Effective Screening Methods for Lipoxygenase Isozymes in Soybean Seeds (콩 lipoxygenase 효소의 효율적인 검정법)

  • Normal soybean seed contains three lipoxygenase isozymes called L-1, L-2, and L-3, respectively, which are responsible for the generation of undesirable grassy-beany flavors. Simple and effective methods for the detection of lipoxygenase isozymes were developed in soybean seeds. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) has been tried in separating these isozymes. It was done effectively on 7.5% separating gel and 4.5% stacking gel. However, no reliable method has been developed specifically for separating L-3, L-13 and L-23. Visual judging methods were based on the bleaching activities of lipoxygenase in contact with methylene blue and ${\beta}$-carotene. Sodium linoleate bleaching method was adopted to determine L-1 and L-2. Carotene bleaching and spectrophotometric methods were used to determine L-3. These systems were very rapid within one minute, furthermore only required a small piece of cotyledon (below 10 mg) and the other part could be used for generation advance after analysis. It was demonstrated that 200 seed samples could be analyzed per day by one laboratory assistant. The combination of visual judging methods and electrophoresis is suitable for breeding programs. It took 6.5 hours for analysis of 100 seed samples by one person.

Studies on Carbohydrate Accumulation in Aspergillus niger During the Differentiation (검정곰팡이의 분화(分化)에 따른 총탄수화물(總炭水化物) 및 트레하로즈량(量)의 변동(變勳)에 대한 연구(硏究))

  • The changes of total carbohydrates and trehalose levels during differentiation of A. niger were studied. Aspergillus niger was cultivated in Czapeck-Dox medium by the method of surface culture and shake culture. Total carbohydrates and trehalose were fractionated and determined by the method of Trevelyan and Harrison (1956). Total carbohydrates and trehalose accumulated in A. niger during sporulation. The influences of nutritional starvation on the levels of cellular carbohydrates in A. niger, which was cultivated in each nitrogen, phosphate and glucose limited Czapeck-Dox medium, were studied. The levels of total carbohydrates and trehalose in A. niger cultivated in nitrogen limited medium increased much than those cultured in full medium. The total carborates and trehalose levels in A. niger cultivated in phosphate limited medium increased, but the increases were less than those cultured in nitrogen limitted medium. In glucose limited medium, any changes of total carbohydrates and trehalose levels were not found. It is considered that the biochemical mechanisms responsible for the changes of total carbohydrates and trehalose levels may be related with differentiation of Aspergillus niger.

  • 소표본 분할표 자료에서 적합도 검정통계량들의 카이제곱 근사 적용 가능에 대하여 많은 연구가 진행되었다. 소표본에서 세 가지 검정 통계량(피어슨 카이제곱 $X^{2}$, 일반화 가능도비 $G^{2}$, 그리고 역발산 I(2/3) 검정통계량)에 관하여 비교한 Rudas(1986)의 연구를 확장하여, 최근에 제안된 차이측도(BWHD(1/9), BWCS(1/3), NED(4/3) 검정통계량)를 포함시켜 비교 분석하였다. 독립모형의 이차원 분할표, 조건부 독립모형과 한 변수 독립 모형을 따르는 삼차원 분할표에 대한 모의실험을 통하여 생성된 90과 95 백분위수와 이에 대응하는 95% 신뢰구간을 살펴보고 실제 백분위수와 비교하였다. 그 결과 $X^{2}$, I(2/3), 그리고 BWHD(1/9) 검정통계량이 유사한 결과를 나타내었고 이 통계량들이 기존에 제안된 검정통계량들보다 적은 표본크기에서도 카이제곱 근사방법에 적용 가능함을 발견하였다.

Study on the partially premixed flames produced by a coflow burner as temperature calibration source (동축류 버너에서 생성된 부분 예혼합 화염을 이용한 화염 온도 측정 검정원 연구)

  • We investigated a uniform temperature zone, produced by double flame structure of a coflow CH4/air partially premixed flame, to be used as a temperature calibration source for laser diagnostics. A broadband N2 CARS(coherent anti-Stokes Raman spectroscopy) system with a modeless laser was used for temperature measurement. When the stoichiometric ratio was 1.5, we found the uniform temperature zone in radial direction of the flame of which the averaged temperature was 2110 K with standard deviation 24 K. In the stoichiometric ratio range between 2.0 and 2.5, we found very stable temperature-varying zones in vertical direction at the center of the flame. The size of the zone was approximately 15 mm and it covered a temperature range from 300 K to 1900 K. We also suggest that this zone can be used as a calibration source for 2-D PLIF(planar laser induced flurescence) temperature measurement.

  • To provide standard data of temperature and species concentration in a flame for calibrating the laser based combustion diagnostics, we investigated combustion characteristics of a flat flame burner(Mckennar Product). For various stoichiometric ratios we measured temperature and concentration of OH in the premixed methane/air flame with Coherent anti-Stokes Raman spectroscopy and laser induced fluorescence technique, respectively. Assuming the chemical equilibrium condition at the measured temperature, the mole fraction of the OH radical in the flame was obtained and compared with numerical analysis.

  • In this paper, we study a method to determine the existence of unit roots by using the adaptive lasso. The previously proposed method that applied the adaptive lasso to the original time series has low power when there is an unknown trend. Therefore, we propose a modified version that fits the ADF regression model without deterministic component using the adaptive lasso to the detrended series instead of the original series. Our Monte Carlo simulation experiments show that the modified method improves the power over the original method and works well in large samples.

  • The purpose of this study was to compare the conventional culture method, enzyme immune method and the PCR method using species-specific primer in the analysis on the Salmonella spp. found in domestically distributed pet foods. For the study, Salmonella spp. were detected from 175 samples. From the conventional culture method and the PCR method, two samples (jerky and corn gluten) were determined as positive. Also, from the enzyme immune method, one sample (corn gluten) was test-positive. The study revealed that application of the PCR method with species-specific primer allows better distinguishment between the species of the strain collected from the samples than the conventional culture method and/or the enzyme immune method.

  • 한우 6번 염색체 유전자 지도에서 QTL (quantitative trait loci) 분석을 실시하여 선별된Locus 값을 순열검정(Permutation Test)을 이용하여 유의성 검정을 실시하였다. 한편, 우수경제형질 DNA marker들을 K-평균 군집법을 실시 파악하였다. 이들 QTL과 K-평균법에 의해 한우의 염색체 6번 ILSTS035의 우수 DNA marker 235번을 선별하였다. 선별된 DNA Marker 235번을 출품우에 적용하여 Bootstrap 방법을 이용하여 신뢰구간을 구하여 검정하였다.

  • 99년과 2000년에 KBS를 통해 방송돼 선풍적인 인기를 끌었던‘검정고무신’은 이미 어른 아이 할 것 없이 우리에게 친숙한작품. 현재는 30~40대의 기성세대 된 이들의 어린시절이었던 60~70년대의 추억을 정겹게 묘사한 창작 애니메이션으로 재미뿐 아니라 아련한 추억의 감동까지 동시에 전하고 있기 때문이다. 이러한 화제의 애니메이션 검정고무신이 3년만에 부활해 지난 5월 19일부터 새롭게 안방문을 두드렸다. 원작만화가 92년부터 출간되기 시작해 아직도 인기리에 연재되고 있는 상황에서 재구성된 검정고무신은 시청자들의 관심을 반증하는 것이다. 특히 후속시리즈로 다시 제작된 이 작품은 산학협동으로 제작돼 주목을 받고 있다. 아련한 추억의 세계로 우리를 인도하고 있는 검정고무신을 만나본다.
