Immuno-stimulatory Activities of a High Molecular Weight Fraction from Cynanchum wilfordii Radix Obtained by Ultrafiltration (한외거르기(Ultrafiltration)에 의하여 분리된 백수오 고분자 분획물의 면역증진 활성)

  • The purpose of this study was to investigate the immuno-stimulatory activity of the high-molecular-weight fraction (HMWF) of Cynanchum wilfordii (CW) extracts obtained by ultrafiltration in murine macrophage RAW 264.7 cells and to assess its immuno-stimulatory effect in mice. Ultrafiltration was performed with polyethersulfone membranes (30 kDa cutoff) in a cross-flow filtration system to obtain the HMWF of CW. The results showed that the HMWF increased the production of various cytokines such as tumor necrosis factor-${\alpha}$, interleukin-6, and nitric oxide in dose-ependent manners. In addition, HMWF treatment increased the relative spleen weight as well as splenocyte proliferation induced by concanavalin A or bacterial lipopolysaccharide in mice. Natural killer (NK) cell activity in the HMWF-treated group was significantly increased compared to that in the control group. These results suggest that the HMWF of CW can support the immune system through secretion of macrophage cytokines, thereby enhancing NK cell activity and murine splenocyte proliferation.

Optimization of Microwave Extraction Conditions for Antioxidant Phenolic Compounds from Ligustrum lucidum Aiton Using Response Surface Methodology (반응표면분석법을 이용한 여정자의 페놀계 항산화 성분에 대한 마이크로웨이브 추출조건 최적화)

  • Response surface methodology (RSM) was applied to optimize the microwave-assisted extraction (MAE) conditions for electron-donating ability, total phenol content, and total flavonoid content of Ligustrum lucidum Aiton. Ligustrum lucidum Aiton from different regions was tested, and Ligustrum lucidum Aiton from Haenam was chosen due to its higher total phenolic content, total flavonoid content, DPPH radical scavenging activity and ABTS radical scavenging activity compared to the other samples. Central composite design was used to optimize extraction of Ligustrum lucidum Aiton from Haenam as well as determine the effects of extraction temperature ($X_1$) and extraction time ($X_2$) on dependent variables ($Y_n$). Determination coefficients ($R^2$) of the regression equations for dependent variables ranged from 0.8858 to 0.9517. The optimum points were $131.68^{\circ}C$ for extraction temperature and 5.49 min for extraction time. Predicted values of the optimized conditions were acceptable when compared to experimental values.

Optimization of Solvent Extraction Process on the Functional Components from Portulaca oleracea Using a Response Surface Methodology (쇠비름의 유용성분 환류추출공정의 최적화)

  • Various functional and useful components in Portulaca oleracea were extracted with ethanol and the optimum solvent conditions were set by monitoring of response surface methodology(RSM). A central composite design for optimization was applied to investigate the effects of the three independent variables of extraction temperature, ethanol concentration, and extraction time, on dependent variables including total phenolics, electron-donating ability, brown clolor and total flavonoids of Portulaca oleracea. The content of total phenol was essentially unaffected by extraction time or extraction temperature, but it was highly influenced by ethanol concentration. The maximum total phenol content was 31.70mg/mL obtained at 45.84% of ethanol concentration, $79.66^{\circ}C$, and after 2.67hr of extraction. Electron-donating ability (EDA) was affected by ethanol concentration and the maximum EDA was 74.67mg/mL at 52.95% ethanol concentration, $52.33^{\circ}C$ and 4.84hr of extration time. The browning color was rarely affected by extraction time but, it was highly influenced by ethanol concentration and extraction temperature. The maximum extent of browning color was obtained at 97.75% of ethanol concentraion, $65.88^{\circ}C$ and 2.93hr of extraction time. The content of total flavonoid was significantly influenced by extraction time, and the maximum total flavonoid level was 58.28mg/mL obtained at 96.62% ethanol concentration, $61.87^{\circ}C$ after 3.70hr of extraction. As a result, The optimal conditions for effective extraction were predicted as follows, 70.3% of ethanol concentration, $62.1^{\circ}C$ of extraction temperature and 3.3hr of extraction time.

  • The optimization of extraction conditions of phytoestrogen from pomegranate by hot water was conducted by analyzing the extraction characteristics. The purpose of this study was effective utilization of bioactive components of pomegranate, and the analyzing was performed with response surface methodology (RSM). This study established 10 sections based on the central composite design with the independent variables of extraction temperature (60, 70, 80, 90, $100^{\circ}C$) and extraction time (1, 2, 3, 4, 5 hr) to predict the optimal conditions for extraction of the effective components. The dependent variables were measured for extracted materials, those were, the major components such as chlorogenic acid, kaempferol, $17-{\alpha}-estradiol\;and\;17-{\beta}-estradiol$ content, and regression analysis was performed by SAS program, and optimal conditions for each characteristics were predicted, and the characteristics of extraction were analyzed by response surface methodology. It was found that chlorogenic acid, kaempferol, and $17-{\alpha}-estradiol$ content were greatly affected by extraction temperature. However, $17-{\beta}-estradiol$ content was affected significantly by extraction time. Regression formulas for each variable were elicited from this study, and the chlorogenic acid, kaempferol, $17-{\alpha}-estradiol\;and\;17-{\beta}-estradiol$ content depending on response surface methodology factor were superimposed. It was shown that optimal temperature and extraction time were $98{\sim}100^{\circ}C\;and\;3{\sim}5$ hrs, respectively.

Manufacture of the vol-oxidizer with a capacity of 20 kg HM/batch in $UO_2$ pellets using a design model (설계 모델을 이용한 $UO_2$ 펠릿 20 kg HM/batch용 분말화 장치 제작)

  • Vol-oxidizer is a device to convert $UO_2$ pellets into $U_3O_8$ powder and to feed a homogeneous powder into a Metal Conversion Reactor in the ACP(Advanced Spent Fuel Conditioning Process). In this paper, we propose a design model of the vol-oxidizer, develop the new vol-oxidizer with a capacity of 20 kg HM/batch in $UO_2$ pellets, and conduct a verification for the device. Design considerations include the internal structure, the capacity, the heating position of the device, and the size. The dimensions of the new vol-oxidizer are decided by the design model. We determine a permeability test of the $U_3O_8$ measuring the temperature distribution, and the volume of $UO_2$ and $U_3O_8$. We manufactured the new vol-oxidizer for a 20 kg HM/batch in $UO_2$ pellets, and then analyzed the characteristics of the $U_3O_8$ powder for the verification. The experimental results show that the permeability of the $U_3O_8$ throughout mesh enhance more than old vol-oxidizer, the oxidation time takes only 8 hours when compared with the 13 hours of the old device, and the average distribution of particle size is $40{\mu}m$. The capacities of new vol-oxidizer for a 20 kg HM/batch in $UO_2$ pellets were agree well with the predictions of design model.

Statistical Optimization of Production Medium for Enhanced Production of Itaconic Acid Biosynthesized by Fungal Cells of Aspergillus terreus (Aspergillus terreus에 의해 생합성되는 이타콘산의 생산성 증가를 위한 통계적 생산배지 최적화)

  • Statistical optimization of the production medium was carried out in order to find an optimal medium composition in itaconic acid fermentation process. Itaconic acid utilized in the manufacture of various synthetic resins is a dicarboxylic acid biosynthesized by fungal cells of Aspergillus terreus in a branch of the TCA cycle via decarboxylation of cis-aconitate. Through OFAT (one factor at a time) experiments, six components (glucose, fructose, sucrose, soluble starch, soybean meal and cottonseed flour) were found to have significant effects on itaconic production among various carbon- and nitrogen-sources. Hence, using these six factors, interactive effects were investigated via fractional factorial design, showing that the initial concentrations of sucrose and cottonseed flour should be high for enhanced production of itaconic acid. Furthermore, through full factorial design (FFD) experiments, negative effects of $KH_2PO_4$ and $MgSO_4$ on itaconic acid biosynthesis were demonstrated, when excess amounts of the each component were initially added. Based on the FFD analysis, further statistical experiments were conducted along the steepest ascent path, followed by response surface method (RSM) in order to obtain optimal concentrations of the constituent nutrients. As a result, optimized concentrations of sucrose and cottonseed flour were found to be 90.4g/L and 53.8g/L respectively, with the corresponding production level of itaconic acid to be 4.36 g/L (about 7 fold higher productivity as compared to the previous production medium). From these experimental results, it was assumed that optimum ratio of the constituent carbon (sucrose) and nitrogen (cottonseed flour) sources was one of the most important factors for the enhanced production of itaconic acid.

Dynamic Network Loading Model based on Moving Cell Theory (Moving Cell Theory를 이용한 동적 교통망 부하 모형의 개발)

  • In this paper, we developed DNL(Dynamic Network Loading) model based on Moving cell theory to analyze the dynamic characteristics of traffic flow in congested network. In this paper vehicles entered into link at same interval would construct one cell, and the cells moved according to Cell following rule. In the past researches relating to DNL model a continuous single link is separated into two sections such as running section and queuing section to describe physical queue so that various dynamic states generated in real link are only simplified by running and queuing state. However, the approach has some difficulties in simulating various dynamic flow characteristics. To overcome these problems, we present Moving cell theory which is developed by combining Car following theory and Lagrangian method mainly using for the analysis of air pollutants dispersion. In Moving cell theory platoons are represented by cells and each cell is processed by Cell following theory. This type of simulation model is firstly presented by Cremer et al(1999). However they did not develop merging and diverging model because their model was applied to basic freeway section. Moreover they set the number of vehicles which can be included in one cell in one interval so this formulation cant apply to signalized intersection in urban network. To solve these difficulties we develop new approach using Moving cell theory and simulate traffic flow dynamics continuously by movement and state transition of the cells. The developed model are played on simple network including merging and diverging section and it shows improved abilities to describe flow dynamics comparing past DNL models.

Effect of Antibody Immobilization Method to Magnetic Micro Beads on its Immunobinding Characteristics (자성 미세입자에의 항체 고정화 방법이 면역결합반응에 미치는 영향)

  • Recent technical advances in the biorecognition engineering and the microparticle fabrication may enable us to develop the single step purification using magnetic particle, because of its simplicity, efficacy, ease of automation, and process economics. In this study, we used commercial magnetic particles from Seradyn, Inc. (Indianapolis, USA). It was ca. 2.8 micron in diameter, consisted of polystyrene core and magnetite coating, and its surface had carboxyl groups. The model, capture protein was IgG and anti-IgG was used as the ligand molecule. We studied the different surfaces ('nude', ester-activated, and anti-IgG coated) for their biorecognition of IgG. At a high pH condition, we could reduce non-specific binding. Also anti-IgG immobilized magnetic particle could capture IgG more selectively. We attempted 'oriented immobilization' of anti-IgG, in which the polysaccharides moiety near the C-terminus was selectively oxidized and linked to the hydrazine-coated MP, to improve the efficacy of biorecognitive binding. Using this method, the IgG capturing ability was improved by ca. 2 fold. From the binary mixture of the IgG-insulin, IgG could be more selectively captured. In summary, the oriented immobilization of oxidized anti-IgG proved to be as effective as the streptavidin-biotin system and yet simpler and cost-effective. This immobilization method can find its applications in protein biochips and biotargeting.

A Study on the Lime Stabilization of Livestock Waste (축산폐기물의 안정화 처리에 대한 연구)

  • One of alternative conventional technologies used for treatment of livestock wastes is composting process, and recently some mechanical composting processes are being practiced. It is, however, recognized the composting process also has its own limitations such as longer time requirement, and difficulties to estimate the degree of decomposition, etc. The incomplete compost contains potentially harmful materials to crops and public health due to instabilized organic contents and pathogenic organisms. The purpose of this investigation is to develop an innovative system whereby anxious livestock wastes are thoroughly stabilized and disinfected. Thus the overall management scheme should meet the following requirements. 1. A system should be in a cost-effective and environmentally sound manner. 2. Sludges must be chemically stabilized and bacteriologically safe. 3. Odor-free by product should be applied to crop land. 4. Sludges are sources of fertilizer nutrients and/or soil amendments to enhance crop production. 5. And they can be used as potential pH adjusting agent of the acidified soils. Overall effectiveness of the developed system is experimentally tested to satisfy the preset criteria and requirements. Major experiments are divided into four categories: they are 1. chemical stability test, 2. optimal condition test of stabilization process, 3. bacteriological examination and disinfection tests, and 4. deodorization tests The stabilization process is consisted of the stabilizing reaction process and the drying process. Stabilized wastes is dried by both sun dryer and rotary dryer. It is shown that an additive dosage of about g/kg solid in wastes with a minimum of 5-minutes reaction would be necessary for effective stabilization reaction. In the stabilization reaction process, the pH of wastes is lowered from initial values of 12.3 to 8.6. High pH prevents odor production and kills pathogenic organisms. Organic matter contents in the stabilized wastes are about 50% and the sum of contents of fertilizer elements such as total nitrogen, $P_2O_5$ and $K_2O$ are about 5.3%. The livestock wastes that are stabilized chemically and hygienically can be used as a good soil conditioner and/or organic fertilizer.

Reestablishment of Approval Toxin Amount in Paralytic Shellfish Poison-Infested Shellfish 3. Thermal Resistance of Paralytic Shellfish Poison (마비성 패류독 허용기준치 재설정을 위한 연구 3. 마비성 패류독의 내열성)

  • The purpose of this study was to determine the kinetics of paralytic shellfish poison (PSP) destruction at various temperature. The toxic digestive gland homogenate of blue mussel (Mytilus edulis), PSP crude toxin, gonyautoxin group and saxitoxin group were heated at temperature ranging from 90 to $120^{\circ}C$, and then the toxicities were measured in samples heated for various time intervals. The rate constant (k) of the toxic digestive gland homogenate, PSP crude toxin, gonyautoxin group and saxitoxin group were $3.28{\times}10^{-2},\;1.20{\times}10^{-2},\;5.88{\times}10^{-2}\;and\;2.58{\times}10^{-2}\;at\;120^{\circ}C$, respectively. The decimal reduction time (D-value) of the toxic digestive gland homogenate, PSP crude toxin, gonyautoxin group and saxitoxin group were 70, 192, 39 and 89 at $120^{\circ}C$, respectively. These results indicate that PSP crude toxin is most heat-stable of 4 types of PSP toxins and PSP toxin are more heat-stable than food poisoning bacteria and spores. The retorting condition to reduce PSP toxicity below quarantine limit ($80\;\mu\textrm{g}/100\;g$ in Korea and America, 4 MU/g in Japan) could be calculated by rate constant. For example, the digestive gland homogenate having a initial toxicity of $200\;\mu\textrm{g}/100\;g$ could have toxicity below quarantine limit when heated at $90^{\circ}C$ for 129 min., $100^{\circ}C$ for 82 min., $110^{\circ}C$ for 48 min. and $120^{\circ}C$ for 28 min. These results suggest that commercial retorting condition ($115^{\circ}C$ for 70 min) in Korea is enough to reduce toxicity below quarantine limit from initial toxicity of $200\;\mu\textrm{g}/100\;g$. From these results, the quarantine limit of PSP-infested shellfish for canning can be level up to raw score of $200\;\mu\textrm{g}/100\;g$.
