Antifreeze Activity of Dimerized Type I Antifreeze Protein Fragments (Type I 결빙방지 단백질 조각 이량체의 결빙방지 활성)

  • Antifreeze proteins (AFPs) bind to ice crystals and inhibit their growth. AFPs are essential for the survival of organisms living in subzero environments. Type I AFP (AFP37) isolated from winter flounder is an ${\alpha}$-helical peptide of 37 residues long. In this study, we attempted to develop short AFP fragments with higher activity and solubility. We designed and synthesized N-terminal 15 and 21 residue-long AFPs, designated AFP15 and 21. Also dimerized AFP15 and 21, designated dAFP15N and dAFP21N, respectively, were generated through disulfide bonds between peptides containing CGG residues added to the N-terminus of AFP15 and AFP21 (designated AFP15N and 21N). Their helical contents and antifreeze activities were assessed using circular dichroism (CD) spectroscopy and a nanoliter osmometer, respectively. The helical content of AFP15 AFP21, AFP15N, AFP21N, dAFP15N and dAFP21N was 47, 48, 23.8, 28, 49.1, and 52%, respectively compared to that of wild type AFP37; the antifreeze activity was 8.4, 9.3, 0.05, 5.6, 12.1, 11.2% respectively, compared to that of wild type AFP37. Contrary to our anticipation, the dimerized peptides showed almost the same antifreeze activity as their monomeric counterparts. These results indicate that the dimerized peptides behave as monomeric peptides due to the high rotational freedom of disulfide bonds connecting two monomeric peptides. The star-shaped ice crystals generated by the peptides also demonstrated weak interaction between ice and peptides.

A Study on Stroke Extraction for Handwritten Korean Character Recognition (필기체 한글 문자 인식을 위한 획 추출에 관한 연구)

  • Handwritten character recognition is classified into on-line handwritten character recognition and off-line handwritten character recognition. On-line handwritten character recognition has made a remarkable outcome compared to off-line hacdwritten character recognition. This method can acquire the dynamic written information such as the writing order and the position of a stroke by means of pen-based electronic input device such as a tablet board. On the contrary, Any dynamic information can not be acquired in off-line handwritten character recognition since there are extreme overlapping between consonants and vowels, and heavily noisy images between strokes, which change the recognition performance with the result of the preprocessing. This paper proposes a method that effectively extracts the stroke including dynamic information of characters for off-line Korean handwritten character recognition. First of all, this method makes improvement and binarization of input handwritten character image as preprocessing procedure using watershed algorithm. The next procedure is extraction of skeleton by using the transformed Lu and Wang's thinning: algorithm, and segment pixel array is extracted by abstracting the feature point of the characters. Then, the vectorization is executed with a maximum permission error method. In the case that a few strokes are bound in a segment, a segment pixel array is divided with two or more segment vectors. In order to reconstruct the extracted segment vector with a complete stroke, the directional component of the vector is mortified by using right-hand writing coordinate system. With combination of segment vectors which are adjacent and can be combined, the reconstruction of complete stroke is made out which is suitable for character recognition. As experimentation, it is verified that the proposed method is suitable for handwritten Korean character recognition.

  • Various crosslinked poly(4-vinylpyridines) having different degrees of crosslinking were prepared by radical copolymerizations of 4-vinylpyridine with N, N'-2, 6-pyridinebisacrylamide as a crosslinker. The abilities of these crosslinked polymers to bind methyl orange and butyl orange were investigated at various temperatures in a buffer solution of pH 7. The first binding constants were evaluated from the equilibrium binding amounts. The first binding constants against the temperatures showed bell-shaped curves. Also, the first binding constants against the degree of crosslinking showed bell-shaped curves. When the temperature and the degree of crosslinking of maximum binding in the curves of these binding systems were compared with those of previous systems containing crosslinked poly(4-vinylpyridines) prepared by using N, N'-methylenebisacrylamide, N, N'-tetramethy-lenebisacryamide and divinylbenzene as crosslinkers, respectively, they were varied with the crosslinked poly(4-vinyl pyridines) containing different crosslinkers. These results were discussed in terms of the properties of the crosslinkers.

An Assessment on the Preliminary Coupled Load Analysis Results for Advanced Low Earth Orbit Earth Observation Satellite (고성능 저궤도 지구관측위성의 예비연성하중 해석결과에 대한 평가)

  • In this paper, an assessment on the preliminary coupled load analysis results for advanced Low Earth Orbit Earth Observation satellite was performed. Spacecraft FE-model was converted into Craig-Bampton model consisting of mass matrix, stiffness matrix, acceleration transformation matrix, displacement transformation matrix, and it was delivered to the launch vehicle developer. Launch vehicle developer performed a coupled load analysis with launch vehicle model and spacecraft Craig-Bampton model, and the coupled load analysis results were provided to us. From the assessment on the analysis results, it was verified that spacecraft is safe under launch environment.

  • 쌀 소비량은 생활 수준 향상과 외식 및 식품 산업의 발달로 감소하고, 고품질 쌀을 요구하는 성향이 높아지고 있다. 현재, 고품질 쌀을 생산할 수 있는 기술과 제반 요건은 구비되어 있으나, 수확 후 건조·저장 중에 쌀의 품질이 저하된다. 특히 건조 중 품질 저하는 고온건조에 의한 동할 발생 및 과건조가 주요 원인이다. 이를 해결하기 위해 원적외선 건조 시스템에 관한 연구가 진행 중이다. 본 연구에서는 방사율이 높고, 접착성 및 내열성 등이 강한 곡물 건조기용 방사체 코팅원료를 개발하고, 이 원료를 원적외선 방사체에 코팅한 후 방사체의 형태와 크기에 따라 표면 온도 분포 및 연료 소비량 등을 측정 분석하여 방사체의 특성과 적정 조건을 규명하고자 하였다. 연구 결과를 요약하면 다음과 같다. (1) 세라믹 코팅 원료의 배합비는 세라믹분말 40%와 결합재 60%가 접착력과 가열경화 후 표면이 양호한 것으로 나타났다. (2) 노즐의 유량 및 분무각에 따른 버너의 화염길이 및 폭은 노즐유량이 증가함에 따라 증가하는 경향을 나타내었다. (3) 방사체 위치별 표면온도편차는 방사체 길이가 1350mm인 것이 작은 것으로 나타났고, 열풍유동관 직경이 Rounding type의 경우 76.3mm, Right angle type의 경우 89.1mm일 때 표면 온도편차가 적었다. (4) 연료소비량은 열 풍유동관 직경이 클수록, 방사체 길이가 길수록 증가하는 경향을 나타내었고, Right angle type이 Rounding type에 비하여 연료소비량이 약간 높은 것으로 나타났다.

  • 뛰어난 혈당강하작용을 가지고 있는 oxirane carboxilic acid의 analogue 합성과 관련하여 oxirane carboxilic acid의 기본골격을 용이하게 형성할 수 있고 동시에 다앙한 관능기를 갖는 side chain을 도입할 수 있는 방법을 개발함으로써 다양한 유도체들을 합성하는데 이용하고자 함. 방법 및 결과 $\alpha$,$\beta$-unsaturated ester로부터 dioxirane을 이용한 직접적인 epoxidation을 통해서 보통의 방법으로는 얻기 어려운 oxirane carboxilic acid의 ester를 높은 수율로 합성할 수 있는 방법이 개발되었으며 특히 분자내에 cpoxide 존재하에서도 Mitsnobu 방법을 이용한 O-alkylation에 의해 aryl ether 결합을 형성할 수 있는 방법이 개발되었으며 이들 방법을 이용하여 다양한 유도체들을 합성하였다.

  • 화학 반응은 원자나 분자가 서로 작용하여 결합이 분해되거나 생성되는 동적인 과정이다. 일반적으로 이러한 동적인 과정은 매우 빠른 시간 내에 일어나기 때문에 (보통수 펨토초에서 수 피코초 내에 일어난다) 화학 반응 도중에 나타나는 중간체(transient structure)의 구조 변화나 분자의 움직임을 실시간으로 잡기 위해서는 매우 높은 시간 분해능이 필요하다. 이러한 중간체의 구조 동역학(structural dynamics)은 그 대상이 되는 분자의 화학적/생물적 기능에 매우 중요한 정보를 제공하기 때문에 실시간 분자 동역학 연구는 복잡한 화학적/생물적 시스템의 메커니즘 규명 연구에 필수적이라고 할 수 있다.   • 본 논문에서 제안하는 분절된 화소들의 특징추출 방법은 이진화 영상에서 수직/수평 화소들의 분절점을 탐색하여 추출하는 특징 탐색기이다. 숫자의 구조적인 면을 고려하여 사소한 부분들도 명확한 특징으로 탐지하여 추출하였고, 이러한 방법은 일반적으로 사용하여지는 특징추출방법 몇가지를 선택하여 이용하였고, 제안하는 방법과 결합하여 필기체 숫자를 인식하였다. 인식기를 구현하기 위하여 3개층 구조를 갖는 클러스터 MLP 신경망을 사용하였다 실험 결과 단순히 일반적인 특징만을 활용하여 얻는 인식률 보다 훨씬 향상됨을 보여주었다.

  • To search of a new porcine pheromonal odorants for biostimulation control system technologies to offer a potentially useful and practical way to improve reproductive efficiency in livestock species, the holographic quantitative structure activity relationship (HQSAR) model between odorants, 2-phenoxytetrahydrofurane (A), 2-cyclohexyl-oxytetrahydrofurane (B), derivatives and binding affinity constants (p[Od.]/sub 50/) for porcine odorant-binding protein (pOBP) as receptor of pig pheromones were derivated and disscused. The binding affinity constants of cyclohexyl substituents (A) for pOBP were higher (A>B) than that of phenyl substituents (B). It was revealed that the optimum HQSAR model XI using PLS analyses had a fragment length (5∼8) with chirality at 5 components and hologram length 97 bin, which had a cross-validated q²(predictablities) of 0.916, and a conventional correlation coefficient r² (fitness) of 0.988, respectively. From the atomic contribution, the C3 and C5 atom in 2-oxyfuryl group contributed to binding affinity constants, whereas the central carbon atom in tert-butyl group on the cyclohexyl ring and the C4 atom of furyl group parts showed no contribution.
