Effects of Heated Oil and Vitamin E on Lipid Peroxidative Liver Damage in Rat (가열유와 Vitamin E가 흰쥐 간장내의 과산화적 손상에 미치는 영향)

  • In order to investigate the cellular peroxidative damage due to heated oil intake and the preventive effect of vitamin E on it rats were fed heated corn oil with acid value of 4.02 at the level of 10 Cal% and three different levels of vitamin E that were 0, 40 and 200 mg/kg diet. Control group was fed fresh corn oil and 40mg/kg diet of vitamin E. After ech feeding period of 0, 3 and 6 weeks, liver superoxide dismutase (SOD), glutathione peroxidase (GPX) activities and microsomal content of vitamin E and lipid peroxide (LPO) were measured as well as cellular morphology was examined. SOD activities and LPO contents were higher, while GPX activities and vitamin e contents were lower in heated oil groups than control group. Electromicroscopic observation revealed the loss of inner mitochondrial membrane and cristae and irregular arrangement of nuclear membrane and chromatin in heated oil groups. As dietary vitamin e level was increased, SOD activity and LPO content were decreased, but GPX activity and vitamin E content in the liver increased and cellular peroxidative damage reduced progressively. This phenomena was more remarkable in 6 weeks of feeding than 3 weeks.

Effects of Freeze-Dried Garlic Powder on Lipid Improvement in Rats Fed a High Fat-Cholesterol Diet (고지방-콜레스테롤 식이 급이 흰쥐에 대한 동결건조 마늘분말의 지질개선 효과)

  • This study was carried out to investigate the effects of freeze-dried garlic powder (FDGP) on the lipid metabolism in rats fed a high fat-cholesterol (HFC) diet for 4 weeks to induce hyperlipidemia. The rats were divided into the following four experimental groups: normal diet group (NC), HFC diet group (HFC), HFC+FDGP 5 mg/100 g body weight (BW) group (FGT-1), and HFC+FDGP 15 mg/100 g BW group (FGT-2). Body weight gains and food efficiency ratios were significantly lower in the FGT groups compared to the HFC group, and the food intake was not significantly different in all experimental groups. Serum total lipid, triglyceride, and total cholesterol levels were significantly lower in the FGT groups compared to the HFC group. Total lipid content of the FGT-2 group was reduced by about 9% and 12% compared to the FGT-1 group and HFC group, respectively. The FGT groups also showed about 10% reductions low density lipoprotein (LDL)-cholesterol, atherogenic index (AI), and cardiac risk factor (CRF) compared to the HFC group. Liver function index (alkaline phosphatase, aspartate aminotransferase, and alanine aminotransferase) and lipid components were significantly lower in the FGT groups compared to the HFC group. Thiobarbituric acid reactive substances content of serum and liver tissue was reduced in the FGT groups compared to the HFC group, and was lower in the FGT-2 group compared to the FGT-1 group in serum. Antioxidant activities in serum and liver tissue were in the order of FGT-2> FGT-1> HFC group, significantly. In conclusion, FDGP effectively reduced the lipid components of serum and liver tissue. Feeding FDGP did not increase high density lipoprotein-cholesterol, but reduced LDL-cholesterol content, AI and CRF, especially. This shows that FDGP, will be effective in preventing and improving cardiovascular diseases such as arteriosclerosis and hyperlipidemia.

Protective Effects of Fermented Aloe vera on Carbon Tetrachloride-induced Hepatotoxicity in Sprague-dawley Rats (사염화탄소 유도 간독성에 대한 발효알로에의 보호효과)

  • Aloe vera extract was fermented by Lactobacillus casei. The ability of fermented Aloe vera (FAV) as an antioxidant to protect against $CCl_4$-induced oxidative stress and hepatotoxicity in rats was investigated. The rats were administered orally with various doses of FAV with 50, 100 mg/kg for 14 consecutive days. For this study, we not only tested activity of various plasma enzymes (AST, ALT), which are used as indicators of liver disease, but also checked those change of liver components such as superoxide dismutase and catalase activity. Pretreatment of FAV for two weeks significantly reduced the elevated plasma enzyme activities induced by $CCl_4$. Pretreatment of FAV also restored the hepatic enzyme, malonedialdehyde (MDA) formation. The results indicate that FAV has a protective effect against acute hepatotoxicity induced by the administration of $CCl_4$ in rats, and that the hepatoprotective effects of FAV may be due to both the inhibition of lipid peroxidation and the increase of antioxidant activity.

  • Three antioxidants were isolated: (-)-epigallocatechin and (-)-epigallo catechin gallate from Theae Folium generally called green tea, and gallic acid from Moutan Cortex. They inhibited $Fe^{3+}-ADP/ascorbate-induced$ lipid peroxidation in rat liver microsomal fraction, showing $IC_{50}$, the concentration of antioxidants which is required to inhibit 50% of the peroxidation for microsomal membranes at 1 mg protein/ml, to be $0.2\;{\mu}g/ml$, $0.4\;{\mu}g/ml$ and $2.5\;{\mu}g/ml$, respectively.

Biological Activities of Solvent Fractions from Methanolic Extract of Crataegi fructus (산사(Crataegifructus) 메탄올 추출물로부터 용매분획된 분획물의 생리활성)

  • The purpose of this study is to determine the possibility of using Crataegi fructus as natural health food source. The research was conducted to determine the biofunctional activities of Crataegi fructus extract. Methanolic extract from Crataegi fructus was partitioned by using organic solvents, including n-hexane, ethyl acetate, n-butanol, and water. Ethyl acetate soluble fraction was shown to have the strongest antioxidant activity ($RC_{50}=4.39{\mu}g/m{\ell}$) among the fractions. In the antimicrobial activity assays, ethyl acetate soluble fraction was effective in bacterial inhibition, against the cases of Escherichia coli and Klebsiella pneumonia, with minimum inhibitory concentrations in $125{\mu}g/m{\ell}$. In the anticomplementary activity assays, water soluble fraction was the most effective exhibiting 18.4% inhibitory activity.

Antioxidant Effects of Ginseng Powder on Liver of $Benzo({\alpha})Pyrene-treated$ Mice (벤조피렌을 투여한 마우스 간에서 인삼 분말의 항산화 효과)

  • In order to determine the effects of ginseng powder on the antioxidant enzyme activities of hepatotoxicity in $benzo({\alpha})pyrene[B({\alpha})P]-treated$ mice, the mice were divided into 5 groups. Ginseng powder was injected intraperitoneally once a day for 5 successive days, followed by the administration of $B({\alpha})P$ treatment on the fifth day. We also evaluated the relationship existing between lipid peroxidation and ginseng powder on oxidative stress. The increased activities of superoxide dismutase, catalase, and glutathione peroxidase observed following $B({\alpha})P-treatment$ were reduced as the result of ginseng powder treatment. Whereas, the glutathione content and glutathione S-transferase activity depleted by $B({\alpha})P$ were increased significantly, but the $B({\alpha})P-associated$ elevation of cytochrome P-450 activities and lipid peroxide content were reduced as the result of ginseng powder treatment. These results indicate that ginseng powder may exert a protective effect against $B({\alpha})P-induced$ hepatotoxicity in mice.

Antioxidative Activity and Structural Analysis of the Steroid Compound from Fomitella fraxinea (Fomitella fraxinea 중 Steroid계 화합물의 항산화 활성 및 구조분석)

  • A steroid compound, F-l with antioxidative activity from the fruit bodies of Fomitella fraxinea was isolated by ethyl acetate extraction, silica gel column chromatography, and preparative silica thin layer chromatography. The structure of the compound was determined to be crgosta-7,22-diene-3-one-$16{\beta}-ol$ by IR, NMR, and GC-Mass. The compound exhibited inhibition on lipid peroxidation of rat liver microsomes, with $IC_{50}$ value of $3.8{\mu}g/ml$.

Screening of Natural Products that have Activities against Skin-Aging (피부노화 억제활성을 갖는 천연 추출물의 탐색)

  • To screen antioxidative and whitening effects of some medicinal plants, 70% ethanol extracts were tested for their electron donating abilities to DPPH ( ${\alpha}$, ${\alpha}$-diphenyl- ${\beta}$-picrylhydrazyl) and inhibitory activities against lipid peroxidation, and tyrosinase inhibition in vitro. The electron donating activities of 70% ethanol extracts at the concentration of 100 $\mu\textrm{g}$/ml were in the order of Forsythiae fructus > Polyporaceae > Glycyrrhizae radix > Schizandrae fructus > Ramulus mori > Carthami semen. The order was Forsythiae fructus > Carthami semen > Schizandrae fructus > Polyporaceae > Glycyrrhizae radix when tested at 10 $\mu\textrm{g}$/ml, indicating that the activities depend on the concentration. The electron donating activities of Forsythiae fructus and Carthami semen at the concentration of 1 $\mu\textrm{g}$/ml were 37% and 31% respectively. Lipid peroxidation inhibitory activities were in the order of Artemisia messerschimidtiana > Glycyrrhizae uadix > Rehmaniae radix > Ramulus mori. Tyrosinase inhibitory activities were in the order of Ramulus mori > Artemisia messerschimidtiana > Oyster mushroom. The results suggest that Forsythiae fructus, Artemisia messerschimidtiana and Ramulus mori could be used as materials for anti-skin aging functional cosmetics.

DPPH Radical Scavenger Activity and Antioxidant Effects of Cham-Dang-Gui (Angelica gigas) (참당귀(Angelica gigas)의 DPPH Radical 소거 활성과 항산화 효과)

  • This study was carried out to evaluate the free radical scavenging effect and antioxidant effect of Cham-Dang-Gui (Angelica gigas) on cyclophosphamide (CYP) injected rats. Rats were divided into five groups: CON (normal group), ANS (CYP-injected and normal diet group), AND (CYP-injected and normal diet and Cham-Dang-Gui-treated group), ALS (CYP-injected and low iron diet group), and ALD (CYP-injected and low iron diet and Cham-Dang-Gui-treated group). CYP (30 mg/kg) was intraperitoneally injected to rats for early 3 days. Saline or Cham-Dang-Gui was administrated orally for entire experimental period. DPPH radical scavenger activity was measured by DPPH method, it was shown higher in methanol extract (81.5%) than in water extract (66.3%) of Cham-Dang-Gui. We observed the preventive effects of Cham-Dang-Gui on lipid oxidation of liver and protein oxidation of plasma. Hepatic SOD and catalase activities were significantly higher in CYP-injected group (ANS) than CON group, but SOD activity was slightly lowered in Cham-Dang-Gui treated group than CYP-injected group (ANS). These results suggest that extract of Cham-Dang-Gui could be useful for functional materials to reduce the oxidation of lipids and protein induced by free radicals.

Effects of Boonsimgieum aqua-acupuncture on Serum and Liver Lipid Composition and Antioxidative Capacity in Rats Fed High Oxidized Fat (분심기음약침(分心氣飮藥針)이 과산화지질(過酸化脂質)을 투여한 흰쥐의 혈장(血漿) 및 간장(肝臟)지질구성과 항산화(抗酸化)에 미치는 영향 -태충혈(太衝穴)과 내관혈(內關穴)을 중심으로-)

  • Effects of Boonsimgieum aqua-acupuncture at $Naegweun(HP_6)$ and $Taecheung(LV_3)$ on liver and plasma lipid composition and antioxidative capacity were investigated in rat fed high oxidized fat. Concentrations of plasma triglyceride, total cholesterol and LDL-cholesterol showed a tendency to increase in the high oxidized fat diet group. However these values showed a tendency to decease in aqua-acupuncture groups. HDL-cholesterol showed no significantly different in all the treatment groups. Liver total cholesterol values showed a high in control group, however other treatment groups showed no significantly different. Liver triglyceride concentration showed a high value in control group and $Naegweun(HP_6)$ aqua-acupuncture group showed lower value than $Taecheung(LV_3)$ aqua-acupuncture group. Plasma GOT and GPT values showed a tendency to increase in high oxidized fat diet group. However aqua-acupuncture groups showed a lower values than control group. The concentration of TBARS in liver and plasma showed a high values in high oxidized fat diet group, however these values showed a tendency to decrease in aqua-acupuncture group. Glutathione peroxidase, superoxide dismutase and catalase activity values showed a low values in high oxidized fat diet group, however these values showed a tendency to increase in aqua-acupuncture groups and in aqua-acupuncture groups, these values showed no significantly different.
