  • 결명자의 주용성분인 노르-루브로푸사린의 함량이 높은 추출물을 얻기 위하여 결명자분말을 0~100%의 에탄올수용액으로 추출하여 고농도의 노르-루브로푸사린을 함유하는 결명자 추출물을 제조하였다. 결명자추출물과 주요 함유성분인 노르-루브로푸사린은 항산화활성과 프리라디칼소거 작용을 나타내었으며, H2O2 유도 세포독성에 대해서도 억제적으로 작용하여 cyto-protective effect를 나타내었다. 또한 DNA crosslinking agent 인 mitomycin C 유도 소핵생성에도 결명자 70% 에탄올추출물과 노르-루브로파사린이 매우 높은 억제활성을 나타내었다. 따라서 결명자 추출물과 노르-루브로푸사린은 산소라디칼들에 의한 산화적 손상 및 DNA 손상 등에 억제적으로 작용하는 기전을 활용하여 항산화성 스트레스를 통한 항노화 , 암예방제로서의 응용가능성이 높은 물질로 판단되었다.

Allelopathic Inhibition by Extracts and Volatiles from Leaf and Seed of Sicklepod (Cassia tora L.) (결명자(決明子)(Cassia tora L.) 엽(葉)과 종자(種子)의 추출물(抽出物)과 휘발성분(揮發成分)의 타감성(他感性) 생육저해작용(生育沮害作用))

  • The growth of weeds and some other plants has been considered to be inhibited by sicklepod(Cassia tora L.) sharing the habitat. The study was conducted, for the first time, to propose that this phenomenon is not only due to competition for physical and nutritional conditions but also due to allelopathy. In addition, autotoxicity of sicklepod was examined. The results are summarized as follows. 1. Rice sheath length reduced progressively from 18 % to 36 % with increasing the concentration of treating aqueous extracts of sickle pod seeds, but rice germination was not affected. In contrast, radish hypocotyl length was not reduced by the aqueous extract treatment but radish germination was significantly reduced by 66 % at 1 : 10 and 1 : 5 treatment. 2. Total chlorophyll contents in rice seedling decreased from 50 % to 65 % by treatment of seed aqueous extracts diluted from 1 : 50 to 1 : 5 ratio. 3. Aqueous extracts of sicklepod leaves significantly reduced hypocotyl length and fresh weight in radish and germination in rice, but mung bean was slightly affected by aqueous extracts only in fresh weight. 4. Volatiles from fresh, immature seeds with husk reduced the radish germination and seedling growth and radish root growth appeared to be more sensitive to the exposure to volatiles from fresh immature seeds than both germination and hypocotyl growth. 5. Volatiles from sickepod leaves inhibited germination and growth of radish, rice and mung bean, and seedling growth was more sensitive to volatiles from leaves than germination. 6. Volatiles from sicklepod leaves reduced germination and radicle length of sicklepod itself. 7. Collectively, it is concluded that there are water-soluble and volatile substances responsible for allelopathy in sicklepod.

Effects of Roasted Cassia tora L. Extracts on the Chemical Changes and Microbial Growth (열처리가 결명자의 화학성분 변화 및 추출물의 균체증식에 미치는 영향)

  • The effects of roasting Cassia tora L. were investigated for proximate composition, color, volatile flavor profile, microbial growth and alcohol fermentation. While moisture, protein and fat contents decreased with increasing roasting temperature, fiber and ash contents increased. The L, a and b values of Cassia tora L. extracts decreased with increasing temperature, and only a small difference in total color difference$({\Delta}E)$ was observed. A little difference in major flavor components between raw and roasted treatment was found during roasting. Furfuryl alcohol, a major component of coffee flavor, was separated from Cassia tora L. extracts extracted with ethyl ether. The yeast growth was the highest on the water-extract of Cassia tora L. roasted at $160^{\circ}C$. With increased levels of water-extract at $160^{\circ}C$, S. cerevisiae grew rapidly for 24 hr incubation and the growth rate was higher than the unroasted control group. The growth rate of Bacillus subtilis was the highest in a treatment of 0.5% concentration. Little differences in ${\alpha}-amylase$ produced from Bacillus subtilis were observed among the treatment groups. The total alcohol content increased with increasing roasting temperature during alcohol fermentation.

Effect of the Ethanol Extract of Cassia tora L. on Antioxidative Compounds and Lipid metabolism in Hepatotoxicity of Rats-induced by Ethanol (결명자 에탄올추출물이 알코올 투여 흰쥐의 항산화물질 및 지질대사에 미치는 영향)

  • This study was done to investigate the effects of ethanol extract of Cassia semen (Cassia tora L.) on the activities of hepatic oxygen free radicals metabolizing enzymes and blood lipid profile in rats of hepatotoxicity induced by ethanol. Sprague-Dawley male rats weighing 100~160 g were divides into 5 groups; control grouts (CON), Cassia semen ethanol extracts (200 mg/kg) treated group (CEL), ethanol (10 mL/kg, 35%) treated group (ETH), Cassia semen ethanol extracts (200 mg/kg) and ethanol treated group (CE1 ) and Cassia semen ethanol extracts (400 mg/kg ) and ethanol treated group (CE2), respectively. Compared with ETH, the growth rate of CE1 and CE2 were to be increased tendency, and in blood levels of total cholesterol, LDL-cholesterol and the activities of alanine aminotranferase and asparate aminotranferase elevated by ethanol were significantly decreased (p<0.05). It was observed that the activities of superoxide dismutase, catalase, xanthine oxidase and glutathione peroxidase of rat liver increased by ethanol, were more decreased by the treatment of Cassia semen ethanol extract than the only ethanol-treated group. The content of glutathione depleted by ethanol treatment was increased in CE1 and CE2. TBA-reactants of liver increased by ethanol were decreased in CE1 and CE2, compared with ethanol-treated group. These results suggested that ethanol extract of Cassia semen may influence upon the ability of oxygen free radical detoxication and lowering of blood lipid level on ethanol-induced hepatotoxicity in rat.

Study on the Organoleptic Quality Characteristics of Cassia tora teas by Roasting Conditions (볶음조건에 따른 결명자차의 관능적 품질특성에 관한 연구)

  • The roasting condition and organoleptic characteristics in Cassia tora tea were investigated. Intact Cassia tora seeds were composed of water 11.6%, crude protein 13.1%, crude fat 4.4%, crude fiber 13.8%, N-free extract 47.2% and ash 4.9%. Organoleptic qualities in Cassia tora tea were sweetness, astringency, tartness, bitterness, roasted coffee like, roasted barley like and burnt smell. Organoleptic qualities were investigated by descriptive analysis method, too. Overall acceptability was increased by roasting but it was low because of formation of bitterness and burnt smell at excessive roasting conditions. Sweetness was the most important factor in organoleptic quality of Cassia tora seeds and the optimum condition for the best quality was $210^{\circ}C$, 20 minutes.

  • The purpose of this study was to investigate effects of chestnut inner skin tea, brown rice-green tea and Cassia lora tea on the activation of physiological functions (regional cerebral blood flow, mean arterial blood pressure, proliferation of immunocytes in vitro and in vitro, suppression of cancer cell proliferation) in mouse and rat. We used 8 weeks-old balb/c male mice, 300g ICR rats and L1210 cell lines. Regional cerebral blood flow(rCBF) and mean arterial blood pressure(BP) were measured using Leser-Doppler Flowmetry(LDF) and the proliferation of cells was measured using a colorimetric tetrazolium assay(MTT assay). The experimental results are as follows : 1. rCBF was increased by Cassia tora tea, but decreased by chestnut inner skin tea in rats. 2. BP was increased by brown rice-green tea in rats. 3. Proliferation of mouse thymocytes and splenocytes were significantly increased by chestnut inner skin tea in vitro. 4. Proliferation of mouse thymocytes was decreased by Cassia tora tea and brown rice-green tea in vitro. 5. Proliferation of mouse thymocytes was decreased by Cassia tora tea and brown rice-green tea in L1210 transplanted mice. 6. Proliferation of splenocytes was accelerated by chestnut inner skin tea in L1210 transplanted mice. 7. Proliferation of L1210 cells was inhibited by chestnut inner skin tea and Cassia tora tea in L1210 transplanted mice.

Effect of Cassia tora Ethanol Extract on the Lipid Levels of Serum and Liver in Rats Fed High Cholesterol Diet (결명자 에탄올 추출물이 고콜레스테롤 식이를 섭취한 흰쥐의 체내 지질함량에 미치는 영향)

  • This study was undertaken to investigate the effect of Cassia tora ethanol extract on the lipid levels in serum and liver of rats fed high cholesterol diet. Experimental rats were divided into the following 4 groups: normal diet group, high cholesterol diet group, high choleslerol-0.25% C. tora ethanol extract group and high cholesterol-0.5% C. tora ethanol extract group. After 4 weeks, rats were sacrificed and analyzed the serum lipid profiles, activities of serum alanine aminotransferase (AST), aspartate aminotransferase (ALT), hepatic glucose-6-phosphate dehydrogenase (G6PDH) and malic enzyme (ME). It was also determined the contents of total cholesterol, triglyceride and thiobarbituric acid reactive substances (TBARS) in liver. There was no difference in weight gains between experimental groups. The concentrations of serum total cholesterol, free cholesterol, triglyceride and free fatty acid were tended to be decreased in C. tora groups compared with control group. HDL-cholesterol concentration was significantly decreased in high cholesterol diet group and slightly increased by C. tora ethanol extract feeding. The contents of liver cholesterol and triglyceride were higher in high cholesterol diet group than normal group, but significantly decreased by feeding of C. tora ethanol extract. Supplementation of 0.5% C. tora extract decreased significantly the activities of hepatic G6PDH and ME. Activities of serum AST, ALT and contents of liver TBARS were tended to be increased with high cholesterol diet and reduced by C. tora ethanol extract supplementation but had not significance. These results suggest that C. tora ethanol extract may exert a lipid lowering effect in serum and liver of rats.

Effect of Dietary Fiber Purified from Cassia Tora on the Quality Characteristics of the Bread with Rice Flour (결명자 식이섬유가 쌀 첨가 식빵의 제빵특성에 미치는 영향)

  • This study was accomplished to investigate the effect of dietary fiber purified from Cassia Tora on the quality characteristics of the bread compared to hydroxypropyl methyl cellulose (HPMC) and xanthan gum (XG). The bread-making flour was prepared with wheat flour/rice flour (75/25) and added with 3% various dietary fibers. Dietary fiber of Cassia Tora (CT fiber) showed the highest volume expansion and the volume of bread with XG was the lowest. The values of lightness in both the crust and crumb color were the highest in the bread with XG. There was no significant difference in the score of sensory evaluation about color. As a result of texture measuring by Texture Analyzer, springiness and cohesiveness were not affected by the addition of CT fiber. However the significant decrease of gumminess, hardness and chewiness were observed from the bread containing CT fiber. Overall acceptance scores by sensory evaluation of baked bread with CT fiber were not significantly different from those of control.

Desmutagenic Effect of Water Extract from Cassia tora L. on the Mutagenicity of N-methyl-N-nitro-N'-nitrosoguanidinein in E. coli PQ37 (N-methyl-N-nitro-N'-nitrosoguanidine의 변이원성에 대한 결명자 물 추출물의 항돌연변이 효과)

  • The desmutagenic activity of the water extract of Cassia tara L on the mutagenicity induced by 4-nitroquinoline 1-oxide (4-NQO) and N-methyl-N-nitro-N'-nitrosoguani-dine (MNNG) was studied using the SOS Chromotest with Escherichia coli PQ37. The inhibition rates of water extract of Cassia tara L. at concentration of $100{\mu}g$/assay were 27.5% and 40% against 4-NQO and MNNG. The water extract of Cassia tara L. was separated into methanol soluble and methanol insoluble parts. The methanol soluble part exhibited higher inhibition effects than the methanol insoluble part against the mutagenic activities of 4-NQO and MNNG. Step-wise fractionation of methanol soluble part was done to obtain methanol, ethyl acetate and water fractions. Among these fractions, water fraction had the strongest inhibitory effects of 23.0 and 19.0% against mutagenicities of MNNG and 4NQO, respectively. The results clearly indicated that the water fraction showed much stronger antimutagenicity against MNNG than 4NQO. The inhibition rates of aqueous fraction of methanol-soluble from water extracted Cassia tara L. at concentrations of 1.0, 10, 100 and $250{\mu}g$/assay were 8.0%, 12.0%, 25.5% and 43.0%, respectively. The water fraction showed the inhibitory effects with dose response against the mutagenic activities induced by MNNG.

Effect of Cassia tora Ethanol Extracts on Carbon Tetrachloride-Induced Liver Injury in Rats (흰쥐에 있어서 사염화탄소 유발 간손상에 미치는 결명자 에탄올 추출물의 영향)

  • This study was conducted to investigate the effect of Cassia tora ethanol extracts on carbon tetrachloride($CCl_4$)-induced hepatotoxicity in rats. Male Sprague-Dawley rats were divided into the following 4 groups: normal group, $CCl_4$, treated group, $CCl_4-0.25%$ Cassia tora ethanol extracts group and $CCl_4-0.5%$ Cassia tora ethanol extracts group. Rats were fed with each experimental diet and water for 5 weeks. Liver weights of rats treated only with $CCl_4$, were significantly increased compared to normal group, but not in rats fed diet containing Cassia tora ethanol extracts. Cholesterol and triglyceride contents in serum and liver were also not influenced by either $CCl_4$, treatment or the supplementation of Cassia tora ethanol extracts. $CCl_4$, treatment significantly increased ALP activities, however the supplementation of Cassia tora ethanol extracts significantly decreased the activities of serum ALT, AST, ${\gamma}-GTP$ in dose-dependent manner. Cassia tora ethanol extracts significantly reduced $CCl_4-induced$ elevation of liver TBARS contents. Activities of superoxide dismutase and catalase were decreased by $CCl_4$, treatment, however by the supplement of Cassia tora ethanol extracts slightly increased activities of SOD and catalase. The activity of glutathione peroxidase in groups fed diets containing Cassia tora ethanol extracts was significantly decreased compared to that of the control group. These results suggest that Cassia tora ethanol extracts may exert protective effect against $CCl_4-induced$ liver injury through the prevention of lipid peroxidation.
