Synthesis of Enzyme-Containing PEG Hydrogel Nanospheres for Optical Biosensors (광바이오센서용 효소를 함유한 PEG 수화젤 나노입자의 합성)

  • In this word as the first step to develop optical biosensors for a single cell level analysis, the preparation method of nano-scale polymer hydrogel spheres containing an enzyme was set up and the feasibility of the spheres as optical biosensors was investigated. The horseradish peroxidase (HRP) was encapsulated in the PEG hydrogel spheres by suspension photopolymerization, yielding spheres of the average size of 305 nm. After the polymerization, the incorporation and activity of HRP within the spheres were determined by the production of fluorescence resulted from the enzymatic reaction between HRP and $\H_{2}O_{2}$. The fluorescence emission response of the HRP-loaded PEG hydrogel spheres increased by nearly 300$\%$ as hydrogen peroxide concentration was changed from 0 to 11 nM in the presence of Amplex Red. The results suggest that the method to prepare the PEG hydrogel nanospheres containing an enzyme could be used for developing optical biosensors to measure various analytes in the very small samples like a single cell.

  • PHEAs [${\alpha}$,${\beta}$-poly(2-hydroxyethyl-DL-aspartamides)], a class of poly(amino acid), have been widely studied as biodegradable and biocompatible polymers for potential biomedical and pharmaceutical applications. In this study, we investigated a homogeneous blend of PHEA with poly(N-vinyl pyrrolidone) (PNVP) and its semi-IPN (semi-interpenetrating polymer network) gels. Blend films were prepared by a solution casting method. The resulting blends were totally transparent over the whole composition ranges and the single $T_g$, changing monotonously with composition, was observed by DSC to confirm the miscibility between these two polymers. FTIR was used to discuss the possible hydrogen-bonding interaction between polymers. In addition, semi-IPN type gels were prepared by chemical crosslinking of PHEA/PNVP blend solution using hexamethylene diisocyanate (HMDI) as a crosslinking reagent. The prepared gel was characterized by their swelling property and morphology.

Properties of Poly(acrylic acid) Hydrogel by the Surface Charge of Magnetite Nanoparticles (나노 자철광의 표면전하에 따른 Poly(acrylic acid) 수화젤의 물성)

  • The superparamagnetic nanoparticles were prepared by coprecipitation of $FeCl_3$ and $Na_2SO_3$ with $NH_4OH$ and the surface charge on hydroxyl group by chemisorption was changed depending on pH. We studied correlation between surface charge of magnetite and pH. Using this correlation the properties of poly (acrylic acid) (PAAc) hydrogel embedded with magnetite was studied. The magnetite was characterized by XRD, AFM. and FTIR. The zeta-potential of magnetite was influenced by pH: great positive charge was shown high under the pH 4 and isoelectric point was found at pH 7. The hydrogen bond formed by combining oi PAAc hydrogel and magnetic colloid under pH 4 caused tensile strength to increase, while swelling and elongation at break to decrease. The result confirmed that the magnetic moment was increased proportionally to the content of magnetite.

  • The solid-state electrolyte based on polymer is applicable to various electrochemical devices including supercapacitor, battery, sensor, actuator and has great attention to develop its ionic conductivity from conventional polymer electrolyte by uisng wide range of ionic liquids. The research about ion gel as a solid state electrolyte with the ionic liquid has focused on the wearable and flexible electronic device to use as the high electrical and electrochemical performances, mechanical strength of polymer. In this work, we have investigated and developed solid-state electrolyte based on the ionic liquid and polymer with enhanced ionic conductivity and stability.

  • Poly(N-isopropylacrylamide-co-acrylonitrile) [P(NIPAAm-co-AN)] copolymers with AN content of up to 10 mol% and their hydrogels were synthesized using water as a reaction medium, and the effects of AN unit incorporation on the critical gel transition temperature(CGTT) and swelling behaviors of the hydrogels were investigated. The CGTT of the copolymer hydrogel was $30{\sim}32\;^{\circ}C$, decreasing with increasing AN content. Below CGTT, swelling rate and equilibrium swelling ratio of the copolymer hydrogel decreased with increasing AN content. On the other hand, it exhibited faster deswelling and lower equilibrium deswelling ratio with increasing AN content above CGTT.

Preparation and Characterization of Biodegradable Superporous Hydrogels (생분해성을 갖는 초다공성 수화젤의 제조 및 특성분석)

  • In this study, biodegradable superporous hydrogels(SPHs) with fast swelling and superabsorbent properties were prepared using biodegradable crosslinkers and their physicochemical properties were characterized. A biodegradable crosslinker (PLA-PEG-PLA DA) was synthesized by a ring opening polymerization of D,L-lactide (LA) using hydrophilic poly(ethylene glycol) as a macroinitiator, followed by diacrylation of the end groups for the introduction of polymerizable vinyl groups. Various kinds of hydrogels with different chemical compositions were prepared and characterized in terms of swelling ratio, swelling kinetics, and biodegradation properties. The synthetic results were confirmed by $^1H$-NMR, FT-IR and GPC measurements, and the porous structures of the prepared SPHs and their porosities were identified by a scanning electron microscope and mercury porosimetry, respectively. The physicochemical properties of SPHs could be controlled by varying their chemical compositions and their cytotoxicity were found to be very low by MTT assay.

Properties of Polymer Electrolytes based on PEO-LiClO$_4$ Matrix Fabricated by Sol-Gel Process (솔-젤 법으로 만든 PEO-LiClO$_4$에 기초한 고분자 전해질의 물성)

  • In spite of high ionic conductivity, the polymer gels have poor mechanical properties and high reactivity with lithium metal anode. To solve these problems, the dry solid systems and polymer composites have been intensively studied, due to their good mechanical, thermal, chemical, and electrochemical stability. The objectives of this experiment were to improve ionic conductivity and mechanical properties of the solid polymer electrolytes based on PEO-LiClO$_4$. To obtain higher ionic conductivity and better mechanical properties, ceramic or rubber phase was added in the PEO-LiClO$_4$(8:1) matrix. The results showed that ionic conductivity and mechanical properties were improved. The ionic conductivity of the samples was as high as 10$\^$-5/ S cm$\^$-1/. This value is similar to the best ionic conductivity ever reported in the solid drying system. To obtain better results, we used PEO with various molecular weights (600∼8000) and changed the salt contents. By using DSC, we found that the addition of salt reduced the crystallinity of PEO. The mobility of polymer dependence on salt contents was examined by FT-IR.
