Variations in Nutrients & CO2 Uptake Rates and Photosynthetic Characteristics of Saccharina japonica from the South Coast of Korea (다시마(Saccharina japonica)의 생장에 따른 영양염 및 CO2 흡수율과 광합성 특성 변화)

  • To investigate the contribution of macroalgae to biogeochemical nutrients and carbon cycles, we measured the uptake rates of nutrients and $CO_2$ and characteristics of fluorescence of Saccharina japonica (Laminaria japonica Areschoug) using an incubation method in an acrylic chamber. From January to May 2011, S.japonica was sampled at Ilkwang, one of well-known macroalgae culture sites around Korea and ranged 46~288 cm long and 4.8~22.0 cm wide of whole thallus. The production rate of dissolved oxygen by S. japonica (n=25) was about $6.9{\pm}5.8{\mu}mol\;g^{-1}$ fresh weight(FW) $h^{-1}$. The uptake rate of total dissolved inorganic carbon ($TCO_2$), calculated by total alkalinity and pH, was $8.9{\pm}7.9{\mu}mol\;g^{-1}\;FW\;h^{-1}$. Mean nutrients uptake were $175.6{\pm}161.1\;nmol\;N\;g^{-1}\;FW\;h^{-1}$ and $12.7{\pm}10.1\;nmol\;P\;g^{-1}\;FW\;h^{-1}$. There were logarithmic relationships between thallus length and uptake rates of nutrients and $CO_2$, which suggested that younger specimens (<100-150 cm) were much more efficient at nutrients and $CO_2$ uptake than old specimens > 150 cm. There was a positive linear correlation ($r^2$=9.4) existed between the dissolved oxygen production rate and the $TCO_2$ uptake rate, suggesting that these two factors may serve as good indicators of S. japonica photosynthesis. There was also positive linear relationship between maximal quantum yield ($F_v/F_m$) and production/uptake rates of dissolved oxygen, $TCO_2$ and phosphate, suggested that $F_v/F_m$ could be used as a good indicator of photosynthetic ability and $TCO_2$ consumption of macroalgae. Maximum relative electron transport rate ($rETR_{max}$) of S. japonica increased as thallus grew and was high in distal part of thallus which may be resulted from the increase of photosynthetic cell density per area. The annual $TCO_2$ uptake by S. japonica in Gijang area was estimated about $1.0\sim1.7{\times}10^3C$ ton, which was about 0.02-0.03% of carbon dioxide emission in Busan City. Thus, more research should be focused on macroalgae-based biogeochemical cycles to evaluate the roles and contributions of macroalgae to the global carbon cycle.

  • The co-culture system of microalgae and bacteria enables simultaneous removal of BOD and nutrients in a single reactor if the pair of microorganisms is symbiotic. In this case, nutrients are converted to biomass constituents of microalgae. This review highlights the importance and recent researches using symbiotic co-culture system of microalgae and bacteria in wastewater treatment, focusing on the removal of nitrogen and phosphorus. During wastewater treatment, the microalgae produces molecular oxygen through photosynthesis, which can be used as an electron acceptor by aerobic bacteria to degrade organic pollutants. The released $CO_2$ during the bacterial mineralization can then be consumed by microalgae as a carbon source in photosynthesis. Microalgae and bacteria in the co-culture system could cooperate or compete each other for resources. In the context of wastewater treatment, positive relationships are prerequisite to accomplish the sustainable removal of nutrients. Therefore, the selection of compatible species is very important if the co-culture has to be utilized in wastewater treatment.

Applicability of a Photosynthetic Ciliate, Mesodinium rubrum MR-MAL01 -Usefulness as a Live Prey Species for the Marine Aquaculture of Bivalves- (광합성 섬모류 Mesodinium rubrum MR-MAL01의 응용성(1) 이매패류 종묘생산을 위한 먹이생물 유용성)

  • In a series of feeding experiments using the newly established the first laboratory strain of a temperate photosynthetic ciliate, Mesodinium rubrum MR-MAL01, direct evidence was obtained for ingestion of the cultured M. rubrum cells by the bivalves like the Korean scallop, Chlamys farreri and Manila clam. Ruditapes phil-ippinarum. Each experimental Korean scallop and Manila clam removed up to 9,590 and 23,200 cells min-1, respectively. Small particles almost identical to the ruptured cells of MR-MAL01 culture were observed in the gut of the experimental bivalves, although no intact M. rubrum cell was found. In a 28 days rearing experiment, Isoshrysis galbana (KMCC H-2), a microalga, supported better growth of small Manila clam spat (0.46 mm in shell length) than MR-MAL01 strain. For the large Manila clam spat (0.84 mm in shell length), however. MR-MAL01 strain was a better prey item. Mass culture methods for this photosynthetic marine ciliate may be developed for use as live feed in aquaculture of the spat and broodstock of bivalve and small-mouthed fish larvae.

Biological Co2 Fixation to Antioxidant Carotenoids by Photosynthesis Using the Green Microalga Haematococcus pluvialis (광합성 녹색 미세조류 Haematococcus pluvialis를 이용한 이산화탄소 고정화 및 항산화성 카로티노이드 생산)

  • As one of the $CO_2$ reduction strategies, a biological method was proposed to convert $CO_2$ to useful biomass with antioxidant carotenoids by photosynthetic microorganisms. One of the photoautotrophs, Haematococcus pluvialis is a freshwater green microalga and accumulates the secondary carotenoid astaxanthin during induction of green vegetative cells to red cyst cells. In this study, $CO_2$ fixation and astaxanthin production using H. pluvialis was conducted by photoautotrophic culture in the $CO_2$ supplemented photo-incubator. Maximum growth rate of H. pluvialis was obtained at a 5% $CO_2$ environment on basic N and P conditions of NIES-C medium. The photoautotrophic induction consisted of 5% $CO_2$ supply and high light illumination promoted astaxanthin synthesis in H. pluvialis, yielding an astaxanthin productivity of $9.6mg/L{\cdot}day$ and a $CO_2$ conversion rate of $27.8mg/L{\cdot}day$ to astaxanthin. From the results the sequential photoautotrophic culture and induction process using H. pluvialis is expecting an alternative $CO_2$ reduction technology with a function of valuable biosubstance production.

Seasonal Mineral Nutrient Absorption Characteristics and Development of Optimum Nutrient Solution for Rose Substrate Culture in a Closed Hydroponic System (순환식 수경재배에서 재배시기별 장미의 무기이온 흡수특성과 적정 배양액 조성)

  • This study was performed to develop a suitable nutrient solution for standard rose substrate culture in a closed hydroponic system. 1/4, 1/2, 2/3 and 1 strength of the nutrient solution made by Japan National Institute of Vegetable and Tea Science (JNIVT) were supplied. The photosynthesis rate, quality and growth of cut flower were higher in the 1/2 and 2/3 strength of nutrient solution during high and low temperature period. Based on the above results, optimum nutrient solutions (UOS) were composed by nutrientwater (n/w) absorption ratio with 1/2S ($NO_{3^-}N$ 6.8, $NH_{4^-}N$ 0.7, $PO_{4^-}P$ 2.0, K 3.8, Ca 3.0, Mg 1.2, $SO_{4^-}S$ $1.2me{\cdot}L^{-1}$) at high temperature season and 2/3($NO_{3^-}N$ 9.7, $NH_{4^-}N$ 0.8, $PO_{4^-}P$ 2.2, K 5.0, Ca 3.9, Mg 1.5, $SO_{4^-}S$ $1.5me{\cdot}L^{-1}$) at low temperature season. The results of suitability examination showed that the EC level in newly composed nutrient solution (UOS) was more stable than other nutrient solutions due to its large amount of calcium and potassium. The growth of cut flower cultivated with UOS was higher than those of other nutrient solutions. Especially, the yield of cut flowers in UOS nutrient solution increased 1.4 times than that of other nutrient solution treatments. Consequently, the new nutrient solution investigated in this experiment was suitable for rose cultivation in a closed hydroponic system.

Selection of Filamentous Cyanobacteria and Optimization of Culture Condition for Recycling Waste Nutrient Solution (폐양액 활용을 위한 Filamentous Cyanobacteria의 선발 및 최적배양)

  • The discharge of waste nutrient solution from greenhouse to natural ecosystem leads to the accumulation of excess nutrients that results in contamination or eutrophication. There is a need to recycle the waste nutrient solution in order to prevent the environmental hazards. The amount and kind of nutrients in waste nutrient solution might be enough to grow photosynthetic microorganisms. Hence in the present study, we examined the growth and mass cultivation of cyanobacteria in the waste nutrient solution with an objective of removing N and P and concomitantly, its mass cultivation. Four photosynthetic filamentous cyanobacteria (Anabaena HA101, HA701 and Nostoc HN601, HN701) isolated from composts and soils of the Chungnam province were used as culture strains. Among the isolates, Nostoc HN601 performed faster growth rate and higher N and P uptake in the BG-II ($NO_3{^-}$) medium when compared to those of other cyanobacterial strains. Finally, the selected isolate was tested under optimum conditions (airflow at the rate of $1L\;min^{-1}$. in 15 L reactor, initial pH 8) in waste nutrient solution from tomato hydroponic in green house condition. Results showed to remove 100% phosphate from the waste nutrient solution in the tomato hydroponics recorded over a period of 7 days. The growth rate of Nostoc HN601 was $16mg\;Chl-a\;L^{-1}$ in the waste nutrient solution from tomato hydroponics with optimum condition, whereas growth rate of Nostoc HN601 was only $9.8mg\;Chl-a\;L^{-1}$ in BG-11 media. Nitrogen fixing capacity of Nostoc HN601 was $20.9nmol\;C_2H_4\;mg^{-1}\;Chl-a\;h^{-1}$ in N-free BG-11. The total nitrogen and total phosphate concentration of Nostoc HN601 were 63.3 mg N gram dry weight $(GDW)^{-1}$ and $19.1mg\;P\;GDW^{-1}$ respectively. Collectively, cyanobacterial mass production using waste nutrient solution under green house condition might be suitable for recycling and cleaning of waste nutrient solution from hydroponic culture system. Biomass of cyanobacteria, cultivated in waste nutrient solution, could be used as biofertilizer.

Characterization of Enzymes Against Oxygen Derivatives Produced by Rhodobacter sphaeroides D-230 (Rhodobacter sphaeroides D230이 생성하는 산소 유도체에 작용하는 효소의 특성)

  • The activities of enzymes that act on oxygen derivatives in Rhodobacter sphaeroides D-230 were investigated under various culture conditions. Intracellular SOD activity from the cells grown in aerobic or anaerobic culture conditions was highest at pH 7.0 and pH 8.0, respectively. On the other hand, extracellular SOD activity was highest at pH 6.0. Catalase activity was highest at neutral pH in both cases. Growth of R. sphaeroides D-230 in aerobic or anaerobic culture conditions was inhibited by methyl viologen. As R. sphaeroides D-230 was cul-tured aerobically, SOD activity was increased about 2-fold by addition of iron ion. But $Mn^+2$ had little effect on the SOD activity of R. sphaeroides D-230 grown in aerobically. NaCN, the inhibitor of Cu$.$Zn-SOD, did not inhibit SOD activity. But, $NaN_3$, the inhibitor of Mn-SOD, inhibited SOD activity in anaerobic cultures con-dition. Therefore, R. sphaeroides D-230 produce Mn-SOD in anaerobic condition, although Fe-Sod is produced in aerobic condition. The activity of catalase was induced by methyl viologen, however, extremely inhibited by NaCN and $NaN_3$.

General characters and applications of photosynthetic bacteria (광합성세균의 생리 및 이용에 관한 최근의 동향)

  • In order to investigate the production of aflatoxin in various conditions such as pH, moisture and temperature, 27 smaples were inoculated with Aspergillus flavus, and in addition 3 smaples were inoculated with the mixture of Aspergillus flavus and Bacillus subtilis and cultured under the conditions such as 20.deg.C and 30% moisture contents. The following results were obtained : 1) Aflatoxin production was the highest at pH 5.0 and relatively high at pH 7.0. Its production was decreased significantly when pH reached 9.0. 2) The yield of aflatoxin was shown comparatively high level at 30% moisture contens. The higher moisture contents was, the lower aflatoxin production was. 3) The highest level of aflatoxin production was at 20.deg,C, and comparatively high level was at 30.deg.C. However, its production was fairly low at 40.deg.C. 4) The highest crude aflatoxin production was 5,093ppm (B$_{1}$, 1.912ppm ; B$_{2}$, and the lowest one 2.197 ppm (B$_{1}$, 0.793 ppm : B$_{2}$, 0.185 ppm : G$_{1}$ ,0.102 ppm G$_{2}$, 0.381ppm) at 63% moisture, pH 9.0 and 40.deg.C. 5) When Aspergillus flavus and Bacillus subilis were cultured together under the conditions such as 20.deg.C and 30% moisture, aflatoxin production was decreased by 27% comparing with the culture of Aspergillus flavus alone.

Optimum Culture Conditions for the Growth of Spirulina platensis NIES 39 (Spirulina platensis NIES 39의 성장을 위한 최적배양조건)

  • Recently, as the interest in the accelerated global warming and the food shortage problem is increased, the concerns for microalgae as photosynthetic microorganisms are also increased. Specially, photosynthetic microalgae, Spriulina platensis have been an attractive source for $CO_2$ gas fixations and for a vast array of valuable nutritious compounds. In this paper, to culture the microalgal Spirulina platensis NIES 39 in a batch culture with high mass, optimal conditions for the culture temperature, initial pH, light intensity and concentration of carbon and nitrogen, were tested. At the most favorable culture condition, $35^{\circ}C$, initial pH 9.5, 4500 lux and carbon and nitrogen concentration of 16.8 g/L $NaHCO_3$ and 2.5 g/L $NaNO_3$, the excellent yields of 2.10 g/L biomass and 29.53 mg/L chlorophyll were obtained.

Effect of Nano Bubble Oxygen and Hydrogen Water on Microalgae (나노기포 산소수 및 수소수가 미세조류 배양에 미치는 영향)

  • Microalgae Nannochloropsis oculata (N. oculta) and Chlorella vulgaris (C. vulgaris) are important sources for biodisel because of the high content of neutral lipids. Stable nano bubble is maintained for a long time and therefore it is possible for use in biotechnology. In this study, effects of nano bubble oxygen or hydrogen water on the microalgae growth were characterized. The cell growth in nano bubble water was similar to that of control, and the total lipid content was rather low. But, chlorophyll content of N. oculata grown in nanno bubble oxygen and hydrogen water increased 54% and 30%, and increased 59%, 39% in C. vulgaris. Carotenoid content also increased 21%, 25% in N. oculata and 49%, 29% in C. vulgaris grown in nano bubble oxygen and hydrogen water. From these results, nano bubble water seems to enhance the photosynthetic capacity of microalgae.
