Embryogenic cell suspension culture and plant regeneration in zoysiagrass (Zoysia japonica Steud) (한국들잔디 배아세포의 부유배양과 식물체 재생)

  • Zoysiagrass (Zoysia japonica Steud) is a warm season turfgrass species widely used for sports field and golf courses. Many cultivars are propagated through vegetative methods. This study was conducted to develop an optimum culture medium and culture conditions for embryogenic callus induction and plant regeneration, and to establish a cell suspension culture system for use in zoysiagrass breeding and propagation. The results indicated that adding $Cu^{++}$ at 2.5 mg $L^{-1}$ to the induction medium was optimum for callus induction. Increasing the numbers of sub-culture cycles improved the quality of calli. The optimum dosage for cell suspension culture ranged from 2.5 to 10 mL. The embryogenic callus suspension used in this study had a plant regeneration rate of 58%.

Identification and Characteristics of Sclerotinia homoeocarpa Causing Dollar Spot Disease in Zoysiagrass (한국잔디에 발생한 달라스팟 병의 주요 원인균인 Sclerotinia homoeocarpa의 동정 및 특성)

  • A fungal isolate was newly collected from Zoysiagrass 'Anyang-Joongji' in small circular patches on a fairway ofa golf course in Korea, which seriously occurred during the early summer period of 2005. The isolate presented on PDAmedium, named Scz1, was closely identical to Sclerotinia homoeocarpa, a casual fungus of dollar spot disease, in cool season turf grasses such as creeping bentgrass. Hereby, this study was accomplished to characterize the isolate and compare it with the fungus, named Scb1, isolated from dollar spot-infected creeping bentgrass (Agrostis palustris Huds. cv Penncross). On PDAmedium, individual mycelial appearance of three isolates was very similar except for the pigment. Mycelial pigments of Scz1 and Scz2 (another analogous isolate collected) were light pinkish on the reverse side of PDA medium but that of Scb1 was dark brownish. In a microscopic study, three isolates were barely distinguishable in the appearance of mycelia. As expected, in the temperaturesensitivity assay, all pathogens were very delicate to $32^{circ}C$ above but not to $30^{circ}C$ below, in which was explained to be one of typical characteristics in S. homoeocarpa. In an artificial inoculation assay, disease symptoms including leaf spots in Zoysiagrass were appeared within 6-7 days after inoculation through the hand inoculation method with the isolate-infested soil. Then the fungus was re-identified from the infected leaf tissues. Interestingly, inoculation of isolate Scz1 gave rise to distinct symptoms in only Zoysiagrass but not in creeping bentgrass 'Penncross' and Kentucky bluegrass 'Midnight'. The observation might be involved in host specific pathogenecity of S. homoeocarpa Scz1 to Zoysiagrass. In a chemical sensitivity assay for the isolate, Scz1, showed a high mycelial inhibition against two fungicides, iprodione and propiconazole. All results described above suggest that S. homoeocarpa Scz1 is a primary pathogen of Zoysia dollar spot disease.

Antifungal Activities of Pseudomonas spp. Strains Against Plant Pathogens and Optimization of Culture Conditions (식물병원성 진균에 항균 효과를 지닌 슈도모나스 균주의 항진균 활성 증진을 위한 배양조건의 최적화)

  • To define the optimum conditions for the mass production of four antifungal Pseudomonas spp. isolated from soil, we have investigated culture conditions and effects of various nutrient sources on the bacterial growth and evaluated antagonistic activity against Rhizoctonia solani and Sclerotinia homoeocarpa, plant pathogens. The optimum temperature and pH for the growth of these isolates were determined as pH 7.0 and $20^{\circ}$ or $25^{\circ}C$, respectively. Sucrose, tryptone, and $K_2HPO_4$ generally were more adequate for better growth as carbon, nitrogen and mineral source, respectively. The nutrient sources were also found to be very effective for high antifungal activities against R. solani and S. homoeocarpa. It was elucidated that YUD-F group (P. mandelii and P. fluorescens), which inhabit regions at relatively low temperature, had more broad spectrum and higher antifungal activity than YUD-O group (P. trivialis and P. jessenii) generally against R. solani and S. homoeocarpa. It is thought that the differences of the average temperature in the various habitats of Pseudomonas spp. influence the optimal growth temperature and antifungal activity. Especially, Pseudomonas spp. of YUD-O group showed the better antifungal activity against dollar spot caused by S. homoeocarpa, but showed relatively weaker antifungal activity against brown patch caused by R. solani.

A Study on the Characteristics of Green Design and Construction of Golf Courses in Korea (한국의 골프 코스 그린의 설계 및 시공 특성에 관한 연구)

  • This study was carried out to survey green system, area, green slope, green turfgrass variety, green section, and particle size of green construction materials, and to investigate and evaluate the characteristics of Design and Construction in Korean golf course green for improving the quality of Korean golf course into that of the international golf course held international tournament. The results were as follows. 1. The greens of 129 Korean golf courses consisted of 2(two) green system and 1(one) green system. 2(two) green system was 50.8%, 1(one) green system was 40.7%, and 1+2 green system was 8.5% of them. 2. In 48 Korean golf courses, the green area of 2(two) green system was mostly 400~$600\m^2$(56.5%) and the green area of 1(one) green system was mostly 600~$800\m^2$(47.8%). In 48 Korean golf courses, 1.5~3% green slope appeared the highest frequency(50.0%) and the next was 3~5%(29.4%). 3. Penncross variety was the highest frequency(71.2%). The next was mixed variety (Penncross+Crenshaw, Penn A-1, Pennlinks, or Penneagle/SR 1020+SR 1019) and the frequency of mixed variety was 7.6%. 4. In 48 Korean golf courses, 70~80cm total thickness of green appeared the highest frequency(36.1%), 10~20cm thickness of green mixed sandy layer appeared the highest frequency(43.6%), and 10~20cm thickness of green coarse sandy layer appeared the highest frequency(55.6%). 0~10cm thickness of green gravel layer appeared the highest frequency(67.6%), 20~30cm thickness of green drain layer appeared the highest frequency(52.8%), and 20~30cm width of green drain layer appeared the highest frequency(44.4%). Below 1mm sand diameter used in green mixed sandy layer appeared the highest frequency(46.2%), below 2mm or over 2mm sand diameter used in green coarse sandy layer appeared the highest frequency(31.4%). 20~40mm coarse gravel diameter used in green gravel layer appeared the highest frequency(43.2%) and 0~20mm fine gravel diameter used in green gravel layer appeared the highest frequency(65.8%). 20~40mm gravel diameter used in green drain layer appeared the highest frequency(64.1%).

Growth of Creeping Bentgrass by Application of Compound Fertilizer Containing Microbes (미생물 함유 복합비료 시비에 따른 크리핑 벤트그래스의 생육)

  • Superintendents have been used microbial fertilizers to improve turfgrass growth and quality and to decrease turfgrass diseases in golf course. This study was conducted to investigate the effects of compound fertilizer containing microbe (MF) on the growth and quality of creeping bentgrass with turf color index (TCI), chlorophyll index (ChI), root length, turfgrass density, clipping yield and nutrient content. Treatments were designed as follows; non-fertilizer (NF), compound fertilizer (21-17-17; CF) as control, compound fertilizer (14-6-17) containing microbe. In pot experiment, TCI and ChI of creeping bentgrass in MF plot were similar to those in CF. But clipping dry weight of MF plot increased by 39.1% compared to that of CF plot. At field experiment applied with MF treatment, TCI, ChI, root length, and nutrient content and uptake of creeping bentgrass were similar to those with CF treatment, but turfgrass density with MF higher about 7.9-15.8% than with CF. These results indicated that the application of MF improved growth and quality of creeping bentgrass by enhancing clipping yield and shoot number.

Effect of Head Rice Ratio on Rice Palatability (쌀의 완전미 비율 증가가 식미에 미치는 영향)

  • This experiment aimed to estimate the effect of imperfect rice on rice palatability. Rice cultivar, Ilpumbyeo, was cultivated by direct-seeding on flooded paddy surface with 11 kg/10a nitrogen application. Palatability of harvested rice was measured by NIR spectroscope. Brown rice was divided according to their appearance namely, perfect, discolored, green-kerneled, and immature opaque with a composition ratio of $75.7\%,\;11.0\%,\;8.0\%,\;and\;5.3\%$ respectively. When the perfect brown rice was milled, the grain were composed of head, cracked, and white core & belly, at $64.7\%,\;25.3\%\;and\;10.0\%$ respectively. The milled rice of discolored brown rice had similar composition with the perfect rice. The milled green-kerneled vice, on the other hand, had $36\%$ head rice and $64\%$ white core & belly rice. The immature opaque brown rice, when milled, had $25.3\%$ white core & belly and $74.7\%$ damage & opaque rice. With the respect to grain quality, the viscosity of white core at belly rice and damaged & opaque rice was lower than that of head rice. In contrast, their protein content was a little higher than that of head rice. The palatability value of pure imperfect rice was much lower than head rice. The palatability value of damaged & opaque rice was the lowest among the imperfect rices. When mixed with head rice, the damaged & opaque rice impacted on the deterioration of vice palatability. Mixing $1\%$ each of white core at belly rice and damaged h opaque rice decreased the palatability value by $5\%$ as compared with the head rice.

Comparison of Grain Quality between Chinese Parboiled and Domestic Rice (국내산 쌀과 중국산 찐쌀의 품질 비교)

  • This study was carried out to compare the rice quality between domestic brand rice and imported Chinese rice. In the appearance of grain, head rice rates were $96.1\%$ in domestic brand rice (Ilpumbyeo), $94.1\%$ in Jingguan (Chinese brand rice) and $86.5\%$ in Xiaozhandao(Chinese brand rice). In case of Chinese parboiled rice, the head rice rate was $0\%$ because the cracked and broken rice occupied about $95.5\%$. The low protein contents in Ilpumbyeo and Xiaozhandao as 6.5 and $6.7\%$ show relatively high palatability as 81.9 and 71.4. However, high protein contents in Jingguan and Chinese parboiled rice as 7.5, $7.4\%$ show low palatability as 64.3 and 55.6. In viscosity characteristics, peak viscosity, break down and final viscosity were higher in Ilpumbyeo and Xiaozhandao, midium in Jingguan and lowest in Chinese parboiled rice. And the set back value, which was negatively related with amylose content, was lowest in Ilpumbyeo and highest in Chinese parboiled, suggesting slow deterioration in Ilpumbyeo and rapid deterioration in Chinese parboiled rice. Overall physical components of cooked rice measured by Texture Analyser were higher in Ilpumbyeo than those in Chinese parboiled rice.

Characteristics and Correlation between Green Management Practices and Speed in Korean Golf Courses (한국의 골프 코스 그린의 관리 및 스피드 특성과 상관에 관한 연구)

  • This study is carried out to investigate the cahracteristics of green management practices and green speed(i.e., ball-roll distance) on 129 Golf Courses in Korea, and to explain the effects of managemet practices that affect green speed. Data collected from green-keepers were subjected to frequency, correlation analysis, and multi-regression analysis using SPSSWIN(Statistical Package for the Social Science). The results are as follows. 1. In spring mowing height, 3.5-4 mm appeared the highest frequency(44.4%) and 4-4.5mm mowing height appeared the high frequency(41.0%). In summer mowing height, 4.5-5mm appeared the highest frequency(51.3%). In fall mowing height, 4-4.5mm appeared the highest frequency(41.0%). 2. In N-fertilizing amount of February and November, 0(zero) g/$m^2$ appeared the highest frequency. In N-fertilizing amount, of June and July 0-2 g/$m^2$ appeared the highest frequency. In N-fertilizing amount, of March, May, August, and September 2-4 g/$m^2$ appeared the highest frequency. In N-fertilizing amount, of October 2-4 or 6-8 g/$m^2$ appeared the highest frequency. 3. In spring topdressing times, 3-6 times appeared the highest frequency(52.6%). In spring topdressing amount, more than 2mm appeared the highest frequency(35.9%). In summer topdressing tierms, 0-3times appeared the highest frequency(71.8%). In summer topdressing amount, 0.5-1mm appeared the highest frequency(46.2%). In fall topdressing times, 0-3times appeared the highest frequency(47.4%). In fall topdressing amount, more than 2mm appeared the highest frequency(35.9%). 4. In spring irrigation tiems, 3-4times/a week appeared the highest frequency (30.6%). In spring irrigation amount, the irrigation below 5mm/day under appeared the highest frequency(38.7%). In summer irrigation times, 4-7times/ a week appeared the highest frequency(38.9%). In summer irrigation amount, 5-10mm/a day appeared the highest frequency(45.2%). In fall irrigation times, 2-3times/a week appeared the highest frequency(36.1%). In fall irrigation amount, the irrigation below 5mm/a day under appeared the highest frequency(45.2%). 5. In spring aeration times, 2 times appeared the highest frequency(55.2%). In spring aeration depth, 5-10mm appeared the highest frequency(81.6%). In fall aeration times, 1 time appeared the highest frequency(82.5%). In fall aeration depth, 5-10mm appeared the highest frequency(86.8%). 6. In spring green speed, 1.98-2.28 or 2.59-2.89mm appeared the highest frequency(32.7%). In summer green speed, 1.98-2.28mm appeared the highest frequency (46.9%). In fall green speed, 1.98-2.28mm appeared the highest frequency(38.8%). 7. The factors which affect green speed were mowing height, N-fertilizing, season, topdressing, irrigation, and aeration. Vertical mowing did not affect green speed. The order of the relevant important factors was mowing height >: N-fertilizing > season > topdressing > irrigation > aeration. Mowing height and N-fertilizing were the most important factors in green speed. As mowing height decreased, green sped always increased. As total N-fertilizing amount decreased, green speed increased. In summer, green sped decreased remarkably. As topdressing times increased and the topdressing amount decreased, green sped increased. As irrigation times increased and irrigation amount decreased, green speed increased.

Comparative Study on the Green Speed by different the Types of Putting green Maintenance Equipment (퍼팅그린 관리 장비 유형에 따른 그린스피드 차이 비교)

  • The purpose of this study is to find the suitable putting green maintenance equipment for improving the quality of putting green. This study was carried out to investigate the green speed(i.e., ball roll distance) by different the types of greens mower(between work-behind greens mower and riding greens mower), the types of reel blades(between 9 blades and II blades) and the types of roller(between riding soil sprayer and lightweight roller). Green speed of golf course putting greens is assessed by use of Stimpmeter. The west course of Lakeside Country Club was selected for the case study. The results are summarized as follows; 1. The green speed was faster in 3.8 mm moving height treatment plot using work-behind greens mower than in 3.8 mm moving height treatment plot using riding greens mower right after the moving and even after eight hours had passed, and it was statistically significant at a 95% confidence level. Therefore, work-behind greens mower was judged to be a proper equipment type for the fast green management practice than riding greens mower. 2. The green speed was faster in 3.2 mm mowing height treatment plot using work-behind greens mower equipped with II blades than in 3.2 mm mowing height treatment plot using work-behind greens mower equipped with 9 blades, and this result was statistically significant at a 95% confidence level. Therefore, II-blade was judged to be a proper blade type for the fast green management practice than 9-blade. 3. The difference in green speed (green speed increased after rolling) between a treatment plot mowed at 3.0 mm mowing height with the work-behind greens mower and then rolled a single time with riding soil sprayer and a treatment plot mowed at 3.0 mm mowing height with the work-behind greens mower and then rolled once with lightweight roller was not statistically significant at a 95% confidence level. However, the difference in green speed (green speed decreased after rolling) between two treatment plots measured after eight hours had passed was statistically significant at a 95% confidence level. Therefore, the lightweight roller was judged to be a proper roller type for the fast green management practice than the riding soil sprayer.
