Analysis of Florfenicol in Meats (식육 중 항생제 florfenicol의 분석)

  • Analytical method for determination of florfenicol was developed for estimate veterinary drug residue of unestablished MRLs in meat. The method was validated in correspondence with the CODEX guideline for florfenicol residue in meat. The samples mixed with sodium sulfate were extracted with ethyl acetate. After clean-up, the residue was dissolved in mobile phase and analyzed using high-performance liquid chromatography with fluorescence detector. The calibration curve showed good linearity($r^2=0.9997$) within the concentration range of $0.05{\sim}1.0\;mg/kg$. The limit of detection(LOD) and limit of quantification(LOQ) were validated at 0.012 and 0.039 mg/kg, respectively. The recoveries in fortified meat ranged from 85.6 to 95.6%($1.1{\sim}5.3%$ RSD) at the 0.05 to 0.4 spiking levels. We monitored 150 samples of meats that were purchased in Korea(Seoul, Busan, Daegu, Daejeon and Gwangju). Among tested samples, florfenicol was detected in 1 of pig at the level of 0.040 mg/kg, and below LOQ in 1 of cattle, 2 of pig and 2 of chicken. The residues of florfenicol in the tested samples were within the MRLs.

  • This survey was carried out to determine five sulfonamide(sulfamerazine, sulfamethazine, sulfadimethoxine, sulfamonomethoxine, sulfaquinoxaline) residues in beef, pork, chicken and swine kidney. For this survey, 30 samples of beef, 15 samples of chicken, 10 samples of pork and 10 samples of swine kidney were collected in Chonnam from June, 1992 to June, 1993, and were analyzed by HPLC. The recoveries of sulfamerazine, sulfamethazine, sulfamonomethoxine, sulfadimethoxine, and sulfaquinoxaline in spiked samples between $0.25{\sim}1.00$ ppm were 71.7%, 80.3%, 71.6%, 70.9%, 68.4%, respectively. None of 65 samples which were examined exceeded 0.1 ppm. Of 15 chicken muscle samples, 2 samples exceeded 0.05 ppm in sulfamerazine (0.077 ppm) and sulfamethazine (0.075 ppm), respectively. Of 10 swine kidney samples, 1 sample exceeded 0.05 ppm in sulfadimethoxine (0.052 ppm). And sulfanilamide concentration of swine kidney were higher than pork.

Bacterial Distributions of Escherichia coli and Bacillus cereus etc. Isolated from Dried Seasoned Marine Products in Garak Fishery Wholesale Market in Seoul, 2009 (그대로 섭취하는 수산가공식품 중 조미건어포류에 대한 주요 식중독균류 분포(2009))

  • This study was practiced to survey food-borne bacterial aspects of Escherichia coli and Bacilhus cereus etc. from 210 processed seasoned marine products in Garak fishery wholesale market in Seoul, 2009. Distributions of these bacterial isolates were 28 coliforms, 32 Bacillus spp., 71 Staphylococcus spp., 11 Listeria spp, and 10 Enterococcus spp. in 157 dried squids, 23 dried file fishes, and 20 dried pollacks etc. respectively. Results in 16 kinds of antibiotics susceptibility test by disc diffusion method, special multiple drugs resistance patterns were NOR + LVX + CIP + SAM + VA + S + TE + CF, SAM + C + VA, VA + S + TE, and VA + S in 9 E. coli strains, also, AMC + SAM + CF, SAM + CF, and VA + CF in 21 Bacillus cereus strains respectively. On the basis of the results above, many seasoned dried fishes had multiple drugs resistances, conclusively, we suggest limited guideline and special management on use of antibiotics in floating net cages of fishery farms.

  • The electromyographic activity of four muscles(biceps, supraspinatus, infraspinatus and subscapularis) was mea­sured from non-dominant shoulders of 12 volunteers by six different provocative test for the biceps pathology. The provocative tests were Speed, Yergason, Ludington, Heuter, O'Brien and the abduction-extension test. Each test was performed in a force of 30% of maximal voluntary contraction. The levels of activity of the biceps were higher than those of the other rotator cuff muscles only in Speed's test: 28% in the biceps, 26% in the infraspinatus, 25% in the supraspinatus and 21 % in the subscapularis. The levels of activity of the biceps as a percent of MMT(maximal manual test) were higher in Speed's(42%) and O'Brien's test with the arm supinatecl(42%). Speed's test can isolate the activity of biceps better than the other tests but it is a nonspecific test by which the biceps tendon was also activated within other rotator cuff muscles.

Antibiotic Resistance Patterns of Enterococcus spp. Isolated From Commercial Frozen Foods (시판 냉동식품에서 분리한 장구균의 항생제 내성 양상)

  • From April to December in 2009, microbial investigation is accomplished for 100 frozen foods asked to microbial control team that corresponds with total aerobic viable bacteria, coliform group, Escherichia coli, Enterococcus spp. and antibiotic resistance patterns of Enterococcus spp. isolates are investigated. Average of total erobic viable bacteria numbers is $4.3{\times}10^4CFU/g$. Average of coliform group numbers is $4.3{\times}10^3CFU/g$. Average f Enterococcus spp. numbers is $1.8{\times}10^3CFU/g$. Escherichia coli from 100 frozen foods is not detected and detection ate is 0.0%. 22 Enterococcus spp. are isolated from 100 frozen foods. 12 of 22 Enterococcus spp. strains are identified as E. faecium. 7 of 22 Enterococcus spp. strains are identified as E. faecalis. 2 of 22 Enterococcus spp. trains are identified as E. gallinarum. 1 of 22 Enterococcus spp. strains is identified as E. hirae. Enterococcus spp. solates show a high resistance to erythromycin, rifampin, tetracycline, ciprofloxacin, chlorampenicol, penicillin and susceptibility to vancomycin, ampicillin, gentamicin, strepomycin, linezolid. 15 of 22 Enterococcus spp. strains are multi-resistant and the most frequent multi-resistant pattern is erythromycin-rifampin for 6 Enterococcus spp. strains.

Analysis on Research Trends and Factor of Success to Communities of Practice in Korea (국내의 실천공동체(Communities of Practice)연구 동향 분석 및 성공 요인 분석)

  • When communities of practice(CoP) was mostly informal, has been formulated. Because Knowledge management(KM) was emphasized in the business sector. So it increase interest in the COP. Since the early 2000s, the research interest has thus far grown. At a point in time has the subject, CoP, been researched for ten years, it is significant to review the literatures, the main topics as well as critical factors on the subject. In this study, the trends in the relevant researches and the performance factors from the previous studies are examined. The references for this study are limited to the authorized scholarly and scientific journals. The results therethrough processed show that total 37 researches were conducted between the year 2000 and 2012. These could be classified by the research methods as 16 case studies, 11 studies using CoP as the dependent variable, 4 studies using CoP as an independent variable and 3 theoretical discourses. 7 out of 16 case studies suggest the 14 success factors of CoP; the most influential performance factor is the external community support. 11 studies examined factors influencing the performance of CoP; 13 performance factors were proposed in these studies. The most significant factor influencing the performance of CoP is the team effectiveness.

Comparison of the Real-Time Nucleic Acid Sequence-Based Amplification (NASBA) Assay, Reverse Transcription-PCR (RT-PCR) and Virus Isolation for the Detection of Enterovirus RNA. (엔테로바이러스 검출을 위한 real-time nucleic acid sequence-based amplification (NASBA), reverse transcription-PCR (RT-PCR) 및 바이러스 배양법의 비교)

  • Rapid detection of enterovirus (EVs) is important in the management of aseptic meningitis. We examined the relative efficiency and specificity of the real-time nucleic acid sequence-based amplification (NASBA) comparing with the established reverse transcription polymerase chain reaction (RT-PCR) and viral culture method which were used for the detection of enterovirus RNA in clinical specimens. Of the total 292 samples, 145 were found to be positive to enterovirus RNA by real-time NASBA, 101 were positive by viral culture, and 86 were positive by RT-PCR. 147 samples and 46 samples were determined to be negative and positive by all methods respectively, but 4 samples were positive only by real-time NASBA. To compare the specificity of each method, various clinical samples which were diagnosed for herpes simplex virus (HSV)-1, HSV-2, adenovirus, mumps, and rhinovirus were applied. Except one rhinovirus sample which was false positive to enterovirus RNA by RT-PCR, the other different samples were negative to all three methods. The real-time NASBA procedure can be completed within 5 hours in contrast with 9 hours for the RT-PCR and 3-14 days for the viral culture. From this study, it was suggested that the real-time NASBA assay could be a standardized, rapid, specific, and sensitive procedure for the detection of enterovirus RNA.

Nutritional Components of Cow Feet Jokpyun and Jokpyun Added Chicken and Codfish (소족편 및 소족에 닭과 건대구를 첨가한 족편의 영양성분 - 연구노트 -)

  • This study investigated the nutritional components of jokpyun. Jokpyuns made with cow feet and two kinds of jokpyun were added 30% of chicken and dry codfish to 70% of cow feet, respectively. There was no difference in moisture contents among three jokpyuns. Crude protein content was higher in jokpyun made with cow feet than in those added chicken and dry codfish. Contents of lipid and ash in jokpyuns added chicken and dry codfish were higher than those of jokpyun made with cow feet. The ratio of unsaturated fatty acid in jokpyun added dry codfish was the highest, but, that of linoleic acid was the highest in jokpyun added chicken than the others. Amino acid contents were higher in the order of glycine, proline, alanine, glutamic acid and composition of essential amino acid was similar to soupstock made with beef shank and leg bone. The contents of glycine and alanine in jokpyun added codfish and that of glutamic acid in jokpyun made with cow feet were the highest. Especially, among essential amino acids lysine was the highest in all jokpyuns. Methionine and valine were higher jokpyun added chicken and jokpyun made with cow feet than the others. Calcium and phosphorous were higher in jokpyun added chicken than in others. From these results, jokpyun, especially added chicken, was seemed to be more nutritious than other jokpyuns.

Shelf-life Prediction of ${\gamma}-Irradiated$ Boiled-Dried Anchovies (감마선 조사 건멸치의 저장수명 예측)

  • As a series of studies on the preservation methods for boiled-dried anchovies, determination of sorption properties and shelf-life prediction were made for the samples. Dried anchovies, which were gamma-irradiated at pre-established dose (5 kGy) after packaging in both a polyethylene film (PE, 0.1 mm) and a laminated film $(nylon\;15\;{\mu}m/polyethylene\;100\;{\mu}m,\;NY/PE)$, were subjected to a quality evaluation during 4 months at different storage conditions, such as $15^{\circ}C/68%\;RH,\;25{\circ}C/75%\;RH,\;and\;35^{\circ}C/84%$ RH. The sample showed 5.47% of BET monomolecular layer moisture content and the corresponding water activity, 0.15. The velocity constants of browning reaction and organoleptic changes in the sample were in proportion to storage temperature, and $Q_{10}$, values were ranged from 2.17 to 2.40 in a given packaging and irradiation conditions. In the shelf-life prediction of the stored sample at $25^{\circ}C$, non-irradiated groups packaged in PE and NY/PE were 84 days and 125 days. While 5 kGy-irradiated groups in the same packaging were 126 days and 138 days, respectively. This finding proved the efficacy of laminated-film packaging and irradiation treatment in preserving the quality of dried anchovies.

Monitoring and Safety Assessment of Pesticide Residues on Agricultural Products Sold via Online Websites (온라인 판매 농산물 잔류농약 실태 및 안전성 평가)

  • This study was carried out to monitor the current status of pesticide residues in selling agricultural products via online and assessed their safety in 2014. A total of 124 samples were purchased six times from March to August 2014 twenty online shopping malls randomly. These samples were analysed 208 pesticides by multiresidue method using a GC-ECD/NPD and a LC-MS/MS and confirmed by a GC-MSD. As a result of analysis, residual pesticides samples were 11 (8.9%) such as leek, young radish, welsh onion etc, of which 2 samples (1.6%) such as sesame bud (Chlorothalonil), artemisia (Chlorpyrifos) were violated Korea Maximum Residue limits (MRLs). 11 kinds of pesticides (19 times) were detected in 11 samples. Risk assessment evaluated human health exposure with the ratio of EDI (Estimated daily intake) to ADI (Acceptable daily intake) of pesticides detected. %ADI (the ratios of EDI to ADI) were 0.04~95.70% and some samples represented a fairly dangerous levels. In particular, Chlorothalonil in the sesame bud was shown as a significant risk close to 100% of %ADI. Accordingly, it is recommended to strengthen a safety check on agricultural products in online sales.
