Light-dependent Hydrogen Production in Chlorobium limicola f. thiosulfatophilum NCIB 8327: A Possibility of Regulation via Glutamine Synthetase (Chlorobium limicola f. thiosulfatophilum NCIB 8327에서의 광수소발생 조절 기작에 대하여)

  • Chlorobium liimicola f. thiosulfatophilum NCIB 8327 was grown on modified Pfennig's medium using ammonium chloride. glutamine. glutamate, or dinitrogen gas as nitrogen sources. Except for the case of dinitrogen gas. the extent of gro\\1h was almost the s~me. The specific activity of glutamine synthetase in crude extracts is the highest in the cells which were grown on the medium containing glutamate. hut that of glutamate synthase is uniform for all four nitrogen sources. When the concentration of ammonium ions increases in the reaction mixture. the specific activity of glutamine synthetase in crude extract from the cells grown on glutamate decreases. hut that of glutamate dehydrogenase increases. whereas that of glutamate synthase remains unchanged. When the concentration of methionine sulfoximine increases, the activity of glutamine synthetases decreases rapidly. On the other hand. when the concentration of ammonium ions increases in the reaction mixture gradually. the activity of glutamine synthetase from the cells grown on higher concentration of ammonium ions less decreases. In the presence of light. the activity of glutamine synthetase increases. hut in the dark it decreases gradually. The production of hydrogen in intact cells depends on light. It is inhihited by adding ammonium ions. hut restores immediately hy adding methionine sulfoximine. The produclion of hydrogen in this strain can he mediated by nitrogenase only. and regulated hy glutamine synthetase.

  • A green sulfur bacterium, Chlorobium limicola f. thiosulfatophilum NCIB 8327, was grown in modified Pfennig's medium including glu1amate as a nitrogen source. Glutamine synthetase was isolated through a series of ultracentrifugation. DEAE-Sepharose CL-6B ion exchange chromatography. Sephacryl S-300 gel permeation chromatography, and preparative HPLC. The recovery and purification fold of the enzyme were 2% and 46.3. respectively. The isolated enzyme was homogeneous on UV-Visible spectrum and polyacrylamide gel electrophoretogram. The relative molecular mass of the native enzyme was estimated to be 280,000 by gel permeation chromatography. The enzyme consisted of ten subunits with relative similar molecular mass. 30.000. which was estimated by SDS-polyacrylamide gel electrophoresis. The optimal temperature and pH of the enzyme were $30^{\circ}C$ and 7.0. Km values were 27.9 mM for L-glutamine and 0.92 mM for hydroxylamine-HCr. The enzyme activity was inhibited by alanine. glycine. and tryptophan considerably, but was not affected by asparagine, lysine. leucine. and valine.

  • With rapid developments in technology and communications' many teachers are increasingly exposed to a variety of e-Learning environments that they must develop new competencies and skills to be successful e-Learning teachers. The purpose of this training program, sponsored by HRD Korea (Human Resources Development Services of Korea), is to provide e-Learning teachers with meaningful opportunities for promoting quality e-Learning teaching. This program covers pedagogical issues as well as technical and practical aspects of the e-Learning environments. Before starting the program development the survey and the current e-Learning program assessments were conducted. The training program is divided into three modules as follows: 1) theoretical issues of e-Learning, 2) development of e-Learning contents, and 3) implementation of e-Learning environments. These three modules can be selectively reorganized in response to teacher requirements and demands. ln each module, there are five subtopics that include creative teaching and interaction strategies for promoting the effective e-Learning teaching. ln conclusion, teachers will gain greater understanding of teacher roles in e-Learning instruction, more flexibility in teaching jobs, increased confidence and knowledge to act as e-Learning facilitators through the completion of this training program.

  • Based on the successful use of chatbot technology, this study examined what business values each company is creating. The chatbot service contributes to improving the productivity of the company by helping to answer or respond to the questions of employees inside the company or customers. And in the field of education, Instead of instructor, AI technology responds the questions and feedback of the students to reduce the work of the instructor. In the field of commerce, offline stores provide convenient and new purchasing experiences to customers by providing product purchasing services through artificial intelligence speakers and personalization service. Although chatbot service is creating business value in some business cases, it is still limited to the process of a specific company, and the spread rate is still slowing because the service scope, convenience, and usefulness are not greater than expected. Therefore, some chatbot development service providers is providing an integrated development platform to improve usability, Chatbots have the features and advantages of providing convenience instead of answering human questions. However, there is a disadvantage that the level of communication can be lowered by reducing various human subjective views and giving mainly objective answers. Through this study, we will discuss the characteristics, advantages and disadvantages of chatbot services by comparing them.

  • This paper investigated the plant and raw material of the ready-mixed concrete company which could supply to the second Lotte World on Pusan. the results were summarized as following. Almost plants were mainly using Twin shaft mixer which was 210m$^3$/hr and horizontal type. There was different the number of admixture silos at each plants, and they were separated by types. The mixtures mainly consisted of the ordinary portland cement, fly ash and blast furnace slag. For favorable quality control, each materials had to carry from same factories, and the monitering standard for quality control should be prepared. The coarse aggregates were used with many different producing districts, so they were only used from Y caused by exclusion of quality difference. The crushed, washed and river sands were generally used as fine aggregates, so the fine aggregates which could be possible to supply stable quality were chosen. This study used Poly Carbonic Acid Admixture which was developed to satisfy maintenance of performance till 2 hours and 10MPa at 15 hours.

Study on the Reality of the Private Educational Institute Street and the Spatial Range of its Service in Pyeongchon, Anyang-si (안양시 평촌 학원가의 교육 서비스 실태 및 공간 범위에 관한 연구)

  • The main purpose of this paper is to identify the characteristics of the private educational institute street in Pyengchon, Anyang-si by analyzing the locational determinants of private education institute, the spatial range of it educational service and the spatial behaviour of private education institute students. The research has concerned with the locational determinants of private educational institutes in Pyengchon. Its main locational determinants are a reputation as a region of private educational institution street, access to large residential areas and rents. Also, it has considered the spatial service range of private educational institutes. To this end, it divided private educational institutes into three hierarchies from first sized to third sized private educational institute. The educational service of first sized institutes delivers to within a 10km radius of them, including Suwon. In particular, the service of them is concentrated on within a 6km radius of them. The educational service of second and third sized institutes both delivers to within a 6km radius of them. The service of second sized one is concentrated on within a 2km radius of them, and third sized one is within a 1km radius of them. What is more, it has analyzed the spatial behaviour of private educational institute students.

  • With the export of commercial nuclear power plants to UAE and that of a research reactor to Jordan, as well as the additional construction of domestic nuclear power plants, the demand of nuclear manpower is expected to increase sharply. Accordingly, nuclear manpower development is recently becoming an important issue. Major institutes involved in nuclear programs are well equipped with education and training procedures and resources. However, small and medium sized businesses have difficulties to educate their employees due to their limited resources and capacity for the education. Addressing the difficulties, this study is intended to develop and education course in accordance with the "Systematic Approach to Training (SAT)". For this, a survey is conducted on the need of education in small and medium sized businesses, based on which a pilot course on the basics of radiation is developed and operated. An assessment on the development and operation using a survey regarding participants response has shown high grades of performance, i.e. above 4.0 points (full mark: 5.0 points) on each level of expectancy, satisfaction and lecturers' capacity. The experience from this study will be used to develop other programs of nuclear power and ASME code, which are also identified from the need analysis.

A Qualitative Case Study of Science Core School Curriculum Management (과학중점학교 교육과정 운영에 관한 질적 사례 연구)

  • The Ministry of Education selected and implemented 'science core school' since 2009 as a policy to strengthen science education to produce talented science students. This study judged that it is necessary to examine the current management practice and diagnose problems to propose improvement measures for more successful management of science core school in the future. To this end, we interviewed and observed teachers and students at a high school specialized in science based in Gyeonggi province which was selected as a science core school, as qualitative study methods such as interview and observation to survey and analyze the current management practice of the school. The management outcome was that the school contributed to fostering talented students in natural sciences and engineering because more varied activities were implemented at the school to develop scientific knowledge of students including experiment, excursion, and circle activity. Identified problems were increased amount of private education due to intense competition over school achievement, negligence of extracurricular activities, burdensome workload for teachers of specific subjects, and lack of expertise of math and science teachers. In conclusion, the following improvement measures are suggested for sustainable management of science core schools: greater liberty should be granted to science core schools; more training opportunities should be given to teachers; college admission program should be improved for science core school students; and it is necessary to introduce courses taught by external teachers, and provide systematic support such as increasing administration staff.

The Present Situation and Expansion of the University Museum Public Education Program (대학박물관 사회교육프로그램 운영의 현황과 발전방향)

  • This paper is mainly deals and suggests about a brief history, an expansion and management of the pubic education program which is running by the university museum after careful analysis of the past and current public education program. There are 20 university museums which operate public education program inducing Seoul National University, Ehwa Women's University, Chungbuk National University and Yeungnam University, etc. Yeungnam University museum has been well running the public education program since l990. The public education program named museum university, museum class, culture class, that have been constantly changing and diversify the theme and education method. After careful analysis of the curriculum, it consists of the fields of archaeology, art history, folklore, anthropology, and modern arts. The programs are being held in the classroom and taking field trips to the related sites. Most of the lectures are given by the university professors and students are mainly mid-aged female. I found that the self evaluation and participant evaluation are insufficient for now. Education on demand program development is needed and also it is mandatory set up the curriculum opening committee. A good way to make the public education program is that to establish the sub-cormmitte talk group under KAUM and I feel keenly the necessity of the constant research on the public education program systematically. I guess it is worth to open a program for youth and weekend program for family.

A Study on Satisfaction of Supplementary Education for Radiological Technologist (방사선사 보수교육에 대한 만족도 연구)

  • As the research about supplementary education of radiological technologist who works in Gyoungsangnam province, this study was conducted to draw the improvements by analyzing the satisfaction level and problems of the supplementary education. During November 01, 2016 to December 15, 2016, after we sent out a total of 250 questionnaires for the survey to radiological technologists working at medical institution located in Gyoungsangnam province, 200 questionnaires suitable for research were analysis by using SPSS 18.0 statistical analysis software. As the satisfaction factors, 21 items such as system, subject, help, appropriateness of lecturer selection, professionalism were used. And as the problem factors, 18 items such as place, transportation, diversity, administrative treatment, education promotion, proceed method were used. Consequentially, the satisfaction level(3.12 point) and problems(3.01 point) of the supplementary education were confirmed as normal level. The supplementary education is an essential education that any health and medical service personnel must complete. In order to improve the effectiveness and efficaciousness of the education, the diverse opinions and efforts to improve the education levels must be supported. Futhermore, through the realistic educational program, the continuous education for radiological technologist with excellent qualities and competence should be done.
