Use of Cellulose and Recent Research into Butyrate (섬유소의 이용과 butyrate의 최근 연구)

  • On earth, there are about 5,400 kinds of mammals, of which about 1,000 kinds are herbivores. Among herbivores, about 250 kinds are known to be ruminants. As for cattle and sheep, which are ruminants, fermentation takes places mainly in their rumen; in contrast, for pigs and men, which are non-ruminants, fermentation takes place mainly in their caecum, colon, and rectum. As for the kind and dominance of rumen microorganisms, Bacteroidetes account for 51% and Firmicutes for 43%. As for the dominance of the large intestine microorganisms in men, Firmicutes account for 65% and Bacteroidetes for 25%. Cell wall components are decomposed by microorganisms, and short chain fatty acids (SCFAs) are generated through fermentation; the ratio of acetate, propionate, and butyrate generate is 60:25:15. Butyrate absorbed through the primary butyrate transporter MCT1 (mono carboxylate transports-1) in the intestines activates such SCFA receptors as GPR43 and GPR41. Butyrate has a strong anti-tumorigenic function. Butyrate is characterized by the fact that it has an effect on many cancer cells, contributes to the coordination of functions in the cells, and induces cancer apoptosis. Butyrate activates caspase but inhibits the activity of HDAC (histone deacetylase), so as to induce apoptosis. In addition, it increases p53 expression, so as to induce cell cycle arrest and apoptosis. Anti-inflammation actions of SCFA include the reduction of IL-8 expression in intestinal epithelial cells, the inhibition of NO synthesis, and the restraint of the activity of NF-${\kappa}B$ (nuclear factor ${\kappa}B$), so as to suppress the occurrence of cancers caused by inflammation. Butyrate plays an important role in maintaining physiological functions of intestinal mucous membranes and is used as a cure for inflammatory bowel disease (IBD).

  • Physiological effects of Korean black soybean pigment were investigated. Major anthocyanin pigments of Korean black soybean were extracted with 1% HCl for 24 hours at $4^{\circ}C$. Inhibitory effects of angiotensin converting enzyme ($IC_{50}$) were 0.22 mg/mL (Kumjungkong #1), 0.28 mg/mL (Ilpumkumjungkong) and 0.38 mg/mL (Milyang #95). Inhibitory effects xanthine oxidase ($IC_{50}$) were 0.118 mg/mL (Kumjungkong #1), 0.165 mg/mL (Ilpumkumjungkong) and 0.163 mg/mL (Milyang #95). The cPLA2 inhibitory effects ($IC_{50}$) were $19.7\;{\mu}g/mL$, $10.7\;{\mu}g/mL$ and $25.3\;{\mu}g/mL$. The cytotoxic effects of anthocyanins from Milyang #95 were 66.0% against human colon cell line (HT29), 58.2% against human liver cell line (HepG2) and 64.4% against mouse liver cell line (Hepa), respectively.

  • The cytotoxicity of 1,3-diphenylpropenone derivatives known to inhibit the farnesyl protein transferase (FPTase) was examined against various established tumor cell line, A549 (lung cancer), SKMEL-2 (uterine cancer), HCT-15 (skin cancer), SKOV-3 (brain cancer) and XF-498 (colon cancer) of the 1,3-diphenylpropenone derivatives showing farnesyl protein transferase (FPTase) inhibition activity. And the structure-activity relationship (SAR) between structure of 1,3-diphenylpropenone derivatives as substrate and cytotoxicity was investigated by Free-Wilson analysis as well as Hansch method with tumor cell lines. From the result of Free-Wilson analyses, X-substituents on the benzoyl group have a more important role than Y-substituents on the styryl group. The 2,4-dichloro substituent, 15 and 2,4-dimethyl substituent, 16 showed the highest cytotoxicity (average pI_(50)=5.0). Particulary, the cytotoxicity of X-substituents increased with electronic effect $({\sigma})$ due to weak electron withdrawing group with optimum value $({\sigma}_{opt}=0.22{\sim}0.29})$ whereas that of Y-substituent resulted from various factors such as logP, $B_1$ and R constant.

Fermentation Properties and in vitro Anticancer Effect of Young Radish Kimchi and Young Radish Watery Kimchi (열무김치 및 열무물김치의 발효특성과 in vitro 항암효과)

  • Fermentation properties and in vitro anticancer effect of young radish (YR) kimchi and young radish watery (YRW) kimchi were investigated during fermentation at 5℃. The fermentation of YR kimchi during 2-3 weeks led to the decrease of pH down to pH 4.3, increased acidity, and the highest Leuconostoc sp. counts. YR kimchi showed the acidity of 1.04-1.27% at the pH 4.3, when the kimchi was ripened properly. The fermentation of YRW kimchi during 9 days led to the decrease of pH down to pH 4.3 and the acidity of 0.20%. Inhibitory effects of the juices of YR, YR kimchi, and YRW kimchi on the growth of AGS human gastric adenocarcinoma cells in MTT assay were increased with the added concentration. The juice of YR kimchi had a higher inhibitory effect on the growth of AGS human gastric adenocarcinoma cells than that of YRW kimchi at same concentration. The juice of YR kimchi showed similar inhibitory effects on the growth of AGS human gastric and HT-29 human colon adenocarcinoma cells in MTT assay to baechu kimchi, which the inhibition rates are more than 50%.

Antiproliferative Effect of Extracts from Corydalis heterocarpa on Human Cancer Cells (염주괴불주머니 (Corydalis heterocarpa) 추출물의 암세포 성장 억제 효과)

  • Whole plants of Corydalis heterocarpa were extracted twice with $CH_2Cl_2$ and MeOH in turn. The combined crude extracts were concentrated in vacuo and then partitioned between $CH_2Cl_2$ and $H_2O$. The organic layer was fractionated with n-hexane and 85% aq. MeOH, and the aqueous fraction was also further fractionated with n-BuOH and $H_2O$, successively. Growth inhibition effects of crude extracts and their solvent fractions were evaluated in AGS, HT1080, U-937, MCF-7 and HT-29 human cancer cells using MTT assay. The inhibitory effects of solvent fractions were increased in a dose-dependent manner. Among these tested samples, 85% aq. MeOH fraction showed the most potent inhibitory effect on the growth of human cancer cells. These results suggest that active compounds having much stronger anticancer effect can be isolated from Corydalis heterocarpa.

Inhibitory Effects of Solvent Extracts from Seven Brown Algae on Mutagenicity and Growth of Human Cancer Cells (7종 갈조류의 항돌연변이 및 인체 암세포 증식 억제 효과)

  • 본 연구에서는 해조류의 항돌연변이 및 항암 생리활성물질을 검색하여 발암물질 생성 방지 및 생체 방어 물질로서의 이용 가능성을 검토하고자 Ames test를 이용하여 직접돌연변이원인 MNNG와 간접돌연변이원인 $AFB_1$에 대한 항돌연변이 효과 및 암세포 증식 억제 효과를 알아보고자 하였다. $AFB_1$에 대해서 괭생이모자반(S. horneri)이 실험에 사용된 다른 해조류들 중에서 가장 높은 돌연변이 억제 효과를 보였다. 첨가농도 1.25mg/plate일 때, 괭생이모자반의 acetone+methylene chloride 추출물과 methanol 추출물은 각각 96%, 91%로 실험에 사용된 다른 해조류들의 추출물들 중에서 가장 높았으며 양성 대조군인 다시마의 용매 추출물보다도 높은 돌연변이 억제 효과를 보였다. $AFB_1$과 같은 농도인 0.6mg/plate의 농도의 MNNG를 사용하여 S. typhimurium TA100 균주에 대한 해조류의 항돌연변이성 실험을 한 결과, 간접 돌연변이원인 $AFB_1$에 비해 직접 돌연변이원인 MNNG에 대해서는 다소 항돌연변이 효과가 떨어지지만, 여기서도 실험에 사용된 해조류들의 돌연변이 억제 효과를 살펴볼 수 있었으며 acetone+methylene chloride 추출물의 경우가 methanol 추출물보다 다소 높은 활성을 나타내었다. 항돌연변이 실험에서 효과가 뛰어난 괭생이모자반과 짝잎모자반을 중심으로 인체 암세포(위암세포, AGS 및 결장암 세포, HT-29) 증식억제효과를 살펴본 결과, 용매 추출물을 0.5%, 1% 및 2%의 농도별로 암세포에 처리했을 때 acetone+methylene chloride 추출물과 methanol 추출물은 둘 다 가장 낮은 농도인 0.5%에서부터 농도 의존적으로 암세포 증식 억제 효과가 증가하였다. 이상의 7종의 갈조류 추출물들은 Ames test에서 높은 항돌연변이 효과를 나타냈을 뿐만 아니라 괭생이모자반 및 짝잎모자반은 인체 암세포에 대해서도 높은 증식 억제 효과를 나타냄을 살펴 볼 수가 있었다.

Sudden Death Caused by Clostridium perfringens Type D Enterotoxemia in Feedlot Cattle (비육 우의 Clostridium perfringens D 형 장독 혈증에 의한 폐사)

  • Sudden deaths have occurred in feedlot cattle with marked necro-hemorrhagic enteritis of the jejunum, ileum and colon. Suckling beef calves are the most frequently affected. Over-consumption of large amounts of milk, inadequate colostrum intake, chilling and stress are conducive to the development of enterotoxemia. Enterotoxemia caused by Clostridium perfringens type D mostly occurs following a sudden change of diet, particularly to feeds made richer in order to grow the cattle to market weight in feedlots. During July 2006, sudden deaths of cattle occurred in the Youngcheon regional area of Gyeongbuk province. There were no significant clinical signs except anorexia, depression, intermittent diarrhea and mild respiratory failure. Histological findings revealed a prominent intranuclear inclusion as well as infiltration of the globular leukocytes in various organs including the heart, kidneys, liver, spleen and lymph nodes. Spleen and lymphatic tissues showed lymphatic necrosis and a starry sky appearance. In the submucosa of the small intestines, basophilic aggregation was detected with massive infiltration of the globular leukocytes and eosinophils. Gram staining for the tissue sections containing inclusions of the small intestines revealed a positive histochemical reaction. Taken together, we suggest that Clostridium perfringens type D-induced enterotoxemia is determined to be the cause of sudden death of feedlot cattle.

Bioactive Molecules Produced by Probiotics to Control Enteric Pathogens (프로바이오틱스가 생산하는 생리활성 물질의 장내 유해균 억제 효과)

  • There is a burgeoning number of products on the market that contain probiotics, but do they do you any good? What exactly are probiotics? They have been defined as living organisms that, when ingested in sufficient quantities, provide health benefits beyond basic nutrition. They are often referred to as "friendly bacteria" or "good bacteria." Probiotics have been claimed, amongst other things, to (i) reduce the incidence of colon cancer and other diseases of the colon, such as IBS, (ii) stimulate the immune system, (iii) have anti-hypertensive and anti-cholesterolemic properties, (iv) mitigate against the effect of antibiotics on the intestinal microbiota, and (v) protect against gastrointestinal infections. However, the scientific basis for many of these claims is not well-established. Indeed, the European Food Safety Authority has denied the use of several health claims associated with probiotics, particularly those related to mitigation of diarrhea following consumption of antibiotics. Thus, there is a need for research on the mechanisms of action of probiotics. We have been mainly interested in the use of probiotics to control enteric infections. There are several possible modes of action to explain how probiotics may protect the host from enteric pathogens, including competitive exclusion and immunomodulation. We have shown that probiotics produce bioactive molecules that interfere with bacterial cell-cell communication (also called quorum sensing), and this results in a down-regulation of virulence genes that are responsible for attachment of the pathogen to the gastrointestinal epithelium. These bioactive molecules act on a variety of bacteria, including enterohemorrhagic and enterotoxigenic Escherichia coli, Salmonella, Clostridium difficile and Clostridium perfringens, and there is evidence that they can inhibit the formation of biofilms by Listeria monocytogenes. These bioactive molecules, which are peptidic in nature, can exert their effects not only in vitro but also in vivo, and we have shown that they mitigate against E. coli O157:H7 and Salmonella in mice and Salmonella and E. coli K88 infections in pigs. They can be delivered in foods such as yoghurt and maintain their activity.

Anti-Cancer Effect of Gallic Acid in CT-26 Cells Inoculated Cancer Bearing Balb/C Mice (CT-26 세포 암 유발 마우스에서 Gallic acid의 항암 효과)

  • This study examined the anti-cancer activity of gallic acid(GA) isolated from P. suffruticosa. was analyzed by ESI-MS, $^1H$-NMR, and $^{13}C$-NMR. The anti-cancer activity was evaluated by measuring the cancer size in CT-26 cancer-allograft mice treated with GA(100 mg/kg p.o) for 14 days. The change in body weight, acute toxicity, weight change of the liver and spleen and biomaker of the liver were evaluated in the mice after the GA treatment. As a result, the cancer size of the CT-26 cancer-allograft mice treated with GA decreased significantly compared to that of the cancer mice without significant changes in weight loss (p<0.05) and acute toxicity. The weight of the liver and spleen and ALT, AST and LPO levels increased by cancer were decreased significantly after the GA treatment, and the GSH levels decreased by cancer were increased significantly with the GA treatment (p<0.05). Therefore, GA could be an attractive lead for the development of anticancer agents.

A Case of Eosinophilic Colitis in a 11-year-old Boy (호산구성 대장염 1례)

  • Eosinophilic gastroenteritis, which shows characteristic eosinophilic inflammation, involves any part of the intestine from esophagus to colon. The immunopathogenesis is expected to be associated with IgE-mediated or non-IgE-mediated reaction, but the precise mechanism is not revealed yet. The clinical manifestation is variably dependent on the extent of eosinophilic infiltration. Usually the symptoms, such as abdominal pain, diarrhea, vomiting, wax and wane for a few months result in failure to thrive, which implicates the importance of early detection. Although the diagnosis is made through clinical and histopathologic evidences, we should suspect the illness in a case of anemia, hypoalbuminemia, and peripheral eosinophilia. Recently, we experienced a case of non-IgE-mediated eosinophilic colitis in a 11-year-old male who complained of diarrhea, right lower abdominal pain, body weight loss, was diagnosed with peculiar histopathologic finding of sigmoid colon specimens obtained by sigmoidoscopy.
