Effect of Fermented Brown Seaweed Waste (FBSW) on in vitro Rumen Microbial Fermentation (발효 미역부산물이 반추위 발효특성에 미치는 영향)

  • This study was conducted to investigate the effects of brown seaweed waste (BSW) fermented with DS-01 microbe on in vitro rumen microbial fermentation. In in vitro trial, three different diets supplemented with 2%, 4%, 6% BSW fermented with DS-01 either for one month or two months was tested at 3 h, 6 h, 9 h, 12 h, and 24 h incubation. The chemical composition (CP, EE, CF, and ash) between brown seaweed waste (BSW) and fermented BSW (FBSW) were not different. The contamination of pathogenic microbes was not detected in FBSW. The pH value tended to be higher with 6% level of supplementation of FBSW for one month than other treatments. The pH at 24 h was significantly higher in FBSW than that of treatments without FBSW (p<0.05). In FBSW for two months, the pH value in 6% FBSW at 3 h in vitro fermentation tended to be higher than 2% or 4% FBSW treatments (p=0.0540), but there were no differences in other fermentation times. Although the concentration of $NH_3$-N of BSW fermented for one month was higher than control at 3 h (p<0.05), the volatile fatty acid values were significantly increased in 4 and 6% FBSW fermented for one month at 6 h incubation (p<0.05). In BSW fermented for two months, the volatile fatty acid values were significantly decreased in 6% treatment at 9 h (p<0.05). As a result of in vitro trial, it was recommended that the 2~4% supplementation level of brown seaweed waste fermented with DS-01 microbe for two months could be utilized for in vivo trial in ruminants.

Relationship Between MC1R and ASIP Genotypes and Basic Coat Colors in Jeju Horses (제주마의 기본모색과 MC1R과 ASIP 유전자형 조합의 상관관계)

  • This study was undertaken to reveal the relationship between genetic variations and the basic coat color classification system in Jeju horses. Genetic variations of the melanocortinreceptor 1 (MC1R) and agouti signaling protein (ASIP) genes were investigated using pyrosequencing technique. A nucleotide substitution mutation for MC1R g.901C>T and an ASIP 11-bp deletion mutation were screened. Black horses had MC1R $E^+$/- ($E^+/E^+$ or $E^+/E^e$) and ASIP $A^a/A^a$ genotypes. In contrast, chestnut horse genotypes were MC1R $E^e/E^e$ and ASIP -/-. Thus, black and bay horses have at least one dominant MC1R allele, $E^+$, whereas chestnut horses have homozygous recessive alleles $E^e/E^e$. This suggests that the MC1R genotypes determine chestnut or black/bay coat color, regardless of the genotype distribution of ASIP. In addition, the horses with MC1R $E^+$/- and a dominant ASIP $A^A$/- allele showed bay coat color, but not black, suggesting that the ASIP $A^A$ allele represses black coat color development in the hairs of the body, but not in the mane and all four legs. Pedigree analysis showed a consistent relationship between the genotype distribution of the MC1R and ASIP genes and basic coat color patterns, even in the $F_1$ progeny. The results of this study revealed the relationship between the coat color phenotype and genetic background and suggested that useful information may be provided for molecular breeding of Jeju horses.

Comparison of Breeding Value by Establishment of Genomic Relationship Matrix in Pure Landrace Population (유전체 관계행렬 구성에 따른 Landrace 순종돈의 육종가 비교)

  • Genomic relationship matrix (GRM) was constructed using whole genome SNP markers of swine and genomic breeding value was estimated by substitution of the numerator relationship matrix (NRM) based on pedigree information to GRM. Genotypes of 40,706 SNP markers from 448 pure Landrace pigs were used in this study and five kinds of GRM construction methods, G05, GMF, GOF, $GOF^*$ and GN, were compared with each other and with NRM. Coefficients of GOF considering each of observed allele frequencies showed the lowest deviation with coefficients of NRM and as coefficients of GMF considering the average minor allele frequency showed huge deviation from coefficients of NRM, movement of mean was expected by methods of allele frequency consideration. All GRM construction methods, except for $GOF^*$, showed normally distributed Mendelian sampling. As the result of breeding value (BV) estimation for days to 90 kg (D90KG) and average back-fat thickness (ABF) using NRM and GRM, correlation between BV of NRM and GRM was the highest by GOF and as genetic variance was overestimated by $GOF^*$, it was confirmed that scale of GRM is closely related with estimation of genetic variance. With the same amount of phenotype information, accuracy of BV based on genomic information was higher than BV based on pedigree information and these symptoms were more obvious for ABF then D90KG. Genetic evaluation of animal using relationship matrix by genomic information could be useful when there is lack of phenotype or relationship and prediction of BV for young animals without phenotype.

Comparing Farming Methods in Pollutant runoff loads from Paddy Fields using the CREAMS-PADDY Model (영농방법에 따른 논에서의 배출부하량 모의)

  • BACKGROUND: For Non-Point Source(NPS) loads reduction, pollutant loads need to be quantified for major farming methods. The objective of this study was to evaluate impacts of farming methods on NPS pollutant loads from a paddy rice field during the growing season. METHODS AND RESULTS: The height of drainage outlet, amount of fertilizer, irrigation water quality were considered as farming factors for scenarios development. The control was derived from conventional farming methods and four different scenarios were developed based combination of farming factors. A field scale model, CREAMS-PADDY(Chemicals, Runoff, and Erosion from Agricultural Management Systems for PADDY), was used to calculate pollutant nutrient loads. The data collected from an experimental plot located downstream of the Idong reservoir were used for model calibration and validation. The simulation results agreed well with observed values during the calibration and validation periods. The calibrated model was used to evaluate farming scenarios in terms of NPS loads. Pollutant loads for T-N, T-P were reduced by 5~62%, 8~37% with increasing the height of drainage outlet from 100 mm of 100 mm, respectively. When amount of fertilizer was changed from standard to conventional, T-N, T-P pollutant loads were reduced by 0~22%, 0~24%. Irrigation water quality below water criteria IV of reservoir increased T-N of 9~65%, T-P of 9~47% in comparison with conventional. CONCLUSION(S): The results indicated that applying increased the height of drainage after midsummer drainage, standard fertilization level during non-rainy seasons, irrigation water quality below water criteria IV of reservoir were effective farming methods to reduce NPS pollutant loads from paddy in Korea.

Fish Fauna of the Saemankum Area in the West Coast of Chollabuk-do, Korea (새만금 일대의 어류상)

  • Fish fauna of fishes collected from the Saemankum area in the west coast of Chollabuk-do was studied from October 1996 to April 1999. The specimens collected from the five sites in this area were identified into 107 species belonging to 83 genera and 52 families. Of them the largest group of fishes was order Perciformes, comprising 50 species and 21 families, and showed 46.7% in total numbers of species. Next larger groups were orders Scorpaeniformes and Pleuronectiformes containing 16 and 14 species respectively. In this area, the group comprising a lots of species was the family Gobiidae taking 13 species, and next groups were the families of Sciaenidae (6 species), Engraulidae (5 species) and Pleulonectidae (5 species). The dominant species was Engraulis japonicus which is 23.4 % in total indiyidual numbers, and subdominant species was Thryssa kummazensis which is 18.5%. The important species economically in the Saemankum area were Anguilla japonica, E. japonicus, T. kummalensis, Konosirus punctatus, Sardinella zunasi, Mugil cephalus, Argyrosomus argentatus, Pseudosciaena polyactis, Collichthys lucidus, Paralichthys olivuceus and Cynoglossus joyneri. The species of high frequency in this area from February to June every year were Anguilla japonica, E. japonicus, T. kammalensis, K. punctatus, S. zunasi, Coilia nasus, Syngnatuus schlegeli, P. polyactis, C. lucidus and Pholis fangi. Ophichthus rotundus, Repomucenus leucopoecitus and R. koreanus were Korean endemic species in the west coast of Chollabuk-do. On the other hand, Boleophthalmus pectinirostris was not found during this study, and Periophthalmus modertus, Tridentiger barbatus and Lophogobius ocellioauda sharply decreased in individual numbers in comparison to several years ago.

Optical Characteristic on the Growth of Centric Diatom, Skeletonema costatum (Grev.) Cleve Isolated from Jinhae Bay in Korea (진해만에서 분리한 중심목 규조류 Skeletonema costatum(Grev.) Cleve의 성장에 미치는 광학적 특성)

  • The effects of light quality and irradiance on the growth of centric diatom, Skeletonema costatum (Jinhae Bay strain) were investigated in the laboratory. At 20$^{\circ}C$ and 30 psu, the irradiance-growth curve showed the maximum growth rate of 1.17 day$^{-1}$ with half-saturation photon flux density (PFD) (K$_s$) of 92.4 $\mu$mol photons $m^{-2}s^{-1}$, $\mu$=1.17 (I-5.28)/(I+81.8), (r=0.98), and a compensation PFD (I$_0$) was 5.28 $\mu$mol photons $m^{-2}s^{-1}$. The 10 equated to a depth of 3$\sim$5 m from March to May, 11 m in June and 4 m from July to September in Jinhae Bay. These responses suggested that irradiance at the depth near the surface layer in Jinhae Bay would provide favorable conditions for S. costatum. To assess the effects of light (i.e. wavelengths) on the growth, nine wave-lengths were used ranging from near ultraviolet to near-infrared supplied by light emitting diode. At an irradiance level of 25 $\mu$mol photons $m^{-2}s^{-1}$, S. costatum grew under wavelengths of 405, 470, 505, 525, 568 and 644 nm, but did not grow at 590 and 623 nm; whereas S. costatum grew at all wavelengths at 100 $\mu$mol photons $m^{-2}s^{-1}$. This implies that S. costatum is likely to grow well in enclosed water bodies where suspended particles absorbs most of the blue wavelengths, and dominated by yellow-orange wavelengths.

Comparison of Nuclear Status and Developmental Potential between Polar Body Extruded Oocytes and Non-extruded Oocytes on in vitro Maturation and Development of Porcine Follicular Oocytes (돼지 난모세포의 체외 성숙 후 극체 방출 및 미방출란의 핵형과 배발달율)

  • The objective of this study was carried out to examine the polar body extrusion of in vitro matured porcine follicular oocytes as a non-invasive marker of oocyte quality to know the developmental competence in advance. The porcine oocytes matured for 48 hours were examined the polar body extrusion and some parts were stained. The examined oocytes were matured for additional $16{\sim}18$ hours and activated with 7% ethanol and cultured in $5{\mu}g/ml$ cytochalasin B for 5 hours for diploid formation. The treated oocytes were washed and cultured for 7 days. The polar body extrusion and degeneration rates were varied with $9.9{\sim}52.4%$ and $21.4{\sim}61.8%$ by repetition. The polar body extruded oocytes were shown the polar body chromosome and metaphase II plate by staining. However the non-extruded oocytes were shown expanded nucleus with 39.1%, premature chromosome condensation with 19.6%, metaphase I plate with 10.9 %, metaphase II with 13%, condensed chromatin with 6.5%, and absent nuclear material with 8.7%. The oocytes that were not examined for the polar body extrusion were cleaved 45.0%, and developed to blastocyst stage with 11.3%. In examined oocytes for polar body extrusion,. the polar body extruded oocytes were cleaved 94.2% and developed with 42.5%. This result suggests that discarding of the degenerating oocytes and oocytes that not extruded polar body will be effective for experiments of culturing effect in porcine embryos and embryo biotechnology.

Effect of Mower Conditioner at Different Harvest Stage on the Field Drying Rate and Quality of Rye Hay (수확시기별 Mower Conditioner 처리에 의한 속성 양질 호밀 건초조제 효과)

  • A field experiment was carried out to determine the effects of chemical /mechanical treatments at mowing on the field drying rate and hay quality of rye (Secale cereale L.). The chemical drying agent/mower conditioner ($K_2CO_3$ 2%, conditioning, $K_2CO_3$ 2% + conditioning and control) were treated at different harvest stage (late boot, heading and bloom stage) for hastening hay-making in the spring of 1996. After field dry, square bales were made by hay baler, and the dry matter(DM) loss, visual estimation and nutritive value of rye hay were evaluated after storing two months. The field drying rate of rye was higher with delayed stage of harvest, and mechanical and chemical + mechanical treatment, but the effectiveness of chemical alone was very low. With mower conditioning, the duration of field dry was shortened by 1.5 to 2 days compared with control. The DM loss of rye hay was reduced by late harvest and mechanical, and chemical + mechanical combined treatment, but the efficiency by chemical alone was very low. The visual score (Ieafiness, green color, odor and softness) of hay after storing was high in mechanical and chemical + mechanical, but the score by chemical alone was very low. The nutritive value (ADF, NDF, digestibility, and relative feed value) of hay was also high with treatment of mechanical and chemical + mechanical, but the quality by chemical alone was similar compared with control. The quality of hay was very low when harvested at bloom stage. In conclusion, mower conditioning can enhance the field drying rate of rye, but the drying effectiveness of chemical drying agent was very low. The effect of chemical/mechanical combined treatment was very similar when compared with mechanical alone. Harvest at early heading to heading stage was recommended for high quality rye hay.

  • The effect of Artemisia argyi H. under liquid-state fermentation by Monascus purpureus (AAFM) on cognitive impairments has been studied in a mice model of diabetes-associated cognitive decline induced by streptozotocin (STZ). C57BL/6 mice (9 weeks of age, male) were separated into four groups: a normal control, STZ-induced diabetic mouse group (STZ group), Artemisia argyi H. (AA) 10 group (diabetic mouse+AA 10 mg/kg/day), AAFM 10 group (diabetic mouse+AAFM 10 mg/kg/day). Administration of AA and AAFM significantly improved glucose tolerance, as shown by the intraperitoneal glucose tolerance test (IPGTT), and ameliorated cognitive deficit, as shown by the behavioral tests including passive avoidance, Morris water maze, and Y-maze tests. After behavioral tests, the cholinergic system was examined by assessment of the acetylcholine (ACh) level and acetylcholinesterase (AChE) inhibitory activity, and the antioxidant system was also assessed by measuring malondialdehyde (MDA) and superoxide dismutase (SOD) levels in the brain and liver.

Stream Ecosystem Assessments, based on a Biological Multimetric Parameter Model and Water Chemistry Analysis (생물학적 다변수 모델 적용 및 수화학 분석에 의거한 갑천생태계 평가)

  • This research was to apply a multi-metric approach, so called the Index of Biological Integrity (IBI) as a tool for biological evaluations of water environments, to a wadable stream. For the study, we surveyed 5 sampling locations in Kap Stream during August 2004 ${\sim}$ September 2005. We also compared the biological data with long-term water quality data, obtained from the Ministry of Environment, Korea and physical habitat conditions based on the Quantitative Habitat Evaluation Index (QHEI). We used ten metric systems for the IBI model to evaluate biological stream health. Overall IBI values in Kap Stream averaged 24 (range: 20${\sim}$30, n=5), indicating a "fair ${\sim}$ poor" conditions according to the modified criteria of Karr (1981) and US EPA(1993). Exclusive of 4th survey, average IBI values at the upstream reach (S1 ${\sim}$ S3)and downstream reach (S4 ${\sim}$ S5) were 20 and 24, respectively. However, in 4th survey the averages were 21 and 20 in the upstream and downstream reaches, respectively. This difference was larger in the upstream than in the downstream because of physical condition disturbed during summer monsoon. Values of the QHEI varied from 75(fair condition) to 148 (good condition) and values of QHEI in the S3 were significantly (P=0.001, n=5) lower than other sites. Biochemical oxygen demand (BOD), chemical oxygen demand (COD), total nitrogen (TN) and total phosphorus (TP) were greater by 3 ${\sim}$ 8 fold in the downstream than in the upstream reach. We believe that present IBI approach applied in this study may be used as a key tool to set up specific goals for restoration of Kap Stream.
