Ultrastructure Characterization of Hemcytes in Larvae of Protaetia brevitarsis seulensis (흰점박이꽃무지 유충의 혈구세포에 대한 형태학적 특성)

  • In this study, we used electron microscopic analysis to characterize the hemocytes in the last larva of Protaetia brevitarsis seulensis (Colbe) (Cetoniidae, Coleoptera). Granulocytes (GR), plasmatocytes (PL), oenocytoids (OE), spherulocytes (SP), prohemocytes (PR) and adipohemocytes (AD) were classified based on their size and ultrastructural differences in the circulating hemocytes. Many dark granules (<$1{\mu}m$ in diameter) in the GR's cytoplasm were observed and well-developed mitochondria, endoplasmic reticulum (ER), nucleus, and Golgi complex were also seen. After microorganisms infected, the GRs were morphologically activated and phagocytosed them. Especially, dark granules (lysosomes) were fused themselves and these bigger granules finally agglomerate together with microorganisms. Other hemocytes seem to have no immune functions.

  • The purpose of this study was to analyze the characteristics of hemocytes in the hemolymph of the larvae of Pentodon quadridens bidentulus and the characteristics of the hemocytes responsible for cellular immunity during pathogen infection. Granulocytes, plasmatocytes, oenocytoids, spherulocytes, prohemocytes and adipohemocytes were found in the circulating hemocytes. Among them, granulocyte were observed as cells responsible for immunological phagocytosis during entry of foreign substances. In particular, it was observed that the most active phagocytic action occurred within 12 hours in vivo, and that after 24 hours, the immune activation was reduced and converted to a normal state. Plasmatocytes were occasionally observed as immunological response, but the remaining hemocytes were not related to immunological activation.

  • 연두·금파리에서 혈구 세포가 면역반응의 일환으로 일으키는 결절형성(nodule formation) 현상을 형태학적으로 관찰하고자 본 실험이 수행되었다. 연두금파리 종령 유충 체내에 정상 효모, 고정된 대모, 세포액이 제거된 원형질체의 효모를 각각 주입하여 면역반응 과정을 전자현미경으로 관찰하였다 먼저 정상 효모를 주입하였을 경우, 효모와 접촉한 헐구의 과립방출과 곧 이은 혈구의 파괴 그리고 효모 주위에 혈림프와 함께 점액성응혈(clot)의 형성을 관찰 할 수 있었다 점액성 응혈에는 여러 유형의 파괴된 혈구가 포함되어 있었다. 이같은 면역반응에는 체내 혈구 중 과립혈구(granulocyte)가 가장 활발히 반응였고 응혈의 대부분을 형성하였다. 결절의 중심부인 응혈은 대모와 파괴되지 않은 혈구 대 파괴된 혈구 그리고 방출된 과립들로 차 있었고, 효모 주입 3시간 까지는 이같은 응혈은 계속 확대되었다. 그러나 10시간 이후에는 응혈이 관찰되지 않았으며 이때 체강내에서 효모가 관찰되곤 하였다. 고정된 효모를 주입하였을 경우에는 혈구파괴나 과립방출 등이 관찰되지 않았으며, 고정된 효모에 대한 혈구의 식 세포작용만이 관찰되었다. 이 식세포작용은 주입후 10시간 까지 계속되었다. 원형질 체의 효모를 주입하였을 경우에는 혈구에의한 면역반응이 관찰되지 않았다.

The Fine Structure of Spider (Araneus ventricosus) Hemocytes (산왕거미 (Araneus ventricosus) 혈구의 미세구조)

  • Hemocytes of the spider Araneus ventricosus were investigated with histochemistry, density analysis of percoll gradient, and fine structural examinations using transmission electron microscope. The hemocytes of this spider were classified into two major groups: granulocytes and non-granulocytes. The granular hemocytes were subdivided into three subtypes according to their histochemical properties which are eosinophilic granuloctes(EGs), basophilic granulocytes(BGs) and cyanocytes. The EGs, which have small granules within the cytoplasm comprise about 5% of the total henocytes. However the granules of BGs are larger than those of HGs. The cyanocytes were characterized to contain hemocyanin granules in their cytoplasm. On the other hand, the non-granulocytes were divided into three subtypes; hyaline leucocytes, oenocytoids, and molting homocytes. The hyaline leucocytes are the most abundant and the smallest hemocyte type in this spider. The oenocytoids that have $10{\sim}15{\mu}m$ in diameter are mostly found at the marginal region of the myocardium in the heart tube. The molting hemocytes, which only appeared during the molting period, contains plenty of glycogen particles in their cytoplasm.

  • The fine structural modification of the granulocytes between the molt and intermolt period were investigated by the transmission electron microscopy. The granular hemocytes of the spider Araneus ventricosus were composed of three subtypes: eosinophilic granulocytes (EGs), basophilic granulocytes (BGs) and cyanocytes. Both of the EGs and BGs have electron dense granules within their cytoplasms, however the granules of BGs are larger than those of EGs. During the molt period, some of the EGs have fine structural modification in their cell organelles including formation of phagosomes as a result of active phagocytosis. However, the BGs have no phagosomes, but electron densities of the granules are changed to lower states than the intermolt period. The cyanocyte is the biggest hemocyte among the granulocytes. They contain numerous hemocyanin crystals in the cytoplasm with some electron-lucent vacuoles. During the molt period, some of the cyanocytes are changed to irregular shapes. High magnification electron micrographs reveal that the lattice sub-structure of the hemocyanin crystals are very similar to those of microtubules, and each tubule is composed of approximately 20 filaments with fine fibrillar structure.

  • In Helice tridens tridens, hylaine cells, small granulocytes, and large granulocytes were identified. Features of hyaline cells include a large nucleus in proportion to the cytoiplasm, and weak electron-dense granules of oval shape and vesicles, and endoplasmic reticulum (ER) in the cytoplasm. Small granulocytes have smaller nucleus than that of the hyaline cells, well-developed ER, Golgi complex, and small, round and electron-dense granules in the cytoplasm. Large granulocytes contain large and electron dense granules (ahout 1 $\mu$m) that fused small granules. Hemocytes of Helice tridens tridens differentiated from hyaline cell to large granulocyte granules of hyaline cells have lysosome and make small vesicles from nuclear envelopes. While these vesicles pass through the Golgi complex, they are filled with electron dense matetials, and then fused with the small granules. They eventually matured into large granules. All of hemocytes have the glycogen particles. In the large granulocytes heterogeneouse granules were supposed to occur by disappearance of granules.

Study on the Detection of In vivo and In vitro Hemocytic Pathway in Grasshopper, Euprepicnemis shirakii (메뚜기 체내와 체외에서 혈구생성 경로 추적에 관한 연구)

  • A Differentiating pathway of hemocytes in vitro and in vivo of grasshopper, Euprepocnemis shirakii was described using light and electron microscopes. In the interior of body, the stem cells of the hemopoietic organ differentiated into six types of cells respectively which are prohemoyte, plasmatocyte, granulocyte I, granulocyte II, spherulocyte and oenocytoid. The formation of these hemocytes was derived from the stem cells surrounded by a reticular cell. Hemopoietic tissue cultured in the insect media differentiated different hemocytes, but none of them underwent any mitotic division. Morphological features of the cultured cells in media were essentially the same as those of the hemocytes differentiated from the stem cells in vivo. These results were shown that each stem cell could differentiate into different types of hemocytes. It was confirmed that the stem cells possessed the pluripotent differentiation ability to directly each hemocyte, and that the once formed hemocytes in vivo and in vitro didn t undergo further transformation to other hemocytes. The maintenance of circulating hemocytes in grasshopper had been depended on the widely spreading hemopoietic organ situated in the upper surface of the dorsal alary muscle and located on the first to eighth segments.

  • Light and transmission electron microscopy of Batillus cornutus hemocytes revealed differences that the morphological distinctions between blast-like cell, granulocytes and hyalinocytes. Base on the morphological characteristics of the cells, we identified the eight types of hemocytes and present a categorization of the hyalinocytes into six sub-categories. The hemocytes of B. cornutus were observed basophilic cell under the light microscopy. Blast-like cells had a spherical profile with a central nucleus filling almost the whole cell. Granulocytes were characterized by presenting variable numbers of granules. This cell had spherical shape with diameter 7 ${\mu}m$ and smooth endoplasmic reticula, granules, mitochondria, glycogen granules in the cytoplasm. Hyalinocytes were the most abundant cell type. Especially, hyalinocyte VI had iirregular an amoebal shape and observed autophagosome and heterophagosome in the cytoplasm. From these results, it is concluded that there are eight types of cells in the hemolymph of B. cornutus. Further studies are now needed to identify the role of these hemocytes in the enzymological and immunological response.

The Morphology of the Hemocytes of the Clam, Ruditapes philippinarum (Mollusca: Bivalvia) (한국산 바지락(Ruditapes philippinarum) 혈구의 분류)

  • To understand host-defence mechanism of clam(Ruditapes philippinarum) hemocyte against foreign materials, classification and their seasonal change in the number were performed. clams collected from a farm in Julpo Bay, Gochang, Chollabuk-Do were used in this experiment. Lots of hemocytes were found between the muscle fibers and connetive tissue of posterior adductor muscle. Hemocytes of R. philippinarum were classified into granulocytes and agranulocytes. Granulocytes were composed of three types, basophilic granulocyte, acidophilic granulocyte and fibrocyte in accordance with the staining affinities of their cytoplasmic granules. Fibrocyte has filopodia and vesicle in endoplasm and bigger than other granulocytes in size. Agranulocytes were less in the number and smaller in the size compared to those of granulocytes. Hyalinocytes had no granule in their cytoplasm. The nucleus located in the center of the cell was oval or spherical shaped. In electron microscopic observation, granulocytes and hyalinocytes contained electron-dense vesicles and some small lucid vesicles in their cytoplasm, respectively. Granulocytes phagocytosed more zymosan particles than hyalinocytes. Acidophilic granulocytes showed higher phagocytic ratio than basophilic granulocytes. Total hemocyte numbers showed the highest at April to August and the lowest at October to December. In the composition of each hemocyte, basophilic granulocytes were always more than acidophilic granulocytes and hyalinocytes.

  • Ultrastructure of the larval hemocytes in Sericinus montela Grey was observed with the transmission and scanning electron microscope. The 5 cell types, according to Breh lin and Zachary(1986), identified in the hemocytes of the last instar S. montela larva were; prohemocyte, plasmatocyte, granular cell, spherule cell and oenocytoid. Prohemocytes could be easily charaterized by a relatively large nucleus nd poorly developed cytoplasmic organelles. Plasmatocytes were relatively large spindle-shaped or oval cells with fibrous bundles. The spindle-shaped plasmatocytes, especially, were characterized by a well developed, elongated nucleus. Granular cells had various granules, either morphologically or in electron density, and highly developed cytoplasmic organelles in the cytoplasm. They had numerous cytoplasmic processes. Spherule cells were characterized by cytoplasmic spherules containing fine materials, which are released into the hemolymph upon maturity of the cell. Oenocytoids consisted of electron dense materials and the organelles in cytoplasm were poorly developed.
