Genetic Variation Analysis of Early-heading Plant (Oryza sativa L.) Lines Derived from Gamma-ray Irradiation (방사선조사 유래의 조기출수 식물(Oryza sativa L.) 계통의 유전적 변이 분석)

  • This study was carried out to evaluate genetic variation of early-heading rice (Oryza sativa L. cv. Dongjin 1) lines derived from gamma-ray ($^{60}Co$, 300 Gy) irradiation. The average heading date of the 5 early-heading lines in $M_7$ and $M_8$ generation was faster than that of untreated control as 11 (line ${\gamma}$-2), 10 (line ${\gamma}$-5), 6 (${\gamma}$-1 line), 5 (${\gamma}$-3) and 4 days (line ${\gamma}$-4), respectively. According to ISSR analysis, polymorphic rate of the early-heading lines (from 5.9% to 23.4%) was higher than that of control (4.3%). The result indicates that the gamma-ray promote variation at DNA level. When genetic variations of rps16-trnK region were evaluated by nucleotide analysis, nucleotide length of the rps16-trnK region was 664 bp in all the early-heading lines and control. Out of 5 sites of nucleotide transposition detected in the region, however, 2 sites were appeared only in the early-heading lines.

  • Two varieties of potatoes, Irish cobbler and Shimabara stored for seven and nine months respectively by irradiation combined with natural low temperature (year-round temperature change:$2{\sim}17^{\circ}C)$ on a batch scale were investigated on the suitability for processing of potato chip. Nine months after storage, irradiated potatoes (Irish cobbler) tended to maintain somewhat-better texture and sensory quality than untreated in potato chip processing. Peel rate, closely related to potato chip yield, of untreated potatoes were $20{\sim}25%$ higher than those of irradiated and Agtron color determination of potato chip from both irradiated were commercially acceptable. Preservation of potatoes by irradiation combined with natural low temperature was evaluated as an alternative method of the supply for raw materials of potato chip processing in the off season in Korea.

  • The effects of gamma irradiation on microbiological and chemical qualities of four kinds of poultry feeds were investigated. The viable counts of total bacteria in the samples were $10^5\;to\;10^6/g$. They were reduced by 2 to 3 log cycles after 3 to 5 kGy irradiation, and were completely eliminated with irradiation of 7 kGy. Coliforms and enteric pathogens were contaminated in high levels in all samples, ranging from $1.2{\times}10^4\;to\;1.7{\times}10^5/g\;and\;4.0{\times}10\;to\;2.6{\times}10^3/g$, respectively, They were sterilized by 3 to 5 kGy irradiation. Fungi, ranging from $10^{2}\;to\;10^4/g$, mainly osmophiles were identified as Aspergillus and Penicillium. They were eliminated to a undetectable level by 5 to 10 kGy irradiation. Six kinds of species, including Aspergillus flavus, were potential mycotoxin producers. Chemical components, such as proximate compositions, and mineral contents were not affected by the gamma irradiation. However, TBA values and amino acid content seemed to be affected by gamma irradiation.

  • The meat samples of carp (Cyprinus carpio, Linne) and rainbow trout (Salmo gairdnerii) packaged in aluminum pouches with polyethylene adjuvant were exposed to gamma radiation of doses up to 1.5 Mrad for the purpose of determining optimum dose range required to bring about a significant storage-life extension at refrigerated temperatures. The maximum permissible dose for carp was determined to be 1.5 Mrad and that for rainbow trout 0.2 Mrad, while the optimum dose was 0.25 Mard and 0.05 Mrad, respectively. By irradiating them at each optimum dose, the practical storage-life of carp could be extended from one week to five at both $0^{\circ}\;and\;5^{\circ}C$ and that of rainbow trout from one week to 3-4 weeks at $0^{\circ}C$ and from 3 days to 14 days at $5^{\circ}C$. The carp meat suffered from extensive drip loss during the post-irradiation storage and it could be reduced effectively by dipping the samples into 10% polyphosphate solution prior to the radurization treatment. The rainbow trout was highly radiosensitive, while carp appeard to one of promising species to be radurization treated for the purpose of extending storage-life at refrigerated temperatures.

Effect of Gamma Irradiation on the Softening of Dried Fernbrake at Different Moist-Heating Conditions (습열 조리 조건에 따른 방사선 조사 건고사리의 연화도)

  • Effect of gamma irradiation on the softening of dried fernbrake at different moist-heating conditions was investigated. Dried fernbrake packaged in controlled atmosphere $(CO_2\;25\%,\;N_2\;75\%)$ was irradiated up to 7 kGy with $\gamma-ray$. Hardness of dried fernbrake was significantly decreased at the irradiation dose of above 5 kGy. Hardness of cooked fernbrake was significantly decreased with increasing irradiation dose level as well as heating temperature and time. $DT_{50}$ value (The heating time required to reach the $50\%$ reduction of hardness) for irradiated fernbrake was shortened according to dose level as well as heating temperature and heating time: $DT_{50}$ value of control: 23 min, 5 or 7 kGy: $2.2\~5.0$ min at $60^{\circ}C$. Also, the activation energy for softening of irradiated fernbrake ($1.85\~1.88$ kcal/mole for $5\~7kGy$) decreased compared to control (4.30 kcal/mole). Moisture content, swelling and rehydration rate of gamma irradiated fernbrake during moist heating increased according to the irradiation doses. Sensory results showed that scores of off-flavor and odor in irradiated fernbrake upto 7 kGy were not significantly different from control. Based on these results, gamma irradiation was effective for the reduction of cooking time and the activation energy for softening of dried fernbrake with increasing dose levels.

A Study on the Energy and Time Characteristics of $BaF_2$ Scintillation Detector ($BaF_2$ 검출기의 시간과 에너지 특성연구)

  • he scintillation detector having $BaF^2$ crystal with 3.6cm dia${\times}$2.0 cm thick was provided. The energy and timing characteristics were measured and compared with NaI(Tl) scintillation detectors, which widely used in unclear medicine. In order to measure the energy spectrum, the radioactive sources used were $^{22}Na,\;^{54}Mn,\;^{57}Co,\;^{137}Cs$ and the source to detector distance was 7cm. For the timing characteristic, NaI(Tl)(1" ${\times}$ 1")-$BaF^2$ and NaI(Tl)(3" ${\times}$ 3")-$BaF^2$ timing coincidence systems were prepared and the used source was $^{22}Na$ emitting 511keV annihilation photons. For the 511keV gamma-ray emitted from $^{22}Na$, It was revealed that the timing response of the $BaF^2$ detector was faster than NaI(Tl)(1" ${\times}$ 1") and NaI(Tl)(3" ${\times}$ 3") detector used in this experimental investigation. The energy characteristics of the $BaF^2$ detector had a good values for about 500keV energy range.

Synergistic Effects of Low Dose Gamma Irradiation and Growth Regulators on Seed Germination, Growth and Photosynthesis in Rice (Oryza sativa L.) (벼의 종자 발아와 생육 및 광합성에 대한 저선량 감마선과 생장조절물질의 상승작용 효과)

  • To investigate the synergistic effects of low dose gamma irradiation and growth regulators on the growth and photosynthesis in rice (Oryza sativa L.), laboratory and greenhouse experiments were conducted using 4-year-old rice seeds. In the laboratory experiment, the germination rate was increased in 0.001 ppm IBA treatment, showing the synergistic effect of gamma irradiation and growth regulators. The seedling growth was increased by treatment of GA₃ and IBA, the irradiated groups having higher than the non-irradiated ones. Particularly, it was remarkable in 0.001 ppm IBA. In greenhouse experiment, seedling growth was increased in response to a combination of gamma irradiation and 0.001 ppm IBA. Effective quantum yield of PSⅡ(Ф/sub PSⅡ/) and photochemical quenching (qP) were increased, while non-photochemical quenching (qN) was decreased by 0.001 ppm IBA. A synergistic effect of gamma irradiation and IBA was only found in seedling growth. The present results suggest that low dose gamma irradiation and growth regulator could synergistically stimulate seedling growth.

A Study on Projection Image Restoration by Adaptive Filtering (적응적 필터링에 의한 투사영상 복원에 관한 연구)

  • This paper describes a filtering algorithm which employs apriori information of SPECT lesion detectability potential for the filtering of degraded projection images prior to the backprojection reconstruction. In this algorithm, we determined m minimum detectable lesion sized(MDLSs) by assuming m object contrasts uniformly-chosen in the range of 0.0-1.0, based on a signal/noise model which provides the capability potential of SPECT in terms of physical factors. A best estimate of given projection image is attempted as a weighted combination of the subimages from m optimal filters whose design is focused on maximizing the local S/N ratios for the MDLS-lesions. These subimages show relatively larger resolution recovery effect and relatively smaller noise reduction effect with the decreased MDLS, and the weighting on each subimage was controlled by the difference between the subimage and the maximum-resolution-recovered projection image. The proposed filtering algoritym was tested on SPECT image reconstruction problems, and produced good results. Especially, this algorithm showed the adaptive effect that approximately averages the filter outputs in homogeneous areas and sensitively depends on each filter strength on contrast preserving/enhancing in textured lesion areas of the reconstructed image.

  • In the field of healthcare, the conventional sterilization treatments have been replaced by irradiation methods which are in accordance with internationally well established quality standards. The quality control in radiation sterilization assures that the absorbed dose of the irradiated material is in agreement with its requirements and standards. The electron beam irradiation requires technical assessments of more process parameters than gamma irradiation does. Korea has witnessed wide uses of electron accelerators since early 2000 but there hasn't been research experiences relating to quality system in accordance with international standards. The new large scale e-beam irradiation system with the specification of 10 MeV, 8 kW was installed and operated in 2008 by Seoul Radiology Services Co. It consists of the electron accelerator, product handling system, safety, documentation and control subsystems into an integrated system to meet the requirement of the Good Manufacturing Practice such as process quality assurance and management of product tracking records. To implement the international standard such as EN ISO11137, it is necessary to understand the purposes aimed in the standard and carry out the tests following the procedures required. This study presented the specification of the e-beam facility and showed what its design requirements and features are. The test results on a variety of process parameters were presented and validated it they are within the required limits.

Studies on the Preservation of Pork Sausage by Gamma Radiation -Part 1. On the Enhancement of the Preservative Effect of Antiseptics by Gamma Radiation- (감마선 조사에 의한 Pork Sausage의 저장에 관한 연구 -제 1 보 방부제와 방사선과의 상승효과에 대하여-)

  • This experiment was conducted to investigate the combined effects of radiation and antiseptics on the keeping qualities of pork sausage, which was treated with potassium sorbate and AF-2(2-(-2-furyl)-3-(5-nitro-2-furyl)-acrylamide), and then followed by gamma radiation of 0.25, 0.5, and 0.75 Mrad. Amounts of treated antiseptics were a quarter, half, and full levels of their maximum permissible concentration. Irradiated and unirradiated sausages were stored for 50 days at $5^{\circ}C\;and\;25^{\circ}C$, and their changes in rancidity, volatile basic nitrogen, bacterial counts, pH, and sensory analysis were examined during the storage period. The results obtained are as follows: 1) Preservative effects of antiseptics were manifested at cold storage; antiseptics treatment of a quarter-level and unirradiation following low-temperature storage showed the same good keeping qualities as the combined treatment of full-level antiseptics and radiation of 0.25 Mrad following high-temperature storage. 2) There did not appear to recognize irradiation-odor, while color and odor were deteriorated intensively by storage temperature. Sausage irradiated with 0.75 Mrad has shown slightly noticeable off-odor at the end of storage at $25^{\circ}C$. 3) The most suitable radiation dose was considered to be 0.5 Mrad, which could extend the storage life about $2{\sim}3$ times longer than untreated.
