Sox4, a transcription factor, consists of three functional domains: an HMG-box domain as a DNA binding domain, serine rich region as a transactivation domain and glycine rich region (GRR), an unknown functional domain. Although Sox4 is known to be functionally involved in heart, B-cell and reproductive system development, its physiological function remains to be elucidated. We used pGEX expression system to develop a simple and rapid method for purifying Sox4 protein in suitable forms for biochemical studies of their functions. Unexpectedly, we observed that full-length Sox4 appears to be protease-sensitive during expression and purification in E. coli. To map the protease-sensitive site in Sox4, we generated various constructs with each of functional domains of Sox4 and purified as the GST-Sox4 fusion proteins using glutathione beads. We found that the specific cleavage site for the proteolytic enzyme, which exists in E. coli, is localized within the novel GRR of Sox4. Our study suggest that the GRR of Sox4 may a target for the cellular protease action and this cleavage in the GRR may be involved in regulating physiological function of Sox4. Additionally, our study may provide a useful method for investigating the proteolytic cleavage of the target molecule in E. coli.

This study focuses on the methodology of an ecological interpretation of Korean traditional landscapes through both life-oriented philosophy and traditional Korean philosophy which are similar to ecology. Also, useful tools for discovering an ecological technique of formation based on the ecological thoughts in ancestors' life are shown. Ecological key words as interpretative tools on the traditional landscape replaced ecological concepts in Korean culture and landscape. There are 'Bonsung(本性; the original nature)', 'Chungjeol(中絶; moderation)', 'Hyoyul(效率; efficiency)', 'Sangsaeng(相生; symbiosis)', 'Jasaeng(自生; self-generation)', 'Chunghwa(中和; neutralization)', 'Bangtong(旁通; communication)', and 'Byuntong(變通; variableness)'. For the case study, the concepts of 'spatial structure', 'constructive elements in the traditional gardens', and 'structural elements in the dwelling houses' were extracted from $\ulcorner$Imwonkyeongjeji$\lrcorner$ as an interpretative subject. As a result, Jeongon house, Jongtaek of Choi's family(the first incoming resident) showed us an ecological technique of formation by interpretation on the composing elements. Namely, they are natural dwelling houses in harmony with natural conditions and delicate relational styles. Five kinds of ecological characteristics were exposed. They are: 1. land use method following natural features('本性' '相生' '中和'), 2. physical and spatial elements in a body with nature('中絶' '相生' '中和'), 3. sustainable circulation system by recycling limited resources('效率' '自生' '旁通'), 4. use of natural materials based on the regional climate ('中絶' '效率' '自生') and 5. plane and structural decision by microclimate('效率' '自生' '變通'). Consequently, the dwelling houses and their traditional gardens aimed at the consuming space of the efficient resources by utilizing and circulating natural energy more than different types of the traditional spaces.

Certain complex forces are existing at the other side of a canvas. These are visual arrangements and within the arrangement, even the simple combination of dots and lines create more than visual patterns. No matter what form that visual art has taken, it is performed in a certain space and it is constucted with various visual elements. For effective communication, visual elements must form a stable sturcture by extablishing organic relationship among each other. These requires an understanding of the human visual characteristics and psychonology, because human sight senses the same biological and psychological visual elements differently in its force and weight, according to the position within a given space. Although the structure which controls such force and weight exists within a peice of work, it is a internal plan of the work that actually controls the communication through a man\ulcorner psychological reactions towards the horizontal and virtical structures within it. Moreover it is a visual statement that effectively expresses a theme or a message. This thesis has studied visual structures through the analysis of art pieces regarding of these kind of a visual picture plane, characteristics of a picture plane formed by a structure of force existing on the other side of the picture and the theory of visual balance. In addition, the aim of this study is to help designers who deals with visual image works to understand the visual structures and psychological recognitions and to apply these picture plane compositions at their real work by recognizing the psychological power within the construction elements.

Learner-oriented education and distance education based on Information & Communication technology are educational countermeasure to various demands of Information age. Especially Web has the characteristics of providing proper environments for implementing constructivism study theory. Yet, existing web-based evaluation system is implemented by HTML and Java script so it has low interaction and imperfect educational effects. Therefore, We evaluated students' study understanding ability of real-time distance education system and analyzed evaluation results to offer useful information for level study considering individual variation. In this thesis, with these results, we designed and implemented interworking and multimedia databank system without time and space limitations on Client/Server environment.

DDA (Discontinuous Deformation Anlaysis) is one of the latest numerical analysis which has merits of both FEM and DEM. In this research a new edge-to-edge contact algorithm was applied on DDA. With adoption of new edge-to-edge contact state definition, sub-algorithm was improved about open-close iteration, contact state judge, contact detecting, and friction forces acting on joints. Newly applied DDA was verified based on two different cases. The DDA results show good agreement with numerically predicted one.

Image compression is the application of Data compression on digital images. The (DCT) discrete cosine transform is a technique for converting a time domain to a frequency domain. It is widely used in image compression. First, the image is divided into 8x8 pixel blocks. Apply the DCT to each block while processing from top to bottom from left to right. Each block is compressed through quantization. The space of the compressed block array constituting the image is greatly reduced. Reconstruct the image through the IDCT. The purpose of this research is to understand compression/decompression of images using the DCT method.

Dramatic changes have so far been made in the system of central places over the past century. The development of transport has particularly exercised principal influences on the changes of the system of central places. This paper is to examine how and why the development of transport enables such changes to take place in the system of central places, through conducting model research. This paper suggests some models that development of transport be taken as an independent variable and that the division of market area between the central places be as the development of variable. The suggested models show that the development of transport extends the market area of higher central places but reduces the market area of lower central places.

국립고흥청소년우주체험센터는, 우리나라 청소년들에게 우주에서 벌어지는 제반 현상과 천체의 실상을 접하게 하고 또 그 현상과 천체 이면에 자리한 기본 원리를 체험과 실험을 통하여 스스로 터득하게 함으로써 저들을 우주시대를 이끌어갈 창조적 인재로 양성함을 목적으로, 2010년 7월 9일에 개원한 특성화 체험 국립 시설입니다. 우리나라 청소년들이 센터에 설치된 주요 우주체험장비 7종을 활용하여 우주왕복선과 우주정거장에서의 임무수행을 경험하며, 우주왕복선 지상통제센터 요원의 역할 및 우주선이나 항공기의 직접 조종도 체험 할수 있다. 또한 달의 저중력 상황을 체험해 볼 수 있다. 지름 15미터의 천체투영관이 12월 말에 완공될 예정이다. 하늘전망대에는 구경 14인치 광학망원경과 다수의 중소형 망원경들이 설치된다. 또한 우주인 기초체력단련훈련을 할 수 있는 야외 활동 공간을 확보하고 있다. 센터의 우주체험시설과 천문학 관련 시설들은 앞으로 청소년의 우주에 대한 관심을 유발하고 국민들의 과학에 대한 이해를 돕는데 크게 기여할 것으로 기대된다. 현재 4개의 과정으로 우주체험캠프 프로그램을 구성하여 운영하고 있다. 또한, 2009년 개정 지구과학 교육과정과 연계한 우주체험프로그램을 개발 중이다. 여기서는 본 센터의 우주체험시설물 소개와 이들을 활용한 체험활동과 교육 현황 및 계획을 소개하고자 한다.

본 논문은 언어장애인을 위한 통신보조기기의 구현에 관한 연구이다. 통신보조기기에 적용되는 어휘는 사용자의 환경을 고려하여 선택되어야 하며 사용자의 환경에는 연령, 교육정도, 가족관계, 자주 이용하는 장소, 장애의 종류와 정도 등 사용자의 정신적, 육체적 능력을 모두 포함하여야 한다. 본 논문에서는 손의 사용이 가능하고, 어휘와 의미심볼의 관게를 이해할 수 있으면서, 음성표현이 부자유스러운 어린이를 사용자로 한정하였다. 사용자에 의해 발훼된 어휘는 중심어휘와 특정환경을 나타내는 장소 도메인에서의 사용어휘로 나눌 수 있다. 중심어휘는 장소에 구애받지 않고 일상생활에서 널리 쓰이는 어휘를 말하며, 장소 도메인에서의 사용어휘로 나눌 수 있다. 중심어휘는 장소에 구애받지 않고 일상생활에서 널리 쓰이는 어휘를 말하며, 장소 도메인에서의 사용어휘는 특정 장소에서 빈번히 사용하는 어휘를 말한다. 발췌된 어휘는 휴대용 통신보조기기로서의 공간적 제약을 극복하기 위하여 어휘를 명사, 동사, 조사로 나누어 좀더 많은 문장을 만들 수 있도록 하였으며, 동적 시스템과 정적시스템의 장점을 고려하여 장소 도메인 별 어휘로 나누면서 의미함축의 원리를 도입하였다. 또한 어휘의 인식이 쉽도록 의미심볼과 어휘를 대응하여 표현하였고, 시스템의 기능 혹은 어휘분류에 따른 화면의 배경색을 다르게 설정하여 쉽게 사용자가 선택할 수 있도록 하였으며 , 사용자의 선택에 의해 구성된 문장은 화면표시관에 보여진 후 음성으로 표현하였다.

When we put a construction on the space organization form of Seyeon-jong garden through a consequence of drawing analysis, we were able to certify that a certain space construction principle was applied as following. First, the arrangement and organization of space construction element embodied systematic order y clearing the character of space, and heightening the degree of cognizance with the means of functionability, scale, height, decorationability, an inclination of artificiality and nature, systemetic approchability. Second, the space construction element of Seyeon-jong garden pursued a harmony of human work and nature for creating a various atmosphere and visual quality. Third, Seyeon-jong garden constructed the space by establishing the measurement of human scale that introduced a conception of near, middle, distant view on the visual angle. Forth, Yun go san introduced the natural shape of 'The song of five friends' as a construction element, and represented the symbolism that he had seeked for throught he arrangement method, christening, composing technique of the space construction element for expressing ideal space which is described in 'The four season song of fishermen' spatially at Seyeon-jong garden. As the results of this study, that Yun go san created his own ideal space that he longed for at Seyeon-jong garden with applying fixed design system, and he realized his will that he strived to form a complete whole in perfect harmony.
