Development of a Method for Producing Liposome Ascorbic acid with Increased Bio-absorption (생체 흡수율이 증가된 liposomal ascorbic acid 제조법 개발)

  • Various methods are known for preparing liposomes, the simplest being the Bangham method which has been widely used. Although it is possible to produce liposomes effectively on a small experimental level with this approach, large-scale production cannot be easily performed due to difficulties in removing the organic solvent and the size of the reactor required to form the lipid film. On the other hand, emulsion can mass produce tons of liposomes with uniform particles but has the disadvantage of a significantly low capture rate. This study therefore developed an optimal liposome processing method using heat with improved capture rate and stability, and bio-absorption experiments were performed by oral administration to SD rat alongside capture rate, particle size, and zeta potential. Through the heating method, a small and uniform liposome of about 214 nm was formed and the capture rate was 38.67%, confirming that the liposome prepared by heating has a higher capture rate than the 26.46% achieved through emulsion. Comparing blood concentrations, it showed a 1.5 to 2 fold increase in all groups, gradually decreasing until 4-12 hr. The highest blood concentration of ascorbic acid powder was about 12.017 ㎍/ml, the emulsion liposome 13.871 ㎍/ml, and the heating liposome 16.322 ㎍/ml, thereby showing an improved absorption rate.

Biological Control of Vegetable Insect Pests with Entomopathogenic Nematodes (곤충병원선충을 이용한 채소해충의 생물적 방제)

  • Entomopathogenic nematodes, Steinernema carpocapsae, was able to invade and kill the several lepidopteran pests including the beet armyworm, Spodeptera exigua Hubner, which was the most effective target host. The beet armyworms treated with the effective nematode concentrations were died within 48 hrs. The lethal effect of the nematode was varied among the developmental stages of the host. The fifth instar larvae of the beet armyworm was more vulnerable to the nematode than the third instar larvae. Pupae was, however, refractory to the nematode. All three bioessays (topical application, filter paper test, and soil treatment) showed the positive correlation between the number of the treated nematodes and the mortality of the host. Topical application was the most effective and fast-acting method so that it gave the lethal effect 2 days earlier than did filter paper test at the same number of the treated nematodes. Soil treatment required higher number of the nematodes to get the effective lethality than did filter paper test. The fifth instar larvae of the beet armyworm expressed the specific hemolymph proteins of 5 to 10 kDa in response to nematode infection.

The Trial Status and Expectation Degree on Internet Diet Program by Female University Students (여대생의 인터넷 다이어트 이용실태 및 기대도 분석)

  • This study was to investigate the trial status of weight control with internet. A survey was carried out by self-questionnaires with 500 female university students in Seoul. The average height, weight, and PIBW(Percentage of Ideal Body Weight) of subjects were 162.2$\pm$4.8cm, 51.9$\pm$6.1kg and 93.7$\pm$11.0%. It appeared that 50.6% of subjects were abnormal range of body weight : in particular 43.4% of the subjects were underweight. While 80.7% of subjects have deeply interested in weight control, 63.4% of them have attempted diet. It showed that 92.9% of subjects used internet, 54.3 % of them knew internet diet site, however only 39 of subjects used internet diet program. Most subjects used internet diet program in order to acquire diet information. According to PIBW, the overweight group is higher frequency of joining internet diet program than the others group. Therefore, education of weight control was needed with internet in abnormal weight group.

Report of Eight Unrecorded Fungi from the Fire Blight Burial Control Soil in Korea (과수화상병 매몰방재지 토양에서 분리된 8종의 국내 미기록 진균 보고)

  • Fire blight, a plant disease caused by Erwinia amylovora, continues to cause considerable damage to fruit-bearing trees belonging to the Rosaceae family, such as apples and pears. In Korea, to hinder its rapid spread diseased apple and pear trees have been buried since its first occurrence. To determine the safety of this control method, the buried plant materials and adjacent soil have been investigated. In the process of investigating the microbiota of the buried soil, Bisifusarium allantoides, Botryotrichum domesticum, Microascus verrucosus, Paraphoma pye, Phaeosphaeria culmorum, Ramophialophora globispora, Sordaria tomentoalba, and Striaticonidium brachysporum were identified as unrecorded species in Korea. To report the above eight species as Korean unrecorded species, we report their morphological and molecular features in this study.

Anti-Inflammatory Active Polysaccharide from Postbiotics of Cordyceps militaris Mycelium-Liquid Culture (동충하초(Cordyceps militaris) 균사체 액체발효 포스트바이오틱스로부터 항염증 활성다당 분리)

  • To investigate the anti-inflammatory activity of submerged culture using Cordyceps militaris mycelium, culture-including mycelia was extracted and lyophilized into postbiotics (hot-water extract; CM-HW). HW was fractionated into crude polysaccharide (CM-CP) by ethanol precipitation, and CM-CP was further dialyzed into CM-DCP by dialysis with running water using 12~14 kDa dialysis tube. When the cytotoxicity of subfractions against cells was assessed, no subfraction had a cytotoxic impact that was substantially different from the control groups. In an inflammatory model using LPS-stimulated RAW 264.7 cells, CM-DCP significantly decreased IL-6 and MCP-1 production levels compared to the LPS-control group. CM-DCP also inhibited IL-6 and IL-8 secretion in HaCaT keratinocytes stimulated with TNF-α and IFN-γ. In the meanwhile, the neutral sugar content and mannose ratio of anti-inflammatory CM-DCP were higher than the other fractions, and CM-DCP contained β-1,3/1,6-glucan of 216.1 mg/g. High pressure size exclusion chromatography revealed that CM-DCP contained molecules with a molecular weight range of 5.6 to 144.0 kDa. In conclusion, postbiotics of C. militaris mycelium significantly promoted anti-inflammatory activity, suggesting that neutral polysaccharides including Glc and Man contribute to the anti-inflammation in RAW 264.7 or HaCaT cells.

Evaluation of fitness according to application of glass fiber reinforcement for lower jaw complete denture (하악 총의치 전용의 유리섬유 보강재 적용에 따른 적합도 비교 분석)

  • Purpose: The purpose of this study is to evaluate the fitness of lower jaw compete denture with glass fiber. Methods: Lower jaw edentulous model was selected as the master model. Ten study models were produced using Type IV stone(n=10). Lower jaw trial dentures were produced by the wax denture base and artificial teeth. Conventional complete denture (CD) group was fabricated by excluding glass fiber reinforcement (n=5). Glass fiber complete denture (GD) group was fabricated with glass fiber reinforcement (n=5). The lower jaw trial complete denture was invested using a plaster. PMMA resin was injected and curing was performed. The CD and GD groups measured the fit using silicone replica technology. The measured data was verified by t-test. Results: The anterior area of the CD group showed the smallest value. There was a statistically significant difference in the anterior area of the CD group and the GD group (p<0.05), but there was no statistically significant difference in the posterior area (p>0.05). Conclusion : Complete denture with glass fiber showed low fitness and further study is needed to apply it clinically.
