Effects of Supplemention of Rehmannia radix on Performance and Physiological Status in Broiler Chicks (건지황 첨가가 육계의 성장과 생리적 변화에 미치는 영향)

  • The present study was conducted to investigate the effects of dietary supplemental dry powder of Rehmannia radix (RR) root on the growth performance and physiological status of broiler chicks. The treatments consisted of corn-soybean meal control diet, and RR 0.5, 1, and 2% diets. A total of 160 Arbor Acre male broiler chicks were randomly allotted to 16 pens ; four pens per treatment and 10 birds per pen. The growth performance, feed intake and carcass weight did not show any differences among the treatment groups, but feed conversion was significantly lower (P< 0.05) in RR 1% group than that of the control group. The muscle color was lighter (P < 0.05) in RR lof group than those of the other treatment groups. The blood concentrations of protein, glutamic-oxaloacetic-transaminase (GOT) and glutamic- pyruvic-transaminase (GPT) were not different (P > 0.05) among the treatment groups, but the blood total cholesterol content was significantly decreased (P < 0.05) in RR 2% group than those the other treatment groups. The blood concentration of sugar was lower (P <0.05) in RR 2% group than those of the other treatment groups, and hemoglobin concentration was decreased (P <0.05) by addition RR. Crude fat content of thigh muscle was higher in RR 1% group than those of the other treatment groups, and bone hardness of broiler chicks fed RR was higher (P < 0.05) than that of the control group. The saturated fatty acid content of the thigh muscle tende to be high in control and RR 0.5% groups, and unsaturated fatty acid content tende to be high in RR 1 and 2% groups than those of the other treatment groups

Comparison of Biological Activities on Rehmannia Radix and R. Radix Preparata produced in Korea (국내산 건지황과 숙지황의 생리활성 비교)

  • We investigated the biological activities on ethanol extracts of R. glutinosa and R. glutinosa Preparata. The result of anti-mutagenicity suggested that the ethanol extract of R. glutinosa Preparata showed stronger activity than that of R. glutinosa ethanol extract. All of the ethanol extracts showed over 50% growth inhibition of several cancer cell lines. Especially, 61% of the cell growth of Hep3B was inhibited by adding 1g/ l of ethanol extract of R. glutinosa Preparata. In hypoglycemic activities and controlling blood pressure, 63.69% of ${\alpha}-glucosidase$ and 56.58% of ACE activities were inhibited by adding 1g/ l of ethanol extract of R. glutinosa Preparata. In general, the ethanol extract of R. glutinosa Preparata showed higher biological activities than those of R. glutinosa.

Characteristic Component of Rehmanniae Radix Preparata Compared to Rehmanniae Radix and Rehmanniae Radix Crudus (숙지황, 건지황 및 생지황 중 숙지황의 특이성분 검색)

  • Rehmanniae Radix Preparata is manufactured with Rehmanniae Radix accoding to KP V. For quality control of Rehmanniae Radix Preparata, its standard component is required. The methanol extracts of Rehmanniae Radix crudus, Rehmanniae Radix, Rehmanniae Radix preparata were divided into the three groups of ether, butanol and aqueous fraction by liquid-liquid separation. In the comparative TLC of ether fraction, the characteristic component of Rehmanniae Radix preparata was found. The ether fraction was evaporated and separated on the silica gel column with chloroform-methanol and further separated on the preparative silica gel TLC with chloroform-methanol-water. The component was illucidated as 5-(hydroxymethyl)-2-furancarboxaldehyde(5-HMF). 5-HMF was not found in Rehmanniae Radix crudus and found in Rehmanniae radix in much less Quantities than Rehmanniae Radix Preparata.

Quality Characteristics of Rehmannia radix Preparata with Pre-soaking Solvents (침지용매에 따른 숙지황의 품질특성)

  • This study was performed to investigate the effects of pre-soaking methods on the preparation of Rehmannia radix Preparata (R.P). The R. radix L (R.L) was soaked in distilled water and traditional Korean wine for 24 hr, then the soaked R.L was treated with a traditional nine-time steaming process. Next, catalpol, 5-hydroxymethyl-2-furaldehyde (5-HMF), polyphenols, flavonoids, antioxidant activities, and ACE inhibition activity were analyzed for the R.P produced by the different methods. The catalpol content of the R.L was 631.4 ppm, but the content decreased as steaming increased to 8-9 times. The 5-HMF, polyphenol, and flavonoid contents of the R.L were 0.12 mg/g, 5.09 mg/g, and 0.83 mg/g, respectively, and these increased gradually with increasing steaming times. As the steaming times of the distilled soaking water increased, the antioxidant activities of 1 mg/mL increased from 19.44% to 75.60% at 14 times of steaming. The ACE inhibition activities of 1 mg/mL of the distilled soaking water increased from 28.70% to 94.78% at 10 times of steaming, but decreased afterward.

Enhancement of Alcohol Fermentation Yield by Adding the Extract of Dried Rehmannia glutinosa Liboschitz (건지황 추출물을 이용한 알콜 발효 수율 증진)

  • The juice extract of Rehmannia glutinosa Liboschitz was used to improve the productivity of ethanol in alcohol fermentation process using a 5 L fermentor under batch and fed-batch cultivations. For batch cultivation, both cell density and ethanol production were increased as the extract of R. glutinosa was increased, showing 11.8 (g/L) of maximum cell density and 0.092 (% /hr) of maximum alcohol productivity in adding 30% (v/v) of the extract. However, in adding more than 40% of the extract both cell growth and ethanol production were dropped. The cell growth was severely inhibited in 50% addition. It was found that fed-batch cultivation in adding 30% of the extract of R. glutinosa was an effective process than batch cultivation, yielding up to 30% cell growth and ethanol production. This ethanol productivity was also 30-40% higher than that obtained from a conventional alcohol fermentation. It can tell that dried R. glutinosa Liboschitz is to be used for both enhancing the yield of alcohol fermentation and utilizing biologically active substances possibly transported from R. glutinosa Liboschitz into fermented broth.

Changes of the Constituents in the Rehmanniae Radix Preparata during Processing (숙지황 제조과정에 따른 성분함량 변화)

  • This study was performed to obtain the good processing in the Relunanniae Radix Preparata. The contents of the Relunanniae Radix and the Relunanniae Radix Preparata produced through different processes were analyzed in the 5-hydroxymethyl-2-furaldehyde (5-HMF), sugar, total nitrogen, crude lipid and ash. 5-HMF was not detected in the Relunanniae Radix, but detected in the Relunanniae Radix Preparata. 5-HMF content was increased gradually with processing times (1∼9 times) and increased expressly in the Relunanniae Radix Preparata steamed for 7 times. Sucrose, fructose and glucose were contained in the Relunanniae Radix, but sucrose was not detected and fructose and glucose were increased largely in the Relunanniae Radix Preparata steamed for 1 time. Fructose and glucose were decreased gradually with processing times (2∼9 times), but the gap of decrease was insignificant. Total nitrogen was changed slightly and crude lipid was decreased slowly with processing times. The ash was suitable to KPⅧ rules (less than 6.0%). From this analysis we found out the content of 5-HMF from Relunanniae Radix Preparata steamed more than 7 times was suitable to KPⅧ rules (more than 0.1%).

The Study for the Production of Rehmannia Glutinosa and the Import Movement and the Improving Methods of Distribution (국내 지황 생산 및 수입 동향과 유통 개선방안에 관한 연구)

  • Rehmannia glutinosa is a perennial herb belonging to the family Scrophulariaceae and is used in traditional oriental medicine. This study was carried out current status of breeding of R. glutinosa, its problems, and the matters to be improved were made. Based on the research data published so far, this study summarized the characteristics of breeds cultivated in Korea and compared and analyzed the status of cultivation area, production, import, and price changes for 15 years from 2008 to 2021. The cultivated area of R. glutinosa is gradually decreasing, and the amount of import is increasing and decreasing repeatedly. Amount of import has declined significantly. There was a lot of disinfection and disposal in the middle of import, and dried Jihwang and Sukjihwang were imported without distinction. R. glutinosa must be cultivated and supplied as a variety desired by the industry, cultivation techniques need to be taught to producers. In order to clearly identify the amount of import, import code of dried Jihwang and Sukjihwang must be classify. In addition, it is necessary to develop and supply harvesting agricultural machines for mechanization. For the stable income of producers, supply control through contract cultivation will ensure stable farm income.

Heavy Metals and Pesticides with Maturing Process of Rehmanniae Radix Preparata (숙지황 제조과정에서 중금속과 잔류농약)

  • To obtain the risk assessment of hazardous materials with maturing process of Rehmanniae Radix Preparata, the residual heavy metals and pesticides in samples are surveyed. Concentration of As, Cd and Pb during the maturing process of Rehmanniae Radix Preparata of Korea are $0.147{\pm}0.007\;mg/L,\;0.025{\pm}0.001\;mg/L\;and\;0.193{\pm}0.021\;mg/L$, respectively. Concentration of As, Cd and Pb during the maturing process of Rehmanniae Radix Preparat of China are $0.0145{\pm}0.014\;mg/L,\;0.023{\pm}0.002\;mg/L\;and\;0.299{\pm}0.013\;mg/L$, respectively. Concentration of As, Cd and Hg remain unchanged during the maturing process of Rehmanniae Radix Preparata. Concentration of As, Cd and Pb in Rehmanniae Radix Preparat on the Korea are 3.53%, 6.66% and 5.16% for Korea Food & Drug Administration (KFDA) advisory level, respectively. The residual pesticides, such as aldrin, $BHC-{\alpha},\;BHC-{\beta},\;BHC-{\gamma},\;BHC-{\delta}$, captan, chlorfenapyr, chlorpyrifos, DDD, DDE, DDT-o,p, DDT-p.p, dieldrin, endosulfan-sulfate, $endosulfan-{\alpha}$, $endosulfan-{\beta}$, fenitrothion, penthoate, permethrin, quintozene, tolyfluanid and vinclozolin, are not detected in Rehmanniae Radix Preparat on the Korea and China. The hazardous materials in Rehmanniae Radix Preparata on the Korea and China are verified the safety of the residual heavy metals and pesticides compare with Korea Food & Drug Administration (KFDA) advistory level.

Changes in Quality of Rehmanniae radix Preparata with Heating Conditions (열처리조건에 따른 숙지황의 품질변화)

  • This study was performed to develop a more simple and convenient heat treatment process for increasing heating temperature than the traditional 9-times steaming process for Rehmanniae radix Preparata (R. radix P.). The R. radix was heated to various temperatures (110, 120, 130, 140 and $150^{\circ}C$) for different lengths of time (1, 2, 3, 4 and 5 hr). Chromaticity, free sugar, amino acid, catalpol and 5-HMF (5-hydroxy-methyl-2-furaldehyde) content of water extracts of R. radix P. were analysed. With increasing heating temperature and time, the L, a and b-value of samples gradually decreased. Fructose and glucose contents were increased to $120^{\circ}C$ for 5 hr (5.21 and 6.67%, respectively) with no changes afterward. Sucrose content was not detected after $130^{\circ}C$. Total amino acid was increased to $140^{\circ}C$ and decreased afterward. The maximum total amino acid content was 4,172 mg/100 g at $140^{\circ}C$ for 2 hr (R. radix L.: 3,029 mg/100 g). The catalpol was decreased to $130^{\circ}C$ for 1 hr and not detected afterward. The 5-HMF content increased with increasing heating temperature and time. The maximum content of 5.HMF was 2.5% in $150^{\circ}C$ for 5 hr. Hence, it can be suggested that the heat treatment at $130^{\circ}C$ for 2 hr or more is the most optimum processing conditions, instead of the traditional 9.times steaming process.
