the recovered metals were nickel (41 wt%) and cadmium (12.9 wt%). Benzene and toluene, which were not detected with the other secondary waste batteries, were detected in the gaseous product. The results of this study can be used as basic data for future research on the dry recycling process of spent secondary batteries.

Assessment of Pollution Characteristics of Surface Sediments from Lake Andong(II): Studies on the Nutrient and Heavy Metal Release Characteristics from Sediments in Andong Dam (안동댐 퇴적물의 오염도 평가(II): 안동댐 퇴적물에 대한 영양염류 및 중금속 용출 특성 연구)

  • Leaching chracteristics of Andong-dam sediment was conducted for heavy metal and nutrients. Five mixed sediment samples were prepared and leaching was conducted under aerobic and anaerobic condition for 60 days. Cd, Cu, Pb, Cr, Zn, Hg, As, Fe, Mn, phosphorus, and nitrogen were analyzed at each sampling time. The leaching rate of phosphorus was higher in anaerobic condition comparing with that of under aerobic condition. Some samples showed higher than the water-quality level IV. In case of As and Cd which showed highest contamination level in the sediment, leached concentration were 0.028 mg/L and 0.003 mg/L in maximum, respectively. The leached concentration is below than the lake water quality standard of Korea. Other heavy metals including Cu, Pb, and Cr also showed similar trend. Five step sequential extraction showed that easily extractable 1-2 step portion such as ion-exchangeable and adsorbed one was less than 10% and the most of the portion was residual. For As and Cd, the residual portion were 80% and 95% respectively indicating the risk by the heavy metal leaching into the lake for a short period was not high in comparing with the contamination levels.

Gas Sorption Analysis of Metal-organic Frameworks using Microresonators (마이크로진동자 기반 금속유기골격체의 기체 흡탈착 분석)

  • Metal-organic frameworks (MOFs) are porous materials with nano-sized pores. The degree of gas adsorption and pore size can be controlled according to types of metal ions and organic ligands. Many studies have been conducted on MOFs in the fields of gas storage and separation, and gas sensors. For rapid and quantitative gas adsorption/desorption analyses, it is necessary to form various MOF structures in uniform films on a sensor surface. In this review, some of representative direct methods for uniformly synthesizing MOFs such as MIL-53 (Al), ZIF-8, and Cu-BDC from anodized aluminum oxide, zinc oxide nanorods, and copper thin films, respectively on the surface of a microresonator are highlighted. In addition, the operation principle of quartz crystal microbalance and microcantilever, which are representative microresonators, and the interpretation of signals that change when gas is adsorbed to MOFs are covered. This is intended to enhance the understanding of gas adsorption/desorption analysis of MOFs using microresonators.

Production and Characterization of the Beneficial β-glucuronidase Inhibitor from non pathogenic wild yeast, Candida oleophila WP5-19-1, and its effects on gut microbes (비병원성 야생효모 Candida oleophila WP5-19-1로부터 장내유용세균 유해 효소 β-glucuronidase 저해제의 생산 및 특성)

  • We aimed to produce a potent β-glucuronidase inhibitor from wild yeast that could inactivate toxic substances in the colon. Culture supernatants and cell-free extracts of non-pathogenic wild yeasts were prepared and their β-glucuronidase inhibitory activities were measured. Cell-free extract from Candida oleophila WP5-19-1 showed the highest β-glucuronidase inhibitory activity (49.0%). The β-glucuronidase inhibitor was maximally produced (IC50 value; 8.4 mg) when C. oleophila WP5-19-1 was cultured in potato dextrose medium containing 5% dextrose (initial pH; 6.0) at 30℃ for 24 hours. β-glucuronidase inhibitor of C. oleophila WP5-19-1 was partially purified by trypsin hydrolysis, ultrafiltration (3 kDa), and Sephadex G-50 filtration. The partially purified β-glucuronidase inhibitor was stable from 30℃ to 60℃ and at pH 6.0 9.0, and showed residual inhibitory activity of about 80%.

Characteristics of Stream and Soil Contamination from the Tailing Disposal and Waste Rocks at the Abandoned Uljin Mine (울진 폐광산의 매립광미와 폐광석에 의한 주변 토양 및 수계의 오염특성)

  • Physicochemical characteristics of stream water, leachate, mine water and groundwater were investigated to estimate the influences of the tailing and waste rock from the abandoned Uljin mine area. Total extraction analysis and mineralogical studies were carried out to understand sulfide weathering and to determine the distributions of trace elements in the soil affected by mine waste (tailing, waste rock and leachate). The pH and EC value of the leachate from the tailing disposal ranged 2.9-6.0, $99{\sim}3,990{\mu}S/cm$, respectively, and the concentrations of dissolved major (up to 492 mg/l Ca; 83.8 mg/l Mg; 45.2 mg/l Na; 44.7 mg/l K, 50.8 mg/l Si) and trace elements (up to $826,060{\mu}g/l$ Fe; $131,230{\mu}g/l$ Mn; $333,600{\mu}g/l$ Al; $61,340{\mu}g/l$ Zn; $2,530{\mu}g/l$ Cu; $573{\mu}g/l$ Cd; $476{\mu}g/l$ Pb) were relatively high. The stream water showed the variation of dissolved metal concentrations in seasonally and spatially. The dissolved metal contents of the stream water increased by influx the leachate from the tailing disposal, but these of the down stream have been considerably decreased by mixing of dilute tributaries. The dissolved metal concentrations of the stream water at dry season (as February) were lower than these at rainy season (as May and July). These represent that the amounts of the leachate varied with season. However, stream water could not be effectively diluted by confluence with uncontaminated tributaries, because the flux of tributaries and streams reduced at dry season. Thus attenuations by dilution had been dominantly happened in rainy seasons. The order of accumulations of trace element in soils compared with background values revealed Mn>Fe>Pb>Cu>Zn. Sulfide minerals were mainly pyrrhotite, sphalerite and galena and chalcopyrite. Pyrrhotite was rapidly weathered along the edge and fractures, and results in the formation of Fe-(oxy)hydroxides, which absorbed a little amount of Zn.

Analysis on Characteristics of Aquatic Ecosystem Environment in Junam Reservoir (주남저수지의 수생태환경 특성에 대한 분석)

  • The purpose of this study was to offer the raw data for conservation and restoration of aquatic ecosystem by surveying and analysing a water quality, sediment, benthic macroinvertebrate, fish and hydrophyte in Junam reservoir. In the result of analysing the water quality, pH was 8.2~8.4, 6.0~7.5mg/${\ell}$ in COD, 10.0~10.3mg/${\ell}$ in SS, 8.3~11.5mg/${\ell}$ in DO, 0.1mg/${\ell}$ in T-P and 1.2~1.3mg/${\ell}$ in T-N. In case of the sediments, Cd was 0.47~0.52mg/kg, 7.08~7.43mg/kg in Cu, 0.22~0.32mg/kg in As, 0.02~0.03mg/kg in Hg, 6.20~7.45mg/kg in Pb, 32.80~39.70mg/kg in Ni, 513.0~543.0mg/kg in F and 137.0~140.0mg/kg in Zn. $Cr^{6+}$ was not detected. The benthic macroinvertebrates were 432 individuals including 3 phyla, 5 classes, 9 orders, 26 families, 33 genera and 39 species, and the ecological score of benthic macroinvertebrate community(ESB) was 25. The fishes were 8 families and 14 species, and the invasive alien species were 2 taxa including Micropterus salmoids and Lepomis macrochirus. The hydrophytes were 63 taxa including 28 families, 42 genera, 56 species, 1 subspecies and 6 varieties. The emergent plants were 38 taxa(60.3%), 5 taxa(7.9%) in floating-leaved plants and each 10 taxa(15.9%) in free-floating plants and submerged plants.

Assessment of Water Pollution by Discharge of Abandoned Mines (휴폐광산 지역에서 유출되는 하천수의 오염도 평가)

  • Several metalliferous and coal mines, including Myungjin, Seojin and Okdong located at the upper watershed of Okdong stream, were abandoned or closed since 1988 due to the mining industry promotion policy. Thus these disposed an enormous amount of mining wastes without a proper treatment facilities, resulting in water pollution in the downstream areas. Acid mine drainage (AMD) and waste water effluents from the closed coal mines were very strongly acidic showing pH ranges of 2.7 to 4.5 and had a high level of Total Dissolved Solids (TDS) showing the ranges of 1,030 to 1,947 mg/L. Also heavy metal concentrations in these samples such as Fe, Cu, Cd and anion such as sulfate were very high. Concentrations of water soluble heavy metals in the Okdong streams were in the orders of Fe>Al>Mn>Zn>Cu>Pb>Cd, indicating Fe from the AMD and waste water effluents contributed greatly to the quality of water and soil in the lower watershed of Okdong stream. Copper concentrations in the effluents from the tile drainage of mine tailings dams were highest during the raining season. Water Pollution Index (WPI) of the surface water at the upper stream of Okdong river where AMD of the abandoned coal mines was flowed into main stream were in the ranges of 16.3 to 47.1. On the other hand, those at the mid stream where effluents from tailings dams and coal mines flowed into main stream were in the WPI ranges of 10.6 to 19.5. However, those at the lower stream were ranged from 10.6 to 14.9. These results indicated that mining wastes such as AMD and effluents from the closed mines were the major source to water pollution at the Okdong stream areas.
