Study on Adsorption Equilibrium, Kinetic and Thermodynamic Parameters of Murexide by Activated Carbon (입상 활성탄에 의한 Murexide의 흡착 평형, 동력학 및 열역학 파라미터에 관한 연구)

  • The equilibrium, kinetic and thermodynamic parameters of adsorption of murexide by granular activated carbon were investigated. The experiment was carried out by batch experiment with the variables of the amount of the adsorbent, the initial concentration of the dye, the contact time and the temperature. The isothermal adsorption equilibrium was best applied to the Freundlich equation in the range of 293 ~ 313 K. From the separation factor (${\beta}$) of Freundlich equation, it was found that adsorption of murexide by granular activated carbon could be the appropriate treatment method. The adsorption energy (E) obtained from the Dubinin- Radushkevich equation shows that the adsorption process is a physical adsorption process. From the kinetic analysis of the adsorption process, pseudo second order model is more consistent than pseudo first order model. It was found that the adsorption process proceeded to a spontaneous process and an endothermic process through Gibbs free energy change ($-0.1096{\sim}-10.5348kJ\;mol^{-1}$) and enthalpy change ($+151.29kJ\;mol^{-1}$). In addition, since the Gibbs free energy change decreased with increasing temperature, adsorption reaction of murexide by granular activated carbon increased spontaneously with increasing temperature. The entropy change ($147.62J\;mol^{-1}\;K^{-1}$) represented the increasing of randomness at the solid-solution interface during the adsorption reaction of murexide by activated carbon.

Mechanical Properties Evaluation of 3D Printing Recycled Concrete utilizing Wasted Shell Aggregate (패각 잔골재를 활용한 3D 프린팅 자원순환 콘크리트의 역학적 성능 평가)

  • The volume of shells, a prominent form of marine waste, is steadily increasing each year. However, a significant portion of these shells is either discarded or left near coastlines, posing environmental and social concerns. Utilizing shells as a substitute for traditional aggregates presents a potential solution, especially considering the diminishing availability of natural aggregates. This approach could effectively reduce transportation logistics costs, thereby promoting resource recycling. In this study, we explore the feasibility of employing wasted shell aggregates in 3D concrete printing technology for marine structures. Despite the advantages, it is observed that 3D printing concrete with wasted shells as aggregates results in lower strength compared to ordinary concrete, attributed to pores at the interface of shells and cement paste. Microstructure characterization becomes essential for evaluating mechanical properties. We conduct an analysis of the mechanical properties and microstructure of 3D printing concrete specimens incorporating wasted shells. Additionally, a mix design is proposed, taking into account flowability, extrudability, and buildability. To assess mechanical properties, compression and bonding strength specimens are fabricated using a 3D printer, and subsequent strength tests are conducted. Microstructure characteristics are analyzed through scanning electron microscope tests, providing high-resolution images. A histogram-based segmentation method is applied to segment pores, and porosity is compared based on the type of wasted shell. Pore characteristics are quantified using a probability function, establishing a correlation between the mechanical properties and microstructure characteristics of the specimens according to the type of wasted shell.

  • The cytochrome P450s (P450s) metabolizing natural products are among the most versatile biological catalysts known in plants, but knowledge of the structural basis for their broad substrate specificity has been limited. The activity of p-coumarate 3-hydroxylase (C3H) is thought to be essential for the biosynthesis of lignin and many other phenylpropanoid pathway products in plants however, all attempts to express and purify the protein corresponding C3H gene have failed. As a result, no conditions suitable for the unambiguous assay of the enzyme are known. The detailed understanding of the mechanism and substrate-specificity of C3Hdemands a method for the production of active protein on the milligram scale. We have developed a bacterial expression and purification system for the plant C3H, which allows for the quick expression and purification of active wild-type C3H via introduction of combinational mutagenesis. The modified cytochrome P450 C3H ($C3H_{mod}$) could be purified in the absence of detergent using immobilized metal affinity chromatography and size exclusion chromatography following extraction from isolated membranes in a high salt buffer and catalytically activated. This method makes the use of isotopic labeling of C3H for NMRstudies and X-ray crystallography practical, and is also applicable to other plant cytochrome P450 proteins.

Stabilization and Release Behavior of W1/O/W2-Type Multiple Emulsions Using Various Block Copolymer Emulsifier and Stabilizer (다양한 Block Copolymer를 유화제 및 안정화제로 사용한 W1/O/W2-Type 다중에멀젼의 방출거동 및 안정성)

  • A new approach to obtain stable $W_1/O/W_2$ multiple emulsions has been studied ; The basis of the interfacial interaction between a PCL-PEO-PCL triblock copolymer and a lipophilic emulsifier in the dispersed oil phase was examined. $W_1/O/W_2$ multiple emulsions were prepared by the two-step method. Arlacel P-l35 was used as a liphophilic emulsifier and Synperonic PE/F 127 as a hydrophilic one. Eutanol-G was used as an oil phase. NaCl was encapsulated within the multiple emulsion droplets as the internal marker and its release rate studies were carried out. The suability of the multiple emulsions have been assessed by measuring Separation Ratios(%) and microscopic observations. The release of NaCl was significantly reduced in $W_1/O/W_2$ multiple emulsions containing PCL-PEO-PCL triblock copolymer(2k-4k-2k or 6k-4k-6k) in the oil phase. It may be concluded that the copolymer and the emulsifier form effective interfacial complex to enhance stability and to control the release rate. The effective diffusion coefficients of the NaCl were estimated as $2.64{\times}10^{-15}s$and $3.23{\times}10^{-16}gcm^2/s$ for the $W_1/O/W_2$ multiple emulsion containing 1.2 wt % of PCL-PEO-PCL triblock copolymers with compositions of 2k-4k-2k and 6k-4k-2k, respectively. The rate of release decreased with the increase of the initial concentration of NaCl. The results were examined in view of Higuchi mechanism. A kinetic model which is similar to the model for release of dispersed drugs from a polymeric matrix was found to be suitable for the release of NaCl from $W_1/O/W_2$ multiple emulsions.

Preparation of 40 wt.% Ag-coated Cu Particles with Thick Ag Shells and Suppression of Defects in the Particles (두꺼운 Ag shell이 형성되는 40 wt.% Ag 코팅 Cu 입자의 제조 및 입자 내 결함 억제)

  • To prepare the Cu-based filler material indicating enhanced oxidation resistance property and Ag content, Ag-coated Cu particles was fabricated by Ag plating of 40 wt % on the spherical Cu particles with an average size of $2{\mu}m$ and their oxidation behavior was also evaluated. In the case that ethylenediaminetetraacetic acid was used alone, the fabricated particles frequently showed broken structures such as delamination at Ag shell/core Cu interface and hollow structure that are induced by excessive galvanic displacement reaction. As a result, fraction of defect particles increased up to 19.88% after the Ag plating of 40 wt.%. However, the fraction in the 40 wt.% Ag-coated Cu particles decreased to 9.01% and relatively smooth surface and dense microstructure in the Ag shell were also observed with additional usage of hydroquinone as a complexing agent. Ag-coated Cu particles having the enhanced microstructure did not show any weight increase by oxidation for exposure to air at $160^{\circ}C$ for 2 h, indicating increased oxidation resistance property.

Stripping of Asphalt Pavements and Antistripping Addities (도로포장 구조물에서의 스트리핑 현상과 스트리핑 방지제의 이용방안)

  • Physico-chemical properties of asphalt, aggregate, and asphalt-aggregate mixture that might influence stripping were summarized in Table 1, based on the fundamental theories concerning stripping. It was found that although physical properties of aggregate affected stripping, there was no strong correlation between the physical properties of aggregate, such as pore volume and surface area, and the stripping propensity of the aggregate. Chemical and electrochemical properties of aggregate surface in the presence of water were most important factors for stripping. All mineral aggregates tested in this study imparted distinctive pH values to the contacting water and possessed distinctive electrochemical properties as measured by zeta potential. It was found that aggregates which had relatively higher surface potential in water and/or which imparted relatively higher pH to the contacting water were more susceptible to stripping. The functionalities contained in antistripping additives tested were primary and secondary amines and those of organic nitrogen compounds. The functionalities were determined by examining their infrared spectra. Based on the interfacial energy concept, the contact angle of an asphalt drop on an aggregate surface immersed in water related to the stripping propensity. The contact angle and stripping propensity were markedly reduced by the presence of an antistripping additive. In general, all the additives tested improved stripping resistance to some extent, depending on their concentration in the asphalts. The optimum dosage of an additive varied with different asphalts, as well as different aggregates. All antistripping additives tested in this study lost their effectiveness and failed to function to some extent when maintained for hours in a hot asphalt.

RGP lens 다목적 용액의 습윤성에 관한 비교 연구

  • We measured the effect of wettability of six MPSs for RGP(rigid gas permeable) lens. The used MPSs(multipurpose solutions) were OPTI-SOAK(ALCON), SOLO care hard(CIBA Vision), Total care(ALLERGAN), Simplicity(BOSTON), Wetting and Soaking Sol.(Bausch & Lomb) and Aquas-multi(Saehan). These MPSs keeps hydrophilic property of lens surface and increase the effect of cleaning or increase the effect of preservative effect. To compare with the effect of wettability we followed the way of contact angle measurement which was general way to measure wettability and compared lens which was conducted by each MPS made by different companies. As a control, 0.9% NaCl solution and artificial tears were used. The degree of the effect of wettability was decided by contact angle. It is hydrophilic property nearby $0^{\circ}$ of contact angle and it is closed by hydrophobic property as it increases. The results showed that every lens was nearby hydrophilic property within $25\;^{\circ}-36\;^{\circ}$. Also, it was differed by various factors. The surface tension showed various differences between 19.8 and 31.3 mN/m. In the viscosity, MPSs represented the highest viscosity between $4^{\circ}C$ and $20^{\circ}C$. It was much higher than compared with the viscosity of soft lens MPS. This experiment could be used to grasping the interaction between solutions used to MPS and the natural endowments of lens and to considering the relations of different factors effecting the wettability.

Hydrolysis of Lactose in Milk by Microencapsulated ${\beta}-Galactosidase$ (Microencapsulated ${\beta}-Galactosidase$에 의한 우유 유당의 가수 분해)

  • The nylon (poly 6, 10) microcapsules containing ${\beta}-galactosidase$ were obtained by the interfacial polymerization of 1, 6-diaminohexane and sebacoyl chloride with ${\beta}-galactosidase$ from Escherichia coli. They were generally spherical and had a mean diameter of $80{\mu}$ with 45 % of the activity recovery. In particular, there was no transport hamper of lactose through the membrane of microcapsules. The characteristics of the microencapsulated enzyme were similar to those of soluble enzyme optimal pHs, $7.0{\sim}7.2$ for the soluble and $7.3{\sim}7.5$ for the microencapsulated ; optimal temperatures, $50^{\circ}C$ for both ; apparent $K_m,\;3.33{\times}10^{-4}(on ONPG),$ $2.86{\times}10^{-3}$ M(on lactose) for the soluble and $5.28{\times}10^{-4}$ (on ONPG), $4.25{\times}10^{-3}$ M (on lactose) for the microencapsulated ; activation energies, 8.94 for the soluble and 9.78 Kcal/mole for the microencapsulated enzyme. Using this microencapsulated ${\beta}-galactosidase$, hydrolyses of lactose and milk lactose were carried out and 80 % of 5 % lactose solution and 70 % of lactose in skim milk were hydrolyzed in 40 hr at $27^{\circ}C$. The reusability and operational stability showed that the remaining activity was 50 % of the original activity after 5 runs and 120 hr of total operating time at $27^{\circ}C$.

Characterization of Phenanthrene Degradation by Sphingomonas sp. HS362 (Sphingomonas sp. HS362에 의한 Phenanthrene 분해특성)

  • A phenanthrene-degrading bacterium HS362, which is capable of using phenanthrene as a sole carbon and energy source, was isolated from oil contaminated soil. This strain is a gram negative, rod shaped organism that is most closely related to Sphingomonas paucimobilis based on biochemical tests, and belongs to the genus Sphingomonas based on fatty acids analysis. It exhibited more than $99.2{\%}$ nucleotide sequence similarity of 16S rDNA to that of Sphingomonas CF06. Thus, we named this strain as Sphingomonas sp. HS362. It degraded $98{\%}$ of phenanthrene after 10 days of incubation when phenanthrene was added at 500 ppm and $30{\%}$ even when phenanthrene was added at 3000 ppm. Sphingomonas sp. HS362 could also degrade low molecular weight PAHs(Polycyclic aromatic hydrocarbons) such as indole and naphthalene, but was unable to degrade high molecular weight PAHs such as pyrene and fluoranthene. The optimum temperature and pH for phenanthrene degradation were $30^{\circ}C$ and $4{\~}8$, respectively. Sphingomonas sp. HS362 could degrade phenanthrene effectively in the concentration range of NaCl of up to $1{\%}$. Its phenanhrene degrading ability was enhanced by preculture, suggesting the possibility of induction of phenanthrene degrading enzymes. Starch and surfactants such as SDS, Tween 85, and Triton X-100 were also able to enhance phenanthrene degradation by Sphingomonas sp. HS362. It carries five plasmids and one of them, plasmid p4, is considered to be involved in the degradation of phenanthrene according to the plasmid curing experiment by growing at $42^{\circ}C$.

Partial Purification and Some Properties of Carboxymethyl Cellulases from Alkalophilic Cephalosporium sp. RYM-202 (호알칼리성 Cephalosporium sp. RYM-202가 생산하는 carboxymethyl cellulase의 부분정제 및 특성)

  • An alkalophilic Cephalosporium sp. RYM-202 capable of producing cellulase components was isolated from soil. This organism grew best at an initial pH 9.0 and produced cellulase maximal at an initial pH 9.5-10.0. Three carboxymethyl cellulases(CMCases), P-I-I, P-I-II and P-II-I, were partially purified by DEAE-Sephadex A-50 ion exchange column followed by Sephadex G-150 gel filtration. The optimum pH values for activity were 7.5 for P-I-I, 8.0-9.5 for P-I-II and 7.5-10.0 for P-II-I. All CMCases were stable between pH 4.5 and 12.0. Temperature optima for activity ranged between 40 and $60^{\circ}C$ and more than 50% of the maximum activity was observed at $20^{\circ}C$ for both of P-I-I and P-II-I. The activity of CMCases was significantly stable in the presence of various laundry components, such as, surfactants, chelating agents and alkaline proteinases.
