  • It is well known that mesenchymal stem cell(MSCs) can be differentiated into fibroblasts, chondrocytes, and osteoblasts and that they develop into fibrous tissue, cartilage, or bone, as a result of mechanical stimulation. In this study, we developed a bioreactor system, which is composed of a reactor vessel that provides the required cell culture environment, an environment controlling chamber to control the media, a gas mixer, and a reactor motion control subsystem to apply mechanical stimuli to the cells. For the MSC culture, We used a poly-urethane (PU) scaffold, with a collagen coating to ensure improved cohesion ratio. Then, we transferred the cultivated MSCs in the PU scaffold, cultured the cells in the bioreactor system, and confirmed the proliferation, differentiation, and ossification processes, resulting from mechanical stimuli.

  • Purpose: The aim of this study was to study the effect of hydroxyapatite (HA) coating crystallinity on the proliferation and differentiation of human osteosarcoma cells. Materials and methods: Surface roughness of the titanium disks increased by anodizing treatment and then HA was coated using ion beam-assisted deposition (IBAD). HA coating was crystallized by heat-treated at different temperature ($100^{\circ}C$, $300^{\circ}C$, $500^{\circ}C$, $800^{\circ}C$). According to the temperature, disks were divided into four groups (HA100, HA300, HA500, HA800). With the temperature, crystallinity of the HA coating was different. Anodized disks were used as control group. The physical properties of the disk surface were evaluated by surface roughness tests, XRD tests and SEM. The effect of the crystallinity of HA coating on HOS cells was studied in proliferation and differentiation. HOS cells were cultured on the disks and evaluated after 1, 3, 5, and 7 days. Growth and differentiation kinetics were subsequently investigated by evaluating cell proliferation and alkaline phosphatase activity. Results: Regardless of the heat-treated temperature, there is no difference on the surface roughness. Crystallinity of the HA was appeared in the groups of HA500, HA800. HOS cells proliferation, ALP activity were higher in HA500 and HA800 group than HA100 and HA300. Conclusion: Within the results of this limited study, heat treatment at $500^{\circ}C$ of HA coating produced by IBAD has shown greater effect on proliferation and differentiation of HOS cells. It is considered that further in vivo study will be necessary.

  • Effects of the water extract from Kalopanax pictus NAKAI on insulin-like action and glucose uptake in 3T3-L1 cells were investigated. The bark of K. pictus NAKAI was treated with hot water and the extract was freeze-dried. Total extract of K. pictus NAKAI was fractionated into 6 fractions with increasing gradients from 0 to 100% MeOH on Amberlite XDA-4. Treatment of 3T3-L1 fibroblasts with 1 ${\mu}g/ml$ and 10 ${\mu}g/ml$ of K. pictus NAKAI total extracts significantly increased the differentiation of the cells. When co-treated with inducers such a dexamethasone, 1-methyl-3-isobutylxanthine and insulin, the differentiation was increased at 1 ${\mu}g/ml$ of total extract, but not at 10 ${\mu}g/ml$. In 3T3-L1 adipocytes, glucose uptake was increased by 3.3 times with addition of 0.3 and 3 ${\mu}g/ml$ of Fr. 1 (0-10% MeOH) and Fr. 3 (30% MeOH) at 3 ng/ml insulin. In conclusion, K. pictus NAKAI contains such compounds that play a role of insulin-like action and insulin sensitizer.

  • In vitro culture of Toxoplasma gondii in HL-60 cells and cell-mediates immunity against Toxoplasma in dimethylsulfoxide(DMSO) -induced HL-60 cells, i.e., differentiation into granulocytes, were pursued. HL-60 calls were treated with various concentrations of DMSO, and 1.3%(v/v) for 3 day incubation was chosen as the optimal condition icy differentiation into granulocytes. The degree of differentiation was assayed in physiological and functional aspects in addition to morphological point. When treated with 1.3% DMSO for 3 days, HL-60 cells did not synthesiar DNA materials beyond background level, and showed active chemotactic response to chemotactic peptide, formal-methionyl-leucyl-phenylalanine(FMLP). Morphologically promyelocytes of high nuclearlcytoplasmic(NIC) ratio changed to granulocytes of relatively low WJC ratio. The relationships between HL-60 cells or DMSO-induced HL-60 cells and Toxoplasma were examined after stain with Giemsa and Buorescent dye (acridine orange). HL-60 cells did not show any sign of torso- plasmacidal activity but showed intracellular proliferation of Texoplasma to form rosette for 72 hr co-culture. In contrast, OMSO-induced HL-60 cells phagocytosed Toxoplasma within 1 hr, and performed a process of intracellular digestion of Toxoplasma thereafter. With the above results, it is suggested that phagosome-Iysosome fusion is one of the critical events for the parasitism by Toxoplasma or for susceptibility of host cells. The in vitro culture system of this study has offered a defined condition to study the protozoan parasite-host cell interactions.

  • The Skaergaard intrusion is widely considered a type example of a strongly fractionated, layered intrusion that has undergone extensive in situ igneous differentiation. The Intrusion, therefore, should be a good locality for modeling trace element vriation in a closed system. Previous studios (Haskin and Haskin, 1968; Faster et al., 1974), however, have suggested thats the rare earth elements in whole rocks and mineeral separates from the Intrusion did not fellow the expected trend for closed system crystatllization. Trace element modeling using published distribution coefficients, modal abundances of the coexisting minerals, and the concentration of trace elements In whole rocks and mineral separates from the Skaergaard Intrusion, reveals that the rare earth elements were significantly Influenced by the crystallization of abundant apatite in the Layered Series suring the final stages of crystallization. The results of trace element modeling also suggcsts that apatite, which appears sporadically in the UBS, is not a primary liquidus phase in these samples as previously suggested (Naslund, 1984) but an interstitial phase that (lid not directly effect trace element abundances In the evolving magma As the Skaergaard magma coaled convection, or convected as small Isolated cells during the final stages of differentiation, an elebated $P_{H2O}$ Induced by accumulation of volatile elements near the roof of the magma chamber ingibited or delayed the precipitation of primary apatite in the UBS If the Skaergaard differentiation Is modeler assuming primary apatite crystallization In the upper par of the LS where abundant modal apatite is present, and only late stage crystallization of apatite In the UBS where apatite Is less abundant, rare earth elements abundances follow a closed system variation trend. These results rule but any differentiation model for the Skaergaard Intrusion that Includesvolumetrically significant injections or discharges of magma Into or out of the chamber during the final 20% of the crystallization history.

  • The lymphocyte component of the immune system is divided into B lymphocytes and T lymphocytes. B lymphocytes produce antibodies (humoral immunity) via maturation into plasma cells, and T lymphocytes kill other cells or organisms (cellular immunity). A traditional immunological paradigm is that B lymphocyte and T lymphocyte interactions are a one-way phenomenon, with T lymphocytes helping to induce the terminal differentiation of B lymphocytes into immunoglobulin class-switched plasma cells. A deficiency of T lymphocytes was reported to result in defective B lymphocyte function. However, evidence for a reciprocal interaction between B and T lymphocytes is emerging, with B lymphocytes influencing the differentiation and effector function of T lymphocytes. For example, B lymphocytes have been shown to induce direct tolerance of antigen-specific CD8+ T lymphocytes and induce T lymphocytes anergy via transforming growth factor-beta (TGF-β) production. The present study showed that LPS-stimulated B lymphocytes inhibited the differentiation of Th1 lymphocytes by inhibiting the production of interleukin-12 (IL-12) from dendritic cells. An interaction between the B lymphocytes and dendritic cells was not needed for this inhibition, and the B lymphocytes did not alter dendritic cell maturation. B lymphocyte-derived soluble factor (BDSF) suppressed the LPS-induced IL-12p35 transcription in the dendritic cells. Overall, these results point to a novel B lymphocyte- mediated immune suppressive mechanism. The findings cast doubt on the traditional paradigm of immunological interactions involving B lymphocyte and T lymphocyte interactions.

  • The Gadaeri granite near Ulsan mine is an oval-shape isolated granitic body, and is genetically related to the iron-tungsten mineralization. The Gadaeri granite exhibits calc-alkaline and I-type characteristics, and generally shows the micrographic texture which indicates the shallow depth of emplacement. Consideration of the stratigraphic thickness of Ulsan formation and minimum-melt compositions suggests that the bulk magma crystallized at pressure of 0.5~2.0 kbar under water saturated condition. The evolutionary trend observed in the studied rocks represents that feldspar fractional crystallization has been a major magmatic process at the Gadaeri granite pluton. Different chemical characteristics between the Gadaeri and the Masan-Kimhae granites cannot be explained by fractional crystallization or different degrees of partial melting, and it reflects that the magma source for Gadaeri granite was different from that of the Masan and Kimhae granites.

  • This study reports the results about the petrography and geochemical characteristics of 10 representative volacanic rocks. The Cretaceous volcanic rocks distributed in the vicinity of the Kageo island composed of andesitic rocks, dacitic welded tuff, and rhyolitic rocks in ascending order. Sedimentary rock is the basement in the study area covered with volcanic rocks. Andesitic rocks composed of pyroclastic volcanic breccia, lithic lapilli tuff and cryptocrystallin lava-flow. Most dacitic rocks are lapilli ash-flow welded tuff. Rhyolitic rocks consists of rhyolite tuff and rhyolite lava flow. Rhyolite tuff are lithic crystal ash-flow tuff and crystal vitric ash-flow tuff with somewhat accidental fragments of andesitic rocks, but dacitic rocks. The variation of major and trace element of the volcanic rocks show that contents of $Al_2O_3$, FeO, CaO, MgO, $TiO_2$ decrease with increasing of $SiO_2$. On the basis of Variation diagrams such as $Al_2O_3$ vs. CaO, Th/Yb vs. Ta/Yb, and $Ce_N/YB_N$ vs. $Ce_N$, these rocks represent mainly differentiation trend of calc-alkaline rock series. On the discriminant diagrams such as Ba/La and La/Th ratio, Rb vs. Y + Nb, the volcanic rocks in study area belongs to high-K Orogenic suites, with abundances of trace element and ternary diagram of K, Na, Ca. According to the tectonic discriminant diagram by Wood, these rocks falls into the diestructructive continental margin. K-Ar ages of whole rocks are from andesite to rhyolite $97.0{\pm}6.8~94.5{\pm}6.6,\68.9{\pm}4.8,\61.5{\pm}4.9~60.7{\pm}4.2$ Ma, repectively. Volcanic rocks in study area show well correlation to the Yucheon Group in terms of rock age dating and geochemcial data, and derived from andesitic calc-alkaline magma that undergone low pressure fractional crystallization dominated plagioclase at <30km.

  • This paper examines the characteristics and determinants of welfare attitudes in Korea. Based on Survey data, this study focuses mainly on how social-economic position variables, welfare status and welfare compliances influence attitudes towards welfare. Welfare attitudes can be classified by five areas as follows: legitimacy of welfare functions, affordability of welfare costs, need-based welfare principles, responsibility of welfare provision and adequacy of welfare spending. There are the major findings. First, attitudes towards welfare is identified state-friendly or pro-welfare attitudes. Second, among the social-economic position variables, welfare status and welfare norms variables, welfare compliance has statistically the most significant effects on welfare attitudes. Third, the level of pro-welfare attitudes is largely determined by welfare norms and images espoused by individuals. These findings suggest that we can not apply cleavages thesis which are developed by studies on the support for welfare state.
