Effect of Feeding High Carbohydrate-Low Fat Fermented Feed on the Meat Quality Characteristics in Finishing Pigs (저탄수화물 고지방 발효사료 급여가 돈육의 육질 특성에 미치는 영향)

  • A total of ninety swine ($79.0{\pm}2.2\;kg$) were employed for 58 d to determine the meat quality of pigs fed fermented agricultural by-products (FAB) mainly consisting of brewers grain shell. FAB was replaced with commercial feed at dietary levels of 20%, 40%, 60%, 80%, and 100% (T1) and 30%, 60%, 100%, 100%, and 100% (T2) at 1, 2, 3, 4 and 5-9 wk, respectively. Compared with the control (CON) feed, FAD feed had lower moisture and nitrogen-free extract content, and higher crude fat, crude fiber, and total calorie (p<0.05). The protein content, amino acid profile, and pH values of pork loin were not affected by dietary treatment. However, higher moisture, crude ash, and meat cholesterol, and lower fat, were found in CON compared with treatment (p<0.05). FAB treatment significantly improved drip loss and cooking loss value (p<0.05), and increased the CIE $L^*$ values of loin and back-fat surface, whereas it decreased the CIE $a^*$ values of loin surface (pp<0.05). The results indicate that dietary FAB affected meat cholesterol and fat content, and improved drip loss and cooking loss, but had no affect on amino acid composition.

Identification of Mycobacterium Tuberculosis in Pleural Effusion by Polymerase Chain Reaction (PCR) (흉막삼출액에서 Polymerase Chain Reaction (PCR)을 이용한 결핵균의 검출에 관한 연구)

  • Background: By amplifying small amount of DNA, polymerase chain reaction (PCR) can be used for the detection of very small amount of microbial agent, and may be especially useful in certain cases which are difficult to be diagnosed microbiologically or serologically. Tuberculous pleurisy is a disease that can be diagnosed in only 70% of cases by conventional diagnostic tools, and PCR would be a very rapid, easy, and sensitive diagnostic method. Method: The specificity and sensitivity of PCR to detect Mycobacterium tuberculosis DNA were evaluated using various strains of Mycobacteria. To evaluate the diagnostic usefulness of PCR in tuberculous pleurisy, we used PCR to detect Mycobacterium tuberculosis DNA in pleural fluid. The amplification target was 123 base pair DNA, a part of IS6110 fragment, 10~16 copies of which are known to exist per genome. The diagnostic yield of PCR was compared with conventional methods, including pleural fluid adenosine deaminase (ADA) activity. Also, the significance of PCR in undiagnosed pleural effusion was evaluated prospectively with antituberculosis treatment. Results: 1) Using cultured Mycobacterium tuberculosis and other strains, PCR could detect upto 1 fg DNA and specific for only Mycobacterium tuberculosis and Mycobacterium bovis. 2) Using pleural effusions of proven tuberculosis cases, the sensitivity of PCR was 80.0% (16/20), and the specificity 95.0% (19/20). 3) Among 13 undiagnosed, but suspected tuberculous effusion, the positive rate was 60% in 10 improved cases after antituberculosis medications, and 0% in 3 cases of proven malignancy later. 4) Adenosine deaminase level of proven and clinically diagnosed tuberculous pleurisy patients was significantly higher than that of excluded patients, and correlated well with PCR results. Conclusion: We can conclude that PCR detection of Mycobacterium tuberculosis in pleural effusion has acceptable sensitivity and specificity, and could be an additional diagnostic tool for the diagnosis of tuberculous pleurisy.

A STUDY ON THE BONDING OF COMPOMER TO DECIDUOUS DENTIN (컴포머와 유치 상아질의 결합에 관한 연구)

  • The purpose of this study was to evaluate the bonding of compomer to deciduous dentin which is known to have been developed to improve the weak properties of glass ionomer cement and composite resin. 120 sound primary molars were used for the shear bond strength test and another 24 for the scanning electron microscopic evaluation. Each material was ailed into polyethylene mold attached to exposed dentinal surface($3{\times}4mm$ in diameter) of sample blocks. Shearbond strength was measured using Universal testing machine and data were analyzed statistically with Oneway-ANOVA and Scheffe test. Scanning electron microscopic observation was performed in order to evaluate the pattern of distribution and penetration of resin tags and hybrid layer. Compomer groups(II-V) showed significantly higher bond strength values than glass ionomer group(I)(p<.05). Etching-compomer groups(III, V) showed the significantly higher bond strength than non-etching compomer groups(II, IV)(p<.05), but slightly lower values than composite resin group(VI) with no statistically significant difference(p>.05). No significantly different bond strength was found between compomer groups of different bonding system(p>.05). Scanning electron micrographs showed more irregular distribution of short and thin resin tags in non-etching compomer groups(II, IV) whereas the more regular and intimate distribution of long and thick tags in etching compomer groups(III, V) and composite resin group(VI). The evaluation of hybrid layer also showed more regular formation of thicker layer in etching compomer groups(III, V). Based on the results of present study, the use of compomer as an esthetic restorative material for primary molars might be justified.

Clinical Trial of Nasal Flumazenil Administration (플루마제닐의 경비 투여)

  • Flumazenil is a competitive antagonist of benzodiazepines. It is usually administered intravenously. However, if the intravenous route is not available then other routes of drug administration should be considered. This study was designed to evaluate the reversal effects of flumazenil after nasal administration. Twenty-five young, healthy adult volunteers participated in this clinical trial. The dosage of 0.08mg/kg midazolam was administered intravenously to induce deep sedation. Ten minutes after midazolam administration, 0.5mg of flumazenil was dropped nasally, over a period of one minute. Blood samples were taken to measure the concentration of midazolam and flumazenil at 0, 5, 10, and 20min after nasal administration of flumazenil, using High Performance Liquid Chromatography. The degree of sedation was evaluated with sedation score and bispectral index (BIS), Statistical analysis was performed by multivariate ANOVA and correlation analysis (P<0.05). Peak serum flumazenil concentration was reached in 10min. Sedation score decreased after midazolam administration and showed a significant increase after flumazenil administration. However, BIS decreased during the first 10min after midazolam administration and then no significant changes after flumazenil administration. There were two instances representing rapid and complete reversal of midazolam after intranasal administration of flumazenil. In conclusion, intranasal flumazenil administration may be effective in some patients when intravenous route is not available in condition of benzodiazepine overdose.

Development of Quality Assurance Program for the On-board Imager Isocenter Accuracy with Gantry Rotation (갠트리 회전에 의한 온-보드 영상장치 회전중심점의 정도관리 프로그램 개발)

  • Positional accuracy of the on-board imager (OBI) isocenter with gantry rotation was presented in this paper. Three different type of automatic evaluation methods of discrepancies between therapeutic and OBI isocenter using digital image processing techniques as well as a procedure stated in the customer acceptance procedure (CAP) were applied to check OBI isocenter migration trends. Two kinds of kV x-ray image set obtained at OBI source angle of $0^{\circ},\;90^{\circ},\;180^{\circ},\;270^{\circ}$ and every $10^{\circ}$ and raw projection data for cone-beam CT reconstruction were used for each evaluation method. Efficiencies of the methods were also estimated. If a user needs to obtain an isocenter variation map with full gantry rotation, a method taking OBI image for every $10^{\circ}$ and fitting with 5th order polynomial was appropriate. However for a mere quality assurance (QA) purpose of OBI isocenter accuracy, it was adequate to use only four OBI Images taken at the OBI source angle of $0^{\circ},\;90^{\circ},\;180^{\circ}\;and\;270^{\circ}$. Maximal discrepancy was 0.44 mm which was observed between the OBI source angle of $90^{\circ}\;and\;180^{\circ}$ OBI isocenter accuracy was maintained below 0.5 mm for a year. Proposed QA program may be helpful to Implement a reasonable routine QA of the OBI isocenter accuracy without great efforts.

Effects of Woori Black Pig Sire on Growth Performance, Body Shape, and Retail Cut Yield of Crossbred Pigs (우리흑돈 종모돈이 삼원교잡종 돼지의 성장, 체형 및 부분육 생산수율에 미치는 영향)

  • To improve the relatively low economic efficiency of the Korean native pig, the Korean National Institute of Animal Science developed a novel pig breed, the Woori black pig (W), by crossing Korean native and Duroc (D) pigs. This study was conducted to evaluate the effects of W as a terminal sire on growth performance, body shape, and retail cut yield of crossbred pigs. By using a completely randomized design, 32 crossbred pigs were allotted to one of two treatment groups based on terminal sire. The two groups were LYD [(Landrace × Yorkshire) × D sire] and LYW [(Landrace × Yorkshire) × W sire]. The experimental assessments were conducted over 53 days. The terminal sire breed had no significant effect on body weight (BW) at d 53, or on BW gain, average daily gain, or days to reach a 90 kg body weight. Moreover, there were no significant differences in body length, body height, or chest depth between the two groups. However, there was a significant difference (p < 0.05) in backfat thickness between the LYD (17.29 mm) and LYW (18.96 mm) groups. Loin yield of crossbred pigs in the LYW group (13.11%) was significantly lower (p < 0.05) than that in the LYD group (13.85%). By contrast, the Boston butt yield was significantly higher (p < 0.05) in the LYW group (8.99%) than in the LYD group (8.21%). In conclusion, these results suggest crossbred pigs sired by a Woori black pig had growth performance, shape, and retail cut yield (except loin yield) Ed. Note: I assume the lower loin yield is a negative factor so I included this wording. similar to those sired by a Duroc pig. The results showed no overall negative effect Ed.   • For the development of low temperature anode-supported planar solid oxide fuel cell, the planar anode supports with the thickness of 0.8 to 1 mm and the area of 25, 100 and $150\;cm^2$ were fabricated by the tape casting method. The strength, porosity, gas permeability and electrical conductivity of the planar anode support were measured. The porosity of anode supports sintered at $1400^{\circ}C$ and then reduced in$H_2$ atmosphere was increased from $45.8\%\;to\;53.9\%$. The electrical conductivity of the anode support was $900 S/cm\;at\; 850^{\circ}C$ and its gas permeability was 6l/min at 1 atm in air atmosphere. The electrolyte layer and cathode layer were fabricated by slurry dip coating method and then had examined the thickness of $10{\mu}m$ and the gas permeability of 2.5 ml/min at 3 atm in air atmosphere. As preliminary experiment, cathode multi-layered structure consists of LSM-YSZ/LSM/LSCF. At single cell test using the electrolyte layer with thickness of 20 to $30{\mu}m$, we achieved $300\;mA/cm^2$ and 0.6V at $750^{\circ}C$

Comparisons of Growth, Heading and Grain Filling Characteristics between Wet-hill-seeding and Transplanting in Rice (벼 무논점파재배와 이앙재배의 생육, 출수 및 등숙특성 비교)

  • Direct seeding of rice is a time- and laborsaving method, compared to machine transplanting. However, the area planted to direct-seeded rice is decreasing because of instability of seedling establishment, lodging, occurrence of weedy rice, and deterioration of grain quality. A newly developed wet-hill-seeding of rice, a type of direct seeding method, has shown features of growth similar to machine transplanted rice. This study was carried out to compare tillering pattern, heading characteristics and grain filling traits between transplanting and wet-hill-seeding in rice. Tillering of wet-hill-seeded rice was more vigorous than that of transplanted rice showing maximum tiller number 30~100% higher than transplanted rice. Wet-hill seeded rice showed later heading and shorter heading duration than transplanted rice. To find out the relationship between heading characteristics and grain filling traits, every panicle was tagged for flowering date, and the panicles on each heading date were inspected for grain filling traits. Wet-hill seeded rice and transplanted rice exhibited no significant difference in culm length, panicle length, the percentage of grain filling and perfect brown rice on each heading date during total heading period. Therefore, we conclude that wet-hill-seeding method is not inferior to machine transplanting in terms of seedling establishment, growth, grain filling and head rice yield.

Effects of Fermented Feedstuff with Wet Brewer’s Grain and Soybean on Fattening Performance and Carcass Grade in hanwoo Steers (맥주박 발효사료 및 대두의 급여가 한우 거세우의 육성성적 및 도체등급에 미치는 영향)

  • This study was conducted to examine the feeding effects of FFWBG (fermented feedstuff with wet brewer’s grain) and GS(grinding soybean) in Hanwoo steers. For this research, total twenty steers(average body weight : 455.7$\pm$25.9kg) were grouped into control(formula feed), T1(formula feed:FFWBG=60:40), T2(formula feed:GS=80:20), T3(formula feed:FFWBG:GS=50:40:10), and T4(formula feed: FFWBG:GS=40:40:20), each treatment was allocated to four steers. There was a tendency to be increased average daily gain in T1, T2, T3 and T4 compared with the control even though no statistical significance was found. The feed conversion of T1, T2, T3 and T4 were beneficially better than that of control(P<0.05; 8.25, 9.44, 6.69 and 7.71 vs 11.90, respectively). The blood urea nitrogen concentration of T4 was higher than that of control(P<0.05), but the creatinine concentration of T4 was lower than that of control(P<0.05). The glucose concentration of T1, T2, T3 and T4 were higher than that of control(P<0.05; 59.39, 62.44, 62.17 and 54.00 vs 51.00mg/$d\ell$, respectively). The back-fat thickness of T3 was thicker than that of control, T1, T2 and T4. The rib-eye area of T1, T3 and T4 was wider than that of control. Appearances percentage of ‘A’ ranked meat quantity were 66, 33, 75 and 33% in control, T1, T2 and T4, respectively.

Effects of Polyclonal Antibody Candidate to Adipocytes for Reducing Body Fat on Body Weight, Fecal Digestibility and Blood Metabolites in Pigs (체지방 감소 다클론 후보 항체가 비거세돈의 체중, 분 소화율 및 혈액 대사물질에 미치는 영향)

  • Twelve pigs were used to investigate the effects of polyclonal antibody candidate against abdominal (AAb) and subcutaneous adipocyte membrane proteins (SAb) on body weight, fecaldigestibility and blood metabolites. When AAb and SAb developed by Choi et al. (2010) were injected to pigs, the numerical increase in BW (body weight) occurred at 4 weeks post-treatment, but BW for an entire period was also increased, indicating that the BW increase may not be affected by the antibodies injection. Antibodies treatment did not affect (P>0.05) fecal digestibility of dry matter, crude protein, crude fat and crude fiber. Fecal digestibility of crude ash for control (no treatment) at 2 weeks decreased, and that for non-immunized serum treatmentgroup at 4 weeks post-treatment increased, respectively (P<0.05). However, fecal digestibility of crude ash for AAb and SAb groups did not significantly change. At 4 weeks after the antibodies treatment, blood urea N concentration for AAb and SAb groups was significantly increased (P<0.05). However, these increases may not be caused by the antibodies treatment because similar pattern in blood urea N concentration occurred before the antibodies treatment. Antibodies treatment did not affect concentration of plasma glucose and triglycerides (P<0.05). Compared with control, concentration of plasma total cholesterol for AAb and SAb groups at 4 weeks post-treatment was significantly (P<0.05) decreased. This may suggest that body fat reduction possibly occurs. In conclusion, the AAb and the SAb developed by Choi et al. (2010) may have safety in nutritional physiological metabolism in pigs. Further study on in vivo fat reduction of the antibodies against abdominal and subcutaneous adipocytes of pigs should be required for fat-reduced pork production.
